A gold nanoparticles modified carbon paste electrode (GN-CPE) has been used for the determination of atenolol (ATN) in drug formulations by cyclic voltammetry (CV), differential pulse voltammetry (DPV) and chronocoulometric methods. The results revealed that the modified electrode shows an electrocatalytic activity toward the anodic oxidation of atenolol by a marked enhancement in the current response in buffered solution at pH 10.0. The anodic peak potential shifts by -80.0 mV when compared with the potential using bare carbon paste electrde. A linear analytical curve was observed in the range of $1.96\;{\times}\;10^{-6}$ to $9.09\;{\times}\;10^{-4}\;mol\;L^{-1}$. The detection limit for this method is $7.3\;{\times}\;10^{-8}\;mol\;L^{-1}$. The method was then successfully applied to the determination of atenolol in tablets and human urine. The percent recoveries in urine ranged from 92.0 to 110.0%.
A hexavalent chromium (Cr (VI)) is one of the hazardous substances regulated by the RoHS. The determination of Cr (VI) in various polymers and printed circuit board (PCB) has been very important. In this study, the three different analytical methods were investigated for the determination of a hexavalent chromium in Acrylonitrile Butadiene Styrene copolymer (ABS) and PCB. The results by three analytical methods were obtained and compared. An analytical method by UV-Visible spectrometer has been generally used for the determination of Cr (VI) in a sample, but a hexavalent chromium should complex with diphenylcarbazide for the detection in the method. The complexation did make an adverse effect on the quantitative analysis of Cr (VI) in ABS. The analytical method using diphenylcarbazide was also not applicable to printed circuit board (PCB) because PCB contained lots of irons. The irons interfered with the analysis of hexavalent chromium because those also could complex with diphenylcarbazide. In this study, hexavalent chromiums in PCB have been separated by ion chromatography (IC), then directly and selectively detected by inductively coupled plasma atomic emission spectrometry (ICP-AES). The quantity of Cr (VI) in PCB was 0.1 mg/kg.
B형 간염 바이러스(Hepatitis B Virus, HBV)는 만성감염, 간암의 원인이 되는 등 가장 큰 공중보건문제 중의 하나이다. 그리하여 감염 초기에 바이러스의 DNA 농도를 측정하여 관찰 하는 것이 중요하다. 본 연구에서는 기존의 Real Time-PCR과 새로운 방법인 Micro-PCR를 이용하여 HBV를 검출하였다. 단국대학교 병원에서 HBV 감염 환자의 혈청 샘플 120개를 얻은 후 분석하였고 각각 장비에서의 검출한계와 재현성, 민감성, 특이성, 분석시간을 비교해 보았다. 그 결과 Micro-PCR과 Real-Time PCR은 높은 검출한계와 재현성, 민감성, 특이성을 가지고 있었다. 그러나 Micro-PCR은 Real-Time PCR보다 빠른 시간 안에 증폭이 가능하며 조작하는데 있어 간편하였고 적은 양의 시약이 소모됨을 알 수 있었다. 그러므로 Micro-PCR은 신뢰성 있고 빠른 임상적 진단이나 각종 검사가 요구되는 곳에서 이용 가치가 높은 장비라 할 수 있겠다.
Since the unprecedented hydrogen fluoride leak accident in 2012, there has been growing demand for customized technical information for rapid response and chemical accident management agencies including the Ministry of Environment, the National Emergency Management Agency, and the National Police Agency need more information on chemicals and accident management. In this regard, this study aims to provide reliable technical data and guidelines to initial response agencies, similar to accident management technical reports of the US and Canada. In this study, we conducted a questionnaire survey and interviews on initial response agencies like fire stations, police stations, and local governments to identify new information items for appropriate initial response and improvements of current guidelines. We also collected and reviewed the Canada's TIPS, US EPA's hydrogen fluoride documents, domestic and foreign literature on applicability tests of control chemicals, and interview data, and then produced items to be listed in the technical guidelines. In addition, to establish database of on-site technical information, we carried out applicability tests for accident control data including ① emergency shut down devide, safety guard, shut down valve, ground connection, dyke, transfer pipe, scrubber, and sensor; ② literature and field survey on distribution type and transportation/storage characteristics (container identification, valve, ground connection, etc.); ③ classification and identification of storage/transportation facilities and emergency management methodslike leak prevention, chemicals control, and cutoff or bypass of rain drainage; ④ domestic/foreign analysis methods and environmental standards including portable detection methods, test standards, and exposure limits; and ⑤ comparison/evaluation of neutralization efficiency of control chemicals on toxic substances.
Cowpea mild mottle virus (CPMMV) is a global plant virus that poses a threat to the production and quality of legume crops. Early and accurate diagnosis is essential for effective managing CPMMV outbreaks. With the advancement in isothermal recombinase polymerase amplification and lateral flow strips technologies, more rapid and sensitive methods have become available for detecting this pathogen. In this study, we have developed a reverse transcription recombinase polymerase amplification combined with lateral flow strips (RT-RPA-LFS) method for the detection of CPMMV, specifically targeting the CPMMV coat protein (CP) gene. The RT-RPA-LFS assay only requires 20 min at 40℃ and demonstrates high specificity. Its detection limit was 10 copies/µl, which is approximately up to 100 times more sensitive than RT-PCR on agarose gel electrophoresis. The developed RT-RPA-LFS method offers a rapid, convenient, and sensitive approach for field detection of CPMMV, which contribute to controlling the spread of the virus.
Microplastics with a size of less than 5 mm have emerged as an important environmental and food safety issue, as they have been detected not only in marine but also in terrestrial ecosystem and drinking water. Although many studies have been conducted on the exposure of microplastics and the effects on human health, the lack of standardized experimental methods for microplastics has been reviewed as a major problem. In order to overcome this, European countries such as the Netherlands and Germany are conducting a project to develop detection methods for microplastics as well as to establish the risk assessment methodologies for microplastics. Being the microplastics suggested to have a substantially potential risk on human health, reliable risk assessments should be conducted considering the various sources of microplastics, chemical pollutants and biological factors. In addition, international standards and regulations should be applied.
The transition from aromatics to heteroaromatics is very attractive since it provides an extremely large structural variety, the chemical functionality as well as the possibilities for electronic tuning of the fullerene properties. A synthesis of heterofullerenes in macroscopic quantities is unknown however the spectrometric detection of $C_{59}B$ has been reported. The electronic structures of $C_{(60-2x)}(BN)_x$ systems, isoelectronic with $C_{60}$ have been explored by Extended Hukel, AM1 and ab initio methods. The polyhedral assembly energy are 7.7 kcal greater than $C_{60}$ when one B-N unit is substituted with C-C unit. The assembly energies are getting bigger if more B-N unit is introduced. We focus on HOMO-LUMO energy gap and the stability effects in $C_{(60-2x)}(BN)_x$ with different compositions of $(BN)_x$ moiety. The bonding properties of the substituent atoms were investigated in detail.
Surface-Enhanced Raman Scattering (SERS) spectroscopy is a multifaceted surface sensitive methodology which exploits spectroscopy-based analysis for various applications. This technique is based on the massive amplification of Raman signals which were feeble previously in order to use them for appropriate identification at qualitative and quantitative in chemical as well as biological systems. This novel powerful technique can be utilized to identify pathogens such as bacteria and viruses. As far as SERS is concerned, one of the most studied problems has been functionalization of SERS active substrate. Metal colloids and nanostructures or microstructures synthesized using noble metals such as Au, Ag and Cu are considered to be SERS active. Silver and gold are extensively used as SERS active substrates due to chemical inertness and stability in air compare to copper. However, use of Cu as a suitable alternative has been taken into account as it is cheap. Herein, we have synthesized air-stable copper microstructures/nanostructures by chemical, electrochemical and microwave-assisted methods. In this paper, we have also discussed the use of as synthesized copper micro/nanostructures as inexpensive yet effective SERS active substrates for the fast identification of micro-organisms like Staphylococcus aureus and Escherichia coli.
The existing technological schemes for processing rhenium-containing raw materials involve the recovery of Re from solutions, which can be effectively achieved by anion exchange method. The application of anion exchange also allows to study rhenium state in aqueous solutions and to develop analytical control methods. The present work is focused on investigation of ion exchange equilibrium in the analytical system Re(VII)-HCl-$SnCl_2$-KSCN-anion exchanger by means of sorption-desorption method as well as by electron, IR- and diffuse reflection spectroscopy. It was shown that rhenium can be quantitatively recovered from this system. It is proposed to use the sorption-spectroscopic method for Re(VII) determination in aqueous solutions. The calibration curve is linear in the concentration range of 0.5-20.0 mg/L (sample volume is 25.0 mL) and the detection limit is 0.05 mg/L. The presence of Mo(VI), Cu(II), Fe(II, III), Ni(II), Zn(II) as well as $K^+,\;Na^+$ do not hinder the solid-phase determination of rhenium. Rhenium (VII) determination by diffuse reflection spectroscopy was carried out in model solutions as well as in samples of river-derived water and in solutions obtained after the dissolution of spent catalysts.
Objective: Detection of adulteration in processed meats is an important issue for some countries due to substitution of beef with a cheaper source of protein like poultry. In this study, the presence of chicken meat was investigated using real-time polymerase chain reaction (real-time PCR) and enzyme-linked immunosorbent assay (ELISA) techniques to verify adulteration of fermented sausage samples. Methods: A total of 60 commercial samples were collected from 20 establishments in three replicates including 10 fermented sausage manufacturers and 10 butchers to investigate the presence of chicken meat with the sequential use of real-time PCR and ELISA techniques. In addition, pH, moisture content, water activity and color values of the samples were determined. Results: Both real-time PCR and ELISA showed agreement on the presence or absence of chicken meat in 55 out of 60 fermented sausage samples and chicken meat was identified with both methods in 16 samples. Five samples produced inconsistent results for the presence of chicken meat in the first run. Nevertheless, the presence of chicken meat was verified with both methods when these samples were analyzed for the second time. In addition, the average physico-chemical values of the fermented sausage samples tested positive for chicken meat were not significantly different from some of those fermented sausage samples tested negative for the chicken meat. Conclusion: The sequential use of real-time PCR and ELISA techniques in fermented sausages could be beneficial for the government testing programs to eliminate false negatives for detection of adulteration with chicken meat. Furthermore, consumers should not rely on some of the quality cues including color to predict the adulteration of fermented sausages with chicken meat since there were no statistical differences among some of the samples tested positive and negative for chicken meat.
