  • In order to evaluate the change in plasma and fecal mineral content of calves to 6 days of age, and to clarify the relationship between electrolyte status and fecal consistency in calves at 6 days of age, data were collected from 52 Holstein calves. Fecal DM and Na of calves decreased at 6 days of age, but fecal Ca, P, Mg, K, Fe and Zn increased. Plasma Ca, inorganic P, Mg, Na, and Fe as well as blood hematocrit and hemoglobin of calves decreased at 6 days of age, but plasma alkaline phosphatase increased. Colostral Ca, P, Mg, Fe and Zn decreased with time postpartum, but colostral Na and K were not affected. The high plasma Na and K of calves at 6 days of age were influenced by the values at birth. There were negative correlations between fecal DM and fecal Na, and fecal K, of calves at 6 days of age, negative correlations between fecal DM and plasma Na, with plasma K. Plasma and fecal Na as well as plasma and fecal K of calves at 6 days of age were increased by the occurrence of diarrhea. These results suggest that the electrolyte status of calves at 6 days of age is adversely affected by the occurrence of diarrhea, and high plasma K and Na of calves at 6 days of age may be partly due to the electolyte status of calves at birth.

  • Eight healthy Holstein bulls, 4-6 years old were used to study the effect of season of the year on the biophysical characteristics of semen. Semen was collected twice a week by AV (artificial vagina) over one-year period. The analyses revealed that all the basic seminal traits studied were differed significantly due to season, except the ejaculate volume and consistency and the percentage of swollen spermatozoa in a hypo-osmotic fructose-citrate solution. Ejaculates collected during hot summer season had significantly lower sperm motility, concentration and total counts, and higher percentage of dead spermatozoa than those collected during winter time. Warm spring had moderate semen quality. The temperature-humidity index was calculated and it was associated (p < 0.01) negatively with the ejaculate pH, sperm concentration and total counts, and positively with the % of dead sperms. Ejaculate volume, percentage of swollen spermatozoa, individual motilities did not correlate significantly with the change in temperature-humidity index values. The total live, motile spermatozoa per ejaculate during both the winter and spring seasons showed significant increase of about 37% and 32% respectively over the summer season. Also, rectal temperatures of the bulls were elevated during the hot summer season, while the values of blood hemoglobin and packed-cell volume were decreased.

  • Gum is known as natural polymer. Biodegradable films were prepared by solution blend method in the weight ratio of natural gums(Xanthan, Locust bean, Guar) for the purpose of useful bioimplants. The possibility of bioimplants, which prepared from natural gums as a skin substitute was evaluated by measuring biodegradability. This biodegradable films were inserted in the back of rats and their of biodegradability were investigated by hematological change evaluation as a function of time to the biotransformation. Rats implantation test results showed that Guar induced increments of monocyte and basophil after 48 hours of implantation. And Locust bean showed increase of monocyte and neutrophile counts after 48 hours of implantation. And Xanthan induced decrease of monocyte and neutrophile at 24 hours after implantation. Guar and Locust showed high hemoglobin contents and hematocrit after 48 hours of implantation. Guar and Locust induced some suspects of incompatibility in the tissue after 48 hours, but three were little effects to the skin inflammation at 24 hours. These values of biodegradable films, which prepared from prepared from natural gums measured in this were some satisfiable results at short period with those of ideal skin bioimplants.

  • A study was conducted to observe the effects of dexamethasone sodium 21 phosphate injection in different amounts, intervals and times on the fluctuation of the parasitized erythrocytes in the peripheral blood of the T. sergenti carrier calves. A total of 31 calves including 15 Holstein and 16 Korean native male calves aged 6-12 months (125-300kg) was used. Results observed were as follows. 1. The calves injected 0.2mg/kg body weight two and three times in 48 hour intervals and 5 times in 24 hour intervals observed that 2 out of 2 heads, 3 out of 6 heads, and 4 out of 5 heads, respectively were substantially increased parasitized erythrocytes in the blood for 3-11 days after the first injection and continued to 2-10 days. 2. However, the calves injected once with 0.1mg/kg body weight and all control calves showed no visible change of the parasitized erythrocytes in the blood during the experimental period. 3. The animals injected over 2 times with 0.2mg/kg body weight tended to be decreased the number of erythrocytes, hemoglobin concentration and PCV for 2-3 days after first injection and continued about one week. 4. All animals injected the dexamethasone were observed the neutrophilic leukocytosis, eosinopenea and lymphopenea until 48 hours after injection.

  • Rosiglitazone is a thiazolidinedione-class antidiabetic drug that reduces blood glucose and glycated hemoglobin levels. We here investigated the interaction of rosiglitazone with Kv3.1 expressed in Chinese hamster ovary cells using the wholecell patch-clamp technique. Rosiglitazone rapidly and reversibly inhibited Kv3.1 currents in a concentration-dependent manner (IC50 = 29.8 µM) and accelerated the decay of Kv3.1 currents without modifying the activation kinetics. The rosiglitazonemediated inhibition of Kv3.1 channels increased steeply in a sigmoidal pattern over the voltage range of -20 to +30 mV, whereas it was voltage-independent in the voltage range above +30 mV, where the channels were fully activated. The deactivation of Kv3.1 current, measured along with tail currents, was also slowed by the drug. In addition, the steady-state inactivation curve of Kv3.1 by rosiglitazone shifts to a negative potential without significant change in the slope value. All the results with the use dependence of the rosiglitazone-mediated blockade suggest that rosiglitazone acts on Kv3.1 channels as an open channel blocker.

  • The retention behavior of proteins was investigated by using reversed-phase chromatography (RPC), comparing to the retention behavior of small molecules in RPC. The evaluation was carried out on a SynChropak RP-P($C_{18}$) column with 0.1% aq. TFA-organic solvent modifier such as acetonitrile, isopropanol, and ethanol. The Z value (the number of solvent molecules required to displace the solute from the surface) was a general index for the characterization of protein retention as a function of organic concentration over a range of temperature between 5 and 70$^{\circ}C$. Van't Hoff plots provided the basis for evaluating the enthalpic and entropic changes associated with the interaction between protein and the stationary phase. Z values did not change significantly at the range of temperature showing the consistent ${\Delta}H^{\circ}$ and ${\Delta}S^{\circ}$ values. From these investigation, it was concluded that the retention behavior of proteins in RPC was able to be predicted by the retention parameters applied to small molecules. Furthermore, myoglobin and hemoglobin in RPC as stated above showed a similar retention behavior regardless of their molecular weights.

  • The purpose of this study was to investigate the anthropometry, biochemical parameters and electrolytes concentrations of the Total Parenteral Nutrition (TPN) patients according to their nutritional status at the time of admission. Thirty-three patients in the Intensive Care Unit at S University Hospital were the subjects of this study. Their nutritional status was classified as At-risk I (Mild PCM, n = 13), At - risk II (Moderate PCM, n = 9) and At-risk III (Marasmus + kwashiorkor + severe PCM, n = 11) . Anthropometeric, biochemical and dietary assessments were performed. The Patients intake of calories (75.02%) and protein (53.15%) was insufficient compare with Korean RDA requirements. The body weight and the Body Mass Index (BMI) in the At-risk III group were significantly lower than in the other groups. The percentage of body weight loss and change of body weight (kg) were significantly higher than in the other groups. The subjects were malnourished as indicated by nutritional related parameters such as serum total protein, albumin, total lymphocyte count (TLC), hemoglobin and hematocrit. Serum total protein, albumin and TLC levels were lower at the time of admission before TPN administration. But after TPN administration, they increased. The electrolyte concentrations did not show any differences following TPN administration. The nutritional status of the patients could be affect by the duration of TPN administration and the number of days of the patients hospitalization. The patients who require nutritional support need the continuous follow-up care and monitoring by a nutritional support team. (Korean J Community Nutrition 8(4) : 574-583, 2003)

The possibility of bioimplants, which prepared from natural polymers as a skin substitute and food wrapping materials were evaluated by measuring the biodegradability. these biodegradable films were inserted in the back of rats and their biodegradability was investigated by hematological change as a function of time. Rats study showed that low-Chitosan induced increments of monocyte and basophil after 48 hours of implantation. And medium-Chitosan showed increase of lymphocyte and decreased neutrophil counts after 48 hours of implantation. Low, medium Chitosan showed high hemoglobin contents, medium and high Chitosan showed high hematocrit value after 48 hours of implantation. As a result, medium, high-Chitosan induced potential incompatibility in the tissue after 48 hours, but there was little effects to the skin inflammation. The values of biodegradable films, which prepared from natural polymers measured in this study were some satisfiable results at short period with those of ideal skin bioimplants and artificial skin.

  • The objective of this study was (a) to examine the change of bloodchemistry and (b) to investigate the secretion trend of endocrinological substance in a dog model after laser-beam radiation at different level of frequency (250 Hz, 2,000 Hz, 5,000 Hz and 8,000 Hz). The experimental groups were divided into four groups were divided into four groups on the basis of the level of frequency radiated and stimulated for five minutes. Before stimulation and after a lapse of time (10-minute, 30-minute and 60-minute) all dogs were checked the following parameters; cortisol, ACTH, RBC, hemoglobin, hematocrit, WBC, Ca, P, ALT, AST and creatinine. The results were as follow: ten minutes after stimulation, cortisol level of 250 Hz group, 2,000 Hz group, 5,000 Hz group and 8,000 Hz group was increased 18.8%, 20.5%, 23.2% and 309%, respectively. Ten minutes after stimulation, ACTH level of 250 Hz group, 2,000 Hz group, 5,000 Hz group and 8,000 Hz group was increased 26.7%, 26.6%, 30.5% and 29.5%, respectively. It began to decrease and at 30-minute after stimulation resturned to pre-stimulation level at 60-minute after stimulation. In blood examination RBC, hemaoglobin, hematocrit and WBC level showed slight increase and decrease and returned to pre-stimulation level at 60-minute after radiation. In bloodchemical examination serum calcium, inorganic phosphorus, ALT, AST and creatinine level were within normal physiological ranges.
