Enzymatic Properties of Barley $\alpha$-Amylase Chimeric Enzymes Produced by Staggered Extension Process (Staggered Extension Process를 통해 제조한 보리 알파아밀라제 Chimera 효소의 특성)

  • Barley malt produces two different $\alpha$-amylase isozymes (AMY1 and AMY2), which share up to 80% of amino acid sequence identity with each other. However, their enzymatic properties differ remarkably. In this study, five chimeric enzymes between AMY1 and 2 were constructed by staggered extension process (StEP) technique, and their enzymatic properties were characterized. According to the results, chimeric AMY-D2, D8, and E12 showed the mixed or intermediate types of calcium-dependent activity between AMY1 and 2. Meanwhile, only AMY-E10 chimera could be significantly inhibited by barley $\alpha$-amylase/subtilisin inhibitor (BASI) protein. Chimera AMY-C6 showed the same calcium-dependency as AMY1, while AMY-E10 was closely similar to AMY2. As a result, it can be proposed that some amino acid residues in the region II, III, and IV of barley $\alpha$-amylases can play very important roles in the interaction with BASI, and those in III, V, VI, and VII may partly affect on the calcium-dependent activity.

  • The prediction of the separation trajectories of external stores released from a military aircraft is an important task in the aircraft design area having the objective to define the operational and release envelopes. This paper presents the results obtained for store separation by employing commercial softwares, FLUENT and CFD-FASTRAN. FLUENT treats the rigid body motion by employing a remeshing scheme. CFD-FASTRAN uses Chimera(overset) grid and interpolations. It was found that, for the prediction of the trajectories and behavior of the stores separated from the wing, both codes show the good agreement with the experimental results.

  • A dynamic simulation on the missile staging system is conducted with numerical techniques. Both Euler equations and Navier-Stokes equations are numerically solved respectively. The dynamic simulation of two moving bodies is fully integrated into the computational fluid dynamics solution procedure. The Chimera grid scheme is applied in this simulation for unsteady supersonic flow analysis with dynamic modeling. The objective of the study is to investigate the problem pertaining to possible unstability in missile staging. In addition, the computational comparison between in viscid and viscid flow solvers is also performed in this study.

  • A numerical analysis was made to investigate the intensity diminution of a simple silencer for high pressure blast flow fields. Reynolds-Averaged Wavier-Stokes equations were solved for an axisymmetric computational domain constructed by multi block Chimera grids. A blast flow field without the silencer was also calculated to validate the present numerical method. The evolution of high pressure blast flow fields was observed by depicting calculated contours of pressure and Mach number. It was found that the tested silencer could achieve 76 percent intensity diminution.

  • A numerical analysis was made for the unsteady flow fields of rotor system of a Tilt-Rotor aircraft in cruise mode. The Reynolds-averaged thin-layer Navier-Stokes equations were discretized by Roe's upwind differencing scheme and integrated in time by the LU-SGS algorithm. The computational domain of the rotor system was constructed by six multi-block Chimera grids. Simulated unsteady flow fields of rotating blades were studied in several different view points.

  • A computational study has been performed using a chimera scheme to study the various operating processes inside a ballistic range. The compression flow fields in the pump tube and projectile motion in the launch tube are captured for various piston masses and diaphragm rupture pressures. The effect of a shock tube in between the pump tube and launch tube is analyzed. The results are compared with available experimental data. It is noted that, by adding a shock tube in between the pump tube and launch tube, the peak pressure in the ballistic range can be reduced without appreciable reduction in the velocity of the projectile.

  • A numerical analysis was made In investigate the pressure diminution of a silencer with baffles for high pressure blast flow fields. Reynolds-Averaged Navier-Stokes equations were solved for an axisymmetric computational domain constructed by multi block Chimera grids. A blast flow field was calculated for the silencers that are with one and three baffles. The evolution of high pressure blast flow fields was observed by depicting calculated contours of pressure. It was found that the tested silencer could achieve 97.7 percent pressure diminution.
