동위원소희석 질량분석법을 이용한 간장 중의 안식향산, 메틸파라벤, 부틸파라벤의 분석 (An analysis of benzoic acid, methyl paraben and butyl paraben in soy sauce using isotope dilution liquid chromatography mass spectrometry)

  • 안식향산, 메틸파라벤, 부틸파라벤은 식품, 의약품뿐 만 아니라 화장품 분야에서도 사용되는 보존료이다. 그러나 이들 보존료들의 여러 가지 독성이 보고되면서, 한국을 비롯하여 여러 나라들에서 식품 중의 이들 보존료의 사용을 규제하고 있다. 본 연구에서는 간장 중의 안식향산, 메틸파라벤, 부틸파라벤의 정확하고 정밀한 분석을 위하여 세가지 분석물질의 동위원소를 내부표준물질로 이용하는 동위원소 희석 액체크로마토그래피 질량분석법 (Isotope Dilution Liquid Chromatography Mass Spectrometry, ID-LC/MS)을 개발하였다. 메틸, 부틸 파라벤보다 pKa가 낮은 안식향산을 고려하여 acetic acid로 pH 4.0으로 조정한 5 mM ammonium acetate를 이동상 용매로 사용하여 C18 컬럼으로 분리하였다. 질량분석 조건으로는 전기분무이온화법으로 음이온을 생성하여 음이온 모드에서 분석하였으며, 방해물질로부터 선택성을 향상하기 위해 선택반응분석법 (Selected Reaction Monitoring)을 이용하여 분석하였다. 간장의 색깔과 간장 중의 여러 가지 방해물질들을 제거하기 위하여 C18 카트리지를 이용하여 정제하였다. 최적화된 조건과 방법을 이용하여 싱가포르 Health Science Authority(HSA)가 제공하는 간장 기준시료를 분석하였다. 본 연구원에서 측정한 결과값은 HSA가 제공하는 기준값과 불확도 내에서 일치하였다.

남해안 통영지역 가두리양식장 해수-퇴적물 경계면에서의 chemical fluxes (Chemical Fluxes at the Sediment-Water Interface Below Marine Fish Cages on the Coastal Waters off Tong-Young, South Coast of Korea)

  • 1995년 6월, 남해안 통영지역 가두리양식장 해수-퇴적물 경계면에서 입자상유기물의 수직유입량과 용존산소의 소모량, 영양염류의 용출량을 관측하였다. 입자상유기물의 수직유입량은 저층고정식 sediment trap을 이용하였으며, 용존산소의 소모량과 영양염류의 용출량은 benthic chamber method로 측정하였다. 가두리양식이 연안부영양화에 미치는 영향과 가두리에서 유출된 입자상유기물의 확산강도를 정량하기 위해, 가두리 아래(수심 약 18 m, Cage Site)와 가두리에서 수평으로 약 100 m 가량 떨어진 곳(수심 약 32 m, Control Site)의 해수-퇴적물 경계면에서, 암모니아와 인산염, 규산염의 용출량을 비교하고, 탄소와 질소와 인의 mass balances를 추정하였다. 관측결과, 가두리정점(Cage Site)으로는 6400 mg C $m^{-2}d^{-1}$의 입자상유기물이 유입되었고, 동시에 230 mmol $O_2\;m^{-2}d^{-1}$ 이상의 용존산소가 소모되었다. 따라서 탄소의 경우, 가두리 아래 해저면으로 공급되는 유기 입자의 약 40%에 달하는 양이 해수-퇴적물 경계면에서 분해되며 (ca. 2400 mg C $m^{-2}d^{-1}$), 나머지 약 60%는 퇴적되어 매몰되는 것으로 보인다. (ca. 4000 mg C $m^{-2}d^{-1}$) 그러나 대비정점(Control Site)에서는 가두리정점에 비해 상대적으로 적은 양의 유기물유입과(ca. 4000 mg C $m^{-2}d^{-1}$), 낮은 용존산소소모율이 관측되었다(75 mmol $O_2\;m^{-2}d^{-1}$). 관측결과는 가두리에서 투기되는 대부분의 입자상유기물이 가두리 아래 해저면에 집중적으로 퇴적되고 있음을 보여주며, 가두리 부근 해저면으로 확산되는 입자상유기물의 양은 가두리에서 멀어질수록 급속히 감소함을 시사한다.

Influence of Ammonia Solvation on the Structural Stability of Ethylene Cluster Ions

  • The stable structures of pure ethylene and mixed ethylene-ammonia cluster ions are studied using an electron impact ionization time-of-flight mass spectrometer. Investigations on the relative cluster ion distributions of $(C_2H_4)_n(NH_3)_m^+$ under various experimental conditions suggest that $(C_2H_4)_2(NH_3)_3^+$ and $(C_2H_4)_3(NH_3)_2^+$ ions have the enhanced structural stabilities, which give insight into the feasible structure of solvated ions. For the stable configurations of these ionic species, we report an experimental evidence that both $(C_2H4)_2^+(C_2H_4)_3^+$ clusters as the central cations provide three and two hydrogen-bonding sites, respectively, for the surrounding $NH_3$ molecules. This interpretation is based on the structural stability for ethylene clusters and the intracluster ion-molecular rearrangement of the complex ion under the presence of ammonia solvent molecules.

헤더-채널 분기관에서의 헤더 입구 형상이 2상 유동 분배에 미치는 영향에 대한 실험적 연구 (Effect of Inlet Geometries on the Two-Phase Flow Distribution at Header-Channel Junction)

  • The main objective of this work is to experimentally investigate the effect of inlet geometries on the distribution of two-phase annular flow at header-channel junctions simulating the corresponding parts of compact heat exchangers. The cross-section of the header and the channels were fixed to $16mm{\times}16mm$ and $12mm{\times}1.8mm$, respectively. Experiments were performed for the mass flux and the mass quality ranges of $30{\sim}140kg/m^2s$ and 0.3~0.7, respectively. Air and water were used as the test fluids. Three different inlet geometries of the header were tested:no restriction (case A), a single 8 mm hole at the center (case B), and nine 2 mm holes around the center (case C) at the inlet, respectively. The tendencies of the two-phase flow distribution were different, in each case. For cases B and C (flow resistance exists), more uniform flow distribution results were seen, compared with case A(no flow resistance), due to the flow pattern change to mist flow from annular flow at the inlet, and the flow recirculation near the end plate of the header.

A pentacyclic triterpenoid possessing analgesic and anti-inflammatory activities from the fruits of Dregea volubilis

  • In present study evaluate the analgesic and anti-inflammatory activity of the compound obtained from the petroleum ether (40 - 60$^{\circ}C$) extract of the fruits from Dregea volubilis in Swiss albino mice and in Wister albino rats respectively. Dried and crushed fruits of Dregea volubilis were extracted by petroleum ether (40 - 60$^{\circ}C$), the proper solvent system was developed by TLC and subjected to column chromatography for obtaining the pure compound/s. IR, MASS, NMR (PMR, C13 NMR and DEPT) spectroscopic analysis were done to elucidate the structure of the compound/s. The petroleum ether (40 - 60$^{\circ}C$) extract of the fruits of Dregea volubilis led to isolation of a pentacyclic triterpenoid designated as taraxerone and characterized as D- friedoolean- 14- en, 3 one. Taraxerone had been screened for analgesic activity in Swiss albino mice and anti-inflammatory activity in Wister albino rats at the dose of 5 mg/kg body weight orally and exhibit significant analgesic and anti-inflammatory properties.

EFFECT OF INTAKE PORT GEOMETRY ON THE IN-CYLINDER FLOW CHARACTERISTICS IN A HIGH SPEED D.I. DIESEL ENGINE

  • Recently, the HSDI (High Speed Direct Injection) diesel engine has been spotlighted as a next generation engine because it has a good potential for high thermal efficiency and fuel economy. This study was carried out to investigate the in-cylinder flow characteristics generated in a HSDI diesel engine with a 4-valve type cylinder head. The four kinds of cylinder head were manufactured to elucidate the effect of intake port geometry on the in-cylinder flow characteristics. The steady flow characteristics such as coefficient of flow rate $(C_{f})$, swirl ratio (Rs), and mass flow rate (m,) were measured by the steady flow test rig and the unsteady flow velocity within a cylinder was measured by PIV. In addition, the in-cylinder flow patterns were visualized by the visualization experiment and these results were compared with simulation results calculated by the commercial CFD code. The steady flow test results indicated that the mass flow rate of the cylinder head with a short distance between the two intake ports is $13\%$ more than that of the other head. However, the non-dimensional swirl ratio is decreased by approximately $15\%$. As a result of in-cylinder flow characteristics obtained by PIV and CFD calculation, we found that the swirl center was eccentric from the cylinder center and the position of swirl center was changed with crank angle. As the piston moves to near the TDC, the swirl center corresponded to the cylinder center and the velocity distribution became uniform. In addition, the results of the calculation are in good agreement with the experimental results.

Purification and Characterization of Heat-Tolerant Protease Produced by Bacillus polyfermenticus SCD

  • A protease produced by Bacillus polyfermenticus SCD was purified and characterized as a new detergent material. The protease was purified from supernatant produced by B. polyfermenticus SCD, by ammonium sulfate precipitation, ion-exchange chromatography on a DEAE-Sephadex A-50, and finally gel filtration chromatography on Sephadex G-50. The molecular mass of this enzyme was 44 kDa based on SDS-PAGE. The optimum temperature and pH were $50^{\circ}C$ and pH 8.0. The ranges of its stability to the pH and temperature were 7.0 to 9.0 and under $40^{\circ}C$, respectively. The enzyme was highly stable in the presence of the surfactants like Triton X-100 (0.1%), showing a 2-fold increase in its proteolytic activity. However, the enzyme was slightly inhibited by the chelating agent EDTA (1 mM). The enzyme has a maximum activity at $50^{\circ}C$ and the activity can be increased by surfactants such as Triton X-100 and Tween 80.

LC-ESI-MS/MS를 이용한 용담사간탕의 주요 성분 분석 (Quantitative Analysis of the Marker Constituents in Yongdamsagan-Tang using Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry)

  • Yongdamsagan-tang has been used to treat the urinary disorders, acute- and chronic-urethritis, and cystitis in Korea. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS/MS) method was established for simultaneous analysis of the 20 bioactive marker compounds, geniposidic acid, chlorogenic acid, geniposide, liquiritin apioside, acteoside, calceolarioside B, liquiritin, nodakenin, baicalin, liquiritigenin, wogonoside, baicalein, glycyrrhizin, wogonin, glycyrrhizin, wogonin, saikosaponin A, decursin, decursinol angelate, alisol B, alisol B acetate, and pachymic acid in traditional herbal formula, Yongdamsagan-tang. Chromatographic separations of all marker compounds were conducted using a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) at $45^{\circ}C$ using a mobile phase of 0.1% (v/v) formic acid in water and acetonitrile with gradient elution. The MS analysis was performed using a Waters ACQUITY TQD LC-MS/MS coupled with an electrospray ionization source in the positive and negative modes. The flow rate was 0.3 mL/min and injection volume was $2.0{\mu}L$. The correlation coefficient of 20 marker compounds in the test ranges was 0.9943-1.0000. The limits of detection and quantification values of the all marker components were 0.11-6.66 and 0.34-19.99 ng/mL, respectively. As a result of the analysis using the optimized LC-ESI-MS/MS method, three compounds, geniposidic acid (from Plantaginis Semen), alisol B (from Alismatis Rhizoma), and pachymic acid (from Poria Sclerotium), were not detected in this sample. While the amounts of the 17 compounds except for the geniposidic acid, alisol B, and pachymic acid were $0.04-548.13{\mu}g/g$ in Yongdamsagan-tang sample. Among these compounds, baicalin, bioactive marker compound of Scutellariae Radix, was detected at the highest amount as a $548.13{\mu}g/g$.

  • In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS/MS) method was established for simultaneous determination of the 25 marker components, including chlorogenic acid, gallic acid, oxypaeoniflorin, homogentisic acid, methyl gallate, caffeic acid, 3,4-dihydroxybenzaldehyde, paeoniflorin, albiflorin, liquiritin, nodakenin, ferulic acid, ginsenoside Rg1, liquiritigenin, coumarin, cinnamic acid, benzoylpaeoniflorin, ginsenoside Rb1, cinnamaldehyde, paeonol, glycyrrhizin, 6-gingerol, evodiamine, rutecarpine, and spicatoside A in traditional Korean formula, Onkyung-tang. All analytes were separated on a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) at $45^{\circ}C$ using a mobile phase of 0.1% (v/v) formic acid in water and acetonitrile with gradient elution. The MS analysis was carried out using a Waters ACQUITY TQD LC-MS/MS coupled with an electrospray ionization (ESI) source in the positive and negative modes. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}L$, respectively. The correlation coefficient of all analytes in the test ranges was greater than 0.98. The limits of detection and quantification values of the 25 marker compounds were in the ranges 0.03-19.43 and 0.09-58.29 ng/mL, respectively. As a result, methyl gallate, 3,4-dihydroxybenzaldehyde, evodiamine, and rutecarpine were not detected in this sample and the concentrations of the 21 compounds except for the above 4 compounds were $33.09-3,496.32{\mu}g/g$ in Onkyung-tang decoction. Among these compounds, paeonol was detected at the highest amount as a $3,496.32{\mu}g/g$.

THEORETICAL STUDY ON OBSERVED COLOR-MAGNITUDE DIAGRAMS

  • From $B\ddot{o}hm$-Vitense's atmospheric model calculations, the relations, [$T_e$, (B-V)] and [B.C, (B-V)] with respect to heavy element abundance were obtained. Using these relations and evolutionary model calculations of Rood, and Sweigart and Gross, analytic expressions for some physical parameters relating to the C-M diagrams of globular clusters were derived, and they were applied to 21 globular clusters with observed transition periods of RR Lyrae variables. More than 20 different parameters were examined for each globular cluster. The derived ranges of some basic parameters are as follows; $Y=0.21{\sim}0.33,\;Z=1.5{\times}10^{-4}{\sim}4.5{\times}10^{-3},\;age,\;t=9.5{\sim}19{\times}10^9$ years, mass for red giants, $m_{RG}=0.74m_{\odot}{\sim}0.91m_{\odot}$, mass for RR Lyrae stars, $m_{RR}=0.59m_{\odot}{\sim}0.75m_{\odot}$, the visual magnitude difference between the turnoff point and the horizontal branch (HB), ${\Delta}V_{to}=3.1{\sim}3.4(<{\Delta}V_{to}>=3.32)$, the color of the blue edge of RR Lyrae gap, $(B-V)_{BE}=0.17{\sim}0.21=(<(B-V)_{BE}>=0.18),\;[\frac{m}{L}]_{RR}=-1.7{\sim}-1.9$, mass difference of $m_{RR}$ relative to $m_{RG},(m_{RG}-m_{RR})/m_{RG}=0.0{\sim}0.39$. It was found that the ranges of derived parameters agree reasonably well with the observed ones and those estimated by others. Some important results obtained herein can be summarized as follows; (i) There are considerable variations in the initial helium abundance and in age of globular clusters. (ii) The radial gradient of heavy element abundance does exist for globular clusters as shown by Janes for field stars and open clusters. (iii) The helium abundance seems to have been increased with age by massive star evolution after a considerable amount (Y>0.2) of helium had been attained by the Big-Bang nucleosynthesis, but there is not seen a radial gradient of helium abundance. (iv) A considerable amount of heavy elements ($Z{\sim}10{-3}$) might have been formed in the inner halo ($r_{GC}$<10 kpc) from the earliest galactic co1lapse, and then the heavy element abundance has been slowly enriched towards the galactic center and disk, establishing the radial gradient of heavy element abundance. (v) The final galactic disk formation might have taken much longer by about a half of the galactic age than the halo formation, supporting a slow, inhomogeneous co1lapse model of Larson. (vi) Of the three principal parameters controlling the morphology of C-M diagrams, it was found that the first parameter is heavy clement abundance, the second age and the third helium abundance. (vii) The globular clusters can be divided into three different groups, AI, BI and CII according to Z, Y an d age as well as Dickens' HB types. BI group clusters of HB types 4 and 5 like M 3 and NGC 7006 are the oldest and have the lowest helium abundance of the three groups. And also they appear in the inner halo. On the other hand, the youngest AI clusters have the highest Z and Y, and appear in the innermost halo region and in the disk. (viii) From the result of the clean separations of the clusters into three groups, a three dimensional classification with three parameters, Z, Y and age is prsented. (ix) The anomalous C-M diagrams can be expalined in terms of the three principal parameters. That is, the anomaly of NGC 362 and NGC 7006 is accounted for by the smaller age of the order of $1{\sim}2{\times}10^9$ years rather than by the helium abundance difference, compared with M 3. (x) The difference in two Oosterhoff types I and II can be explained in terms of the mean mass difference of RR Lyrae variables rather than in terms of the helium abundance difference as suggested by Stobie. The mean mass of the variables in Oosterhoff type I clusters is smaller by $0.074m_{\odot}$ which is exactly consistent with Rood's estimate. Since it was found that the mean mass of RR Lyrae stars increases with decreasing Z, the two Oosterhoff types can be explained substantially by the metal abundance difference; the type II has Z<$3.4{\times}10^{-4}$, and the type I has higher Z than the type II.
