Cloning, Expression, and Characterization of a New Phytase from the Phytopathogenic Bacterium Pectobacterium wasabiae DSMZ 18074

  • The soft rot bacterium Pectobacterium wasabiae is an economically important pathogen of many crops. A new phytase gene, appA, was cloned from P. wasabiae by degenerate PCR and TAIL-PCR. The open reading frame of appA consisted of 1,302 bp encoding 433 amino acid residues, including 27 residues of a putative signal peptide. The mature protein had a molecular mass of 45 kDa and a theoretical pI of 5.5. The amino acid sequence contained the conserved active site residues RHGXRXP and HDTN of typical histidine acid phosphatases, and showed the highest identity of 48.5% to PhyM from Pseudomonas syringae. The gene fragment encoding the mature phytase was expressed in Escherichia coli BL21 (DE3), and the purified recombinant phytase had a specific activity of 1,072$\pm$47 U/mg for phytate substrate. The optimum pH and temperature for the purified phytase were pH 5.0 and 50$^{\circ}C$, respectively. The $K_m$ value was 0.17 mM, with a $V_{max}$ of 1,714 $\mu$mol/min/mg. This is the first report of the identification and isolation of phytase from Pectobacterium.

Development of a position sensitive CsI(Tl) crystal array

  • A position-sensitive CsI(Tl) crystal array coupled with the multi-anode position sensitive photomultiplier tube (PS-PMT), Hamamatsu H8500C, has been developed at the Institute of Modern Physics. An effective, fast, and economical readout circuit based on discretized positioning circuit (DPC) bridge was designed for the 64-channel multi-anode flat panel PSPMT. The horizontal and vertical position resolutions are 0.58 mm and 0.63 mm respectively for the 1.0 × 1.0 × 5.0 ㎣ CsI(Tl) array, and the horizontal and vertical position resolutions are 0.86 mm and 0.80 mm respectively for the 2.0 × 2.0 × 10.0 ㎣ CsI(Tl) array. These results show that the CsI(Tl) crystal array with low cost could be applied in the fields of medical imaging and high-resolution gamma camera.

Korean-Chinese Person Name Translation for Cross Language Information Retrieval

  • Named entity translation plays an important role in many applications, such as information retrieval and machine translation. In this paper, we focus on translating person names, the most common type of name entity in Korean-Chinese cross language information retrieval (KCIR). Unlike other languages, Chinese uses characters (ideographs), which makes person name translation difficult because one syllable may map to several Chinese characters. We propose an effective hybrid person name translation method to improve the performance of KCIR. First, we use Wikipedia as a translation tool based on the inter-language links between the Korean edition and the Chinese or English editions. Second, we adopt the Naver people search engine to find the query name's Chinese or English translation. Third, we extract Korean-English transliteration pairs from Google snippets, and then search for the English-Chinese transliteration in the database of Taiwan's Central News Agency or in Google. The performance of KCIR using our method is over five times better than that of a dictionary-based system. The mean average precision is 0.3490 and the average recall is 0.7534. The method can deal with Chinese, Japanese, Korean, as well as non-CJK person name translation from Korean to Chinese. Hence, it substantially improves the performance of KCIR.

  • 중국 시장에서는 한국 화장품의 점유율이 급격히 증가하고 세계 시장에서의 입지가 점차 안정되고 있다. 중국 소비자들이 한국 화장품에 열광하는 이유는 다음과 같다. 우선 한국 화장품은 아시아계 중국 소비자에게 적합하다. 둘째, 한국 화장품의 스타 효과는 의심할 여지가 없는 효과를 발휘하여 한국 화장품이 빠르게 발전하고 중국 시장을 빠르게 점유하고 있다. 한국 화장품에 대한 중국 소비자의 만족도는 한국 화장품 시장에 직접적인 영향을 미친다. 따라서 본 연구의 연구 대상은 '중국 소비자'로 정하고, '한국 화장품에 대한 중국 소비자의 만족도에 영향을 미치는 요인'에 초점을 맞추고 있다. "한국 화장품에 대한 중국 소비자 만족도" 모델을 구축하여 관련 영향 요인을 찾아내고 가설에 대해 시뮬레이션을 하여 중국 소비자가 한국 화장품에 대한 만족도에 실제로 영향을 미치는 요인을 분석하여 제안한다.

Wearable Intelligent Systems for E-Health

  • Due to the increasingly aging population, there is a rising demand for assistive living technologies for the elderly to ensure their health and well-being. The elderly are mostly chronic patients who require frequent check-ups of multiple vital signs, some of which (e.g., blood pressure and blood glucose) vary greatly according to the daily activities that the elderly are involved in. Therefore, the development of novel wearable intelligent systems to effectively monitor the vital signs continuously over a 24 hour period is in some cases crucial for understanding the progression of chronic symptoms in the elderly. In this paper, recent development of Wearable Intelligent Systems for e-Health (WISEs) is reviewed, including breakthrough technologies and technical challenges that remain to be solved. A novel application of wearable technologies for transient cardiovascular monitoring during water drinking is also reported. In particular, our latest results found that heart rate increased by 9 bpm (P < 0.001) and pulse transit time was reduced by 5 ms (P < 0.001), indicating a possible rise in blood pressure, during swallowing. In addition to monitoring physiological conditions during daily activities, it is anticipated that WISEs will have a number of other potentially viable applications, including the real-time risk prediction of sudden cardiovascular events and deaths.

  • The heat transfer characteristics between liquid lead bismuth eutectic (LBE) and helium are of great significance for the two-loop cooling system based on an accelerator-driven system (ADS). This paper presents an experimental study on the resistance characteristics and heat transfer performance in a LBE-helium experimental loop of ADS. Pressure drops in the LBE loop, the main heat transfer, and the coupled heat transfer characteristics between LBE and helium are investigated experimentally. The temperature of LBE has a significant effect on the LBE thermo-physical properties, and is therefore considered in the prediction of pressure drops. The results show that the overall heat transfer coefficient increases with the increasing helium flow rate and the decreasing inlet temperature of helium. Increasing the LBE Reynolds number and LBE inlet temperature promotes the heat transfer performance of main heat transfer and thus the overall heat transfer coefficient. The experimental results give an insight into the flow and heat transfer properties in a LBE-helium heat exchanger and are helpful for the optimization of an ADS system design.

  • Chinese construction industry has been enjoying a good opportunity for development which appeals to lots of foreign investment. However, the difference in socioeconomic environment and legal environment brings difficulties to foreign enterprises. This paper analyses the institution environment in Chinese construction market. The main objective is to form an insight of Chinese construction environment, and to identify critical factors that may block the way of foreign enterprises.

Exploring the adoption of IPD practices in Chinese construction industry

  • Integrated Project Delivery (IPD) is a procurement method that has been proved to improve construction project performance. However, in China implementation of IPD practices in construction projects is unknown though some researchers have studied the problems and constraints in adoption IPD. The purpose of this study was to explore IPD adoption in Chinese construction industry. Critical components of IPD implementation were reviewed, and questionnaires were distributed to collect industry views. The results revealed that IPD uptake is still low. In particular, the liability waiver and shared risks and rewards have been rarely used. In addition, co-location, value engineering method and the new compensation approach have also been hardly adopted. Some practices related to early involvement of key parties were adopted. Surprisingly, the findings indicate that the client has been continuously involved in the projects. The findings may imply that the legal issues and problems of contractual frameworks are still constraining IPD implementation in Chinese construction industry.

  • In this review paper, we report the characteristics of the critical current density in FeAs based superconductors which is newly discovered by Hosono group of Tokyo Institute of Technology on 2008. Since the many specimens in present stage are not single crystals, there are two kinds of critical current density observed in the specimens which are so-called local and global critical current densities. Therefore, it is necessary to evaluate both kinds of critical current densities. The history effect in which the global critical current density shows different values in increasing and decreasing magnetic field is also observed when the specimens have the local and the global critical current densities. The wire which critical current is 180 A is successfully developed with using the knowledge of abovementioned characteristics of two kinds of critical current densities and the history effect.

  • The purpose of this study was to investigate Chinese men's perception and satisfaction on Korean apparel products. The 1000 Questionnaires were distributed to Chinese men, and returned 863 questionnaires were analyzed by mean and percentage with SPSS 10.0. The results of this study were as follows: Chinese men had more perception on their own brand and Italy brand than UK, Korean, and Japan brand. Over age fifties of Chinese men preferred Korean brand to the other ages. Chinese men perceived positively Korean fashion products as more trendy, very creative, high-class, qualitative, expensive and creditable, otherwise, there were little famous brands among Koran apparel products. Chinese men satisfied on Korean apparel products positively, specially, age forties and fifties, resident from Harbin, income 2 and 3 level, and young casual mild group showed higher satisfaction on Korean apparel products than the others.
