Pharmacophore-Based Comparative Molecular Similarity Indices Analysis of CRTh2 Antagonists

  • Chemoattractant Receptor Homologous molecule expressed on Th2 cells (CRTh2) is a chemoattractant receptor with seven transmembrane helices targeted for inflammatory diseases such as asthma and allergic rhinitis. In this study, pharmacophore based Comparative Molecular Similarity Indices Analysis (CoMSIA) were performed on the series of 2-(2-(benzylthio)-1H-benzo[d]imidazol-1-yl) acetic acids derivatives. Initially, GASP module was used for generation of pharmacophore models using five highly active compounds from the dataset. Among the generated pharmacophores, the best pharmacophore model was selected based on fitness score and was used as template for the alignment of compounds which was used for CoMSIA analysis. The best predictions were obtained utilizing steric, hydrophobic and H-bond acceptor parameters showing a $q^2$=0.559 and $r^2$=0.730. 15 test set compounds was used to investigate the predictive ability of the CoMSIA model. Contour maps suggested that presence of bulky substituents and H-bond acceptor atoms at $5^{th}$ position of benzene ring will increase the activity of the compounds. The results obtained from this study will be useful to design more potent CRTh2 antagonists.

Monocyte Chemoattractant Protein-1 (MCP-1)/CCL2 Induces the Chemotactic Activity of Human Eosinophils

  • Eosinophil is an improtant leukocyte in the development of various inflammatory diseases. Monocyte chemoattractant protein-1 (MCP-1) acts as a key regulator on monocyte movement, and activation of T cells and NK cells. However, the role of MCP-1 in eosinophils remains to be solved. In the present study, we examined the effect of MCP-1 on eosinophil migration, using human eosinophilic EoL-1 cells as an in vitro model of eosinophils. The surface expression of CCR2 in EoL-1 cells was little detected but MCP-1 strongly induced EoL-1 cell migration in a dose-dependent manner. Increased chemotactic activity due to MCP-1 was blocked by pertussis toxin, a $G_i/G_o$ protein inhibitor and U73122, a phospholipase C (PLC) inhibitor. These results suggest that MCP-1 activates $G_i/G_o$ protein and PLC and this signal pathway is involved in eosinphil movement. This finding supports the elucidation of pathogenic mechanism of eosinophilic inflammation such as asthma and atopic dermatitis.

주화성에 의한 활동성 정자 분리용 마이크로 바이오칩에 관한 연구 (A Study About Microbiochip for Separation of Motile Sperm by Using Chemotaxis)

  • This paper presents a new microchip which can separate motile sperm by chemotaxis. The microchip was developed to create longitudinal concentration gradient in the microchannel due to diffusion. Linearly good concentration gradient of chemoattractant was generated without any fluid control devices. In sperm separation experiment with the developed microchip, mouse sperm was used as sample and acetylcholine was selected as chemoattractant. Human tubal fluid (HTF), buffer solution, was introduced into the microchannel of the microchip and attractants diluted in ratio of 1, 1/2, 1/4, 1/8, 1/16, 1/32 and 1/64 including control (DI water) were dropped in each outlet by $2\;{\mu}l$ volume with micropippet. After 5min, $1\;{\mu}l$ sperm solution was dropped into inlet of the chip. After 10 min, when sperms reached to the outlet by chemotaxis, we counted sperms in each outlet by using microscopy. Consequently, we could separate progressive motile sperm with the new microchip. In the experiment, the most sperms were isolated at the outlet dropped with 1/16 diluted solution. The optimal concentration gradient to induce chemotaxis was about 0.625 mg/ml/mm.

CBP7 Interferes with the Multicellular Development of Dictyostelium Cells by Inhibiting Chemoattractant-Mediated Cell Aggregation

  • Calcium ions are involved in the regulation of diverse cellular processes. Fourteen genes encoding calcium binding proteins have been identified in Dictyostelium. CBP7, one of the 14 CBPs, is composed of 169 amino acids and contains four EF-hand motifs. Here, we investigated the roles of CBP7 in the development and cell migration of Dictyostelium cells and found that high levels of CBP7 exerted a negative effect on cells aggregation during development, possibly by inhibiting chemoattractant-directed cell migration. While cells lacking CBP7 exhibited normal development and chemotaxis similar that of wild-type cells, CBP7 overexpressing cells completely lost their chemotactic abilities to move toward increasing cAMP concentrations. This resulted in inhibition of cellular aggregation, a process required for forming multicellular organisms during development. Low levels of cytosolic free calcium were observed in CBP7 overexpressing cells, which was likely the underlying cause of their lack of chemotaxis. Our results demonstrate that CBP7 plays an important role in cell spreading and cell-substrate adhesion. cbp7 null cells showed decreased cell size and cell-substrate adhesion. The present study contributes to further understanding the role of calcium signaling in regulation of cell migration and development.

치수세포의 Substance P 자극 시 Interleukin-8과 Monocyte chemattractant protein-1의 분비에 관한 연구 (The effect of Substance P on the secretion of Interleukin-8 and MCP (Monocyte Chemoattractant Protein)-1 from the Human Dental Pulp Cells)

  • 본 연구는 치수 염증 시 IL-8과 MCP-1 분비에서 neuropeptide의 역할에 대해 관찰하고자 발거된 건전한 치아를 수직 파절시켜 치수조직을 채취하여 배양된 치수세포 및 혈관내피세포(ECV 304세포)를 각기 다른 농도의 Substance P(SP)로 12시간 자극하였고, 24시간 동안 4시간 간격으로 시간대별로 자극하였으며, 또 치수세포를 Calcitonin gene-related peptide (CGRP)로 12시간 자극하였다. 이들 세포를 SP길항제 (Spantide)로 15분간 차단한 후 SP로 12시간 재 자극하였으며, SP와 CGRP혼합액을 12시간 자극하였다. 상기의 실험 후 부유물로 ELISA를 시행하여 IL-8과 MCP-1의 분비 량을 측정하였다. 치수세포는 SP로 자극 시 IL-8이 현저히 증가한 반면, CGRP는 효과가 없었으며, SP와 CGRP를 혼합자극 시 시너지 효과 또한 없었고, Spantide는 치수세포의 IL-8과 MCP-1의 분비를 차단시켰다. 치수세포를 SP로 24시간 동안 4시간 간격으로 자극 시 8시간 후 최대의 IL-8은 분비량 나타내었으며, 8시간과 12시간 사이에서 최대의 MCP-1 분비량을 나타내었다. ECV 304세포를 SP로 자극 시 IL-8과 MCP-1 분비량이 미약하게 증가하였으며, Spantide는 ECV 304세포의 IL-8과 MCP-1 분비를 억제시켰다.

Aggregatibacter actinomycetemcomitans Strongly Stimulates Endothelial Cells to Produce Monocyte Chemoattractant Protein-1 and Interleukin-8

  • Aggregatibacter actinomycetemcomitans is the most important etiologic agent of aggressive periodontitis and can interact with endothelial cells. Monocyte chemoattractant protein-1 (MCP-1) and interleukin-8 (IL-8) are chemokines, playing important roles in periodontal pathogenesis. In our current study, the effects of A. actinomycetemcomitans on the production of MCP-1 and IL-8 by human umbilical vein endothelial cells (HUVEC) were investigated. A. actinomycetemcomitans strongly induced the gene expression and protein release of both MCP-1 and IL-8 in a dose- and time-dependent manner. Dead A. actinomycetemcomitans cells were as effective as live bacteria in this induction. Treatment of HUVEC with cytochalasin D, an inhibitor of endocytosis, did not affect the mRNA up-regulation of MCP-1 and IL-8 by A. actinomycetemcomitans. However, genistein, an inhibitor of protein tyrosine kinases, substantially inhibited the MCP-1 and IL-8 production by A. actinomycetemcomitans, whereas pharmacological inhibition of each of three members of mitogen-activated protein (MAP) kinase family had little effect. Furthermore, gel shift assays showed that A. actinomycetemcomitans induces a biphasic activation (early at 1-2 h and late at 8-16 h) of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and an early brief activation (0.5-2 h) of activator protein-1 (AP-1). Activation of canonical NF-${\kappa}B$ pathway ($I{\kappa}B$ kinase activation and $I{\kappa}B-{\alpha}$ degradation) was also demonstrated in these experiments. Although lipopolysaccharide from A. actinomycetemcomitans also induced NF-${\kappa}B$ activation, this activation profile over time differed from that of live A. actinomycetemcomitans. These results suggest that the expression of MCP-1 and IL-8 is potently increased by A. actinomycetemcomitans in endothelial cells, and that the viability of A. actinomycetemcomitans and bacterial internalization are not required for this effect, whereas the activation of protein tyrosine kinase(s), NF-${\kappa}B$, and AP-1 appears to play important roles. The secretion of high levels of MCP-1 and IL-8 resulting from interactions of A. actinomycetemcomitans with endothelial cells may thus contribute to the pathogenesis of aggressive periodontitis.

Streptozotocin으로 당뇨가 유도된 C57BL/6 생쥐 지방조직에서의 염증성 사이토카인 유전자의 이상발현 (Altered Gene Expression of Inflammatory Cytokines in Adipose Tissue of Streptozotocin-induced Diabetic C57BL/6 Mice)

  • 본 연구를 통하여 streptozotocin 주사에 의한 당뇨 유발이 일반식이와 고지방식이로 키운 C57BL/6 수컷생쥐의 지방조직에서의 염증성 사이토카인 유전자 발현에 미치는 영향을 조사하였다. 네 그룹의 당뇨생쥐(일반식이 또는 고지방식이로 키운 16주령 또는 26주령 생쥐)와 네 그룹의 비당뇨 대조군을 포함한 모두 73마리의 생쥐가 이 실험에 사용되었다. Real-time PCR을 이용하여 지방조직에서의 tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)와 monocyte chemoattractant protein-1 (MCP-1)의 유전자 발현량을 측정한 결과, TNF-${\alpha}$ mRNA는 당뇨 유발에 의해 증가하는 양상을 보였다. 특히, 16주령의 일반식이 생쥐의 경우 비당뇨 대조군에 비해 당뇨가 유발된 실험군에서 유의한 증가가 관찰되었다. MCP-1 mRNA 발현은 STZ처리에 따른 당뇨유발에 의해 감소하는 경향을 나타내었다. 특히, 16주령 고지방식이의 당뇨 실험군에서의 발현이 비당뇨 대조군에서의 발현량의 26%에 해당할 정도로 큰 감소를 나타내었다. 또한, MCP-1의 발현은 인슐린 농도와 유의한 상관관계가 있음이 확인되었다. 이들 실험결과는 당뇨 모델 생쥐에서 지방조직의 염증성 사이토카인이 이상발현되고 있음을 나타내며, 비만, 인슐린저항성, 및 당뇨에서의 저준위 염증상태와 지방조직에서의 염증성 사이토카인 발현 조절의 기작을 밝히는데 유용한 정보를 제공할 것으로 기대된다.

한국인 천식환자의 Monocyte chemoattractant protein 1(MCP-1) 유전자 다형성에 대한 분석 (Analysis of Monocyte Chemoattractant Protein 1(MCP-1) Polymorphism in Korean Patients with Asthma)

  • Background : Monocyte chemoattractant protein-1(MCP-1), one of the CC chemokines, appears to play a significant role in asthma pathogenesis. It was reported that polymorphism in the MCP-1(-2518 A/G promoter) was associated with asthma in Caucasians, but the association of this polymorphism and asthma patients in the Korean population has not yet been clarified. Objective : We investigated the possible association between 2 polymorphisms (-2518 A/G promoter and Cys35Cys) and asthma patients in a Korean population. Materials and Methods : DNA samples were obtained from 86 Korean asthma patients and 270 healthy controls. MCP-1 genomic variants (-2518 A/G promoter and Cys35Cys polymorphism) were detected by PCR-RFLP. Level of MCP-1 was measured by ELISA for each genotype (n=8) (AA, AG, GG) and allele types of -2518 A/G promoter polymorphism for control subjects. Results : The Cys35Cys polymorphism was associated with asthma patients in Korean population [genotype distribution ($X^{2}=16.011$, P<0.001)]. Comparison of the two groups revealed no detectable differences in genotype and allele frequencies of the -2518 A/G polymorphism. Haplotype frequencies analysis revealed significant difference $(X^{2}=51.70$, P<0.001). MCP-1 serum level of subjects with G genotype of -2518 A/G promoter polymorphism was statistically higher than that with AA genotype (P<0.05). Conclusion : Our data indicate that no association exists between the MCP-1 -2518 A/G polymorphism and asthma susceptibility in the Korean population. However, it is noteworthy that the high prevalence of the -2518 G allele in the Korean population suggests a potentially important ethnic variation in the regulation of MCP-1 production. This variation must be considered in gene-association studies in different ethnic populations.

Although the lipid mediator leukotriene B$_4$ (LTB$_4$) is known to be a principal chemoattractant for recruiting neutrophils to inflamed sites across the airway epithelium, the precise signaling mechanism involved remains largely unknown.   • It has been proposed that G protein interacts with receptor via multiple interaction sites. With regard to this, C-terminus of the G${\alpha}$ subunit is clearly not the only structural determinant on the G proteins that is critical for receptor coupling selectivity, but the extreme N-terminus of Ga subunit and other structural elements were proposed to be responsible for dictating the interaction with receptors.
