  • Carbofuran(CF) is one the most widely used carbamate pesticides in the world applied for insect and nematode control. Due to its widespred use in agriculture and households, contamination of food, water, and air has become serious, and consequently adverse health effects are inevitable in humans, animals, wildlife and fish, it has reported that CF alone or in combination with other carbamate insecticides influences the level of reproductive and metabolic hormones such as thyroxine and corticosterone, and results in impairment of endocrine.   • In vitro 소핵시험(vitMNT)은 유전독성의 유망한 대체시험법 중 하나로, OECD에서 TG로 채택되어 화학물질의 등록에 사용되고 있다. 본 시험에서는 CHL cell을 사용한 vitMN test에서 소핵을 판별하기 위한 최적화된 세포질 조건을 찾고자 하였으며, 양성대조물질로 MMC와 Col을 사용하고 세포 염색을 위해 giemza 용액을 사용하였다. 시험결과, 세포현탁을 위해 사용되는 고정액의 acetic acid의 농도는 1%로 하는 것이 band의 두께와 세포질의 퍼짐성 측면에서 적당하였다. 또한 세포의 깨짐을 최소화하는 최종 고정액의 적하시간은 현탁 후 1~4시간이었다. 이러한 결과는 vitMNT에서 소핵관찰의 신속성, 용이성 및 정확성을 확보하는데 도움이 될 것이다.

  • We performed chromosomal aberration assay in Chinese Hamster Lung (CHL) cells in vitro to evaluate theclastogenicity of 11 synthetic chemicals which were listed in Toxicity Evaluation Program of Ministry of Environment of Republic of Korea in 1996. All of the chemicals were carried out MTT assay to determine the 50% cell growth inhibition concentration. All compounds were tested with and without metabolic activation system. Benzoyl chloride revealed positive result at $43\;\mu\textrm{g}/m{\ell}$ in the presence of metabolic activation system, and at 30.8, 61.5 and $123\;\mu\textrm{g}/m{\ell}$ in the absence of metabolic activation system. And p-phenoxy ethanol was observed as positive with the metabolic activation system, but negative without metabolic activation system. Especially 2-propyn-l-ol showed high frequency of pulverization and showed critical difference of cytotoxicity between with and without S9 mixture. Pulverizatiuon is not included in the frequency of structural aberration in our criteria. Dicyclopentadiene, methacrylic acid, aa-dimethylbenzyl hydroperoxide, benzylbutyl phthalate, and p-chlorophenal were revealed negative results.esults.

  • To evaluate the genotoxicity of CKD-602, an anticancer agent the in viかo reverse mutation assay using Salmonella typhimurium, the Chromosome aberration assay using Chinese hamster lung (CHL) cells and the in vivo micronucleus assay using bone marrow cells of ddY mice were performed. In the reverse mutation assay, CKD-602 did not induced mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535, and TA 1537 strains with and without metabolic activation. In the chromosome aberration test using CHL cells, there was an increased incidence of structural aberrations induced by CKD-602 without metabolic activation during 24 and 48 hours, but CKD-602 did not induce chromosome aberration with metabolic activation. The in vivo induction of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ddY mice. At 24 hours after treatment with CED-602 by i.p. once, there was an increased incidence of micronucleated polychromatic erythrocytes in bone marrow of ddY male mice.

  • To evaluate the genotoxicity of SKP-450, an antihypertensive agent the in vitro reverse mutation assay using Salmonella typhimurium, the Chromosome aberration assay using Chinese hamster lung (CHL) cells and the in vivo micronucleus assay using bone marrow cells of ddY mice were performed. In the Reverse mutation test, SKP-450 did not induced mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 with and without metabolic activation. In the chromosome aberration assay using CHL cells, there was no increased incidence of structural and numerical aberrations with and without metabolic activation. The in vivo induction of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ddY mice at 30 hours after treatment with SKP-450 by p.o once. The results showed no increased incidence of micronucleated polychromatic erythrocytes in bone marrow of ddY male mice treated with SKP-450.

Antioxidant Property of Vitamin C - in Comparison with Vitamin B1

  • Various aspects of antioxidant activity in vitamin C were evaluated in this study. Relatively high level of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was detected in vitamin C, but not in non-antioxidative vitamin, vitamin B1. Vitamin C also reduced the production of lipid peroxidation in Chinese hamster lung fibroblast (V79-4) cells with $IC_{50}$ value of $4{\mu}g/ml$. Vitamin B1 showed comparable reduction in lipid peroxidation products ($IC_{50}$ value was about $10{\mu}g/ml$). It was shown that vitamin C also dose-dependently enhanced the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in V79-4 cells, and these effects were not observed in vitamin Bl-treated cells. Our data suggest that well-known antioxidant vitamin C involved in direct activation of SOD, CAT and GPX.

  • (R)-JG-381, a R form of alkylglycidic acid derivative, was examined for mutagenicity in the reverse mutation test on bacteria, chromosomal aberration test on cultured mammalian cells and micronucleus test in mice. In the reverse mutation test on bacteria using Salmonella typhimurium strain TA98, TA100, TA102, TA1535, TA1537 with or without a metabolic activation system (S9 mix), (R)-JG-381 did not affect the revertant colonies but significantly increased revertant colonies in one test strain, TA98, compared with the vehicle control. In the chromosomal aberration (CA) test using cultured Chinese Hamster Lung fibroblast(CHL) cells, the number of aberrant cells was clot increased in the presence or absence of 59 mix at concentration of the (R)-JG-381 0.025 $\mu$l/m1 to 0.1 $\mu$l/m1, compared with vehicle control. In the micronucleus (MN) test, micronucleated polychromatic erythrocytes in the (R)-JG-381-treated mice were not different from those of the vehicle-treated mice.

  • Pendimethalin [N-(1-ethyl-propyl)-2, 6-dinitro-3, 4-xylidine, $C_{13}$H$_{19}$N$_3$O$_4$, M.W. = 281.3, CAS No. 40481-42-1]는 제초제의 일종으로, 본 연구에서는 박테리아 복귀 돌연변이 시험과 포유동물 세포를 이용한 염색체 이상 시험 및 마우스를 이용한 in vivo 소핵 시험을 수행하여 pendimethalin의 유전독성을 평가하였다. 박테리아 복귀 돌연변이 시험에서 pendimethalin은 Salmonella thphimurium TA98, TA1537 균주의 경우, 대사 활성계 존재와 부재시,TA100의 경우는 대사 활성계 부재시에만 313∼5,000 $\mu\textrm{g}$/p1a1e의 범위에서 농도의존적인 돌연변이율의 증가를 보여주었고, TA1535의 경우에는 대사 활성계 존재시 약간의 돌연변이가 증가되는 것을 관찰할 수 있었다. 그러나 대사 활성계 부재시 TA1535와 대사 활성계 존재시 TA100균주의 경우에는 돌연변이 유발능을 관찰할 수 없었다. 한편 포유동물 세포인 Chinese hamster lung(CHL) fibroblast를 이용한 염색체 이상 시험에서 pendimethalin은 대사 활성계 존재 및 부재시 2.32∼9.28 $\mu\textrm{g}$/ml 농도에서 clastogenicity를 보이지 않았다. 또한 203∼810 mg/kg의 pendimethalin을 구강 투여한 마우스의 골수세포를 이용한 in vivo소핵 시험의 결과에서도 통계적으로 유의한 소핵 유발능을 관찰할 수 없었다.다.

투명 양이온 계면활성제 ASCO EAQ80에 대한 급성 경구 독성시험 및 유전 독성시험에 관한 연구 (Acute Oral and Genetic Toxicity Study of ASCO EAQ80, a Novel Cationic Surfactant)

  • 본 연구는 투명한 섬유유연제 제조용 양이온 계면활성제인 ASCO EAQ80의 개발 완료 후 이 제품의 안전성과 관련된 평가를 받기 위하여 실시되었다. 시험은 급성 경구투여 독성시험과 유전 독성시험으로 나뉘어 진행되었으며, 랫드를 이용한 경구 단회 투여 독성시험 결과, $LD_{50}$는 5000 mg/kg를 초과하는 것으로 판단되었으며, Globally Harmonized Classification System의 기준에 의해 Category 5 또는 Unclassified로 분류되었다. 시험물질인 ASCO EAQ80의 복귀돌연변이 유발성에 대해서, 살모넬라균(TA98, TA100, TA1535, TA1537) 및 대장균(WP2uvrA (pKM101))을 이용하여 대사활성화 및 비대사활성화의 경우에서 변이원성 시험을 실시하였고, 그 결과는 모두 음성으로 판정되었다. 또한, 염색체이상 유발성 여부를 검색하기 위하여 Chinese Hamster Lung (CHL/IU) 배양세포를 이용하여 염색체이상시험을 수행하였으며, 시험물질인 ASCO EAQ80은 단시간처리법 및 연속처리법의 경우 대사활성계 적용여부에 관계없이 Chinese Hamster Lung (CHL/IU) 배양세포에 대해 염색체이상을 유발하지 않는 것으로 확인되었다.
