SELECTIVE TOXICITY OF CHRONIC LEAD INGESTION TO CENTRAL CATECHOLAMINERGIC NERVOUS SYSTEM IN RATS

  • The selective toxicity of lead was tested in central catecholaminergic nervous system of postnatally lead exposed rats. Three groups of animals were prepared; 1) rats exposed to low dose of lead (0.05%PbAc); 2) rats exposed to high dose of lead(0.2%PbAc); 3) age-matched normal control rats. At2, 4, 6 and 8 weeks of age brain and body weight gain, and lead concentrations in brain tissues were measured. At the same ages tyrosine hydroxylase and Na-K ATPase activities were measured in the 4 brain areas of each animal. Body weight gain was decreased after 6 weeks of age in rats exposed to high dose of lead. Concentrations of lead in whole brain tissues were increased from 0.37 to 0.83 (ng/mg wet tissue) in these animals. in lead exposed rats, tyrosine hydroxylase activities were higher but Na-K ATPase activities were lower than those of age-matched control animals. Brain areas where tyrosine hydroxylase activities were detected without concomitant changes of Na-K ATPase activities were pons-medulla (2 weeks of age) and telencephalon (6 weeks of age) in rats exposed to low dose of lead, and those in rats exposed to high dose of lead were midbrain (4 and 6 weeks of age). These data indicate that catecholaminergic nervous system in the brain areas described above could selectively be affected by lead.

KI-60606의 일반약리작용시험 (General Pharmacology of KI-60606)

  • In this study general pharmacological profiles of KI-60606 on the central nervous system, the cardiovascular system and the other organs were investigated. The dosages given were 0,5, 10 and 25 mg/kg and drugs were administered intravenously. The animals used for this study were mice, rats, cats and guinea pigs. KI-60606 showed no effects on general behavior, motor coordination, spontaneous locomotor activity, hexobarbital-induced hypnosis time, body temperature, analgesic activity, anticonvulsant activity and contraction of nictitating membrane in cats. Furthermore KI-60606 showed no effects on blood pressure, heart rate, LVP (left ventricular peak systolic pressure), LVEDP (left ventricular end diastolic pressure), LVDP (left ventricular developing pressure), DP(double product), CFR(coronary flow rate), smooth muscle contraction using guinea pig ileum and gastric secretion at all dosage tested except the increase of gastrointestinal transport and urinary $K^+$ excretion.

  • To investigate the experimental effects of Chunwangboshimdan which have been widely used for aterial or auricular flutter, neurosis, insomnia and disease caused by cardiac malfunction, we have done the effects on the central nervous and cardiovascular system. The result as follow; It inhibited the convulsion induced by caffeine, the writhing syndrome induced by acetic acid and the spontaneous movement. Furthermore it prolonged the sleeping time induced by thiopental-Na in mice. It also showed the negative inotropic action on the isolated heart of frog, the vasodilative action as to peripheral bloods of rabbit ear and the hypotensive action in the anesthetized rabbit.

  • In trauma patients, cognitive impairment may develop due to several causes: traumatic brain injury such as intracranial hemorrhage, diffuse axonal injury, hypoxic brain injury or reperfusion injury, the psychologic disorder, such as acute stress disorder, post-traumatic disorder or delirium. We describe a 62-year-old male with post-trauma cognitive impairment due to a primary central nervous system lymphoma.

  • Neuropathic orofacial pain (NOP) is initiated or caused by a priomary lesion or dysfunction in the peripheral nervous system or the central nervous system. NOP is divided into paroxysmal and continuous by duration of pain. NOP is important for dentists, because they can occur after dental procedures, such as extractions, endodontic treatment, and dental implant insertion. Dentists should have concern about NOP, because it is difficult to diagnose and treat.

  • The cutaneous network of unmyelinated nerve fibers is extremely dense, and closely interacts with the many cell types present in dermis and epidermis, including keratinocytes, fibroblasts, Langerhans cells, and melanocytes. Cell communication involves various neuroendocrine factors, with cell differentiating and proliferative activities, or inflammatory properties. Thus, nervous cells in the skin not only create a sensory system connected to the central nervous system, but also mediate many of the biological activities of the skin.(omitted)

  • Breathing is controlled by complex interactions between the central and peripheral nervous systems in conjunction with the respiratory system. Neurological diseases predispose patients to nocturnal desaturation and pneumonia due to respiratory dysfunction, which increases mortality, daytime sleepiness and fatigue, and reduces the quality of life. Respiratory function tests are required to identify respiratory function decline and to consider compensatory management. This review summarizes the characteristics of several respiratory function tests and their applications to neurological diseases.

General Pharmacology of Artesunate, a Commonly used Antimalarial Drug: Effects on Central Nervous, Cardiovascular, and Respiratory System

  • Artesunate, a semi-synthetic derivative of artemisinin, is used primarily as a treatment for malaria. Its effects on the central nervous system, general behavior, and cardiovascular, respiratory, and other organ systems were studied using mice, rats, guinea pigs, and dogs. Artesunate was administered orally to mice at doses of 125, 250, and 500 mg/kg and to rats and guinea pigs at 100, 200, and 400 mg/kg. In dogs, test drugs were administered orally in gelatin capsules at doses of 50, 100, and 150 mg/kg. Artesunate induced insignificant changes in general pharmacological studies, including general behavior, motor coordination, body temperature, analgesia, convulsion modulation, blood pressure, heart rate (HR), and electrocardiogram (ECG) in dogs in vivo; respiration in guinea pigs; and gut motility or direct effects on isolated guinea pig ileum, contractile responses, and renal function. On the other hand, artesunate decreased the HR and coronary flow rate (CFR) in the rat in vitro; however, the extent of the changes was small and they were not confirmed in in vivo studies in the dog. Artesunate increased hexobarbital-induced sleeping time in a dose-related manner. Artesunate induced dose-related decreases in the volume of gastric secretions and the total acidity of gastric contents, and induced increases in pH at a dose of 400 mg/kg. However, all of these changes were observed at doses much greater than clinical therapeutic doses (2.4 mg/kg in humans, when used as an anti-malarial). Thus, it can be concluded that artesunate is safe at clinical therapeutic doses.

  • In the article were observed the epidemiological aspects of malignant tumors of the central nervous system (MT CNS) in Kazakhstan in a retrospective study for the years 2004-2011. The material of the study was consolidated accounting data of oncology centers on patients with MT CNS (C70-72) with first time established diagnosis. Calculated were crude, age, standardized (world standard), aligned and predicted incidence of MT CNS among both male and female populations. It was found that over the studied period, there were 4,604 cases of MT CNS. The average annual crude incidence rate of MT CNS in total population was $3.7{\pm}0.1^0/_{0000}$. Trends in aligned incidence rates in the whole country had a tendency to increase (T=+0.9%). Defined levels of morbidity MT CNS in the whole population in different regions of Kazakhstan: low up to $2.87^0/_{0000}$, the average from 2.87 to $4.45^0/_{0000}$ and high from $4.45^0/_{0000}$ and above on the basis of which was given the space-time estimate. Age and sex differences in MT CNS incidence were also clearly established.

  • General pharmacological effects of SB-31$^{R}$, the extracts of Pulsatilla koreana, were investigated in mice, rats and guinea-pigs. Intravenous injection of SB-31 (3 and 6 ml/kg) produced almost no effect on central nervous system no effects on the general symptom and behaviors of mice, spontaneous locomotor activity, pentobarbital- induced sleeping time , rotared performance , electroshock and pentylenetertrazole -induced seizures, acetic acid-induced writhing and normal body temperature in mice. SB-31 showed little effects on the spontaneous movement of the isolated ileum and contraction induced by agonists in isolated ileum, suggesting no influence on autonomic nervous system. Administration of SB-31 also did not show any effect on blood pressure in conscious rats. However, a slight decrease in heart rate was observed at high doses (6 and 10 ml/kg) of SB-31 in conscious rats. Similarly, a slight increase in respiratory rate was observed at 6 m1/kg of SB-31 in anesthetized rats. SB-31 did not produce any effect at the dose of 3 ml/kg, but showed a tendency to increase the urinary volume at 6 ml/kg, and produced a decrease in urinary excretions of N $a_{+}$and $K_{+}$at 6 ml/kg. However, transport capacity within the gastrointestinal tract and the secretion of the gastric juice were not influenced by 6 ml/kg of SB-31. In conclusion, these results suggest that SB-31 did not pro-duce any acute effects on the central nervous system, autonomic nervous system, respiratory and circulatory systems, digestive system and kidney function at the dose of below 3 ml/kg.ml/kg.
