A Study of Cultural Aspects of Kimchi in 「Banchandeungsok」 (「반찬등속」에 기록된 김치의 식문화적 고찰)

  • "Banchandeungsok" is recipe book written by a woman from a noble family who lived in Cheongju during the late 1900s. The book has 9 among 46 Kimchi recipes. In this study, the literatures from 1700 to 1900s, "Jeungbosanlimkyeongje", "Gyuhapchongseo", "Siuijeonseo", "Buinpilji", "Bogamrok", and "Josenmussangsinsikyorijebeob" were reviewed to examine the significance of "Banchandeungsok" Kimchi in the aspect of food culture. The results are as follows. "Banchandeungsok" Kimchies were considered as typical type of Kimchi of the late Joseon, and the main ingredients include Chinese cabbage, white radish, and cucumber. Salted-fish juice was not used for making Mukimchi, Bachujjanji, Jjanji in "Banchandeungsok", and gallic, ginger, green onion were used as main spice. Therefore, "Banchandeungsok" Kimchies belongs to Damjeo (light salting) type, which is characterized by light and clear flavor of present Cheongju Kimchi. While salted-fish juice was not used for "Banchandeungsok" Kimchies, yellow corbina was used in the most of "Banchandeungsok" Kimchies. Various forms of pepper were used in "Banchandeungsok" Kimchies - green pepper, peper leaf, minced, sliced shredded and powdered red pepper. Two kinds of Kkakdugi were introduced in "Banchandeungsok", which is the first record in the literatures. A typical type of stuffed cucumber pickles and unique type of cucumber Kimchi appeared in "Banchandeungsok".

Analysis of Bronze Artifacts and Gold Ornaments Excavated from Xiongnu Tombs No. 2~4 at Duurlig Nars in Mongolia (몽골 도르릭나르스 흉노 무덤 2~4호분 출토 청동 및 금제 유물 분석)

  • A purpose of this research is to find out characteristic of bronze artifacts and gold ornaments excavated from Xiongnu tombs No. 2~4 at Duurlig Nars in Mongolia through scientific analysis of them. The Tombs are comparatively small. There were still lots of relics remaining although the tombs had been already robbed. Also the tombs are evaluated important since the origin of them show coexisting of chinese and northern style. First of all, an analysis result about bronze vessels found in this site, they have high lead(Pb) content and relatively low tin(Sn) content, as compared with the Bronze Han Mirror and End-fittings of Bronze Parasol Rib. Especially in case of bronze tray and bronze lamp from the no. 2 tomb and also bronze cauldron from the no. 4 tomb contain only 1wt% of tin which means binary alloy composition(Cu-Pb). Also, in the case of gold ornaments found in the no. 2 tomb, they have comparatively high purity. And the research suppose that the high possibility of that they were used soldering using alloy of Au-Cu or diffused bonding(using malachite and copper oxide) for joining gold grains of gold granulation ornament. Further scientific research and analysis in Mongolia and other countries will provide more clues to solve mystery of Xiongnu culture.

Cisplatin and Extract of Tissue Cultured Mountain Ginseng-Induced Apoptosis in Human Cervical Cancer Cells (인체 자궁암세포에서 cisplatin과 산삼배양근추출물에 의한 apoptosis유도)

  • Mountain ginseng is a perennial crop rarely found in the deep mountains of Korea. The medicinal effect of the mountain ginseng is well known as a panacea in traditional Chinese medicine for a long time. But scientific studies to elucidate the medicinal effect of the mountain ginseng have never been made on account of lack of sample. Recently an improved method of adventitious root culture system through the use of bioreactor has been developed in Panax ginseng that seems to be a reliable way of commercialization of root derived secondary metabolites. This experiment was conducted to evaluated chemotherapeutic effect against human cervical cancer cells by cisplatin (CDDP) and extract of tissue cultured mountain ginseng (ETCMG). CDDP and ETCMG-induced apoptotic cell death in human cervical cancer cell line, HeLa was confirmed by the analysis of cell growth, morphological changes, DNA fragmentation, flow cytometry showed that ETCMG is an inducer of apoptosis and synergizes with CDDP. These results suggest that ETCMG present evidence of anticancer effect and could have a possibly natural therapeutic potential in cervical cancer patients.

  • 본 논문은 얼굴 인식 또는 표정 인식 분야에 있어서 중요한 특징을 나타내는 얼굴과 얼굴의 주요소인 눈과 입, 눈썹의 영역 추출 및 그의 윤곽선·추출에 관한 방법을 제시한다. 얼굴요소의 영역 추출은 엣지 정보와 이진화 영상을 병합하여 이용한 프로젝션 분석을 통하여 얼굴 및 각 얼굴요소를 포함하는 최소포함사각형(MER: Minimum Enclosing Rectangle)을 추출하였다. 얼굴 영상에 관련된 윤곽선 연구에는 가변 템플릿(Deformable Template), 스네이크(Snakes: Active Contour Model)를 이용하는 연구들이 이루어지고 있는데 가변 템플릿 방법은 수행시간이 느리고 추출된 윤곽선의 모양이 획일 된 모양을 갖는 특성이 있다. 본 논문에서는 사람마다 얼굴요소의 모양의 개인차가 반영되고 빠른 수렴을 할 수 있는 스네이크 모델을 정의하여 눈, 입, 눈썹, 얼굴의 윤곽선 추출 실험을 하였다. 또한 스네이크는 초기 윤곽선의 설정이 윤곽선의 추출 곁과에 큰 영향을 미치므로, 초기 윤곽선의 설정 과정이 매우 중요하다. 본 논문에서는 얼굴 및 각 얼굴요소를 포함하는 각각의 최소 포함 사각형(MER)을 추출하고, 이 추출된 MER 내에서 얼굴 및 각 얼굴요소의 일반적인 모양을 초기 윤곽선으로 설정하는 방법을 사용하였다. 실험결과 눈, 입, 얼굴의 MER의 추출은 모두 성공하였고, 눈썹이 흐린 사람들의 경우에만 눈썹의 MER추출이 졸지 않았다. 추출된 MER을 기반으로 하여 스네이크 모델을 적용한 결과, 눈, 입, 눈썹, 얼굴의 다양한 모양을 반영한 윤곽선 추출 결과를 보였다. 특히 눈의 경우는 1차 유도 엣지 연산자에 의한 엣지 와 2차 유도 연산자를 이용한 영점 교차점(Zero Crossing)과 병합한 에너지 함수를 설정하여 보다 더 나은 윤곽선 추출 결과를 보였다. 얼굴의 윤곽선의 경우도 엣지 값과 명도 값을 병합한 에너지 함수에 의해 비교적 정확한 결과를 얻을 수 있었다.잘 동작하였다.되는 데이타를 입력한후 마우스로 원하는 작업의 메뉴를 선택하면 된다. 방법을 타액과 혈청내 testosterone 농도 측정에 응용하여 RIA의 결과와 비교하여 본 바 상관관계가 타액에서 r=0.969, 혈청에서 r=0.990으로 두 결과가 잘 일치하였다. 본 실험에서 측정된 한국인 여성의 타액내 testosterone농도는 107.7$\pm$12.0 pmol/l이었고, 남성의 타액내 농도는 274.2$\pm$22.1 pmol/l이었다. 이상의 결과로 보아 본 연구에서 정립된 EIA 방법은 RIA를 대신하여 소규모의 실험실에서도 활용할 수 있을 것으로 사려된다.또한 상실기 이후 배아에서 합성되며, 발생시기에 따라 그 영향이 다르고 팽창과 부화에 관여하는 것으로 사료된다. 더욱이, 조선의 ${\ulcorner}$구성교육${\lrcorner}$이 조선총독부의 관리하에서 실행되었다는 것을, 당시의 사범학교를 중심으로 한 교육조직을 기술한 문헌에 의해 규명시켰다.nd of letter design which represents -natural objects and was popular at the time of Yukjo Dynasty, and there are some documents of that period left both in Japan and Korea. An Anti-Tumor Effects of Dioscorea bulbifera L. on Stomach Cancer Cells (황약자(黃藥子)의 위암세포(胃癌細胞)에 대(對)한 항암(抗癌) 효능(效能) 검색(檢索))

  • Background : Nowadays a lot of research is based on natural substances or materials world wide since many kinds of side effects are accompanied by anti tumor chemotherapy. In Chinese medicine, Dioscorea bulbifera L is widely used to treat many kinds of cancer, but in Korea it is rarely used. Therefore, we need to scientifically identify anti tumor effects of Dioscorea bulbifera L. Objective : We aimed to identify anti tumor effects of Dioscorea bulbifera L on the stomach cancer cells through molecular biological methods. Materials & Methods : We used AGS, a stomach cancer cell from American Type Culture Collection. We injected the boiled extract of Dioscorea bulbifera L 5 ul(sample 1), 10ul(sample 2) to cultured media(ml) for 0,6,12,18,24 hours. We measured the killing effect on stomach cancer cells through Tryphan blue exclusion test and suppressive effect on viability of stomach cancer cells via MTT assay. Results : Tryphan blue exclusion test showed that each test group killed more stomach cancer cells than the controlled group with a dosage-dependent, but not significantly. MTT assay showed that each test group had a more suppressive effect on viability of stomach cancer cells than the controlled group without a dosage-independent, but not significantly. The cell cycle analysis via flow cytometry showed that the test group extended cell cycle, and there was no peak in M phase, the number of sub G1, G0, G1 phase cells increased a little, but not significantly. Conclusion : This experiment showed that Dioscorea bulbifera L. has an anti-tumor effect, but not significantly. This is in vitro experiment and basic experiment on Dioscorea bulbifera L. We hope more progressive researches on Dioscorea bulbifera L. will be conducted and its anti tumor will be more accurately identified.

The production and characterization of anti-Naegleria fowleri monoclonal antibodies (Naegleyiu fowleri에 대한 단세포군 항체의 생산과 그 특성에 관한 연구)

  • Naegleria fowleri, a free-living amoeba commonly found in moist soil and fresh water, enters the body via the nasal mucosa and migrates along the olfactory nerve to t he brain, where it causes acute amoebic meningoencephalitis. In the present study 7 clones secreting monoclonal antibodies (McAbs) against N. fowleri were produced and the effector function of them was investigated. Their isotopes were IgGl (Nf 1, Nf 154), 19G3 (Nf 137) and 19A (Nf 1, Nf 2, Nf 256, Nf 279). Five McAbs (McAb Nf 2, Nf 279, Nf 27, Nf 154, Nf 137) were specific for N. fowleri by ELISA and recognized the antigenic determinants located on the trophoBoite surface by IFAT and immunoperoxidase stain. These aye McAbs had capacity to agglutinate N. fowleri trophozoites and inhibited the growth of the amoeba in culture medium. McAb Nf 2 inhibited proliferation of trophozoites in vitro significantly. Also the cytotoxicity of JV. fowleri against CHO cell was reduced in the presence of McAb Nf 2 and McAb Nf 154. From these results McAb Nf 2 was confirmed to weaken the virulence of the amoeba among 7 screened McAbs.

Proteinase activity in the isolates of Trichomonas vaginalis according to their pathogenicity (질트리코모나스의 병원성과 단백 분해 효소와의 상관성)

  • Ten axonic isolates of Trichomonns uaginolis were subcutaneously injected to the BALB/c mice in order to assess their pathogenicity by means of so-called "mouse assay" method. All the isolates revealed neutral and acid proteinase activities both in their Iysates and in culture media, but the specific activities of both proteinases in the severely pathogenic group were significantly higher than the mildly pathogenic group (p < 0.05). In the SDS-PAGE system in which the electrophoretic gels contained 0.4% gelatin as the substrate, five different handing patterns of trichomonal proteinases were detected, and the patterns were closely related with the pathogenicity of the isolates of T. vosinalis. All five bands might be regarded as cysteine proteinases group in the inhibitor assays. The cytotoxicity of the Iysates of T. vaginalis to the target Chinese hamster ovarian (CHO) cell line was also significantly different according to the pathogenicity of the isolates, and generally lower in the Iysates treated with cysteine proteinase inhibitors than in the control Iysates. In summarizing the results, it might be considered that the proteinases of T.vaginalis showing five electrophoretic banding patterns are closely related with the pathogenicity and cytotoxicity of the isolates of T. voginolis.

Identification and Biological Activity of Two New Phytotoxins Isolated from Botrytis cinerea (Botrytis cinerea로부터 분리한 두 개의 새로운 phytotoxin의 구조 결정 및 생물활성)

  • We discovered two novel phytotoxins produced by the pathogenic fungus, Botrytis cinerea. Among the twenty-five B. cinerea isolates, which were obtained from various host plants in 1994 and 1996, twenty-two showed strong or moderate pathogenicity on five plants such as cucumber, tomato, red pepper, tobacco and Chinese cabbage. The culture filtrate of the B. cinerea 2-16 strain showed the most potent phytotoxic activity in a tobacco leaf-wounding assay. Two novel phytotoxins were isolated from the liquid cultures of B. cinerea 2-16 by ethyl acetate extraction, flash silica gel column chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, preparative TLC and subsequently preparative HPLC. Their chemical structures were determined to be 3-O-acetyl botcinol and 3-O-acetyl botcinolide, respectively, by mass and NMR spectral analyses. These two phytotoxins caused leaf necrosis in a leaf-wounding bioassay, and significant electrolyte leakage from leaf tissues of tobacco. In the two bioassays tested, 3-O-acetyl botcinol exhibited stronger phytotoxic activity than 3-O-acetyl botcinolide. This is the first report on the production of both 3-O-acetyl botcinol and 3-O-acetyl botcinolide from B. cinerea.

Consideration of Making Techniques for Red Painted Roof Tiles from Presumed Site of Daetongsa in Gongju Using Nondestructive Analysis (비파괴 분석을 활용한 공주 대통사 추정지 출토 주칠흔 기와의 제작기술 검토)

  • The Daetongsa temple is the earliest temple to be constructed during the era of the Three Kingdoms in ancient Korea. The main architect, purpose, and name of the temple have been confirmed through ancient literature and archeological materials carved in the Chinese letter, Daetong, excavated around Gongju. However, the location and range of the temple have remained elusive and were discussed in various studies. In this study, we examine the roof tiles obtained from the presumed site of the Daetongsa temple. The tiles were found to contain traces of red paint (red pigments) on their surface and analyzed using nondestructive techniques. The results imply that roof tiles were made using clay tablets and wooden cylinders, with latticed cloth in between. Additionally, some wooden cylinders appeared to comprise numerous wooden plates tied together by strings. The clay tablets used to make the roof tiles were produced from the source clay via the sorting process. The traces of red paint on the surface of the roof tiles were verified to be traditional pigments used for painting wooden buildings. These pigments were extracted from red ocher or red clay (Seokganju), mainly consisting of iron oxide. In the literature, the location of provenance sites for Seokganju is estimated to be far from Gongju. However, the materials for extracting the red pigments were relatively easy to source because most rocks comprised iron oxides. Therefore, it is necessary to discuss the provenance of the red pigments around the presumed site of Daetongsa.

Design and Implementation of Luo-kuan Recognition Application (낙관 인식을 위한 애플리케이션의 설계 및 구현)

  • In oriental paintings, there is Luo-kuan that expressed in a single picture by compressing the artist's information. Such Luo-kuan includes various information such as the title of the work or the name of the artist. Therefore, information about Luo-kuan is considered important to those who collect or enjoy oriental paintings. However, most of the letters in the Luo-kuan are difficult kanji, kanzai, or various shapes, so it is difficult for the ordinary people to interpret. In this paper, we developed an Luo-kuan search application to easily check the information of the Luo-kuan. The application uses a search algorithm that analyzes the captured Luo-kuan image and sends it to the server to output information about the Luo-kuan candidates that are most similar to the Luo-kuan images taken from the database in the server. We also compared and analyzed the accuracy of the algorithm based on 170 Luo-kuan data in order to find out the ranking of the Luo-kuan that matched the Luo-kuan among the candidates. Accuracy Analysis Experimental Results The accuracy of the search algorithm of this application is confirmed to be about 90%, and it is anticipated that it will be possible to develop a platform to automatically analyze and search images in a big data environment by supplementing the optimizing algorithm and multi-threading algorithm.
