  • A simple and economical method for separation of lactate and malate dehydrogenase isozymes is described in detail. The method is based on cellulose acetate strip electrophoretic separation of the isozymes, tetrazolium reduction to purple formazan. Resolution is as good as in the experiment using expensive equipments.

  • The hemoglobin phenotype and the gene frequencies of 44 Formosan deer(Cervus nippon) in Kyung-Gi area were examined by using cellulose acetate and starch gel electrophoresis. 1. The method of cellulose acetate electrophoresis was simplier, more clear and preserative than starch gel electrophoresis. 2. The hemoglobin phenotype was appeared 3 types as $Hb^F$ $Hb^{FS}$ and $Hb^S$. The frequencies of appearance were $Hb^F$ 47.7%, $Hb^{FS}$ 47.7% and $Hb^S$ 4.5%, respectively. 3. The genetic factors of hemoglobin were observed as $Hb^F$ and $Hb^S$ and the rates of gene frequencies were 71.6% and 28.4%, respectively.

  • Each ferritin molecule consists of light subunit 19,000 dalton and heavy subunit 22,000 dalton. Twenty-four protein subunit about $440,000{\sim}500,000$ dalton apoferritin which contained $20{\sim}30%$ Fe as ferric hydroxyphosphate polymer form. Horse spleen-derived ferritin consists of 90% light subunit. These genetic characteristics of ferritin preparations were able to determine by cellulose acetate electrophoresis, but these ferritin preparations contained other components to be disturbed during refining, extraction and making finish products and have difficulties in deciding to be just. So, this study was performed to establish the scientific method for determine the quality of ferritin preparations with immunodiffusion methods which has high specificity between heterogeneous proteins.

  • Serum protein and hemoglobin patterns were obtained by cellulose acetate electrophoresis for several anurans. Differences were found both in the number of components from individual animal hemoglobins and in the mobility of these components. Under the conditions employed, toad and R. plancyichosenica had a single component. Other frogs showed 2-component pattern. Sera of all anurans examined contain albumin but no preablumin. Relatively few assayable proteins are found in serum from B. buro gragraizans and differ thereby from most serum patterns examined for frogs, which show complex serum protein bands. Different species have dissimilar patterns although some of bands are apparently homologous between species.

  • To observe the changes of serum proteins according to the process of Babesia gibsoni(B gibsoni) infection, the babesia protozoa($10^8/kg$) were inoculated into the cephalic vein of healthy dogs. The serum proteins of experimentally infected dogs were separated by using cellulose acetate electrophoresis. The results obtained were as follows; 1. Cellulose acetate electrophoresis was fractionated to total 6 of bands such as, albumin, ${\alpha}_1$, ${\alpha}_2$, ${\beta}_1$, ${\beta}_2$ and $\gamma$-globulin. 2. The concentration of total protein was shown a decreasing tendency after B gibsoni infection. Albumin and A/G ratio were lowered through all periods of the infection, but they were not significant changes. 3. The level of ${\alpha}_1$-globulin was significantly(p<0.05) incresed in early stage of the infection. 4. The levels of ${\alpha}_2$ and total $\alpha$-globulin were shown highly significant decreases (p<0.01) through all periods of the infection. 5. The levels of ${\beta}_1$ and total $\beta$-globulin had highly significant changes (p<0.01) that was increased in early stage of infection and decreased later. 6. The level of $\gamma$-globulin was seen to be constantly increased through all periods of infection. It was a highly significant change (p<0.01). 7. Plasma protein: fibrinogen (PP:F) ratio was shown a temporally significant increase (p<0.05) following the decrease in early infection.

Study on Extraction of Mucopolysaccharide-protein Containing Chondroitin Sulfate from Chicken Keel Cartilage

  • The objective of this study was to investigate technical methods for extraction of mucopolysachharide-protein containing chondroitin sulfate from keel cartilage of chickens. The chemical composition of chicken keel cartilage was determined. For the preparation of mucopolysaccharide-protein from lyophilized chicken keel cartilage, hot water extraction and alcalase hydrolysis methods were examined. Results showed that the optimum condition of hot water extraction was incubation for 120 min with a yield of 40.09% and chondroitin sulfate content of 28.46%. For alcalase hydrolysis, the most effective condition was 2% alcalase in 10 volumes of distilled water for 120 min. The yield of hydrolysate was 75.87%, and chondroitin sulfate content was 26.61%. For further separation of chondroitin sulfate from the alcalase hydrolysate, which has a higher yield than that of hot water, 60% ethanol precipitation was performed. The yield of the ethanol precipitate was 21.41% and its chondroitin sulfate content was 46.31%. The hot water extract, alcalase hydrolysate and ethanol precipitate showed similar electrophoretic migration with standard chondroitin sulfate (chondroitin sulfate A), using cellulose acetate membrane electrophoresis. These results indicated that a significant amount of mucopolysaccharide-protein containing chondroitin sulfate could be acquired form chicken keel cartilage. Therefore, keel cartilage in chicken may provide an inexpensive source of chondroitin sulfate for commercial purposes.

  • The alcohol dehydrogenase (ADH) isozyme of the four strains of Drosophila melanogaster in Korea was studied by the starch gel electrophoresis and the results are given below: 1. The numbers of bands, staining intensity and mobility are considerably variable in the three strains, Chunchun, Jinju and Sinchon-Seoul, but relatively uniform in the Taijun strain. 2. It is suggested that the Taijun strain may be homogeneous and the other three strains, on the contrary, heterogeneous for the ADH constitution. 3. The electrophoretic patterns are observed to be different between males and females in all strains used which is hard to intereret. 4. The starch gel electrophoresis is to have a resolving power superior to cellulose acetate electrophoresis.

  • Lactate dehydrogenase isozymes in heart, kidney, liver and skeletal muscle of 15 species of vertebrate animals belonging to 5 classes were separated by cellulose acetate electrophoresis and the levels of them were measured and compared with each other. Lactate dehydrogenase isozyme patterns were different from each other among animal species and among tissues. The activity of LDH$_{5}$ was superior in anaerobic tissues such as liver and skeletal muscle, and the activity of LDH$_{1}$ was superior in aerobic tissues such as heart and kidney. The level of LDH of vertebrate animals of the 5 classes has found approximatry increasing in the following order: Pisces>Amphibia>Reptelia

  • Cellulose acetate electrophoresis was used for differential diagnosis of radicular cysts and granulomas. Sixteen periapical lesions were excised from sixteen teeth with radiographic evidence of periapical pathosis, and two normal periodontal tissues were surgically removed from two wisdom teeth. The tissue samples were separated into two parts. Half was prepared for cellulose acetate electrophoresis, and the electrophoretic patterns were scanned with Helena densitometer. The other half was examined histologically, and the histologic diagnoses were then compared with the electrophoretic patterns and with the x-ray findings. The results were the following: 1. The histopathologic results showed the presence of 13 granulomas (81.8%) and 3 cysts (18.3%). 2. An albumin pattern, alpha-globulin pattern, beta-globulin pattern, fibrinogen pattern and gamma-globulin pattern were found in all eighteen cases. 3. All eighteen cases were divided into normal periodontal tissue group, dental granuloma group, and radicular cyst group by the histopathologic diagnosis. The electrophoretic pattern of dental granuloma group and that of normal periodontal tissue group resembled each other. The two groups showed the highest percentage in beta-globulin fraction, and the beta-globulin of dental granuloma group(47.17%) was higher than that of normal periodontal tissue group (40.54%) by 6.63%. On the contrary, beta-globulin fraction of radicular cyst group (28.00%) was much lower than those of the granuloma group (47.17%) and normal periodontal tissue group (40.54%), and alpha-globulin of radicular cyst group (34.16%) was much higher than those of the granuloma group (20.04%) and normal group (19.58%). 4. Five cases (31.3%) of the sixteen periodontal lesions showed different results between histopathologic diagnoses and radiographic findings.

  • Acidic glycosaminoglycans were isolated from the bovine gingiva and analyzed by chemical methods and by two dimensional electrophoresis on cellulose acetate strip. The average total amount of acidic glycosaminoglycans-expressed as glucuronic acid-was 0.36% of dry gingival tissue. By using colorimetric analysis with two dimenional electrophoresis, the distribution of dermatan sulfate was calculated to be 33% of whole acidic glycosaminoglycans, chondroitin sulfate A to be 26% ad hyaluronic acid to be 38%, respectively.
