Antigenicity Study on IDC7181, a Quaternary Ammoniopropenyl Cephalosporin Possessing Two Vinyl Groups (Cephalosporin계 항생제 IDC7181에 대한 항원성시험)

  • Antigenic potential of a novel cephalosporin, IDC7181 was examined a active systemic anaphylaxis (ASA) and passive cutaneous anaphylaxis (PCA) test, in guinea pig and mouse-rat models, respectively. In ASA test, IDC7181 induced the signs of restlessness in a few animals immunized with a high dose (100 mg/kg) of IDC7181 alone or in combination with Freund's complete adjuvant (FCA). In PCA test, only one of ten sera from the animals immunized with a high dose (100 mg/kg) of IDC7181 in the absence or presence of FCA showed positive reaction. The positive reaction, induced by IDC7181, which may be due to $\beta$-lactam ring, in ASA and PCA tests were negligible in comparison with those of traditional cephalosporins. Taken together, it is suggested that IDC7181 do not cause immunological problems in clinical dosage.

Effects of Nutrients and Culture Conditions on Morphology in the Seed Culture of Cephalosporium acremonium ATCC 20339

  • The objective of this study was to investigate the effects of nutrients and culture conditions on morphology during the seed culture of C. acremonium ATCC 20339 Morphological factors such as hyphal length number of tips number of arthrospores were observed to investigate the relationship between seed morphology and CPC production. During the time course of seed culture, hyphal length was shortened and the number of arthrospores increased rapidly On the other hand the number of tips deceased rapidly and this was closely related to the hyphal length Mixed nitrogen sources of 3% solybean meal and 1% cotton seed flour were determined as the proper organic nitrogen sources, in terms of the morphological factors in the seed culture. This fact was proven in batch culture for the production of Cephalosporin C. It was also found that a proper agitation speed enhanced the morphological differentiation of C. acremonium ATCC 20339, thus improving the production of Cephalosporin C.

  • The elongation of the chain length at 2'position of aminothiazolylacetamido group of cephalosporin antibiotics was achieved. The derivatives with elongated side chain at 2'position were successfully synthesis and their in vitro activities were evaluated. It is indicated that there is an optimum range of the chain length in order to exhibit higher biological activities. This result could open a possibility for the development of the cephalosporin antibiotics with higher activities by further variation of the substituents at other positions. The cephalosporin derivatives with a simple (relatively non-functionalized) side chain as well as with a linear extended side chain were also prepared. Low activities of these derivatives led to a conclusion that either simple (non-functionalized) change or linear elongation of the side chain does not enhance the antibacterial activities.

  • Using the polyclonal antibody for Xanthomonas citri ampicillin acylase raised in Pseudomonas-free Balb/c mice, the immunochemical similarity of several types of penicillin acylases including Erwinia aroideae penicillin V acylase, Escherichia coli penicillin G acylase, Pseudomonas melanogenum and Acetobacter turbidans ampicillin acylases, and Pseudomonas cephalosporin acylase was examined. Among tested, only P. melanogenum ampicillin acylase showed the cross-reactivity with the antibody.

  • Using cephalosporium acremonium ATCC 14553, we studied on the important factors in fermentation. Those factors were :carbon and nitrogen sources, methionine effect, absorbent effect, fermentation variables in a batch fermentation variables in a batch fermentation, effect of reuse of its cellular biomass as a reusable nutrient, and 2, 4-dinitrophenol effect on cephalosprin C production.

  • Purpose: To evaluate the effectiveness of new management policies on the incidence of invasive Candida infections Methods: This observational study involved a retrospective analysis of the patients' medical records. In total, 99 very low birth weight infants, who were admitted to the neonatal intensive care unit at Ajou University Hospital from January 2010 to December 2011, were enrolled for the study. Period I, defined as the period before the revision of management policies, comprised 57 infants; whereas, period II, defined as the period after the implementation of new management policies, comprised 42 infants. The new management policies entailed a reduction in antibiotic and histamine type 2 receptor blocker (H2 blocker) use, duration of central venous catheterization, and duration of endotracheal intubation. Results: There was a significant overall decrease in the use of antibiotics including 3rd generation cephalosporin and H2 blockers (P<0.05), and a significantly lower incidence of invasive Candida infections in period II as compared to period I (0/42 vs. 6/57, respectively; P=0.037). Comparison between infants with invasive Candida infections (n=6) and those without (n=93) showed that gestational age (odds ratio [OR], 0.909; 95% confidence interval [CI], 0.829 to 0.996; P=0.042) and the duration of 3rd generation cephalosporin use (OR, 1.093; 95% CI, 1.009 to 1.183; P=0.029) were statistically significant risk factors. Conclusion: The new management policies effectively decreased overall use of antibiotics, especially 3rd generation cephalosporin, and H2 blockers, which led to a significantly lower incidence of invasive Candida infections.

Prophylactic antibiotics in intra-oral bone grafting procedures: a prospective, randomized, double-blind clinical trial

  • Objectives: This study was conducted in order to assess the efficacy of 1st generation cephalosporin as use as a single-dose preoperative prophylactic antibiotic for surgical wound infections resulting from intra-oral bone grafting procedures. Materials and Methods: A total of 23 patients who were to undergo intra-oral bone graft procedures participated in this study. After randomization, 2 grams of 1st generation cephalosporin was orally administered to both the experimental and placebo groups one hour prior to surgery in a double-blind fashion. Post-operatively, the experimental group (12 patients) was orally administered placebo three times a day for three days. The control group (11 patients) was orally administered 1st generation cephalosporin three times a day for three days. The postoperative course was observed for one month including the clinical parameters associated with infection. Results: Postoperative infections were noted in 1 out of 11 patients in the experimental group. No infections occurred in the control group. Conclusion: There was no significant difference in the incidence of postoperative infections between the two groups. Two grams of 1st generation cephalosporin administered orally one hour before surgery served as an effective prophylactic antibiotics therapy for intra-oral bone graft surgery.

The Improvement of Cephalosporin C Production by Fed-batch Culture of Cephalosporium acremonium M25 Using Rice Oil

  • The objective of this study is to improve cephalosporin C (CPC) production byoptimization of medium and culture conditions. A statistical method was introduced to optimize the main culture medium. The main medium for CPC production was optimized using a statistical method. Glucose and corn steep liquor (CSL) were found to be the most effective factors for CPC production. Glucose and CSL were optimized to 2.84 and $6.68\%$, respectively. CPC produc­tion was improved $50\%$ by feeding of $5\%$ rice oil at day 3rd and 5th day during the shake flask culture of C acremonium M25. The effect of agitation speeds on CPC production in a 2.5-L bio­reactor was also investigated with fed-batch mode. The maximum cell mass (54.5 g/L) was obtained at 600 rpm. However, the maximum CPC production (0.98 g/L) was obtained at 500 rpm. At this condition, the maximum CPC production was improved about $132\%$ compared to the re­sult with batch flask culture.
