The Effects of Acidic Electrolytic Water on the Development of Barley Chloroplast (산성 전해수가 보리(Hordeum vulgae L.) 엽록체의 발달에 미치는 영향)

  • To investigate the effects of strong acidic electrolytic water on the chloroplast, barley leaves were treated with strong acidic electrolytic water(pH 2.5). And to investigate the effects of weak acidic electrolytic water on the chloroplast development, etiolated barley leaves were treated with weak acidic electrolytic water(pH 6.5) during greening period. Chl contents, Fo, Fv, and Chl fluorescence quenching coefficient in barley leaves were measured during and after treatment of acidic electrolytic water. The following results were obtained. Chl a, b, and carotenoid were decreased with treatment of strong acidic electrolytic water. Chl contents were significantly decreased than that of the control after 5 min. These results provide evidence that the strong acidic electrolytic water dissimilate the Chl and so that the value of Fo was slightly increased. The strong acidic electrolytic water damaged PS II because Fo was increased and Fv, Fm, and Fv/Fm ratio were decreased. qP, qNP and qE were decreased. On the other hand qI was increased than that of the control. But Chl content and Chl fluorescence patterns were a little changed as the pH increase over 4.0 Chl a, b, and carotenoid were increased with treatment of weak acidic electrolytic water during greening period. Chl contents were significantly increased than that of control after 12 hours greening. These results provide evidence that the weak acidic electrolytic water accelerated the chlorophyll synthesis. And the weak acidic electrolytic water accelerated PS II development because Fv, Fm, qP and Fv/Fm ratio were increased than that of the control.

Formation of Chlorophyll-Protein Complexes in Greening Rape Cotyledons (Greening에 따른 유채 자엽의 엽록소-단백질 복합체 형성)

  • The formation of chlorophyll-protein complexes (CP-complexes) during the greening of rape cotyledons (Brassica napus cv. Yongdang) was investigated by the SDS-polyacrylamide gel electrophoresis. The total chlorophyll content and Chl a/b ratio were also determined. In addition, the effects of dark treatment on the CP-complex patterns during greening have been examined with respect to their photosynthetic electron transport activity. Greening has brought about the increasein total chlorophyll content and the decrease in Chl a/b ratio, but there have been no changes in Chl a/b ratio after 24 hrs of greening. The light-harvesting chlorophyll a/b-protein complex (LHCP-complex0 was predominant during the initial greening period. Thereafter, the amout of chlorophyll a-protein complex (CP I-complex) was gradually increased. Twenty-four-hr dark treatment immediately after illumination for 6 hrs and 12 hrs resulted in the increase of the Chl a/b ration and the CP I complex, otherwise the decrease of the LHCP-complex. The LHCP/CP I ratio was gradually decreased with further greening, and appeared no change after 48 hrs illumination. The investigation of the photosynthetic electron transport activity indicated that photosystem (PS) II activity (H2Olongrightarrowp-PD*+FeCy**) did not change, but the activity of PS I was increased suddenly due to the dark treatment. The data suggests that the increase of CP I-complex may result in that of P-700, that is, the increase of PS I activity.

Contribution of Phytoplankton and Zooplankton to Total Organic Carbon (TOC) in the Reservoir-river-Seonakdong River, Busan (서낙동강에서 동·식물플랑크톤의 총유기탄소 기여율 변동 분석)

  • Carbon biomass of plankton community, Total Organic Carbon (TOC) and Chlorophyll a (chl.a) concentration were examined in the SeoNakdong river from January to December in 2014, to assess composition of phyto- and zoo-plankton variation, to certify the correlation between chl.a and TOC and to determine the level of contribution of plankton carbon content to TOC in the reservoir-river ecosystem. The correlation level between TOC and chl.a was low in the year 2014 but exceptionally was highly correlated only during the period with cyanobacterial bloom. The high level of contribution of plankton carbon content to TOC was attributed to cyanobacterial carbon biomass from May to November and to Cladocera carbon biomass from March to May, November and December despite of its low abundance. These results suggest that there were inter-relationships between phytoplankton, zooplankton and TOC and also subtle consistency of their properties through the year. These patterns should be discussed in relation to the physiochemical and biological characteristics of the environment, as well as to allochthonous organic matters from non-point pollution sources.

Disassembly of Chlorophyll-Protein Complexes in Arabidopsis thaliana during Dark-Induced Foliar Senescence

  • The disassembly of Chl-protein complexes during dark-induced senescence (DIS) was investigated using detached third and fourthleaves of 21$\pm$1 day-old Arabidopsis thaliana. Although Chl content decreased linearly after 1 d, a significant decrease of photochemical effeciency (Fv/Fm) was observed after 2 d. In experiments using native green gel electrophoresis of Chl-protein complexes combined with additional two-dimensional SDS-PAGE analysis, we could observe the degradation of both photosystems after 2 d. Although light-harvesting complex(LHC) for PSI (LHCI) was degraded first in PSI complex, small PSII apoproteins including CP47/CP43 and D1/D2 apoproteins were degraded first in PSII complexes. LHC for PSII (LHCII) trimers were stable until 4 d. The level of LHCII monomers was increased until 3 and decreased thereafter, resulting in the increase of free pigments. These results suggest that the disassembly process of PSI is different from that of PSII.

The Effect of NaCl on the Greening of Etiolated Leaves of Barely (Hordeum vulgare L.) Seedings (NaCl이 황백화된 보리(Hordeum vulgare L.) 잎의 녹화에 미치는 영향)

  • The effects on photosynthesis of NaCl(0, 0.2, 0.4, 0.6, 0.8 or 1.0 M) were examined in etiolated barley seedlings. Chlorophyll(Chl) a, Chl b and carotenoid contents, Chl a fluorescence and quenching coefficients of Chl fluorescence have been determined in the primary leaves of etiolated barley seedlings cultivated under low light(60 $\mu$$m^{-2}\;s^{-1}$). Chl a, b, and carotenoid contents were decreased remarkably in comparison with the control at 0.4 M NaCl. However, the value of Fo and Fv were decreased at 0.6 M NaCl and the ratio of Fv/Fm were deceased at 1.0 M NaCl. Chlorophyll synthesis was seriously inhibited from 0.4 M NaCl, and the photosynthetic electron transport system was inhibited from 0.6 M NaCl. Quantum of photosystem II reaction center was inhibited at 1.0 M NaCl. The effects of NaCl on the Chl content were raised in a 6 hrs, but the effects of NaCl on the value of Fo, Fv and Fv/Fm were raised in 30 hrs. The value of qP was decreased in comparison with the control at all concentrations, but there was a small change in the value qE. These results provide evidence that NaCl inhibited effects of various concentration of NaCl were inhibited quinone redox, however, proton gradient between thylakoid membranes was little damaged.

Biomass of Primary Producer in the Ch$\check{o}$nsu Bay -Relationships between Phytoplankton Carbon, Cell Number and chlorophyll- (천수만 일차생산자의 생물량 -식물플랑크톤 탄소량과 세포개체수 및 클로로필과의 관계-)

  • In order to study the biomass of primary producer, phytoplankton is collected monthly September 1985 to August 1986 in Ch$\check{o}$nsu Bay. Phytoplankton carbon contents which are calculated from phytoplankton volume were ranged from $26.7{\mu}gC/l$ to $960.7{\mu}gC/l$, and average carbon contents of each month lie in the range of $58.6-684.7{\mu}gC/l$(annual mean $208.5{\mu}gC/l$). For net plankton analysis with the carbon contents, cell numbers, and chlorophyll concentrations show a close correlation, while for nanoplankton the correlation was low, indicating that nano-fraction includes a significant portion of picoplankton. Also, the multiple regression analysis with carbon content, cell number, and chlorophyll concentration to size fraction well illustrate the prime importance of the net-fraction in phytoplankton group. C/Chl-a ratios ranged from 9.1 to 100.5, average rations of net- and nanoplankton are 111 and 6.4, respectively. The greater net plankton faction is, the higher C/Chl-a ratio is, however in case of high nanoplankton portion C/Chl-a ratio show low level. These results indicate that the difference of C/Chl-a ratio per phytoplankton cell size be main factor for the variation of C/Chl-a ratio in Ch$\check{o}$nsu Bay. As C/Chl-a ratio fluctuates greatly in coastal ecosystem, that use of a direct conversion of convert chlorophyll to organic carbon may lead erronous estimation.

Effect of Algal Fraction to Particulate Organic Matter in the Upper Regions of a Brackish Lake Sihwa (시화호 상류 기수역에서 입자성유기물에 대한 조류영향)

  • To estimate the effect of algae to particulate organic matter in the upper regions of brackish Lake Sihwa, temporal and spatial variations of particulate organic carbon (POC) and phytoplankton pigments (chlorophyll a; Chl-a, pheophytin-a; Pheo-a), and their relationships were studied at seven sites of the brackish regions from March to October 2005 and 2006. POC concentration varied from 1.0 to $76.6mgL^{-1}$ (mean $7.4mgL^{-1}$), with maximal concentrations occurring in the middle parts of the study area in spring of 2005 and 2006. Concentrations of Chl-a and Pheo-a varied from 1.3 to $942.9{\mu}gL^{-1}$ (mean $71.0{\mu}gL^{-1}$) and $1.4{\sim}1,545.5{\mu}gL^{-1}$ (mean $59.9{\mu}gL^{-1}$), respectively, and corresponded closely with variation in POC. During the study period Pheo-a concentration was 44.2% of total Chl-a, implying that non-living or inactive phytoplankton is also the important part of phytoplankton-derived POC in brackish regions of Lake Sihwa. From the positive linear relationships between POC and phytoplankton pigments (POC with Chl-a (r=0.93), total Chl-a (r=0.88), and Pheo-a (r=0.81)), it is suggested that phytoplankton was a significant component of POC in the upper regions of brackish Lake Sihwa. On the other hand, the ratios of POC/Chl-a and POC/total Chl-a (Chl-a+Pheo-a) were 82.9 and 35.9, respectively. The ratio of POC/total Chl-a is similar to those reported in previous studies, including 40~60 in estuaries. This study suggests that Pheo-a concentration is considered in estimation of POC concentration from phytoplankton pigments in aquatic systems with high content of Pheo-a, like an upper region of blackish Lake Sihwa.

Changes in Chloroplast Ultrastructure and Thylakoid Membrane Proteins by High Light in Ginseng Leaves

  • Ultrastructural changes in Panax ginseng C. A. Meyer mesophyll chloroplasts and variation of thylakoid membrane protein in responce to the light intensity were studied in leaves of two-y-old plants exposed to two different light intensities under field coditions. The leaves were allowed to function for three months after emergence under two contrasting light conditions. The ginseng chloroplasts of 5% light were filled with highly stacked grana of condensely arrayed thylakoids, so that the stroma space was hardly observed. In contrast, chloroplasts from leaves at 100% sunlight had fewer thylakoid membranes and smaller grana stacks. The number of osmiophilic globules increased. Total Chl content and Chl b content were lower at 100% sunlight than 5% sunlight. The thylakoid membrane proteins in the leaves grown at 100% sunlight showed lower CPIa, LHCII and CP29 than those with 5% sunlight. This effect was most obvious for LHCII. Polypeptides showed major bands at 90, 64, 29-30, 22 and 14 kD, and minor bands at 59, 58, 54, 52, 49, 46, 44, 35, 23, 21 and 18-19 kD. All these bands were lower in intensity in the leaves exposed to 100% sunlight. Moreover, the bands at 58-59, 46-47 and 23 kD disappeared.

Modifications of nutrient regime, chlorophyll-a, and trophic state relations in Daechung Reservoir after the construction of an upper dam

  • Background: Previous numerous studies on watershed scale demonstrated that the constructions of upper dams may influence the below dams due to modifications of flow regime and nutrient inputs. Little is known about how the dam constructions influence the downstream lakes or reservoirs in the regional scale. This study demonstrates how the construction of upper dam (i.e., Yongdam Dam) influences nutrient regime, trophic relations, and empirical models in Daechung Reservoir (DR). Yongdam Dam was constructed at the upstream region of DR in year 2000. Results: The analysis of hydrological variables showed that inflow and discharge in the DR were largely reduced after the year 2000. The construction of upper dam construction also resulted in increases of water temperature, pH and conductivity (as an indicator of ionic content) in the DR. Empirical models of TP-CHL and N:P ratio-CHL suggested that stronger responses of CHL to the phosphorus were evident after the upper dam construction, indicating that algal production at a unit phosphorus increased after the upper dam construction. Mann-Kendall tests on the relations of N:P ratios to TN showed weak or no relations ($t_{au}=-0.143$, z = -0.371, p = 0.7105) before the dam construction, while the relation of N:P ratios to TP showed strong in the periods of before- ($t_{au}=-0714$, z = -2.351, p = 0.0187) and after the construction ($t_{au}=-0.868$, z = -4.270, p = 0.0000). This outcome indicates that TP is key determinant on N:P ratios in the reservoir. Scatter Plots on Trophic State Index Deviations (TSIDs) of "TSI(SD) - TSI(CHL)" against "TSI(TP) - TSI(CHL)" showed that the dominance of clay turbidity or light limitation was evident before the upper dam construction [TSI(TP) - TSI(CHL) > 0 and TSI(SD) - TSI(CHL) > 0] and phosphorus limitation became stronger after the dam construction [(TSI(TP) - TSI(CHL) < 0 and TSI(SD) - TSI(CHL) > 0]. Conclusions: Overall, our analysis suggests that the upper dam construction modified the response of trophic components (phytoplankton) to the nutrients or nutrient ratios through the alteration of flow regime, resulting in modifications of ecological functions and trophic relations in the low trophic levels.

Physiological Differences of Ilex rotunda and Illicium anisatum under Low Light Intensities (다른 광도에서 생육한 먼나무, 붓순나무의 생리적 차이)

  • We examined seedlings of two species (Ilex rotunda and Illicium anisatum) which have a different level of shade tolerance and raised them under different light regimes (full sunlight and 50% shading). After 12 months, we investigated chlorophyll content (Chl. a, Chl. b and Chl. a+b), photosynthetic systems (photosynthetic rate, light compensation point, dark respiration rate and quantum yield), intercellular $CO_2$ concentration and water use efficiency to show acclimation reaction to different light conditions. Seedlings grown under full sunlight showed lower chlorophyll content than those in the shading regime. There was a significant difference between the full sunlight and shade treatments in I. anisatum (shade tolerance species). I. rotunda (intermediate species) showed high photosynthetic rate and water use efficiency over PPFD $1000\;{\mu}mol\;m^{-2}s^{-1}$ to full sunlight. Also, I. anisatum grown under full sunlight showed lower photosynthetic rate and water use efficiency over a range of all PPFD. This result showed that I. rotunda has a more flexible reaction system than that of I. anisatum.
