Evaluation of KiSS1 as a Prognostic Biomarker in North Indian Breast Cancer Cases

  • Background: Breast cancer is the commonest female cancer worldwide and its propensity to metastasize negatively impacts on therapeutic outcome. Several clinicopathological parameters with prognostic/predictive significance have been associated with metastatic suppressor expression levels. The role of metastatic suppressor gene (MSG) KiSS1 in breast cancer remains unclear. Our goal was to investigate the possible clinical significance of KiSS1 breast cancer. Materials and Methods: The study was conducted on 87 histologically proven cases of breast cancer and background normal tiisue. Quantitative reverse transcriptase polymerase chain reaction (qRT PCR) and immunohistochemistry (IHC) were used to investigate KiSS1 at gene and protein levels, respectively, for correlation with several patient characteristics including age, family history, hormonal receptor status, stage, tumor size, nodal involvement and metastatic manifestation and finally with median overall survival (OS). Results: Our study revealed (i) KiSS1 levels were generally elevated in breast cancer vs normal tissue (P < 0.05). (ii) however, a statistically significant lower expression of KiSS1 was observed in metastatic vs non metastatic cases (P = 0.04). (iii) KiSS1 levels strongly correlated with T,N,M category, histological grade and advanced stage (p<0.001) but not other studied parameters. (iv) Lastly, a significant correlation between expression of KiSS1 and median OS was found (P = 0.04). Conclusions: Conclusively, less elevated KiSS1 expression is a negative prognostic factor for OS, advancing tumor stage, axillary lymph node status, metastatic propensity and advancing grade of the breast cancer patient. Patients with negative KiSS1 expression may require a more intensive therapeutic strategy.

  • Objective : From an analysis of a community outbreak of measles in Youngju, Gyeongbuk, in March 2000, the authors evaluated the effectiveness of the measles vaccination and its determinants to Provide an epidemiologic basis for the establishment of a vaccination policy. Methods : Information was collected regarding the vaccinations from the health records of four primary schools and through a questionnaire surrey of the parents of students in two middle and two high schools (N=4638). Measles cases were surveyed from the patient list of each school and from case reports in the public health center. The attack rate and vaccine effectiveness of measles was evaluated by school; grade; frequency, region, and institution of vaccination. Results : The attack rate cf measles, 6.3% among the total subjects, was higher in middle school students (15.8%) than in primary (2.0%, p<0.05) and high school students (8.9%, p<0.05). The attack rate of the unvaccinated group was 4.6-fold higher than the vaccinated group in primary schools (p<0.05). Vaccine effectiveness was 83.2% among lower graders of primary schools, 34.0% among higher graders of primary schools,26.1% in middle schools, and -7.0% in high schools. In multiple logistic regression analysis, grade in school (odds ratio, 0.77; 95% CI=0.67-0.87) and frequency of vaccination (odds ratio, 0.57: 95% CI=0.37-0.58) were significant predictors of the outcome. Conclusions : We concluded that the explosive outbreak of measles in this area resulted from both inadequate vaccination coverage and secondary failure of vaccination. There was no evidence of any effect of the cold chain system on the vaccine failure.

크실렌 노출로 인한 요중 메틸마뇨산 배설에 미치는 유전자 다형성 연구 (A Study on Polymorphism Affecting Excretion of Urinary Methylhippuric Acid due to Xylene Exposure)

  • Objectives : The purpose of this study was to investigate the effect of genetic polymorphism of cytochrome P450 2E1 (CYP2E1) and aldehyde dehydrogenase 2 (ALDH2) on the xylene metabolism. Methods : Among 247 workers, 116 were occupationally exposed to xylene and 131 were not. Workers exposed to xylene had different work such as spray, touch-up, mix & assist, and pre-treat. Questionnaire variables were age, sex, use of personal protective equipment, smoking, previous night's drinking and work duration. The urinary methylhippuric acid was measured in the urine collected in the afternoon and corrected by urinary creatinine concentration. The genotypes of CYP2E1 and ALDH2 were investigated by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) methods with DNA extracted from venous blood. Results : 1. The urinary concentrations of o-, m-, and p-methylhippuric acid and total methylhippuric acid in the exposed group were significantly higher than those in the non-exposed group (p<0.001). 2. In multiple regression analysis, the urinary methylhippuric acid concentration was significantly influenced by exposure grade (Job-exposure matrixes), smoking, drug use and kind of protective equipment (p<0.1). 3. Genetic polymorphism of CYP2E1 and ALDH2 did not affect urinary methylhippuric acid level in the exposed group (p>0.05). Conclusions : Exposure grade, smoking, drug use and kind of protective equipment affected urinary methylhippuric acid level, whereas genetic polymorphism of CYP2E1 and ALDH2 did not. However, further investigation for the effect of genetic polymorphism on the metabolism of xylene with a larger sample size is needed.

기초공통개념으로서 에너지에 대한 3~9학년 학생들의 문항 반응 분석 (Item Response Analysis of Energy as a Cross-Cutting Concept for Grades 3 to 9)

  • 이 연구에서는 기초공통개념으로서 에너지에 대한 학습발달과정연구의 기초 자료를 제공하기 위하여 3~9학년 학생들의 평가 문항 응답 결과를 분석하였다. 검사 문항은 전기 회로, 낙하 물체의 역학적 에너지, 물질의 상태 변화, 용해 현상, 생물체의 생명 현상, 먹이 사슬, 태양과 지구의 복사평형, 및 물의 순환계에서 에너지에 대한 이해를 조사하는 순위 선다형 문항으로 구성되었다. 학생들의 응답 결과는 학년별, 선택지 수준별 응답 빈도에 따라 교차분석을 실시하였고, Rasch 모델을 적용한 문항반응 분석으로서 Wright map 및 DIF 분석을 수행하였다. 연구 결과, 8가지 주제들에 대하여 에너지 이해의 발달과정은 현상과 에너지의 관련성을 인식하거나 에너지의 종류를 파악, 에너지의 이동과 전달을 인식, 에너지가 다른 형태로 전환 및 변환됨을 인식, 에너지의 보존을 인식하는 순서로 진행된다는 경험적 근거를 확보할 수 있었다. 전체 학년에 걸쳐서 공통적으로 8개 주제에 대하여 에너지 보존에 대한 이해가 부족하였다. 학년이 높아짐에 따라 에너지의 전달과 전환에 대한 이해 수준은 주제들마다 다소 차이가 있었다. Rasch 모델을 적용한 문항 반응을 분석한 결과 학생들은 다른 현상의 에너지에 비해 용해 현상의 에너지를 가장 쉬운 과제로 인식하였고 물의 순환 과정에서 에너지 이해를 가장 어려운 과제로 인식하였다. 이 연구의 결과는 에너지의 관점에서 각 주제들이 초중등 과학 교육과정의 어느 학년에, 몇 번째 단원에 어떤 방식으로 배치되어야 할 것인지는 경험적인 연구 자료로서 활용될 수 있으며, 새 과학 교육과정의 내용 체계를 구성할 때 고려할 수 있는 중요한 시사점이 될 수 있었다.

Effects of feeding high-energy diet on growth performance, blood parameters, and carcass traits in Hanwoo steers

  • Objective: Our study aimed to investigate the effects of a 2% increase in dietary total digestible nutrients (TDN) value during the growing (7 to 12 mo of age) and fattening (13 to 30 mo of age) period of Hanwoo steers. Methods: Two hundred and twenty Hanwoo steers were assigned to one of two treatments: i) a control group (basal TDN, BTDN, n = 111 steers, growing = 70.5%, early fattening = 71.0%, late fattening = 74.0%) or high TDN (HTDN, n = 109 steers, growing = 72.6%, early = 73.1%, late = 76.2%). Growth performance, carcass traits, blood parameters, and gene expression of longissimus dorsi (LD) (7, 18, and 30 mo) were quantified. Results: Steers on the BTDN diets had increased (p≤0.02) DMI throughout the feeding trial compared to HTDN, but gain did not differ appreciably. A greater proportion of cattle in HTDN received Korean quality grade 1 (82%) or greater compared to BTDN (77%), while HTDN had a greater yield grade (29%) than BTDN (20%). Redness (a*) of LD muscle was improved (p = 0.021) in steers fed HTDN. Feeding the HTDN diet did not alter blood parameters. Steers fed HTDN diet increased (p = 0.015) the proportion of stearic acid and tended to alter linoleic acid. Overall, saturated, unsaturated, monounsaturated, and polyunsaturated fatty acids of LD muscle were not impacted by the HTDN treatment. A treatment by age interaction was noted for mRNA expression of myosin heavy chain (MHC) IIA, IIX, and stearoyl CoA desaturase (SCD) (p≤0.026). No treatment effect was detected on gene expression from LD muscle biopsies at 7, 18, and 30 mo of age; however, an age effect was detected for all variables measured (p≤0.001). Conclusion: Our results indicated that feeding HTDN diet could improve overall quality grade while minimum effects were noted in gene expression, blood parameters, and growing performance. Cattle performance prediction in the feedlot is a critical decision-making tool for optimal planning of cattle fattening and these data provide both benchmark physiological parameters and growth performance measures for Hanwoo cattle feeding enterprises.

  • Schools should be a safe and comfortable environment for learning and studying. However, school facilities are usually used by many students during restricted time. These conditions such as many people, restricted time and space mold a chain of school accidents. In Korea, the school accidents are increasing annually and many students experience 'big or small incidents' and are exposed to 'potential accidents'. In this study, I investigated the present situation of the elementary schools' accidents in In-Cheon area, and analyzed the various characteristics of them. The accidents in elementary school have tendencies of occuring frequently in higher grade and boy students relatively. As analyzed in the place of accident, classrooms have a set of factors of accident, on the other hand, corridor and staircase show single distinct cause of safety problem. And also, the major portion of accidents is associated with the door in restroom and entrance space. In order to reduce the severity and frequency of school accidents, there should be needed multilateral and interdisciplinary approach. To achieve the safe school environments, this study focused on establishing a foundation of practical directions in designing elementary schools.

지하수 세균 군집의 유전적 다양성 (The Genetic Diversity of Bacterial Communities in the Groundwater)

  • 서울시 소재 지하수 중 중금속으로 오염되어 음용수 이외의 생활 용수로 사용하고 있는 1개 정점과 음용수로 사용하고 있는 1개 정점, 대조군으로 강화도 천연 동굴의 1개 정점을 대상으로 실험을 실시하였다. 지하수 세균군집의 유전적 다양성의 변화를 보기 위해 지하수 세균 군집에서 16SrDNA를 증폭하는 primer로 PCR(polymerase chain reaction)을 실시한 후 ARDRA(amplified ribosomal DNA restriction analysis) 지문 분석으로 비교하였다. 16S rDNA를 증폭하여 제한효소 지문분석을 한 결과 in situ와 음용수에서 유전적 다양성이 상대적으로 크게 나타났다. 지하수 세균 군집의 ARDRA지문 분석은 상이한 환경과 서식지를 반영한 유전적 차이를 빠르게 비교 분석할 수 있었으며 지하수의 오염도에 따른 미생물의 다양성(천연 동굴>음용수>오염수)을 확인할 수 있었다.

The GSTP1 Ile105Val Polymorphism is not Associated with Susceptibility to Colorectal Cancer

  • The glutathione S transferase (GST) family is a major part of cellular defense mechanisms against endogenous and exogenous substances, many of which have carcinogenic potential. Alteration in the expression level or structure of the glutathione-S-transferase (GST) enzymes may lead to inadequate detoxification of potential carcinogens and consequently contribute to cancer development. A member of the glutathione-S-transferase (GST) family, GSTP1, is an attractive candidate for involvement in susceptibility to carcinogen-associated colorectal cancer. An $Ag{\rightarrow}G$ transition in exon 5 resulting in an Ile105Val amino acid substitution has been identified which alters catalytic efficiency. The present study investigated the possible impact of Ile105Val GSTP1 polymorphism on susceptibility to colorectal cancer. in Jordan We examined 90 tissue samples previously diagnosed with colorectal carcinoma, and 56 non-cancerous colon tissues. DNA was extracted from paraffin embedded tissues and the status of the GSTP1 polymorphism was determined using a polymerase chain reaction restriction fragment length polymorphism (RFLP) method. No statistically significant differences were found between colorectal cancer cases and controls for the GSTP1 Ile/Ile, Ile/Val and Val/Val genotypes. The glutathione S-transferase polymorphism was not associated with risk in colorectal cancer cases in Jordan stratified by age, sex, site, grade or tumor stage. In conclusion, the GSTP1 Ile105Val polymorphism is unlikely to affect the risk of colorectal cancer.

Gemcitabine in Treating Patients with Refractory or Relapsed Multiple Myeloma

  • Background: Patients with refractory or relapsed multiple myeloma are considered to have a very poor prognosis, and new regimens are needed to improve the outcome. Gemcitabine, a nucleoside antimetabolite, is an analog of deoxycytidine which mainly inhibits DNA synthesis through interfering with DNA chain elongation and depleting deoxynucleotide stores, resulting in gemcitabine-induced cell death. Here we performed a systemic analysis to evaluate gemcitabine based chemotherapy as salvage treatment for patients with refractory and relapsed multiple myeloma. Methods: Clinical studies evaluating the impact of gemcitabine based regimens on response and safety for patients with refractory and relapsed multiple myeloma were identified by using a predefined search strategy. Pooled response rate (RR) of treatment were calculated. Results: In gemcitabine based regimens, 3 clinical studies which including 57 patients with refractory and relapsed multiple myeloma were considered eligible for inclusion. Systemic analysis suggested that, in all patients, pooled RR was 15.7% (9/57) in gemcitabine based regimens. Major adverse effects were hematologic toxicity, including grade 3 or 4 anemia, leucopenia and thrombocytopenia i. No treatment related death occurred with gemcitabine based treatment. Conclusion: This systemic analysis suggests that gemcitabine based regimens are associated with mild activity with good tolerability in treating patients with refractory or relapsed multiple myeloma.
