The morphological characteristics, carbon and nitrogen concentrations, stable isotope values and photosynthetic rates of Porphyra yezoensis were studied at the main purple lavers production areas on southwestern coast of Korea. The morphological characteristics of leaf length, leaf width and weight of Porphyra blades were between 11.6~16.3 (average 13.8) cm, 4.6~6.3 (average 5.4) cm, $1.1{\sim}2.6(average\;1.86)g\;DW\;m^{-2}$, respectively. Photosynthetic pigment of Chl a concentration of Porphyra blades was between $2.18{\sim}17.77(average\;9.65)mg\;DW\;Chl\;a\;m^{-2}$. Carbon and nitrogen concentrations of Porphyra blades was between $201{\sim}317(average\;240)mg\;DW\;g^{-1}$, $39.8{\sim}50.0(average\;43.5)mg\;DW\;g^{-1}$ and C/N ratio 5.0~6.7 (average 5.5). The range of average ${\delta}^{13}C$ and ${\delta}^{15}N$ values of Porphyra blades was between - 25.6 to - 24.0 (average - 24.7)‰ for ${\delta}^{13}C$, and 1.3 to 4.1 (average 2.1)‰ for ${\delta}^{15}N$. Photosynthetic characteristics of seaweeds measured by pulse amplitude modulation (PAM) fluorometry was used as an indicator of photosynthetic activity. We use Diving-PAM fluorometry to examine photosynthetic rates of the seaweeds Porphyra yezoensis at each station. Maximum quantum yield of Porphyra blades was between 0.46~0.55 (average 0.52), the variance of the effective PS II maximum quantum yield of the station was broadly similar. Maximum relative electron transport rate (rETRmax) of Porphyra blades was between $4.71{\sim}5.84(average\;5.33){\mu}mol\;electrons\;m^{-2}\;s^{-1}$, the changes of maximum relative electron transport rate (rETRmax) of Porphyra yezoensis were similar to those of PS II maximum quantum yield. Photosynthetic efficiency (${\alpha}$) was between 0.027~0.045 (average 0.036). Minimum saturating irradiance ($E_k$) range was $139{\sim}180(average\;156){\mu}mol\;photons\;m^{-2}\;s^{-1}$. Minimum saturating irradiance ($E_k$) made a difference by station within the area on southwestern coast. Carbon and nitrogen concentrations and photosynthetic rates of Porphyra blades production areas on southwestern coast were broadly similar. The photosynthetic characteristics showed low photosynthetic rates because the low maximum quantum yields and low maximum relative electron transport rate.
The photosynthetic characteristics and primary production by phytoplankton in open waters of two wetlands (the Banwol and the Donghwa wetland) of Sihwa Constructed Wetland with different water chemistry were investigated to provide the information for the wetland management considering the water treatment efficiency. During the study period (from March to October, 2005) the primary productivity in open waters ranged from 481 to 11,275 mgC $m^{-2}$$day^{-1}$, which is very high compared with the eutrophic level of 600mgC $m^{-2}$$day^{-1}$. From the analysis of the photosynthesis-irradiance (P-I) model parameters, the photosynthetic characteristics may be affected by different concentration and ratio of nutrient (N and P) between two wetlands. Assimilation number (AN) was higher in the Donghwa wetland (average AN: 8.5gC $gChl^{-1}$$hr^{-1}$) with high P and low N/P ratio than the Banwol wetland (average AN: 5.8gC $gChl^{-1}$$hr^{-1}$) with high N and high N/P ratio. This result indicates that AN may be concerned with phosphorus than nitrogen and low NIP ratio. Positive correlation (R=0.81) was observed between the initial slope and AN, implying that AN was high in case of phytoplankton having more active photosynthesis ability under low light. On the other hand, maximum photosynthesis (Pmax) was related positively with chlorophyll a concentration showing correlation coefficient of 0.47. In this study, considering the high primary production through phytoplankton photosynthesis in open waters of Sihwa Constructed Wetland, the produced organic matter by phytoplankton may affect the water quality within wetland and its efficiency of water treatment. Also, the photosynthetic characteristics may be affected by different nutrient enrichment (especially phosphorus) of wetlands. This study suggests that the production by phytoplankton and its characteristics in open water of constructed wetland for water treatment should be considered to improve the removal efficiency of organic matter.
Seasonal succession and community composition of planktonic ciliates were studied in Kyungan Stream from December 2000 to December 2001. Oligotrichs accounted for 53% of total ciliates. Total abundance of ciliates peaked in spring (30 March, 6 April) and in summer (8 June, 20 July) reaching values up to $1.9\times10^4$ cells $L^{-1}$. Seasonal succession of dominant species occurred obviously. Large-sized $(>50{\mu}m)$ species (Stylonychia sp1, Phascolodon vorticella and Codonella cratera) dominanted from winter to spring. Small sized $(<30{\mu}m)$ species (Vorticella spp., Rimostrombidium hyalinum and Halteria grandinella) dominanted in summer and autumn. Total abundance of large-sized species coincided with the Chl-${\alpha}$ concencetation during the study (r=0.33, p<0.05, n=39). Among the small-sized species Halteria grandinella was a significant relationship with bacterial abundance (r=0.35, p<0.05, n=39).
Mudflats are crucial for understanding the ecological structure and biological function of coastal ecosystem because of its high primary production by microalgae. There have been many studies on measuring primary productivity of tidal flats for the estimation of organic carbon abundance, but it is relatively recent that optical remote sensing technique, particularly hyperspectral sensing, was used for it. This study investigates hyperspectral sensing of chlorophyll concentration on a tidal flat surface, which is a key variable in deriving primary productivity. The study site is a mudflat in Geunso bay, South Korea and field campaigns were conducted at ebb tide in April and June 2021. Hyperspectral reflectance of the mudflat surfaces was measured with two types of hyperspectral sensors; TriOS RAMSES (directionalsensor) and the Specim-IQ (camera sensor), and Normal Differenced Vegetation Index (NDVI) and Contiuum Removal Depth (CRD) were used to estimate Chl-a from the optical measurements. The validation performed against independent field measurements of Chl-a showed that both CRD and NDVI can retrieve surface Chl-a with R2 around 0.7 for the Chl-a range of 0~150 mg/m2 tested in this study.
Red tide has potential to harm marine ecology and aquaculture. Research on detecting red tide using various optical remote sensors has been conducted, but most of existing algorithms for detecting red tide has limitations, especially in shallow coastal waters with high levels of suspended sediment. For enhanced understanding of the optical behavior of red tide waters, analysis on remote sensing reflectance and water constituent is becoming increasingly important. This study analyzed the optical remote sensing data and water quality variables(Chl-a(Spec), SPM, aph, ad, Turbidity, Chl-a(HPLC), Dominant species) of red tide waters. The data were collected from ship-based campaigns. In addition to the research on detecting red tide, the remote sensing reflectance and extinction coefficients for mesodinium and cochlodinium species were also analyzed. Through the analysis, it was possible to estimate the red tide chlorophyll concentration based on a specific wavelength of the remote sensing reflectance. The study found that chlorophyll concentration and phytoplankton absorption coefficient were highly correlated(R2=0.9), and that the REdiff formula provided a more accurate estimate of red tide concentration than the B-G ratio.
In this study, we applied a Particle Swarm Optimization (PSO) algorithm to predict the changes of chlorophyll-${\alpha}$ related to environmental factors in agricultural reservoirs in Korean national scale. Data were obtained from water quality monitoring networks of reservoirs operated by the Ministry of Agriculture and Forestry and the Ministry of Environment of Korea. From the database of the monitoring networks, 290 reservoirs were chosen with variables such as chlorophyll-${\alpha}$ and 13 environmental factors (COD, TN, TP, Altitude, Bank height, etc.) measured in 2002. Based on Carlson's trophic status index, reservoirs were divided into five groups, and most agricultural reservoirs $(TSI_{CHL}\;64.1%,\;TSI_{TP}\;75.5%)$ were in the eutrophic states. The groups were discriminated with environmental variables, showing that COD, DO, and TP were important factors to determine the trophic states. MLP-PSO (Multilayer perceptron (MLP) with PSO for the optimization) was applied for the prediction of chlorophyll-${\alpha}$ with environment factors, and showed high predictability (r=0.83, p<0.001). Additionally, the sensitivity analysis of the MLP-PSO model showed that COD had the strongest positive effects on the concentration of chlorophyll-${\alpha}$, and followed by TP, TN, DO, whereas altitude and bank height had negative effects on the concentration of chlorophyll-${\alpha}$.

This study was conducted to evaluate the practical usefulness of the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PACE) fingerprinting of whole cell proteins far the identification of lactic acid bacteria in Kimchi. SDS- PACE of whole cell proteins of the reference strains and lactic acid bacteria isolated from Kimchi yielded differential banding patterns that were highly specific fingerprints, thus making it possible to identify. Identification of the isolates from Kimchi was achieved by comparing the SDS-PAGE fingerprints of isolates to those of reference strains. In addition, the reliability of SDS-PAGE was examined by comparing the results with those of the APL 50 CHL system assay and 16S rRNA gene sequence. SDS-PACE assay showed a different identity to reference strains, while the APL 50 CHL system and 16S rRNA gene sequence could not distinguish a few strains. Therefore, SDS-PAGE of the whole cell proteins is a specific and a reliable method that will be useful for the identification of lactic acid bacteria in Kimchi to the species level, and can be used as an alternative or complementary identification method.
The growth of Selenastrum capricornutum in culture was investigated as a function of ionic copper concentrations to verify the toxicity and physiological variation. In a $Cu^{++}$ excess culture (E: $Cu^{++}$ of $130{\mu}g/l$), the growth rate (K)(0.32) was lower than that of the control culture (C: $Cu^{++}$ of $0.065{\mu}g/l$) (0.61) after 8 days and the exponential growth rate ($R_E$) was also found to be lower in culture E (1.1) than culture C (2.9). On the contrary, the K of S. capricornutum in trace $Cu^{++}$ culture ($T_1$ and $T_2$, 0.72) after 6 days tended to be more increased than culture C (0.68). From 8 to 14 days of culture, the amounts of chlorophyllls a and b were increased in culture C (chlorophyll a, $106->126{\mu}g/g$ dry wt; chlorophyll b, $158->208{\mu}g/g$ dry wt; chlorophyll b were decreased in culture E (chlorophyll a, $309->235{\mu}g/g$ dry wt; chlorophyll b, $405->352{\mu}g/g$ dry wt). The amounts of chlorophylls in ionic copper trace culture ($T_1$ and $T_2$) [(chl a/b) of $T_1$: $384/620\;{\mu}g/g$ dry wt; (chl a/b) of $T^2$:$320/467{\mu}g/g$ dry wt] were increased more than the culture C($260/387{\mu}g/g$ dry wt). Howeveer, when photosynthetic rates were normalized to the dry weight of algae, the control culture continued to showed higher values than the treated culture ($T_1$). An appropriate amount of ionic copper ($T_1:\;26\;{\mu}g/l$) stimulated the grwoth of S. capricornutum than the ionic copper content of $13\;{\mu}g/l$ ($T_2$), while the excess amount of ionic copper ($130\;{\mu}g/l$) resulted in the highest toxicity to the growth of S. capricornutum.
In order to assess the inorganic and organic pollutents characteristics in marine water, we investigated COD (Chemical Oxygen Demand), Chlorophyll a and Escherichia coli during four seasons at 20 stations of Gwangyang Bay and at 23 stations of Jinhae Bay, Korea. The bay is divided into three zones in Gwangyang and four zones in Jinhae respectively, based on the pollutent levels. In Gwangyang Bay, the high concentration (mean 4.7 mg $L^{-1)$) of COD was recorded during spring season at Zone I, which can be characterized as a semi-enclosed eutrophic area (St. 1~9). Also, Chl. a concentrations were high at Zone I (mean 14.0 ${\mu}g\;L^{-1}$). The colony of E. coli were detected during summer season at Zone II, which is influenced by Seomjin River water. The E. coli may have been entered from the river water in a large pulse during rainy season. On the other hand, E. coli was kept low levels during four seasons at the Zone III, which is influenced indirectly by surface water currents from offshore of the bay. In Jinhae Bay, the high COD and Chl. a were shown during all seasons at Zone I, which is characteristed by semi-enclosed eutrophic area of Masan and Haengam bays. The Zone I also had been shown relatively high E. coli concentration in all seasons. In constrast, other three zones did not show seasonal characteristics of the E. coli concentrations. The present study suggests that E. coli concentrations can be significantly elevated in eutrophic semi-enclosed area.
Filtration rates and fecal production of freshwater bivalve, Unio douglasiae on two kinds of hibernal diatom communities were measured simultaneously in a laboratory. One community is the Han River (HAN), which dominated by Asterionella Formosa. Stephanodiscus hantzschii (ca. 98% of total phytoplankton). The other community is the Ilgam Lake (IL), which dominated by Synedra ulna, Scenedesmus sp. Microcystis aeruginosa (ca. 82%). The HAN water has higher concentrations of nutrient (TN and TP) and chlorophyll $\alpha$ (Chl-$\alpha$), lower turbidity and conductivity than the IL water. Water sampling for the feeding experiment was conducted in the same day (Jan 15, 2008) and similar time (AM 10:00 for HAN, AM 11:00 for IL). Mussels with the similar size ($0.0{\pm}0.5\;cm$) were collected from the Gunsan and Okgu district (Jeonbuk), and starved in a laboratory for 2 days before the experiment. The experiment comprised CON (no addition of mussel), LOW (addition of mussel at 0.3 indiv. $L^{-1}$), MID (1.0 indiv. $L^{-1}$) and HIGH (2.0 indiv. $L^{-1}$), respectively. With the increment of mussel density and time, the concentration of Chl-$\alpha$ in two diatom communities were clearly decreased; Chl-$\alpha$ of HAN gradually decreased after 1 hour of mussel treatment, while that of IL decreased as soon as mussel introduction. In 7 hours of treatment, the former was removed finally up to about 90% of control, while the later was remained as about 50%. Under the presence of mussel, total phytoplankton density was shifted as the similar patterns to that of Chl-$\alpha$ (r=0.705, P<0.0001), however, there showed the drastic differences following a species. Based on the concentration of Chl-$\alpha$, filtration rate of U. douglasiae averaged 0.266 $L\;g^{-1}\;h^{-1}$ (0.115 to 0.442) on HAN and $0.577\;L\;g^{-1}\;h^{-1}$ (0.146 to 1.428) on IL water, respectively. There were no differences in feces production among the mussel density in the HAH water (ANOVA, P>0.5), while in IL water, including lots of seston, the HIGH mussel produced the higher fecal materials, over one hundred times of LOW. These results suggest that freshwater bivalve Unio douglasiae have the alternative potential, as a filter-feeder of seston in turbid lake, and a biological controller of diatom bloom in cold stream.
