Detection of Cymbidium Mosaic Virus and Odontoglossum Ringspot Virus in Seed-Derived Plantlets of Phalaenopsis Imported by One-Step RT-PCR (One-Step RT-PCR 방법에 의한 수입 호접란묘의 심비디움 모자이크 바이러스와 오돈토글로섬 윤문 바이러스의 검정)

  • This experiment was carried out to detect the cymbidium mosaic virus (CymMV) and the odontoglossum ringspot virus (ORSV) in the seed-derived plantlets of Phalaenopsis imported from Taiwan by one-step reverse transcription-polymerase chain reaction (RT-PCR). Simple and rapid crude plant extracts for RT-PCR were prepared. The reverse transcription step was performed at $42^{\circ}C$ for 45 min and the following thermal cycling scheme was used for 36 reaction cycles: template predenaturation at $96^{\circ}C$ for 2 min, template denaturation at $96^{\circ}C$ for 30 s, primer annealing at $60^{\circ}C$ for 30 s, and DNA synthesis at $72^{\circ}C$ for 1 min. Of the 40 seed-derived plantlets of Phalaenopsis imported from Taiwan, all of them were infected with CymMV, but ORSV was not detected.

Outliers and Level Shift Detection of the Mean-sea Level, Extreme Highest and Lowest Tide Level Data (평균 해수면 및 최극조위 자료의 이상자료 및 기준고도 변화(Level Shift) 진단)

  • Modeling for outliers in time series was carried out using the MSL and extreme high, low tide levels (EHL, HLL) data set in the Busan and Mokpo stations. The time-series model is seasonal ARIMA model including the components of the AO (additive outliers) and LS (level shift). The optimal model was selected based on the AIC value and the model parameters were estimated using the 'tso' function (in 'tsoutliers' package of R). The main results by the model application, i.e.. outliers and level shift detections, are as follows. (1) The two AO are detected in the Busan monthly EHL data and the AO magnitudes were estimated to 65.5 cm (by typhoon MAEMI) and 29.5 cm (by typhoon SANBA), respectively. (2) The one level shift in 1983 is detected in Mokpo monthly MSL data, and the LS magnitude was estimated to 21.2 cm by the Youngsan River tidal estuary barrier construction. On the other hand, the RMS errors are computed about 1.95 cm (MSL), 5.11 cm (EHL), and 6.50 cm (ELL) in Busan station, and about 2.10 cm (MSL), 11.80 cm (EHL), and 9.14 cm (ELL) in Mokpo station, respectively.

Investigation of Water Leakage in Seosan A-Region Sea Wall using Integrated Analysis of Remote Sensing, Electrical Resistivity Survey, Electromagnetic Survey, and Borehole Survey (원격탐사, 전기탐사, 전자기탐사 및 시추공영상의 융합적 분석을 통한 서산지역 방조제 누수구역 판별)

  • This study introduces integrated approach on detection of a leakage in a sea wall based on remote sensing, electric resistivity survey, electromagnetic survey, and borehole survey for the Seosan A-Region sea wall. The satellite temperature distribution from Landsat ETM+ data identifies water leakage distribution and period by analyzing temperature mixing patterns between sea water and fresh water. Electric resistivity survey provides both horizontal and vertical anomaly distributions over the sea wall showing below average electric resistivity. Electromagnetic survey(electrical conductivity survey) reveals the potential possible leakage areas with minimal background impact by comparing electrical conductivity values between high and low tides. Borehole image processing system confirmed the locations of anomalies identified from the other survey methods and distributions of vertical fracture zones. The integrated approach identified 41.7% of the sea wall being the most probable area vulnerable to water leakage and effectively approximated both horizontal and vertical distribution of water leakage. The integrated analysis of remote sensing, electric resistivity survey, electromagnetic survey and borehole survey is considered to be an optimal method in identifying water leakage distribution, period, and extent of fractures knowledged from the boreholes.

Analytical method validation and monitoring of levulinic acid in soy sauce and their application to food (간장 및 이를 활용한 식품의 레불린산 분석방법 검증 및 모니터링)

  • This study presents a method validation for extraction and quantitative analysis of levulinic acid in soy sacues using high performance liquid chromatograph-photodiode array detector (HPLC-PDA). The levulinic acid in samples were extracted with distilled water, and then purified with C18 Sep-Pak cartridge. The calibration curves showed good linearity (R > 0.999) in a relatively wide concentration range ($2.5-400{\mu}g/mL$). Mean recoveries and relative standard deviation (RSD) of levulinic acid spiked in soy sauce samples at different spiking levels ($2.5-400{\mu}g/mL$; 6 point). Recoveries were 87.58-97.26 % with RSD less than 15 %, and limit of detection (LOD) and limit of quantification (LOQ) were 0.64 and $1.64{\mu}g/mL$, respectively. According to monitoring result with the established method, levulinic acid was found in 43 of 59 domestic commercial soy sauces, soy sauce based sauces and seasoned meats. The contamination levels were 0.44-1.23 mg/mL for soy sauces, 0.03-0.83 mg/mL for soy sauce based sauces and 8.43-38.94 mg/mL for seasoned meats. The results indicated to be rapidly and accurately qualifying levulinic acid and can be used as a suitable quality control method for soy sauce and soy sauce related commodities.

Online Content Editing System to Edit Broadcasting and Personal Contents (방송 및 개인 콘텐츠 편집을 위한 온라인 콘텐츠 편집 시스템)

  • In this paper, we propose a new online content editing system to edit the broadcasting and personal contents. The proposed editing system consists of the content management server and the content editor, in which the content management server is used for storing the contents, while the content editor is used for editing the contents via the user interface. Unlike the existing content editing methods which edit the downloaded contents, the proposed editing system edits the contents stored in the content management server while the content editor plays them by using the streaming technology. However, it is not effective to edit the whole segments of contents while playing them by using the streaming technology. To resolve the problem, the proposed editing system performs the scene detection of the contents, and the detected scene information is used for playing and editing of contents. After completing the content editing, the edited information is uploaded to the content management server as a metadata format. In the proposed editing system, both scene and edited information are represented only as the metadata format, and the physical content segmentation according to the scene and edited information is not performed. The implementation results show that the proposed editing system provides similar performances with the existing content editing methods which use the content download and editing methods.

Establishment of Pre-Harvest Residue Limit of Fungicides Pyrimethanil and Trifloxystrobin during Cultivation of Persimmon (단감 재배기간 중 살균제 pyrimethanil과 trifloxystrobin의 생산단계 잔류허용기준 설정)

  • BACKGROUND: This study was carried out to establish pre-harvest residue limit of fungicides pyrimethanil and trifloxystrobin in persimmon, based on dissipation and biological half-lives of two fungicides residue. METHODS AND RESULTS: Both pyrimethanil and trifloxystrobin were extracted with acetonitrile, clean-up with $NH_2$ SPE cartridge and residue were analyzed by HPLC/DAD. Limit of Detection was 0.01 mg/kg. Average recovery were $81{\pm}1.62%$, $98{\pm}1.58%$ of pyrimethanil, and $91{\pm}2.94%$, $98{\pm}1.25%$ of trifloxystrobin at fortification levels at 0.1 and 0.5 mg/kg, respectively. CONCLUSION: The biological half-lives of pyrimethanil were 15.6 and 11.6 days at sprayed with recommended and double dosage, respectively. The biological half-lives of trifloxystrobin were 10.4 and 10.3 days at sprayed with recommended and double dosage, respectively. The pre-harvest residue limit of pyrimethanil and trifloxystrobin were recommended as 2.69 and 0.83 mg/kg for 10 days before harvest, respectively.

A 3-D Vision Sensor Implementation on Multiple DSPs TMS320C31 (다중 TMS320C31 DSP를 사용한 3-D 비젼센서 Implementation)

  • High-speed 3D vision systems are essential for autonomous robot or vehicle control applications. In our study, a stereo vision process has been developed. It consists of three steps : extraction of edges in right and left images, matching corresponding edges and calculation of the 3D map. This process is implemented in a VME 150/40 Imaging Technology vision system. It is a modular system composed by a display, an acquisition, a four Mbytes image frame memory, and three computational cards. Programmable accelerator computational modules are running at 40 MHz and are based on TMS320C31 DSP with a $64{\times}32$ bit instruction cache and two $1024{\times}32$ bit internal RAMs. Each is equipped with 512 Kbytes static RAM, 4 Mbytes image memory, 1 Mbytes flash EEPROM and a serial port. Data transfers and communications between modules are provided by three 8 bit global video bus, and three local configurable pipeline 8 bit video bus. The VME bus is dedicated to system management. Tasks between DSPs are distributed as follows: two DSPs are used to edges detection, one for the right image and the other for the left one. The last processor computes the matching process and the 3D calculation. With $512{\times}512$ pixels images, this sensor generates dense 3D maps at a rate of about 1 Hz depending of the scene complexity. Results can surely be improved by using a special suited multiprocessors cards.

Muscle Tissue Distribution Level after Dipping Administration of Streptomycin in Olive Flounder (Paralichthys olivaceus), Rockfish (Sebastes schlegeli), and Red sea bream (Pagrus major) (Streptomycin의 약욕에 따른 양식 어류(넙치, 조피볼락, 참돔)의 근육조직내 잔류량의 변화)

  • The residue depletion of streptomycin was investigated in the olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major) after consecutive three days treatment with dipping water at a dose of 20 g/ton water. Fishes were sampled for muscle on 1st, 2nd, 3rd, 4th, and 5th day after treatment. Streptomycin concentrations were determined by high performance liquid chromatography with tandem mass spectrometry. The recovery rates of streptomycin in muscle samples ranged from 87.2 to 102.3% and from 80.4 to 94.1% for the concentration of 0.05 mg/kg and 0.1 mg/kg, respectively. Streptomycin concentrations detected on the 1st day after treatment were 0.066, 0.058, and 0.073 mg/kg in muscles of olive flounder, rockfish, and red sea bream, respectively. At day 2, residue concentrations of all samples were believed to decrease to lower than 0.05 mg/kg, the detection limit. From results of the present study, a withdrawal period of streptomycin is proposed on 3 days after consecutive three days treatment with dipping administration at a dose of 20 g/ton water to avoid the presence of excessive residues of the edible muscles of olive flounder, rockfish, and red sea bream. The present study showed that residue concentrations of streptomycin decreased to below 0.05 mg/kg after treatment 2nd day.

Factors that Affect on the Necessity of Establishment of Eco-friendly Hospital (친환경 병원 도입필요성에 영향을 미치는 요인)

  • The purpose of this research study is to analyze the influence on the necessity of establishment of eco-friendly hospital by evaluating hospital employees' awareness and their satisfaction level of working environment. Questionnaire investigation was conducted in April 2014 throughout five different medium-size hospitals in Seoul, Gyeonggi and Chungcheongbuk provinces. 213 sets of survey were used to conduct Frequency Analysis, T-test, Dispersion Analysis(ANOVA) and Structural Equation Modeling Analysis. The research concluded that employees with higher level of education, working position and longer term of working experience were more aware about Eco-friendly hospital. In addition, male, doctor, and employees with higher level of education and working position were more satisfied with their working environment. And male, employees with higher level of education and working position were more aware about necessity of establishment of eco-friendly hospital. According to the Structural Equation Modeling Analysis, the awareness of Eco-friendly hospital and the satisfaction level of working environment were the varying factors that influence the necessity of establishment of Eco-friendly hospital. Therefore, it is crucial for hospitals that are preparing for establishing Eco-friendly hospital to raise employees' level of understanding of Eco-friendly hospital and their working environment satisfaction level.

Optimization of the Bone-softening and Fishy Odor-reducing Processing of Mackerel Scomber japonicus Products using Response Surface Methodology (표면반응분석법을 활용한 뼈 연화 및 비린내 저감화 고등어(Scomber japonicus) 가공품의 가공공정 최적화)

  • This study optimized the bone-softening and fishy odor-reducing process for mackerel Scomber japonicus products using response surface methodology (RSM). The RSM showed that the optimum concentrations of doenjang and citric acid for reducing the fishy odor in bone-softened mackerel were 11.8% and 0.04%, respectively, and the optimum immersion time was 52.2 min. The estimated overall acceptance, salinity, and acidity of the products under these optimum conditions were 7.7 points, 1.1%, and 202.6 mg/100 g, respectively, which were similar to the actual measured values of $7.6{\pm}1.2$ points, $1.0{\pm}0.1%$ and $203.2{\pm}3.8mg/100g$. Moreover, the heating temperature and time for bone-softening based on RSM were $107.3^{\circ}C$ and 4.4 h, respectively. The estimated hardness and proportion of skin removed from the product under the optimal conditions were $161.5g/cm^2$ and 0.09%, respectively, which were also similar to the actual measured values of $171.1{\pm}12.6g/cm^2$ and $0.10{\pm}0.02%$. The optimum bone-softening and fishy odor-reducing process for mackerel consisted of the following steps: thawing (${\leq}10^{\circ}C$, 8 h), filleting, washing/dewatering, immersing in an 11.8% doenjang -0.04% citric acid solution for 52 min, washing/dewatering, heating ($107.3^{\circ}C$, 4.4 h), freezing, depanning, internal and external packaging, and X-ray detection treatment.
