A Novel Trp-rich Model Antimicrobial Peptoid with Increased Protease Stability

  • In order to increase protease stability of a novel Trp-rich model antimicrobial peptide, $K_6L_2W_3$ (KLWKKWKKWLK-$NH_2$)and investigate the effect of L-amino acid to peptoid residue conversion on biological functions, we synthesized its antimicrobial peptoid, $k_6l_2w_3$. Peptoid $k_6l_2w_3$ had similar bacterial selectivity compared to peptide $k_66L_2W_3$. The bactericidal rate of $k_6l_2w_3$ was somewhat slower than that of $K_6L_2W_3$. Peptoid $k_6l_2w_3$ exhibited very little dye leakage from bacterial outer-membrane mimicking PE/PG liposomes, as observed in $K_6L_2W_3$, indicating that the major target site of $K_6L_2W_3$ and $k_6l_2w_3$ may be not the cell membrane but the cytoplasm of bacteria. Trypsin treatment of $K_6L_2W_3$ completely abolished antimicrobial activities against Escherichia coli and Staphylococcus aureus. In contrast, the antimicrobial activity of $k_6l_2w_3$ was completely preserved after trypsin treatment. Taken together, our results suggested that antimicrobial peptoid $k_6l_2w_3$ can potentially serves as a promising therapeutic agent for the treatment of microbial infection.

Investigation of morphological changes of HPS membrane caused by cecropin B through scanning electron microscopy and atomic force microscopy

  • Background: Antimicrobial peptides (AMPs) have been identified as promising compounds for consideration as novel antimicrobial agents. Objectives: This study analyzed the efficacy of cecropin B against Haemophilus parasuis isolates through scanning electron microscopy (SEM) and atomic force microscopy (AFM) experiments. Results: Cecropin B exhibited broad inhibition activity against 15 standard Haemophilus parasuis (HPS) strains and 5 of the clinical isolates had minimum inhibition concentrations (MICs) ranging from 2 to 16 ㎍/mL. Microelectrophoresis and hexadecane adsorption assays indicated that the more hydrophobic and the higher the isoelectric point (IEP) of the strain, the more sensitive it was to cecropin B. Through SEM, multiple blisters of various shapes and dents on the cell surface were observed. Protrusions and leakage were detected by AFM. Conclusions: Based on the results, cecropin B could inhibit HPS via a pore-forming mechanism by interacting with the cytoplasmic membrane of bacteria. Moreover, as cecropin B concentration increased, the bacteria membrane was more seriously damaged. Thus, cecropin B could be developed as an effective anti-HPS agent for use in clinical applications.

Antimicrobial activity of sophorolipid biosurfactant

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214. showed antimicrobial activity against Bacillus subtilis. Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm as MIC(minimum inhibitory concentration). Also 100ppm of sophorolipid inhibited 50% of cell growth of plant pathogenic fungus. Botrytis cinerea. However, sophorolipid showed no effect on the Escherichia coli, indicating its selective antimicrobial activity depending on the cell wall structure. Treatment of B. subtilis with sophorolipid increased the leakage of intracellular enzyme, malate dehydrogenase, indicating the possible interaction of sophorolipid with cellular membrane. Between lactone-type and acid-type sophorolipid, the former showed higher antimicrobial activity.

  • MCPs means Micro Cellular Plastics. The micro-cells are generated in the products by the difference of dissolution through the pressure drop after super critical fluid of CO2 or N2 dissolves into polymer. We have developed injection molding process adopting MCPs and applied it to a broad range of injection molded thermoplastic materials and applications. It can prevent the leakage of impact strength and increase the thermal conductivity, moreover regulate the thermal conductivity. Then we can develop the high strength foaming plastics. Also, it can be gained a competitive advantage by utilizing its processing benefits, e.g. the lightweight products and significant reductions in material consumption.

Scanning Electron Microscopy Studies of Saccharomyces cerevisiae Structural Changes by High Hydrostatic Pressure Treatment

  • The structural change and leakage of cellular substances of Saccharomyces cerevisiae attributed by high hydrostatic pressure (HHP) treatment were observed with scanning electron microscopy (SEM). S. cerevisiae (ATCC16664) was inoculated in apple juice for 10 min at $23^{\circ}C$ and the apple juice treated at 138, 207, 276, 345, and 414 MPa pressure for 30 sec at $23^{\circ}C$. Increased of roughness, elongation, wrinkling, and pores on yeast cell surfaces, the yeast cell walls were severely damaged by HHP treatment from 276 to 414 MPa. Inactivation of S. cerevisiae by HHP is dependent on structural changes on the cell walls observed with SEM.

  • The inhibition of chlorophyll formation in cucumber cotyledons by N-phenyl oxadiazolidinedione derivatives Ia-u showed similar trend as their herbicidal activities. In case of oxadiazolidinedione Iq, with a propargyloxy substituent, both the highest herbicidal activity and inhibitory action(pI$_{50}$ = 6.37) were observed. The accumulation of protoporphyrin IX and cellular electrolyte leakage by oxadiazolidinedione Ia, Ik and Iq were well correlated with their inhibition of chlorophyll biosynthesis. These results suggest that the herbicidal activity of oxadiazolidine Ia-u is originated from the inhibition of chlorophyll biosynthesis.

Antimicrobial Activity of Biosurfactants

  • Biosurfactant has antibiological activities. Due to its low toxicity, biosurfactant can be applied to human health care products. Posseing the antibiological activities as well as the surfactant properties implies that biosurfactant can be widely used in the cosmetic industry. In this presentation, applications of biosurfactant have been reviewed including the antimicrobial activities of sophorolipid.   • A non-contact battery charger for cellular phone is designed using half-bridge series resonant converter. This converter utilizes series resonance to reduce the undesirable effect of large leakage inductance of the detachable transformer and ZVS operation can reduce switching loss and switching noise. In this paper, analysis and design procedure of half-bridge series resonant converter with detachable transformer is presented. The input voltage is 85VAC∼270VAC, and the output voltage and current is 4.1V and 800mA, respectively. Furthermore, a method of calculating the secondary current of the transformer to control the battery charging current in the constant current charging mode is proposed. The performance of the charger is verified through experiments.

  • 본 논문에서는 잘 알려진 선형 대수 이론을 이용하여 LFSR을 활용한 LFSM의 구현 방법을 제시한다. 이를 사용하여 셀룰라 오토마타를 MFSR대신 사용하여 Anderson 정보누출과 같은 스트림 암호의 좋지 않은 성질을 제거하고자 하는 시도로는 안전성을 높일 수 없음을 보인다. 또한 LFSM에 기반한 비선형 필터 생성기가 Anderson 정보 누출의 위험성이 있는 형태로 변형 가능한지 확인하는 방법을 제시한다.

  • The aim of this study was to investigate the protective effect of acanthoic acid on liver injury induced by either tertiary-butyl hydroperoxide (tBH) or carbon tetrachloride in vitro and in vivo. Acanthoic acid, (-)-pimara-9(11),15-diene-19-oic acid, is a diterpene isolated from the root bark of Acanthopanax koreanum. In in vitro study, the cellular leakage of lactate dehydrogenase (LDH) with 1.5 mM tBH for 1 j, were significantly inhibited by treatment with acanthoic acid(25 and 5mg/mL). (omitted)
