돈사폐수의 세정산발효시 온도와 전처리의 영향 (Effect of Temperature and Pre-treatment for Elutriated Acidogenic Fermentation of Piggery Waste)

  • The performance of elutriated acid fermentation with slurry-type piggery waste was investigated, especially to evaluate the effects of temperature and pre-treatment. In the first phase, the acid elutriation reactor with piggery waste after centrifugation operated at both mesophilic and thermophilic conditions to evaluate the effect of temperature. Solubilization yield($gVFAs/gSCOD_{prod.}$) and acidification rate($gVFAs/gSCOD_{prod.}$) in the thermophilic digestion were 0.45 and 0.55, which were higher than those of the mesophilic digestion, 0.25 and 0.45. In addition, the acid elutriation reactor at thermophilic temperature is more effective in removing e-coli. In the second phase, the acid elutriation reactor was fed with piggery waste before centrifugation. With piggery wastes before centrifugation, the solubilization yield and the acidificaton rate were 0.40 and 0.80, respectively, which were higher than the rates using piggery waste after centrifugation at both mesophilic and thermophilic conditions. The higher sludge volume reduction of 80% benefits sludge management. Furthermore, economical advantages can be achieved by removing the pre-treatment process, such as centrifugation. Consequently, the treatment with piggery waste before centrifugation proved to be effective. Also, the optimum temperature condition was estimated at mesophilic or thermophilic conditions, considering solubilization yields and acidification rates, though the system should be heated.

  • Purpose: We use the centrifugation of refrigeration state in separation of blood serum, Anti-ds-DNA, Vitamin $B_{12}$/Folate and GAD-Ab assay. However, Cortisol urine and 25-Hydroxyvitamin $D_3$ ($25OHD_3$) are conducted centrifuge at room temperature. This is troublesome that change centrifugation temperature into room temperature due to using of most assays at cold temperature. Therefore when using centrifuge after extraction of Cortisol urine and $25OHD_3$, we conducted researches on effect of the centrifugation temperature in assay results. Materials and Methods: In Cortisol urine, add dichloromethane 1.0 mL in urine $500\;{\mu}L$, mix for 15 minutes, and then centrifuge for 8 minutes at 2600rpm. In $25OHD_3$ add acetonitrile 0.5 mL in serum $200\;{\mu}L$, and then centrifuge for 8 minutes at 2600rpm. Those experiments were conducted centrifuge at room temperature and $4^{\circ}C$. And experiments conducted immediately after centrifugation at $4^{\circ}C$ and standing for 20 minutes after centrifugation $4^{\circ}C$. Results: In Cortisol urine, room temperature result in 1.93, 2.18, 2.43, 9.45, 14.2 (${\mu}g/dL$). Experiments of performing immediately after centrifuge at $4^{\circ}C$ result in 1.8, 2.0, 2.3, 8.1, 13.7 (${\mu}g/dL$). Experiments of performing after 20 minutes result in 2.1, 2.1, 2.7, 9.95, 14.35 (${\mu}g/dL$). On the other hand, the $25OHD_3$ tests conducted at room temperature result in 7.13, 26.6, 35.8, 48.2, 74.8 (ng/dL). Experiments were conducted immediately by pipetting after $4^{\circ}C$ centrifugation result in 7.53, 30.9, 40.3, 61.5, 89.1 (ng/dL) as results are higher than experiments at room temperature. The experiments that conducted centrifuge at $4^{\circ}C$ and then left at room temperature for 20 minutes result in 7.40, 32.4, 41.3, 51.6, 85.6 (ng/dL). Conclusions: Experiments were conducted by using centrifuge at $4^{\circ}C$ are higher or lower than room temperature. The differences between results of standing for 20 minutes after centrifuge at $4^{\circ}C$ and those of centrifuge at room temperature are less than conducting immediately. It is concerned that experiments conducted immediately after centrifuge at $4^{\circ}C$ are incorrect, because tubes become dim due to temperature differences between $4^{\circ}C$ and room temperature. Therefore, it is desirable to centrifuge at room temperature as manual and we should pipet promptly without stopping.

DILUTION AND SHORT-TERM STORAGE OF COCK SPERMATOZOA BY INHIBITION OF MOTILITY WITH FRUCTOSE AT AMBIENT TEMPERATURE

  • A simplified dilutor for cock spermatozoa at ambient temperature was achieved by adjusting the 5% concentration of fructose in isotonic saline. Motility of cock spermatozoa was arrested completely for maximum 6 hrs without affection the survivability of spermatozoa by employing this sugar. To study the effect of high concentration of fructose on fertility, sperm were inseminated with or without fructose at different hrs. Fructose from semen samples was removed by centrifugation. High fertility obtained in the hens inseminated with fructose free sperm (washed). In addition, washed sperm maintained the 85.00% fertility for 6 hrs in winter season ($17-21^{\circ}C$) and 82.67% fertility for 3 hrs in summer season ($31-35^{\circ}C$). Whereas control groups showed 47.33 and 25.33% fertility in winter and summer season respectively. No significant difference was found in percent motility and live counts between the control and washed experimental groups during winter season. However, these measures differed significantly in summer. Washing of cock spermatozoa more than once, high speed centrifugation and more duration for centrifugation proved harmful to fertility. It may be concluded that fructose (5%) can be used as a motility or metabolic inhibitor of spermatozoa for short-term storage of cock semen at ambient temperatures.

  • Indonesia is a tropical country having a temperature range of 25-35$^{\circ}C$ which can affect the skin and causes damages like aging. This aging process is due, at least, to free radical reactions. For this reason, many attempts had been done to find out creams containing natural antioxidant compound which have a potential of free radical scavenger. Kluwek, a fermented form of foot ball fruit or picung (Pangium edule.Reinw), had been proved to contain antioxidant compound in its methanol fraction oil to which antiaging cream was formulated. Stability evaluation was conducted for cream with Kluwek oil compared to base cream, including organoleptic (colour and odour), pH, viscosity, particle size, centrifugation test and flow characteristics either in room temperature (27$^{\circ}C$) or stress condition (4$^{\circ}C$ and 5$0^{\circ}C$) for 8 weeks continuously, and six times cycling test at 4$^{\circ}C$ and 5$0^{\circ}C$ every 24 hours. The results showed that cream with Kluwek oil and base cream were stable at temperature 27 and 4$^{\circ}C$, cycling test and centrifugation test, but not stable at 5$0^{\circ}C$. Free radical evaluation was done by Electron Spin Resonance and the result showed that cream with Kluwek oil had less free radicals compared to base cream.

피브린의 무게측정에 의한 새로운 트롬빈활성측정법 (The Novel Assay Method for Thrombin by Weighing Fibrin Clot)

  • 본 연구는 피브린의 무게를 측정하는 방법에 의하여 간단하고 단순하게 트롬빈활성을 측정하는 방법을 확립하기 위하여 수행되었다. 이 새로운 트롬빈측정 법은 트롬빈이 기질인 피브리노겐과 반응 후 생성된 피브린의 무게를 측정한다. 피브린의 생성은 사용한 트롬빈의 효소활성, 반응온도, 반응시간 및 원심분리의 시간에 의존하였다. 피브린의 생성은 37$^{\circ}C$에서 트롬빈활성이 1.0 unit/ml 이내, 4.0mg/ml의 피브리노겐 농도 및 반응시간이 5분 이내에는 직선적으로 증가하였다. 그리고 생성된 피브린은 3,000$\times$g에서 5분간 원심분리의 경우에 안정하였다. 이 새로운 피브린 측정 법은 트롬빈을 저해하는 천연물질을 탐색하는데 유행하게 사용될 것으로 기대된다.

청징 사과주스의 제조 및 품질특성 비교 (Quality Characteristics of Clarified Apple Juices Produced by Various Methods)

  • 청징 사과주스 제조시 원심분리를 이용한 청징보다는 한외여과 또는 혼합처리에 의하여 효율적인 청징 사과주스를 제조할 수 있는 것으로 나타났다. 원심분리시 회전속도가 높고 저온에서 공정을 행할수록 청징효과가 개선되어 회전속도 증가뿐만 아니라 저온의 효과가 매우 뚜렷했다. 원심분리시 최적조건은 $5^{\circ}C$, 10,000rpm에서 10분이었다. 한외여과시 공정조건에 관계없이 청징효과가 매우 우수한 것으로 나타났는데, 한외여과 최적 공정조건은 투과플럭스와 청징효과를 고려해 볼 때 온도 및 압력이 각각 $45^{\circ}C$, 150 kPa이었다. 혼합공정 후에 청징효과가 뚜렷하게 개선되지 않았는데 이는 한외여과 공정을 거치면서 이미 일정 수준의 청징이 이루어 졌음을 의미한다. 한외여과 공정의 효율성은 색도의 변화에서도 간접적으로 측정할 수 있었는데 한외여과 처리된 시료의 L*값과 ${\Delta}E$는 가장 높고 반면 a*값은 가장 낮은 것으로 나타났다. 전반적으로 비교하여 볼 때 한외여과 방법이 고품질의 청징 사과주스를 제조하는데 우수한 것으로 판단되며 또한 배제액 중의 고형성분은 식이섬유 강화 주스 제조 시에 첨가될 수 있는 이점이 있다.

Assessment of Stability and Safety of Maskne Cosmetic

  • Wearing a mask is still advised since COVID-19 continues to spread. However, masks may also irritate the skin and cause mask acne, often known as "maskne", which is a type of acne mechanica caused by friction between the skin and clothing. Therefore, there is a need to develop an effective maskne cosmetic. In this study, we made the maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 and investigated its stability and safety. To measure stability, a centrifugation test and heat-cool cycling were done, and changes in viscosity and pH were measured for 8 weeks. The Cumulative Irritation Test (CIT, WKIRB-202111-HR-096) was performed and positive reactions were determined by the ICDRG criteria. The results indicated that the samples were stable after centrifugation, temperature cycling, viscosity, and pH tests. In addition, cosmetic safety test results revealed that maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 did not cause any skin responses. These findings indicate that prepared maskne cosmetics' stability and safety were comparable to those of currently available commercial cosmetics.

다시마 추출액의 특성에 미치는 추출온도의 영향 (Effect of Extraction Temperature on Some Quality of Sea Tangle Extract)

  • Aqueous extraction of sea tangle at the temperature range of 60~10$0^{\circ}C$ was studied for temperature effects on soluble solids and protein yields, amino nitrogen, turbidity and viscosity of extracts. The solids and protein yield were increased as the temperature increased and most of solids and protein were extracted during 1 hour. The supernatant ratio after centrifugation showed significantly low for the extraction at 6$0^{\circ}C$. More temperature effects were found on turbidity and viscosity than yields. A significantly higher total amino nitrogen contents was obtained from higher temperature at initial stage of extraction and then the differences of them became to be narrow as the extraction prolonged further. The low values of 24.1% solids and 13.5% protein yields after 2 hours of extraction at 10$0^{\circ}C$ indicated that most of solids in sea tangle are insouble.

Effects of season and single layer centrifugation on bull sperm quality in Thailand

  • Objective: The aim of study was to investigate the effects of season and single layer centrifugation (SLC) before cryopreservation on post-thaw bull sperm quality in Thailand. Methods: Semen was collected from 6 bulls (Bos indicus) in summer, rainy season and winter 2014 through 2016. Semen characteristics, sperm morphology, sperm kinematics, viability, chromatin structure and mitochondrial membrane were evaluated. Meteorological data were available from the local meteorological station; Results: Season had an effect on semen characteristics in the raw ejaculate, with higher proportions of normal spermatozoa and lower abnormalities in winter than in the other two seasons. Sperm kinematics, viability, DNA fragmentation index, and mitochondrial membrane potential were not different between seasons. Sperm samples selected by SLC had greater normal morphology and a lower proportion with bent tails than controls and higher values of progressive motility (PRO), beat cross frequency, linearity, straightness, wobble (WOB), and lower values of slow motility, velocity average path (VAP), velocity curved line, and amplitude of lateral head displacement than controls. In addition, SLC-selection had a favorable effect on PRO, VAP, and WOB that differed among seasons. Conclusion: Our results suggested that these bulls were well adapted to their location, with season having an effect on sperm morphology. Moreover, SLC could be used prior to cryopreservation, regardless of season, to enhance normal morphology and kinematics of bull sperm samples without adversely affecting other parameters of sperm quality. However, there was considerable variation among bulls in DNA fragmentation index, mitochondrial membrane potential and sperm viability. In addition, SLC had a positive effect on sperm morphology and sperm kinematics, which could be expected to influence fertility.

Transglutaminase로 처리한 초고온 살균유 침전물의 전자현미경적 관찰 (Electron Microscopical Observation of Transglutaminase-treated Ultra High Temperature Milk Sedimiment)

  • 초고온-살균 우유를 이용하여 탈지유와 콜로이드성 인산칼슘이 제거된 우유에 TGase를 첨가하여 반응시킨 다음 초고속 원심분리를 실시하고 침전된 카제인 입자들을 동결건조하여 조직의 성상에 대해 주사 전자 현미경을 이용해 관찰, 비교하였다 탈지유는 카제인 입자들이 원심분리 초기(5,000${\times}$g)에는 규칙적으로 회합하였으며 원심분리 속도가 증가하면서 홈을 형성하였고(10,000∼20,000${\times}$g), 40,000${\times}$g에서 다시 회합하였다. 그리고, 100,000${\times}$g에서는 카제인 입자 수의 증가와 함께 규칙적인 회합층을 이루었다. 탈지유에 TGase를 처리하고 1시간 반응시킨 경우에 카제인 입자들의 성상은 입자들끼리 엉켜져서 회합을 하였으며 초고속 원심 분리 속도 증가에 따라 규칙적으로 혹은 불규칙적으로 변형되면서 입자들이 넓어지다가 다시 규칙적으로 카제인 입자들이 회합층을 이루었다. 탈지유에 TGase를 처리하고 8시간 반응시킨 경우 카제인 입자들의 성상은 1시간 반응시킨 경우보다 카제인 입자들이 미세해지면서 규칙적인 층을 형성하였으며, 초고속 원심분리 속도가 증가함에 따라 카제인입자들의 조직이 홈을 형성하면서 불규칙적으로 회합하여 분산된 형태를 나타내다가 다시 비교적 규칙적으로 회합하였다. 콜로이드성 인산칼슘이 유리된 우유에서는 카제인 입자들이 침전되지 않았다. TGase가 첨가되어 1시간 반응시킨 후 콜로이드성 인산칼슘이 유리된 우유에서는 20,000${\times}$g 속도에서부터 카제인 입자들이 침전하였고 그 양상은 작은 낙엽처럼 미세하게 회합층을 형성하였으며, 속도 증가에 따라 카제인 입자들의 조직은 개방되었다. TGase가 첨가된 후 8시간 반응시킨 다음 콜로이드성 인산칼슘이 유리된 시료의 경우 카제인 입자들은 대부분 넓게 회합층을 이루고 불규칙적이었으며 원심분리 속도 증가에 따라서도 큰 변화가 없었다. TGase가 첨가된후 1시간 혹은 8시간 반응시킨 다음 콜로이드성 인산칼슘을 제거하고 원심분리한 현탁액(유청 단백질)을 pH 4.6으로 조정 후 2단계 원심 분리한 침전물의 경우 카제인 입자들은 1시간 반응시킨 시료에서는 대부분의 카제인 입자들끼리 엉켜져 불규칙적인 회합층을 이루고 있는 반면에 8시간 반응시킨 시료에서는 입자들이 미세한 형태로 분산되어 규칙적인 층을 형성하였다.
