β-Nicotinamide mononucleotide improves chilled ram sperm quality in vitro by reducing oxidative stress damage

  • Objective: The present study aimed to investigate the effect of β-nicotinamide mononucleotide (NMN) supplementation on ram sperm quality during storage at 4℃ in vitro. Methods: Tris-citric acid-glucose solution containing different doses of NMN (0, 30, 60, 90, and 120 µM) was used to dilute semen collected from rams and it was stored at 4℃. Sperm motility, plasma membrane integrity as well as acrosome integrity were evaluated at 0, 24, and 48 h time points after storage at 4℃. In addition, sperm mitochondrial activity, lipid peroxidation (LPO), malondialdehyde (MDA) content, reactive oxygen species (ROS) content, glutathione (GSH) content, superoxide dismutase (SOD) activity, and apoptosis were measured at 48 h time point after storage at 4℃. Results: Results demonstrate that the values obtained for sperm motility, acrosome integrity, and plasma membrane integrity in the NMN treatments were significantly higher than control (p<0.05). The addition of 60 µM NMN significantly improved ram sperm mitochondrial activity and reduced LPO, MDA content, and ROS content compared to control (p<0.05). Interestingly, sperm GSH content and SOD activity for the 60 µM NMN treatment were much higher than those observed for control. NMN treatment also decreased the level of Cleaved-Caspase 3, Cleaved-Caspase 9, and Bax while increasing Bcl-2 level in sperm at 48 h time point after storage at 4℃. Conclusion: Ram sperm quality can be maintained during storage at 4℃ with the addition of NMN at 60 µM to the semen extender. NMN also reduces oxidative stress and apoptosis. Overall, these findings suggest that NMN is efficient in improving the viability of ram sperm during storage at 4℃ in vitro.

  • This study describes the experimental study that focuses on the effects that distributor shapes and flow rate variations have an influence on the stratification in a rectangular thermal storage tank. Experiments were carried out under the conditions that the flow rates of working fluid are 20, 10 and 5/min. The storage tank is initially filled with chilled water of 1C, and is extracted through the bottom at the same rate as the return warm water from load is entered through the distributor at the top of the tank. The thermo-cline forms at the top of the storage tank as the warm water enters the tank from the load through the distributor and the thermo-cline thickness increases with time. Emphasis is given to the effects of mixing at the inlet that increases the thermo-cline decay Flow rate variation and inlet distributor shapes are the important parameters in deciding the performance of a storage system. Stratification degree increases with decreasing in inlet flow rate under 10/min. Experiments shows that better thermal stratification can be obtain using the distributor to limit momentum mixing at the inlets and outlets. Also, 12% of improvement in the thermal energy usage has been achieved using the modified distributor discharging same flow rate in each lateral ports.

Quality Change in Precooked Sardine during Frozen Storage (자숙 정어리육의 동결저장중의 품질변화)

  • Qualify changes of the precooked frozen sardine (Sardinops melanosticta) during frozen storage were investigated by measuring extractable protein, expressible drip, available lysine and lipid oxidation as peroxide value. Fresh sardine was dressed, washed in chilled water, cooked in boiling water to have 55Cand70 at the center of the body, frozen at 40C, and finally stored at 20C for 84 days. The quality factor mentioned above were determined in both ordinary and dark muscle at 14 day intervals through the period of storage. When cooked at 70C, the changes in expressible drip were less than that of raw and the one cooked at 55C. In observation of the extractability of muscle protein, no great change in extractable sarcoplasmic protein was observed, the extractable myofibrillar protein, however, showed a tendency to decrease during the period of frozen storage, accompanying the increase of the alkali-soluble protein. That was more excessive in ordinary muscle than dark muscle. Lipid oxidation of dark muscle was faster than that of ordinary muscle. Acid value was not changed, and peroxide value of the samples cooked at 70Cand55 was higher than that of raw at the early stage of the storage, after 40-50 day storage, it became lower than that of raw muscle.

Evaluation of Microbiological, Physicochemical, and Sensory Properties of Galbi-jjim Prepared by Sous-vide and Cookchill Method at Different Temperatures

  • The aim of this study was to evaluate the physico-chemical, sensory, and microbiological properties of ready-to-eat Korean traditional seasoned beef ribs ("galbi-jjim") prepared by sous-vide/cookchill technology during storage at three different temperatures (4, 10, and 20C). Beef short ribs marinated in soy sauce for 24 h at 3C were packed with vegetables under vacuum. Vacuum-packed beef ribs mixed with vegetables were heated at 90C for 90 min in a water bath, and then immediately chilled below 3C within 120 min in an ice slurry. Physicochemical (pH, water activity, TBARS, Lab color, and texture profile), sensory (appearance, odor, flavor, texture, and acceptance) and microbiological (Coliform, Escherichia coli, food-borne pathogenic bacteria) properties of the samples were determined during storage at different temperatures. Results showed that pH, aw, and sensory evaluation of products were not affected in any consistent way as a function of either storage duration or temperature. Coliform, E. coli and food-borne pathogens were not detected during storage at any temperature. However, TBARS significantly increased during storage period (p<0.05). Based on TBARS values, SV/CC "galbi-jjim" can be stored for 15 d, 12 d and 1 d at 4, 10 and 20C, respectively.

  • This study was undertaken to prolonging the freshness in Squid(Todarodes pacificus) on the effects of ice storage methods(tap water ice, electrolytic water ice) using stainless diaphragm electrolytic instrument and also test an efficiency of instrument. Basically, stainless diaphragm electrolytic instrument studied for changes of pH on difference water flow and ampere. The lower water flower and higher ampere made low pH on acid part of electrolytic instrument. Squid samples were stored in tap water ice, acid part of electrolytic water and base part of electrolytic water and used in studying the changes of VBN and skin color through storage. Acid water had strong sterilization effects and VBN was lower levels of acid water than the others. Base water had not sterilization effects. Tap water ice storage was more sterilizing effects than base water ice storage but less effects than acid water ice storage. Lightness of Squid skin showed getting decrease at storage of ice water of all storage methods but acid water ice storage showed more retarding than the other two storage. From these results, it could be suggested that acid electric water ice storage is effective in extendance the shelf-life of squid at chilled storage.

Changes of in Vacuum Packed Pre-aged Venison with Seasoning during Chilled Storage (양념으로 사전 숙성하여 진공포장한 사슴고기의 냉장저장 중 품질 변화)

  • This study was carried out to evaluate the quality characteristics of the vacuum packed pre-aged venison with seasoning during chilled storage. The ham of deer were cut to cube and two seasonings such as red pepper sauce (T1) or soy sauce (T2) were seasoned by the proportions or meat to seasonings (1:1), respectively. The seasoned venison was aging by fill into plastic box at 0±1C for 10 days. And then, the aged meat from each pack was vacuum-packaged and stored at 0±1C for up to 28 days. pH were significantly decreased with storage periods in all treatments (p<0.05). Salinity (%) were no significant differences in T1 during the whole storage, but T2 had significantly high content (1.33%) at the storage days of 28 (p<0.05). Saccarinity (%) of T1 were significantly higher than that of T2 during the whole storage periods (p<0.05). As storage time extended the value of L and b in T1 were increased and those values decreased in T2. a and shear force (g/cm2) or T1 and T2 were increased as the storage period passed In the result of sensory evaluation. all treatment had relatively high score (T16.75\~T27.00) in overall acceptability during the storage period of 28 days. In conclusion, this study suggest that vacuum packed pre-aged venison with seasoning may be useful for new deer meat menu.

Effects of Edible Films Containing Procyanidin on the Preservation of Pork Meat during Chilled Storage

  • Procyanidins, which are natural antioxidants and antimicrobials found in grapes, enhance the quality and extend the shelf life of meat. We explored the effects of edible films incorporating procyanidins on pork loin stored for various times. Procyanidins (0, 0.1, and 0.3%, w/w) were incorporated into the edible films. We assessed meat color, pH, levels of volatile basic nitrogen (VBN) and 2-thiobarbituric acid-reactive substances (TBARS), and microbial populations for 14 d. The chromaticities and pH values of pork loin wrapped in film containing procyanidins (0.1% and 0.3%) generally increased (p<0.05) with storage time. VBN and TBARS levels, and total bacterial and Escherichia coli (E. coli) counts, significantly decreased (p<0.05) in the procyanidin groups. In particular, procyanidins strongly inhibited TBARS formation. Thus, our findings suggest that edible film impregnated with procyanidins inhibits lipid oxidation and microbial growth, thereby enhancing the quality and shelf life of pork meat.

Microbial changes under packaging conditions during transport and comparison between sampling methods of beef

  • This study was performed to evaluate the microbial and temperature changes of boxed beef during transport and distribution under vacuum and modified atmosphere packaging (MAP), and to compare between excision and swab sampling for 15 days. The top round and striploin (quality grade 1) from Hanwoo steers at 2 days post-slaughter were obtained from a local meat processing plants and chilled at 4±2C in a cold room. The boxes were transported under refrigeration (4±2C) to the laboratory within half an hour. Vacuum and MAP packs were subsequently taken out from cool boxes, and microbiological examinations were carried out at 0, 6, 12, and 24 h of storage time. MAP was more effective than vacuum packaging for the inhibition of total aerobic, lactic acid bacteria and Pseudomonas (p < 0.05). Microbial loads of swab methods were slightly lower than those of excision ones (p < 0.05). The results of this study could be utilized by meat consumers in future studies as well as by manufacturers to determine the ideal storage conditions for cool boxed meat, thus ensuring reduced economic losses due to spoilage.

Physicochemical and Sensory Characteristics of Domestic Vacuum Packaged Pork Hams (국내산 진공 포장 냉장 돈육 뒷다리육의 이화학적 및 관능적 특성)

  • This study were carried out for investigation of physicochemical and sensory characteristics of shelf-life of Korean vaccum packaged chilled pork hams for export. The samples were stored at 0±1C (A and B companies) and 2±1C (C and D companies). In the proximate analysis of composition of hams, moistures were 75.36~76.57%, crude proteins 19.26~21.08%, crude fat 1.40~3.69% and crude ash 1.03~1.13%. TBA values were 0.192~0.264 mgMA/kg and did not change much during all storage times regardless of storage temperature. VBN values were 12.14~14.01mg / 100g after 50 days in the storage at 0C, but the samples stored at 2C exceeded 20mg/100g at 50 days. The values of pH were 5.71~6.50, which are higher than those of loins during all storage times. Purge losses were 1.23~1.98% at the initial time and 2.83~10.59% after 50 days. So, the variation among samples appeared to be large. Cooking losses appeared to be 33.86~56.24%. In the result of sensory evaluation of cooked meat, the ham of B company obtained good scores in all items. But, after 20 days, all samples were not significantly different in total acceptability.

  • The heat transfer characteristics of ice storage system of falling film type using sprial coil is investigated. The experimental facilities consisted of a water tank, spiral coils located above the tank, an upper water distributor, and a circulating water pump. Water is distributed uniformally over the spiral coils and it forms falling thin films. In the process of freezing, ice is formed on outside of the spiral coils through recirculation of tank water. In the process of melting, ice is melted with return water from the heat load, while the water is chilled again and drops into the tank. The results of falling film type of ice thermal storage system are as follows. The highly efficient shower flowrates for icing is near 3/min. Icing rates on spiral coils is rosed while brine flowrates is increased. Lower brine temperature is not only increased freezing rates but. also become higher total icing weight and overall heat transfer coefficient. Smaller shower flowrates is obtained lower water temperature on outlet for a long time. The amounts of quantity can be detected more accurately by measuring storage tank weight.
