Possibility of Wood Classification in Korean Softwood Species Using Near-infrared Spectroscopy Based on Their Chemical Compositions

  • This study was to establish the interrelation between chemical compositions and near infrared (NIR) spectra for the classification on distinguishability of domestic gymnosperms. Traditional wet chemistry methods and infrared spectral analyses were performed. In chemical compositions of five softwood species including larch (Larix kaempferi), red pine (Pinus densiflora), Korean pine (Pinus koraiensis), cypress (Chamaecyparis obtusa), and cedar (Cryptomeria japonica), their extractives and lignin contents provided the major information for distinction between the wood species. However, depending on the production region and purchasing time of woods, chemical compositions were different even though in same species. Especially, red pine harvested from Naju showed the highest extractive content about 16.3%, whereas that from Donghae showed about 5.0%. These results were expected due to different environmental conditions such as sunshine amount, nutrients and moisture contents, and these phenomena were also observed in other species. As a result of the principal component analysis (PCA) using NIR between five species (total 19 samples), the samples were divided into three groups in the score plot based on principal component (PC) 1 and principal component (PC) 2; group 1) red pine and Korean pine, group 2) larch, and group 3) cypress and cedar. Based on the chemical composition results, it was concluded that extractive content was highly relevant to wood classification by NIR analysis.

Engineering of Recombinant Escherichia coli Towards Methanol Sensing Using Methylobacterium extroquens Two-component Systems

  • Five genes (mxbDM, mxcQE and mxaB) are responsible for the transcription of methanol oxidation genes in Methylobacterium strains. Among these, MxbDM and MxcQE constitute the two-component system (TCS) regulating methanol metabolism. In this study, we integrated the methanol-sensing domain of MxbD and MxcQ with the EnvZ/OmpR from Escherichia coli. The domain-swapping strategy resulted in chimeric histidine kinases (HK's) MxbDZ and MxcQZ AM1 containing recombinant E. coli. Real-time quantitative PCR was used to monitor OmpC expression mediated by the chimeric HK and response regulator (RR) OmpR. Further, an ompC promoter based fluorescent biosensor for sensing methanol was developed. GFP fluorescence was studied both qualitatively and quantitatively in response to environmental methanol. GFP measurement also confirmed ompC expression. Maximum fluorescence was observed at 0.05% methanol and 0.01% methanol using MxbDZ and MxcQZ AM1, respectively. Thus the chimeric HK containing E. coli were found to be highly sensitive to methanol, resulting in a rapid response making them an ideal sensor.

상백피의 Sanggenon C에 의한 Streptococcus mutans의 생육 및 균부착 저해효과 (The inhibitory Effect of Sanggenon C from the Root-bark of Morus alba L. on the Growth and the Cellular Adherence of Streptococcus mutans)

  • The methanolic extract of the root-bark of Morus alba L.(Mulberry tree) has the potent antibacterial activity against Streptococcus mutans. Its active component was identified to be sanggenon C. The active component had stronger anti-bacterial activity than berberine, having minimum inhibitory concentration(MIC) of $25\;{\mu}g/ml$. Moreover, the inhibitory effect of this component on the cellular adherence of Streptococcus mutans to glass surfaces also was more remarkable than that of berberine in the presence of glucosyltransferase(GTase) and sucrose in vitro. These results indicate that sanggenon C may play an important role in inhibiting plaque formation and caries incidence.

Preparation and Properties of Modified PHEMA Hydrogels Containing Thermo-responsive Pluronic Component

  • To modify and strengthen the properties of PHEMA hydrogel, composite hydrogels containing varying amounts of a Pluronic (PEO-PPO-PEO) component were synthesized by bulk polymerization of HEMA in the presence of Pluronic dimethacrylate under mild photo initiating conditions. The effects of the Pluronic component on gel properties were investigated by measuring the degree of swelling with its temperature responsive behavior, the mechanical properties, and the morphology of the composite hydrogels. With increased Pluronic content, the modified PHEMA hydrogels exhibited an increase in the degree of swelling, and the swelling showed an enhanced thermo-responsive behavior that was completely reversible. In addition, improved mechanical strength and the development of a microporous gel morphology were observed in hydrogels containing Pluronic.

  • A series of pyranopyrazoles, was efficiently synthesized via one-pot, four component reaction of ethyl acetoacetate, hydrazine hydrate, aldehydes and malononitrile in the presence of catalytic amount silicotungstic acid under solvent free condition. NOE experiments confirmed that the product exist exclusively in the 2H form. The present protocol offers the advantages of clean reaction, short reaction time, high yield, easy purification and economic availability of the catalyst.

지역별 에어로졸 화학성분 농도 및 광학특성 비교 (Comparison of the concentration characteristics and optical properties of aerosol chemical components in different regions)

  • The aerosol chemical components in $PM_{2.5}$ in several regions (Seoul, Busan, Daejeon, and Jeju Island) were investigated with regard to their concentration characteristics and optical properties. The optical properties of the various aerosol components (e.g., water-soluble, insoluble, Black Carbon (BC), and sea-salt) were estimated using hourly and daily aerosol sampling data from the study area via a modeling approach. Overall, the water-soluble component was predominant over all other components in terms of concentration and impact on optical properties (except for the absorption coefficient of BC). The annual mean concentration and Aerosol Optical Ddepth (AOD) of the water-soluble component were highest in Seoul (at the Gwangjin site) ($26{\mu}g/m^3$ and 0.29 in 2013, respectively). Further, despite relatively moderate BC concentrations, the annual mean absorption coefficient of BC ($21.7Mm^{-1}$) was highest in Busan (at the Yeonsan site) in 2013, due to the strong light absorbing ability of BC. In addition, high AODs for the water-soluble component were observed most frequently in spring and/or winter at most of the study sites, while low values were noted in summer and/or early fall. The diurnal variation in the AOD of each component in Seoul (at the Gwangjin site) was slightly high in the morning and low in the afternoon during the study period; however, such distinctions were not apparent in Jeju Island (at the Aweol site), except for a slightly high AOD of the water-soluble component in the morning (08:00 LST). The monthly and diurnal differences in the AOD values for each component could be attributed to the differences in their mass concentrations and Relative Humidities (RH). In a sensitivity test, the AODs estimated under RH conditions of 80 and 90% were factors of 1.2 and 1.7 higher, respectively, than the values estimated using the observed RH.

  • 화학공정의 안전하고 효율적인 운전에 관심이 커지면서 공정이상의 원인을 조기에 진단하기 위한 다양한 이상진단방법이 연구되어 왔다. 최근에는 통계적 모델 등 정량적 데이터에 기반한 이상진단방법이 많이 연구되고 있으나, 특정 조업영역에서 얻어진 통계적 모델을 다른 조업영역에 적용하면 오진단이 많아지게 된다. 따라서 공정특성상 다양한 조업영역이 존재하는 화학공정에 데이터기반 방법론을 적용하기에는 어려움이 있어 화학공정의 조업영역 판별법이 요구되고 있다. 이 연구에서는 유클리드 거리(Euclidean distance), FDA(Fisher's discriminant analysis), PCA(principal component analysis)의 통계모델과 이 모델들에 공정변수의 동특성을 반영한 모델을 제안하였다. 6개의 조업모드를 가진 TE(tennessee eastman) 공정에 대한 사례연구를 통해 동특성을 반영한 PCA 모델의 성능이 가장 우수함을 확인하였다.

알루미나 졸과 아크릴레이트 단량체를 이용한 UV경화형 유-무기 하이브리드 하드코팅 막 제조 (Preparation of UV-Curable Organic-Inorganic Hybrid Hard Coating Films Using Alumina Sols and Acrylate Monomers)

  • 본 연구에서는 알루미나 졸과 아크릴레이트 단량체를 이용하여 UV 경화형 유-무기 하이브리드 하드코팅 용액을 제조하였다. Aluminum isopropoxide로부터 얻어진 알루미나 졸에 실란커플링제인 methacryloxypropyl trimethoxysilane(MPTMS)을 첨가하여 제조된 졸을 무기물 성분으로 사용하였다. 유기물 성분으로는 다양한 아크릴레이트 단량체인 pentaerythritol triacrylate(PETA), 1,6-hexanediol diacrylate(HDDA), dipentaerythritol hexaacrylate(DPEHA)를 혼합하여 사용하였다. 이후 무기물 성분과 유기물 성분을 혼합하여 유-무기 하이브리드 코팅 용액을 제조한 후, 이를 polycarbonate 기판 위에 스핀 코팅 후 UV 경화를 실시하여 도막을 형성하였다. 이때 무기물 성분 중의 MPTMS의 첨가량과 UV 경화 시간 변화가 코팅 막의 물성에 미치는 영향을 연구하였다. 그 결과 MPTMS가 0.20 mole 첨가 될 경우 코팅 막은 3H의 우수한 연필경도를 나타내었으며 haze값이 2%로 내마모성도 우수하였다.

Moving Window Principal Component Analysis for Detecting Positional Fluctuation of Spectral Changes

  • In this study, we proposed a new promising idea of utilizing moving window principal component analysis (MWPCA) as a sensitive diagnostic tool to detect the presence of peak position shift. In this approach, the moving window is constructed from a small data segment along the wavenumber axis. For each window bound by a narrow wavenumber region, separate PCA analysis was applied. Simulated spectra with complex spectral feature variations were analyzed to explore the possibility of MWPCA technique. This MWPCA-based detection of the peak shift, potentially coupled with 2D correlation analysis to provide additional verification, may offer an attractive solution.

Principal Component Analysis Based Two-Dimensional (PCA-2D) Correlation Spectroscopy: PCA Denoising for 2D Correlation Spectroscopy

  • Principal component analysis based two-dimensional (PCA-2D) correlation analysis is applied to FTIR spectra of polystyrene/methyl ethyl ketone/toluene solution mixture during the solvent evaporation. Substantial amount of artificial noise were added to the experimental data to demonstrate the practical noise-suppressing benefit of PCA-2D technique. 2D correlation analysis of the reconstructed data matrix from PCA loading vectors and scores successfully extracted only the most important features of synchronicity and asynchronicity without interference from noise or insignificant minor components. 2D correlation spectra constructed with only one principal component yield strictly synchronous response with no discernible a asynchronous features, while those involving at least two or more principal components generated meaningful asynchronous 2D correlation spectra. Deliberate manipulation of the rank of the reconstructed data matrix, by choosing the appropriate number and type of PCs, yields potentially more refined 2D correlation spectra.
