This study was carried out to investigate yield loss due to soybean leaf spot disease caused by Cercospora sojina Hara and to determine the economic threshold level. The investigations revealed highly significant correlations between disease severity (diseased leaf area) and yield components (pod number per plant, total grain number per plant, total grain weight per plant, percent of ripened grain, weight of hundred seed, and yield). The correlation coefficients between leaf spot severity and each component were -0.90, -0.90, -0.92, -0.99, -0.90 and -0.94, respectively. The yield was inversely proportional to the diseased leaf area increased. The regression equation, yield prediction model, between disease severity (x) and yield (y) was obtained as y = -3.7213x + 354.99 ($R^2$ = 0.9047). Based on the yield prediction model, economic injury level and economic threshold level could be set as 3.3% and 2.6% of diseased leaf area of soybean.
This study was conducted to obtain a supply of conidia sufficient for screening mungbean mutant lines for a source of resistance to Cercospora leaf spot caused by Cercospora canescens Ellis and Martin. Abundant sporulation occurred in cultures on mungbean leaf decoction oatmeal agar(MOA) exposed to about 2,500 Lux of fluorescent light. but it did not occur in continuous darkness. The conditions that produced maximum number of conidia was not coincided with those for vegetative growth and pigmentation in culture medium. Removal of aerial mycelium in culture by brushing with sterile water so enhanced the conidial production that oatmeal agar medium(OA) could be useful for production of abundant conidia by the treatment.
Disease incidence of Codonopsis lanceolata was surveyed at the major cultivating fields in Chonbuk province in 1996 to 1997. The main diseases of Codonopsis lanceolata were ovserved as leaf spot caused by Septoria codonopsis, anthracnose by Glomerella cingulata, brown leaf spot by Cercospora sp., rust by Coleosporium koreanum, powdery mildew by Erysiphe sp., Fusarium wilt caused by Fusarium oxyporum, and white root rot by Sclerotium rolfsii. Anthracnose, leaf spot and brown leaf spot occurred severely on leaves from early July to late August. They were caused early fallen leaves. Fusarium wilt and white root rot occurred severely on stem and below the soil line in late August. They resulted in withering to death or chlorosis and fallen of leaves. Disease incidence of Codonopsis lanceolata was also substantially different in occurrence with a method of cultivation in late growth stage. Fusarium wilt and white root rot were more severe with a method of no support cultivation than those with a method of support cultivation with a stick. Fusarium wilt occurred 48.8% in a method of no support cultivation but 3.1% in a method of support cultivation with a stick. And white root rot occurred 18.9% in a method of no support cultivation but 0.3% in a method of no support cultivation with a stick. Thus, it proved that soil-borne diseases could be controlled support cultivation with a stick.
Soybean frogeye leaf spot caused by the fungus Cercospora sojina Hara, has known to lead a severe reduction of crop yield. Since frogeye leaf spot on soybean has recently become a serious problem in Korea, the susceptibility of recent recommended cultivars against C. sojina had been tested. To standardize the disease severity of soybean, the optimum sporulation condition of C. sojina and the disease index were established in this study. Sporulation was maximized on the 10% V8 juice agar with 12 h light and 12 h dark at $25^{\circ}C$. Spore suspension ($10^5spores/ml$) was sprayed on the leaves of soybean (V6 stage), and the disease responses to each isolate were evaluated on 28 days after inoculation. As a result, Daepung, Shinpaldal2ho, Yeonpung and Cheonga showed the resistance reaction to 8, 7, 6, 6 isolates of C. sojina, respectively, whereas Cheongja, Hwangkeum, Taekwang, Daewon, Cheonsang and Sinhwa showed the susceptible reaction to 8 isolates of C. sojina. Breeding the resistant soybean cultivars against C. sojina requires a uniform resistance for screening technique. The disease index of frogeye leaf spot on soybean developed in this study can be effectively used for the accurate field assay to select the frogeye leaf spot resistant soybean.
Bipolaris panici-miliacei, Cercospora fusimaculans, Fusarium moniliforme and Rhizoctonia solani were pathogenic fungi detected from 5 seed samples of common millet (Panicum miliaceum). Morphological characters of B. panici-miliacei were as follows. Conidiophores were dark olivaceous brown, simple, cylindrical, geniculate, and septate. Conidia were fusoid, dark olivaceous brown, tapering gradually toward the ends, straight to slightly curved, 3~13 distoseptate, and 29.4~155.4$\times$10~26 ${\mu}{\textrm}{m}$ in size with dark hilum included within the contour of the basal cell. Seed infection with B. panici-miliacei caused seed rotting, coleoptile spot, and seedling blight of common millet plants. According to the inoculation experiments, B. panici-miliacei showed strong virulence on the young seedlings of common millet, but very weak virulence on the young seedlings of rice (Oryza sativa) and foxtail millet (Setaria italica).
The pathotypes of Cercospora beticola, causal agent of sugar beet leaf spot disease, were identified by application of pathogenicity test using 100 isolates obtained from the provinces with intensive sugar beet cultivation. For the identification of pathotypes, five sugar beet cultivars were used each with different resistance factors. Cultivar reactions were determined by inoculation of cultivars with the isolates under controlled conditions and measuring disease severity on the $15^{th}$ day according to the 1-9 KWS Scale. Based on the reactions of the five cultivars, a total of 15 pathotypes were detected. All employed sugar beet cultivars were resistant to Pathotype no:1 comprising most of the isolates. Genetic diversity of the causal agent was characterized by AFLP reaction. The products acquired at the end of AFLP reaction were detected by means of Beckman CEQ 8800 DNA Capillary Series Analysis and the results obtained were evaluated according to the similarity index UPGMA. For the genetic analysis of C. beticola isolates, 9874 polymorphic fragments of sizes between 100 and 500 bp were analysed which were generated by nine primers. The dendrogram derived from AFLP analysis depicted the existence of five different subgroups. The polymorphism rate among isolates was 91.13% and the dendrogram distribution of the pathotypes obtained by pathogenicity indicated that pathotypes were not discriminated and did not compose any groups.
Lipoxygenase and Kunitz trypsin inhibitor protein of mature soybean [Glycine max (L.) Merr.] seed are main anti-nutritional factors in soybean seed. A new soybean cultivar, "Gaechuck#1" with the traits of black seed coat, green cotyledon, lipoxygenase2,3 and Kunitz trypsin inhibitor protein free was developed. It was selected from the population derived the cross of "Gyeongsang#1" and C242. Plants of "Gaechuck#1" have a determinate growth habit with purple flowers, brown pubescence, black seed coat, black hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on dry weight basis have averaged 39.1% and 16.2%, respectively. It has shown resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. "Gaechuck#1" matured on 5-10 October with a plant height of 50 cm. The 100-seed weight of "Gaechuck#1" was 23.2g. Yield of "Gaechuck#1" was averaged 2.2 ton/ha from 2005 to 2007.

A new soybean cultivar, "Gaechuck#2" with yellow seed coat, lipoxygenase2,3-free and Kunitz trypsin inhibitor protein-free was developed. It was selected from the population derived from the cross between "Jinpumkong2ho" and C242. Plants of "Gaechuck#2" have determinate growth habit with purple flowers, tawny pubescence, yellow seed coat, yellow hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on a dry weight basis were 40.7% and 18.7%, respectively. It has shown a resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. Gaechuck#2 matured in 4 October with plant height of 54cm and a 100-seed weight of 24.4g. Average Yield of Gaechuck#2 was 230 - 250 kg/10a in 2005 - 2007.

Serratia marcescens W1, isolated from cucumber-cultivated soil in Suwon, Korea, evidenced profound antifungal activity and produced the extracellular hydrolytic enzymes, chitinase and protease. In order to isolate the antifungal genes from S. marcescens W1, a cosmid genomic library was constructed and expressed in Escherichia coli. Transformants exhibiting chitinase and protease expression were selected, as well as those transformants evidencing antifungal effects against the rice blast fungus, Magnaporthe grisea, and the cucumber leaf spot fungus, Cercospora citrullina. Cosmid clones expressing chitinase or protease exerted no inhibitory effects against the growth of fungal pathogens. However, two cosmid clones evidencing profound antifungal activities were selected for further characterization. An 8.2 kb HindIII fragment from these clones conditioned the expression of antagonistic activity, and harbored seven predicted complete open reading frames(ORFs) and two incomplete ORFs. The deduced amino acid sequences indicated that six ORFs were highly homologous with genes from S. marcescens generating pyrroloquinoline quinone(PQQ). Only subclones harboring the full set of pqq genes were shown to solubilize insoluble phosphate and inhibit fungal pathogen growth. The results of this study indicate that the functional expression of the pqq genes of S. marcescens W1 in E. coli may be involved in antifungal activity, via as-yet unknown mechanisms.

A new mungbean cultivar "Dahyeon" (Vigna radiata (L.) Wilczek) was derived from the cross between Jeonnam11 and AV-1-3-34-204 at National Institute Crop Science (NICS) and Jeollanamdo Agricultural Research & Extension Services (JARES) in 2006. "ahyeon" has erect growth habit, heart leaflet, green hypocotyl, black and straight pod when matured, and dull green seed surface. The pods number per plant was 21.0, which was 4.0 more than check cultivar "wool" The plant height and thousand seed weight were 57cm and 48 g respectively. The sprout yield was up to 5% more than that of the check cultivar. It was field resistance to mungbean mottle virus, cercospora leaf spot, and powdery mildew. The average yield was 1.90 ton/ha, which was 17% more than the check cultivar.
