Precise counting of cell number stands in important position within clinical and research laboratories. Conventional methods such as hemocytometer, migration/invasion assay, or automated cell counters have limited in analytical time, cost, and accuracy., which needs an alternative way with time-efficient in-situ approach to broaden the application avenue. Here, we present simple coding-based cell counting method using image analysis tool, freely available image software (ImageJ). Firstly, we encapsulated RFP-expressing bacteria in a droplet using microfluidic device and automatically performed fluorescence image-based analysis for the quantification of cell numbers. Also, time-lapse images were captured for tracking the change of cell numbers in a droplet containing different concentrations of antibiotics. This study confirms that our approach is approximately 15 times faster and provides more accurate number of cells in a droplet than the external analysis program method. We envision that it can be used to the development of high-throughput image-based cell counting analysis.
This study was carried out to quantify degree of contribution of harvested wood product (HWP) on mitigation of climate change by valuation of public benefits, environmentally and economically. The potential carbon dioxide emission reduction of HWP was estimated by accounting carbon storage effect and substitution effect. Based on 2014 statistics of Korea Forest Service, domestic HWPs were sorted by two categories, such as wood products produced domestically from domestic and imported roundwood. The wood products were divided into seven items; sawnwood, plywood, particle board, fiberboard (MDF), paper (including pulp), biomass (wood pellet) and other products. The carbon stock of wood products and substitution effects during manufacturing process was evaluated by items. Based on the relevant carbon emission factor and life cycle analysis, the amount of carbon dioxide emission per unit volume on HWP was quantified. The amounts of carbon stock of HWP produced from domestic and from imported roundwood were 3.8 million $tCO_{2eq}$., and 2.6 million $tCO_{2eq}$., respectively. Also, each reduction of carbon emission by substitution effect of HWP produced from domestic and imported roundwood was 3.1 million $tCO_{2eq}$. and 2.1 million $tCO_{2eq}$., respectively. The results of this study, the amount of carbon emission reduction of HWP, can be effectively used as a basic data for promotion of wood utilization to revise and establish new wood utilization promotion policy such as 'forest carbon offset scheme', and 'carbon storage labeling system of HWP'.

Accident rate in the construction industry of South Korea is increasing every year, and it represents the highest percentage among industries. This shows that activities performed to prevent safety accidents in the country are not efficient when it comes to reduce the accident rate. In order to resolve this issue, a model for the prediction of human accidents should be established. In addition, it is required a quantification study based on pattern of human accidents. Therefore, the objective of this study is to quantify uncertainty of human accidents risk and predict how to change in various circumstances by using Monte Carlo Simulation. To achieve the objective, first, pattern of human accidents was defined. Second, insurance claim payout and information of human accidents during 14 years in construction site were collected. Third, descriptive analysis is conducted to determine the characteristics of the accident pattern. Fourth, to quantitatively analyze the pattern of the human accidents, the population of each accident occurrence and payout were estimated. Finally, estimated populations was analyzed according to characteristics of distribution by using Monte carlo simulation. In the future, this study can be used as a reference for developing the safety management checklist in construction site and development of prediction models of human accident.

Many researchers have studied the process of retail evolution in the United States and in Europe. Although extensively studied(e.g., Agergaard, Olsen & Allpass, 1970; Oren, 1989), used in conceptual work(e.g., Cist, 1968), and applied as foundation in empirical work(e.g., HcNair, 1958), some limitations exist as follows: inability to cover multiple types of retail institutions and limited quantification. The purpose of this study is to build a conceptual framework combining existing retail evolution theories to overcome existing limitations. Data collection and analysis followed a qualitative research design, specifically a grounded theory type of design with a constant comparative analysis method. As a result of the study, a conceptual framework was built by synthesizing aspects of retail evolution theories and showed retail institution types in a change process.

Spinosad and spinetoram are widely used insecticides for the control of lepidopteran larvae, leaf miners, and thrips; however, they might also have low toxicity toward beneficial insects like bees. Because these pesticides are easily photolyzed by ultraviolet radiation, the QuEChERS method, with its simple pretreatment procedure, is often used for analyzing residues of spinosad and spinetoram. The present study performed a residue analysis using a modified QuEChERS method by pretreating with ammonium salt. The limit of detection (LOD) of the modified method was 0.05 mg/kg and the limit of quantification (LOQ) was 0.25 mg/kg. The coefficient of determination ($R^2$) for the calibration curve was 0.999. Also, we examined any change in the adhesion of spinosad and spinetoram on the plants depending on a spray volume. The adhesion was approximately 70% when the spray volume was increased from 60 L to 120 L per 10 a whereas the adhesion was approximately 37% when the spray volume was increased from 125 L to 250 L. This showed that the amount of adhesion decreased with the higher spray volume. The efficacy result of spinetoram was that over 90% of Frankliniella occidentalis was controlled with the application volume of 125 L per 10 a. Therefore, the result of this study indicates that control of insects is effective and sufficient with a spray volume of 125 L per 10 a in paprika cultivation facilities.
Pathogen infection in plants induces complex responses ranging from gene expression to metabolic processes in infected plants. In spite of many studies on biotic stress-related changes in host plants, little is known about the metabolic and phenotypic responses of the host plants to Pseudomonas cichorii infection based on image-based analysis. To investigate alterations in tomato plants according to disease severity, we inoculated plants with different cell densities of P. cichorii using dipping and syringe infiltration methods. High-dose inocula (${\geq}10^6cfu/ml$) induced evident necrotic lesions within one day that corresponded to bacterial growth in the infected tissues. Among the chlorophyll fluorescence parameters analyzed, changes in quantum yield of PSII (${\Phi}PSII$) and non-photochemical quenching (NPQ) preceded the appearance of visible symptoms, but maximum quantum efficiency of PSII ($F_v/F_m$) was altered well after symptom development. Visible/near infrared and chlorophyll fluorescence hyperspectral images detected changes before symptom appearance at low-density inoculation. The results of this study indicate that the P. cichorii infection severity can be detected by chlorophyll fluorescence assay and hyperspectral images prior to the onset of visible symptoms, indicating the feasibility of early detection of diseases. However, to detect disease development by hyperspectral imaging, more detailed protocols and analyses are necessary. Taken together, change in chlorophyll fluorescence is a good parameter for early detection of P. cichorii infection in tomato plants. In addition, image-based visualization of infection severity before visual damage appearance will contribute to effective management of plant diseases.
Automated greenhouse production system often require crop growth monitoring involving accurate quantification of plant physiological properties. Conventional methods are usually burdensome, inaccurate, and harmful to crops. A thermal image analysis system can accomplish rapid and accurate measurements of physiological-property changes of stressed crops. In this research a thermal imaging system was used to measure the leaf-temperature changes of several crops according to nutrient stresses. Thermal images were obtained from lettuce, cucumber, and pepper plants. Plants were placed in growth chamber to provide relatively constant growth environment. Results showed that there were significant differences in the temperature of stressed plants and non-stressed plants. In a case of the both N deficiency and excess, the leaf temperatures of cucumber were $2^{\circ}C$ lower than controlled temperature. The leaf temperature of cucumber was $2^{\circ}C$ lower than controlled temperature only when it was under N excess stress. For the potassium deficiency or excess stress, the leaf temperaures of cucumber and hot pepper were $2^{\circ}C$ lower than controls, respectively. The phosphorous deficiency stress dropped the leaf temperatures of cucumber and hot pepper $2^{\circ}C$ and $1.5^{\circ}C$ below than controls. However, the leaf temperature of lettuce did not change. It was possible to detect the changes in leaf temperature by infrared thermography when subjected to nutrition stress. Since the changes in leaf temperatures were different each other for plants and kinds of stresses, however, it is necessary to add a nutrient measurement system to a plant-growth monitoring system using thermography.
This study was carried out to predict the optimal growth site and estimate carbon stocks of Quercus acuta, evergreen broad-leaved trees in warm temperate zone according to climate change. The criterion for the optimal site prediction was created by quantification method with quantitative and qualitative data, collected from growth factors of stands and environmental factors of survey sites of 42 plots in Q. acuta by study relationship between growth of tree and site environmental factors. A program for the optimal site prediction was developed by using GIS engine tools. To prediction of the suitable growth site of Quercus acuta, developed program in this study applied to Wando in Jeollanam-do, distributing a various evergreen bread-leaved trees of warm temperate zone. In the results from analysis of the optimal site prediction on Q. acuta, the characteristics of the optimal site showed as follows; site environmental features of class I (the best site class for Q. acuta) was defined as 401 ~ 500 m of altitude, $21{\sim}25^{\circ}$ of slope with above hillside, residual of deposit convex of slope type with west of aspect. The area and carbon stocks of optimal site prediction by class for Q. acuta in classI showed 147.1 ha (2.5%), total 316.5 tC/ha, total $1,161tCO_2/ha/yr$ of class I, 2,703.5 ha (46.3%), total 5,817.4 tC/ha, total $21,331tCO_2/ha/yr$ of class II, 2,845.5 ha (48.6%), total 6,123.0 tC/ha, total $2,845.5tCO_2/ha/yr$ of class III and 153.7 ha (2.6%), total 330.7 tC/ha, total $1,213.7tCO_2/ha/yr$ of class IV.

Line density index(LDI) was developed to quantify a densely isothermal line rate as standard index in the ocean environment. Theoretical background on the LDI development process restricting index range 0 to 100 was described. And validation test was done for the LDI application condition that total line length is not greater than 1/10 of unit area. NOAA SST(Sea Surface Temperature) data were used for the experimental application of LDI in the South Sea of Korea. Using GIS, $0.1^{\circ}C$ isothermal lines were linearized as vector data form SST raster data, and unit area were built as polygon data. For the LDI calculation, spatial overlapping(line in polygon) was implemented. To analyze the effect of unit area size for the LDI distribution, two cases of unit area size were designed and descriptive statistics was calculated including performing normality test. The results showed no change of LDI's essential characteristics such as mean and normality except for the range of value, variance and standard deviation. Accordingly, it was found that complex structure of thermal front and even smaller scale of front width than unit area size could influence on the LDI distribution. Also, correlation analysis performed between LDI and difference of temperature(${\Delta}T^{\circ}C$), and horizontal thermal gradient(${\Delta}T^{\circ}C/km$) on the front was obtained from linear regression model. This obtained value was compared with the results from previous researches. Newly developed LDI can be used to compare the thermal front regions changing spatio-temporally in the ocean environment using absolute index value. It is considered to be significant to analyze the relationship between thermal front and marine environment or front and marine organisms in a quantitative approach described in this study.

Recently, climate change has gradually accelerated the occurrence of landslides. Among the various effects caused by landslides,debris flow is recognized as particularly threatening because of its high speed and propagating distance. In this study, the impacts of various factors were analyzed using quantification theory(I) for the prediction of debris flow hazard soil volume in Seoraksan National Park, Korea. According to the range using the stepwise regression analysis, the order of impact factors was as follows: vertical slope (0.9676), cross slope (0.6876), altitude (0.2356), slope gradient (0.1590), and aspect (0.1364). The extent of the normalized score using the five-factor categories was 0 to 2.1864, with the median score being 1.0932. The prediction criteria for debris flow occurrence based on the normalized score were divided into four grades: class I, >1.6399; class II, 1.0932-1.6398; class III, 0.5466-1.0931; and class IV, <0.5465. Predictions of debris flow occurrence appeared to be relatively accurate (86.3%) for classes I and II. Therefore, the prediction criteria for debris flow will be useful for judging the dangerousness of slopes.
