  • As Korean ginseng is hybrid, an individual variation is very severe, and it takes long times in new breeding because it is required 4 years to pick the seed. But, transformation technique makes the high-functional breeding in short time. The focus of these ginseng studies is to find and secure the useful gene. And it is urgent to accumulate the fundamental data for the molecular breeding and secure the useful genes. Therefore, transformation and soil acclimatization technique are necessary to molecular breeding in use of the introduction of functional genes. In this study, it add to secure of new regulation gene and useful gene as to accumulate the fundamental data for the place where it will contribute to raise the national competitive power. To analyze the useful genes in large scale, we constructed CDNA libraries with various tissues, species, and treated tissue. EST analysis of ginseng perform in large scale and build the EST database of ginseng. We perform the full length sequencing about the selected lots of clones that include the entire open reading frame of the amino acid residues and construct cDNA chip with the parental EST clones. Establishment of the transformation and a soil acclimatization system throuth the re-introduction of the selected ginseng gene that related with the secondary metabolism and anti-stress into the ginseng.

Low-Temperature Microencapsulation of Sesame Oil Using Fluidized Bed Granulation (Fluidized bed granulation을 이용한 참기름의 저온 미세캡슐화)

  • Top spray-drying method is frequently utilized for flavor encapsulation, but the top spray-dried products frequently suffer from high losses of volatile flavor as the result of a high processing temperature (150-$300^{\circ}C$). In an effort to solve these problems, a low-temperature fluidized-bed granulating method was utilized to encapsulate the flavor. For the encapsulation of sesame oil, oil-in-water emulsions of sesame oil and a mixture of maltodextrin, modified starch, gum arabic, and gellan gum were bottom-sprayed at milder temperatures (70-$100^{\circ}C$) using a fluidized-bed granulator. Sesame oil extracts from microcapsules were obtained via a simultaneous distillation/extraction technique, and the retention of volatile flavor compounds was analyzed via a gas chromatography-mass spectrometry. The retention of volatile flavors of sesame oil per se, spray-dried and fluidized-bed granulated microcapsules after 3-day-storage at $37^{\circ}C$ were 0.8%, 37.2%, and 42.0%, respectively. In addition, the low-temperature fluidized-bed granulation showed higher encapsulation yield and sensory preferences for the application of commercial products (beef rice porridge), as compared to spray drying.

Acceptance of Fashion Forecast as Reflected in the Street Fashion in Korea (스트리트패션에 나타난 한국 소비자들의 패션예측 수용)

  • Forecasting is a critical task for fashion companies because of continuous change in fashion and long process lead-time. Therefore, it is of great importance for both scholars and companies to understand how forecasted fashion styles are accepted by consumers. This research aimed to investigate consumer acceptance of fashion styles in Korea. The study examined and compared oversea collections of women's wear to the street fashion in Korea for seven seasons from 02 s/s to 05 s/s. Information on oversea collections were obtained from the magazine, Fashion Show, and the street fashion information from Seoul Fashion Design Center. The results showed that overall trends presented in oversea collections have been well accepted, while acceptance of specific styles or items varied. During the period of this research, sporty style and feminine style were very strong in the street fashion. Many styles and items were modified and selectively accepted probably because of cultural differences and limitations of mass production. Some styles which were presented in oversea collections were not accepted in Korea, and at the same time some cases were observed only in the street fashion in Korea. The results of this study provide guidelines for Korean apparel companies in merchandise planning and empirical findings to deepen the understanding on Korean society with respect to fashion.

An Investigation on the Long Term Durability of High-strength Shotcrete Using Field and Combined Deterioration Test (현장실험과 복합열화시험을 통한 고강도 숏크리트의 장기내구성 검토)

  • Domestic practices in shotcrete use have developed in many respects even now, but it still has issues about material, construction, quality standard and so on. In overseas, the construction using high strength shotcrete with $39.2{\sim}58.8 MPa$ of compressive strength is becoming common based on the shotcrete technology of high strength and durability. However, domestic shotcrete design strength is low at around 20.6 MPa of compressive strength and a long term durability is also insufficient. In this paper, field tests using high-quality additives and accelerators were performed to obtain the improvement of shotcrete strength and EFNARC standard was used to evaluate the field test results. In addition, deterioration test combined with the freezing-thawing and carbonation was also performed in order to investigate a long-term durability of high-strength shotcrete. As a result of the field test, the promotion ratio of early strength was $90{\sim}97%$ in case of using alkali-free accelerators. And the compressive strength of the shotcrete using Micro-silica fume was $45.2{\sim}55.8MPa$ and flexible strength was $5.01{\sim}6.66MPa$, so the promotion ratio of strength was $37{\sim}79%$ and $17{\sim}61%$ respectively. The promotion effect of strength by silica fine additives ratio of $7.5{\sim}10%$ for cement mass was much superior to the other cases. It was especially examined that using Micro-silica fume reduced deterioration due to mixed steel fiber and improved a long-term durability of shotcrete.

Determination of Color Value (L, a, b) in Green Tea Using Near-Infrared Reflectance Spectroscopy (근적외 분광분석법을 이용한 녹차의 색도 분석)

  • Near infrared spectroscopy (NIRS) is a rapid and accurate analytical method for determining the composition of agricultural products and feeds. The applicability of near infrared reflectance spectroscopic method was tested to determine the color value (L, a, b) of green tea. A total of 162 green tea calibration samples and 82 validation samples were used for NIRS equation development and validation, respectively. In the developed NIRS equation for analysis of the color value (L, a, b), the most accurate equation for L value was obtained at 2, 8, 6, 1 (2nd derivative, 8 nm gap, 6 points smoothing, and 1pointsecond smoothing), and for a, and b value were obtained at 1, 4, 4, 1 (1st derivative, 4 nm gap, 4points smoothing, and 1 point second smoothing) math treatment condition with SNVD (Standard Normal Variate and Detrend) scatter correction method and entire spectrum ($400{\sim}2,500\;nm$) by using MPLS (Modified Partial Least Squares) regression. Validation results of these NIRS equations showed very low bias (L: 0.005%, a: 0.003%, b: -0.013%) and standard error of prediction (SEP, L: 0.361%, a: 0.141%, b: 0.306%) as well as high coefficient of determination ($R^2$, L: 0.905, a: 0.986, b: 0.931). Therefore, these NIRS equations can be applicable and reliable for determination of color value (L, a, b) of green tea, and NIRS method could be used as a mass screening technique for breeding programs and quality control in the green tea industry.

The association between measurement sites of visceral adipose tissue and cardiovascular risk factors after caloric restriction in obese Korean women

  • Quantities as well as distributions of adipose tissue (AT) are significantly related to cardiovascular disease (CVD) risk factors and can be altered with caloric restriction. This study investigated which cross-sectional slice location of AT is most strongly correlated with changes in CVD risk factors after caloric restriction in obese Korean women. Thirty-three obese pre-menopausal Korean women ($32.4{\pm}8.5$ yrs, BMI $27.1{\pm}2.3\;kg/m^2$) participated in a 12 weeks caloric restriction program. Subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) were measured using computed tomography (CT) scans at the sites of L2-L3, L3-L4, and L4-L5. Fasting serum levels of glucose, insulin, triglyceride, total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), leptin and homeostasis model assessment-insulin resistance (HOMA-IR) were observed. Pearson's partial correlation coefficients were used to assess the relationship between AT measurement sites and changes in CVD risk factors after calorie restriction. When calories were reduced by 350 kcal/day for 12 weeks, body weight (-2.7%), body fat mass (-8.2%), and waist circumference (-5.8%) all decreased (P < 0.05). In addition, following caloric restriction, serum levels of glucose (-4.6%), TC (-6.2%), LDL-C (-5.3%), leptin (-17.6%) and HOMA-IR (-18.2%) decreased significantly (P < 0.05) as well. Changes in VAT at the level of L3-L4 were significantly greater than those at other abdominal sites, and these changes were correlated with changes in TC (P < 0.05), LDL-C (P < 0.001), SBP (P < 0.001) and HOMA-IR (P < 0.01). These results show that VAT at L3-L4 had a stronger correlation with CVD risk factors than with other AT measurement sites after caloric restriction.

Effect of Whole Body Vibration Training in Drop Landing Jump (전신진동운동이 드랍랜딩점프 동작에 미치는 영향)

  • Objective: This study was conducted to investigate the effects of WBVE on biomechanical factor analysis of drop landing jumps before and after a four week training program. Methods: Participants were divided into two group: VEG (n=5, age: $25.7{\pm}2.3yrs$, height: $170{\pm}7.6cm$, weight: $69.3{\pm}8.3kg$) and OEG (n=5, age: $24.6{\pm}3.4yrs$, height: $164{\pm}4.9cm$, weight: $58.8{\pm}9.2kg$). Ten infrared cameras (Vicon, UK) with a sampling rate of 100 Hz were used in two GRF measurement systems (AMTI, USA). Results: The variability of the center of mass was largest at the falling phase, and there was a great risk of injury when landing after the jump. Second, the jumping heights of the VEG (vibration exercise group) were higher than those of the OEG (only exercise group), regardless of training period. Third, there were significant differences in the hip joint P1 (flexion) and knee joint P2 (extension) between the exercise groups after 4 weeks of training. However, there were no significant differences among training periods or phases. Conclusion: regardless of training periods, the VEG showed increased jump height compared with the OEG, but the consistency between the jump height and the lower joint power could not be determined.

Conversion Characteristics of Liquid Fuels from Sawdust by Acetone-Solvolysis (아세톤-용매분해반응에 의한 톱밥으로부터 액체 연료물질의 전환 특성 연구)

  • Sawdust, produced as an wood by-product, is usable biomass as liquid fuels if decomposed to monomer unit, because the chemical structure are similar to high octane materials found in gasoline. In this study, parameters of thermochemical degradation by acetone-solvolysis reaction of sawdust such as the effect of reaction temperature, reaction time and type of solvent on conversion yield and degradation products were investigated. The liquid products by acetone-solvolysis from sawdust produced various kind of ketone, phenol and furan compounds. The optimum sawdust conversion was observed to be 88.7% at $350^{\circ}C$, 40min. Combustion heating value of liquid products from thermochemical conversion processes was as high as 7,824 cal/g. The energy yield and mass yield in acetone-solvolysis of sawdust was 60.8% and 36.4 g-oil/100g-sawdust after 40 min of reaction at $350^{\circ}C$, respectively. The major components of the acetone-solvolysis products, that could be used as liquid fuel, were 4-methyl-3-pentene-2-one, 1,3,5-trimethylbezene, 2,6-dimethyl-2,5-heptadiene-4-one, 3-methyl-2-cyclopenten-1-one as ketone compounds.

Influence of Surfactants on Enhancing Transport of Bacteria in Geological Materials (지질매질체내에서 계면활성제가 박테리아 이동 증진에 미치는 영향)

  • This study investigated the effect of surfactants (nonionic surfactant (Tween 20), biosurfactant) on enhancing transport of bacteria (Bacillus subtilis ATCC 6633) in geological materials. Column experiments were performed under various surfactant conditions with columns packed with quartz sand (particle size distribution: 0.5~2.0 mm, mean diameter: 1.0 mm). Bacterial mass recovery, sticking efficiency, and other parameters were quantified from breakthrough curves. Results indicate that bacterial attachment to sand surfaces increased considerably in the presence of mineral salt medium (MSM), especially at the inlet, which was due to the increase of ionic strength by MSM. It was observed that bacterial transport in sand columns was enhanced in the presence of surfactant. Results also show that simultaneous injection of both surfactant and MSM or pre-injection of surfactant was more effective in bacterial transport enhancement than after-injection of surfactant. This study suggests that transport of bacteria in geological materials could be influenced by surfactants and their injection methods.

TA Study on Maximum Oxygen Uptake according to Body Measurement and Vascular Compliance (신체계측치 및 혈관탄성도에 따른 최대산소섭취량에 관한 연구)

  • This study was conducted to investigate the relationship between maximum oxygen uptake and its related factors including body measurement and vascular compliance. The subjects of our study were 43 males and 53 females aged 40 to 59 who were visited in military health center. The main results were that 1.the degree of obesity had significantly negative correlation to vascular compliance in male, and waist-hip ratio in female. 2. The degree of obesity, body mass index, waist-hip ratio and body fat percent had significantly negative correlation to maximum oxygen uptake in male, and waist-hip ratio & body fat percent in female. 3. Maximum heart rate had significantly positive correlation to maximum oxygen uptake in male, and vascular compliance & maximum heart rate in female. 4. In multiple regression analysis which maximum oxygen uptake was as dependent variable, body fat percent, vascular compliance and maximum heart rate were significant variables for both male and female. In conclusion, Maximum oxygen uptake of some rural people age 40 to 59 were affected by various factors such as body fat percent, vascular compliance, and maximum heart rate.
