N-acetylcysteine modulates cyclophosphamide-induced immunosuppression, liver injury, and oxidative stress in miniature pigs

  • Cyclophosphamide, a cytotoxic anticancer agent, induces immunosuppression and has several adverse effects. N-acetylcysteine alleviates oxidative stress, liver injury, and intestinal tissue damage. The present study examined whether N-acetylcysteine modulates the adverse effects of cyclophosphamide in pigs. Miniature pigs (n = 15) were used as an experimental model to evaluate the effects of N-acetylcysteine treatment on immune reactions, liver injury, and oxidative stress after cyclophosphamide challenge. Corn-soybean meal based dietary treatments were as follows: control diet with either saline injection, cyclophosphamide injection, or 0.5% N-acetylcysteine and cyclophosphamide injection. N-acetylcysteine increased the number of immune cells and decreased TNF-α production after cyclophosphamide injection and decreased TNF-α, IFN-γ, NF-κB, and IL-8 expression and increased IL-10 expression in peripheral blood mononuclear cells. Serum levels of alanine transaminase and aspartate aminotransferase decreased, superoxide dismutase activity increased, and malondialdehyde activity decreased following N-acetylcysteine treatment after cyclophosphamide injection. N-acetylcysteine decreases immunosuppression, liver injury, and oxidative stress in cyclophosphamide-challenged miniature pigs. The present study suggests that N-acetylcysteine has therapeutic application in livestock for modulating immune reactions, liver injury, and oxidative stress.

Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.

Effects of Fermented Soybean Meal on Immune Response of Weaned Calves with Experimentally Induced Lipopolysaccharide Challenge

  • The present study was conducted to evaluate the effects of fermented soybean meal (FSBM) on the level of cortisol hormone and immune-related serum proteins in weaned calves after experimentally induced lipopolysaccharide (LPS) challenge. Holstein neonatal calves (n = 21; 8 males and 13 females, BW = $42.2{\pm}6.15$ kg) were randomly allocated to one of two dietary treatments: SBM (control calf starter having soybean meal (SBM) as a main protein source) and FSBM (substitute SBM in control diet with FSBM) groups. All calves were fed milk replacer using an automatic milk-feeder according to step-down milking method and weaned at 7 weeks old. Experimental diets were given to calves ad libitum throughout the experimental period. For LPS challenge, all calves except negative control animals given phosphate buffered saline (PBS), were injected subcutaneously with Salmonella typhimurium LPS on day 7 (D7) after weaning (D0). No significant difference in growth performance and milk intake was observed between SBM and FSBM calves. Feeding FSBM diet resulted in significantly (p<0.05) higher LPS-specific IgG at D12 and D19 and LPS-specific IgA at D19 in peripheral blood. Calves fed with FSBM diet also had significantly (p<0.05) higher concentration of serum haptoglobin (Hp) at D8. Overall concentration of cortisol in FSBM group was considerably lower than that of SBM group. Results from current study indicate that FSBM may provide beneficial effects in alleviating weaning stress and enhance immune status of weaned calves.

  • 본 연구에서는 초고속 자기부상열차의 단면도를 통하여 2-D형상을 모델링하고 이를 기반으로 항력과 유동 특성에 대한 분석을 수행하였다. 유동의 마하수가 0.3 이상임을 고려하여 압축성 모델이 사용되었고, 난류모델은 Menter's k-w SST(Shear Stress Transport)모델을 적용시켰다. 2-D 해석과 자기부상열차의 특성상 열차가 공기중에서 주행하고 있는 것으로 가정하고 공력 특성을 해석하였다.

Analysis of the Stress Effects of Endocrine Disrupting Chemicals (EDCs) on Escherichia coli

  • In this study, three of the representative EDCs, $17{\beta}$-estradiol, bisphenol A, and styrene, were employed to find their mode of toxic actions in E. coli. To accomplish this, four different stress response genes, recA, katG, fabA, and grpE genes, were used as a representative for DNA, oxidative, membrane, or protein damage, respectively. The expression levels of these four genes were quantified using a real-time RT-PCR after challenge with three different EDCs individually. Bisphenol A and styrene caused high-level expression of recA and katG genes, respectively, whereas $17{\beta}$-estradiol made no significant changes in expression of any of those genes. These results lead to the classification of the mode of toxic actions of EDCs on E. coli.

  • The underground construction and mining are facing many geomechanics challenges stemming from, geological complexities and stress-driven rock mass degradation processes. Brittle failing rock at depth poses unique problems as stress-driven failure processes often dominate the tunnel behaviour. Such failure processes can lead to shallow unravelling or strainbursting modes of instability that cause difficult conditions for tunnel contractors. This keynote address focuses on the challenge of anticipating the actual behaviour of brittle rocks in laboratory testing, for empirical rock mass strength estimation, and by back-analysis of field observations. This paper summarizes lessons learned during the construction of deep Alpine tunnels and highlights implications that are of practical importance with respect to constructability. It builds on a recent presentation made at the $1^{st}$ Southern Hemisphere International Rock Mechanics Symposium held in Perth, Australia, in September this year, and includes results from recent developments.

  • The purpose of this study is to optimize the artificial lumbar spine with the most lightweight three - dimensional porous structure while satisfying the safety factor of 3. We have introduced representative volumes to proceed with a local analysis and studied the maximum porosity distribution that meets allowable stress. The maximum porosity was studied at 12 points and the intermediate values were predicted based on this points. If the lumbar spine is designed according to this predicted results, the optimal design will be achieved.

  • In this paper, a composite material analysis program based on the finite element method(FEM) is used. The purpose of this study was to verify whether the composite material analysis program which developed as part of a project of development of softwares and educational contents for structural vibration and composite material analysis that can calculate how similar the macroscopic mechanical behavior of the composite materials actually. Because composite materials are generally anisotropic, analysis of composite laminate is used for the constitutive equations of orthotropic material. For convenience, the unit is ommited in all calculations. To verify the accuracy of the finite element method based program, the deflection and stress distribution of the simply supported composite material laminated plate subjected to a uniform load distribution is compared with exact solution. Size and properties of the composite material laminate used for analysis are fixed variables, and by changing the number of elements and the total thickness of the laminate is compared with the exact solution to the resulting value, respectively.

Stress and language recovery in individuals with aphasia: constraint induced aphasia therapy

  • Objective: Constraint induced aphasia therapy (CIAT) is a more intensive form of language treatment for aphasia as compared to traditional treatments. This study examined whether there are differences in cortisol stress levels between the two methods of aphasia treatment as well as effects on language skills. Design: Randomized controlled trial. Methods: A total of 20 participants with expressive aphasia were randomly placed into one of the two treatment groups. The CIAT group received 10 days of intensive treatment over two weeks. The traditional therapy group received 6 days of treatment over 2 weeks. All participants in each group provided salivary cortisol samples before treatment, at the mid-point of treatment, and at the conclusion of treatment. Language skills were assessed before treatment and at the conclusion of treatment. Results: A significantly higher proportion of individuals in the CIAT treatment group had increased salivary cortisol stress levels when compared to the traditional treatment group at the mid-point of the program (80% versus 30% respectively, p<0.05). There was no significant difference in the proportion of individuals with increased cortisol stress by the end of the treatment. Language scores for word repetition and overall aphasia quotient significantly improved for the CIAT group when compared to the traditional group (p<0.05). Conclusions: The CIAT treatment appears to initially create increased psychophysiological stress as compared to the traditional treatment. In spite of the initial increases in psychophysiological stress, participants appear to become conditioned to the challenge and ultimately have enhanced benefit from CIAT treatment.

The Effect of job Characteristics and Personal Factors on Work Stress, Job Satisfaction and Turnover Intention (간호사의 직무특성과 개인의 성격이 직무스트레스, 직무만족 및 이직의도에 미치는 영향)

  • The present study examined the causal relationships among nurses' job environment /job characteristics(work overload, lack of autonomy, professional role conflict, interpersonal relationships), maturity, job stress, job satisfaction and turnover intention by constructing and testing a theoretial framework. Based on Katz and Kahn's (1978) theory of organizational open system and Kahn, Wolfe, Quinn, and Snoek's (1964) theory of stress, nurses' turnover intention, job satisfaction and job stress were conceived of as outcomes of the interplay between personal characteristics and work environment. Personal aspects associated with outcome variables included professional knowlege and skill, and maturity(challenge, commitment, control, responsibility). The work environment factors involved work overload, lack of autonomy, professional role conflict, and interpersonal relationships (social support). Three university hospitals located in Seoul were selected to participate. The total sample of 443 registered nurses represents a response rate of 96 percent. Linear structural relationships (LISREL) technique was used to test the fit of the proposed conceptual model to the data and to examine the causal relationships among variables. The result showed that both the proposed model and the modified model fit the data excellently, revealing considerable explanatinal power for job stress and job satisfaction. The explanatory power of turnover intention was relatively lower than those of stress and satisfaction. In predicting nurses' stress, satisfaction and turnover intention, the findings of this study clearly demonstrated that professional role conflict might be the most important variable of the all the environmental variables and personal characteristics. The results were dis-cussed, including directions for the future research and practical implications drawn from the research were suggested.
