아토피 피부염을 설명하는 데는 두 가지 패러다임이 있는데, 첫 번째는 outside-inside paradigm이고 두 번째는 inside-ouside paradigm이다. Outside-inside paradigm에 의하면 아토피 피부염을 치료하기 위한 가장 좋은 방법은 피부 장벽 기능을 회복시키는 것이다. 장벽 기능은 각질층의 특징적인 구조에 의해 유지되는데, 각질층은 층상구조의 inter-cellular lipids와 corneocyte로 구성되어 있다. 아토피 피부염 환자, 층판상 어린선 환자의 각질층 내 층상구조는 hexagonal 구조가 많이 존재하고 있음을 보였으며, 이는 Ceramide 1 타입과 long chain fatty acid의 감소 때문이라고 보여진다. 이러한 이유에서 라멜라구조를 가지며 intercellular long periodicity phase 기능을 복원할 수 있는 보습제는 각질층의 intercellular lipid의 층상구조를 복원시킬 수 있을 것이다. 또한 세라마이드와 유사세라마이드를 함유한 보습제는 손상된 피부장벽, 피부질환, 아토피 피부염을 위한 유용한 치료제로 사용할 수 있다. Inside-outside paradigm에 따르면, 아토피 피부염 등이 보조 T cell, lg E, eosinophil 등을 포함한 면역반응과 관계되는데, 특히 Th1/Th2 imbalance의 복원이 아토피 피부염을 치료하는데 효과적일 것이다. 현재 여러 종류의 면역억제제가 소개되고 있지만, 이들은 고유 면역기능을 감소시킬 우려를 가지고 있다. 따라서, 세라마이드 대사산물인 SPC와 S1P는 면역조절 기능 및 상처치유의 기능을 가짐으로써 향후 관심이 집중될 것이다.
본 연구는 꿀벌 꽃가루 추출물(BPW)의 면역 활성에 관하여 알아보기 위하여, 선천면역계의 대표적인 수지상세포와 후천면역계의 대표적인 비장세포에 BPW를 처리 하여 면역세포의 활성능을 관찰하였다. 수지상 세포에 BPW를 처리하여 세포 생존율, 산화질소(II)와 사이토카인($TNF-{\alpha}$, IL-6과 $IL-1{\beta}$) 분비능과 세포 표면분자를 관찰 하였다. 세포 생존율은 수지상 세포에 BPW를 처리하였을 때, 세포 독성을 일으키지 않았으며 주요 면역 활성 인자인 산화질소(II) 분비능을 관찰한 결과, 농도 의존적으로 증가하는 것을 확인하였다. 또한 사이토카인의 분비능을 관찰한 결과, $TNF-{\alpha}$, IL-6과 $IL-1{\beta}$의 함량이 농도 의존적으로 증가하는 것으로 관찰되었다. 또한 활성화된 면역세포의 세포 표면에서 발현되는 CD80과 CD86의 발현과 항원제시에 밀접한 관련이 있는 MHC class I, II의 발현이 유의적으로 증가하는 것으로 관찰되었다. 또한 후천면역에서 중요한 역할을 수행하는 면역 T 세포가 다량 분포하는 지라 세포를 분리하여 BPW를 처리 하였을 때 Th1 세포가 분비하는 사이토카인의 함량이 농도 의존적으로 증가되는 것으로 나타났다. 이러한 결과로 미루어 볼 때 BPW는 선천면역뿐만 아니라 후천면역에 관여하는 다양한 면역세포의 활성화에 직간접적으로 관여하는 것으로 사료된다.

This review suggested that the high rate of glutamine utilization by cells of the immune system serves to maintain a high intra cellular concentration of the intermediates of biosynthetic pathways such that optimal rates of DNA, RNA and protein synthesis can be maintained. In the absence of glutamine, lymphocytes do not proliferate in vitro: proliferation increase greatly as the glutamine concentration increase. Glutamine is synthesized in skeletal muscle. Skeletal muscle and plasma glutamine levels are lowered by sepsis, injury, bums, surgery and endurance exercise and in the overtrained athlete. Background: Ginseng is a commonly used herbal medicine in treating various medical conditions. Chronic gut inflammation is a recognized factor for the development of colorectal cancer (CRC). In this project, Asian ginseng berry polysaccharide preparations were used to assess their effects on CRC and related immune regulation mechanisms. Methods: Ginseng berry polysaccharide extract (GBPE) and purified ginseng berry polysaccharide portion (GBPP) were used to evaluate their activities on human HCT-116 and HT-29 CRC cell proliferation. Interleukin-8 secretion analysis was performed on HT-29 cells. Naive CD4 cell isolation and T-helper cell differentiation were performed and determined using flow cytometry for Th1 and Treg in addition to cell cycle and apoptotic investigation. Results: GBPE and GBPP significantly inhibited interleukin-8 secretion and cancer cell proliferation, inhibited CD4+IFN-γ+ cell (Th1) differentiation, and decreased CD4+FoxP3+ cell (Treg) differentiation. Compared to the GBPE, GBPP showed more potent antiinflammatory activities on the malignant cells. This is consistent with the observation that GBPP can also inhibit Th1-cell differentiation better, suggesting that it has an important role in antiinflammation, whereas Treg cells hinder the body's immune response against malignancies. Supported by cell cycle and apoptosis data, GBPE and GBPP, at various degrees, remarkably enhanced the anticancer activities of 5-fluorouracil. Conclusion: Data from this project suggested that Asian ginseng berry potentially has clinical utility in managing enteric inflammation and suppressing CRC through immunomodulation mechanisms.
Effects of different photoperiod regimens on the cellular and humoral immunity in broiler chickens were studied(Exp 1). Total one hundred ninety two one-day-old commercial broiler chicks(Cobb$\times$Cobb) were raised between constant lighting(CL) and intermittent lighting (1h light: 3h darkness(IL; 1l; 3D) Body weight, feed intake and feed conversion were measured for seven week. Peripheral blood and splenic lymphocyte activities were tested at 3 and 5 wk of age by performing a mitogen cellproliferation assay with a polyclonal T-cell mitogen, concanavalin A (Con A), and B-cell mitogen, lipopolysaccharide (LPS). To investigate the effect of photoperiod on the humoral immunity, chicks were immunized with sheep red blood cell(SRBC) and iinactivated Newcastle disease virus(NDV) vaccine. Total immunoglobulin G(IgG) concentration was also determined. Diurnal change of melatonin was tested in sera. In experiment 2, 0.1ml melatonin were subcutaneously injected from three to five weeks old if immunomodulation effect of lighting regimen was due to the melatonin or not. Injections of melatonin were made at 0700h and the dosage was 10ng (M2), 100ng(M3), 1$\mu\textrm{g}$(M4) per bird daily, respectively. control were quivalent injections of vehicle(M1). Lymphocyte activities were tested and humoral immunities were examined at 5 weeks of age. Blood melatonin concentration was determined at 0h, 1, h, 2h, and 3h posterior to injection at five weeks old. It was higher in CL chicks than IL chickens during the subsequent period of 3 to 5 wk of age. However, weight gain of chicks raised IL were significantly higher at 6 wk of age than CL(P<0.05). Antibody response to NDV was not affected by both photoperiod regimens and melatonin injection, whereas anti-SRMB titer and IgG concentration were enhanced. Lymphocyte activity of chickens raised under IL was sighificantly higher than those of chickens raised under CL. Melatonin injection also increased lymphocyte activity. When peripheral blood lymphocytes were used, proliferation response to LPS and Con A were significantly increased in M2 and respectively. The results of this experiments suggest that IL improved host immune response and melatonin have immunomodulatory roles.
Rising of water temperature due to global warming is a great concern to aquaculturists and fishery biologists. Hence, the present study aimed to investigate the effects of high water temperature on juvenile red spotted grouper, Epinephelus akaara based on the evaluation of stress responses in blood. E. akaara juveniles were exposed to different thermal conditions ($25^{\circ}C$, $28^{\circ}C$, $31^{\circ}C$, and $34^{\circ}C$) for 6 weeks following 2 weeks of acclimation at $25^{\circ}C$. Blood cell morphology and number were examined at three sampling points (2, 7, and 42 days) from a total of 180 fish. Major erythrocytic cellular abnormalities (ECA) observed in blood smears of thermally stressed groups ($31^{\circ}C$ and $34^{\circ}C$) after 6 weeks were echinocytes, teardrop-like cells, swollen cells and vacuolated cells. Both red and white blood cell number (RBC and WBC) were significantly (p<0.05) elevated in $31^{\circ}C$ and $34^{\circ}C$ group after 6 weeks thermal exposure. Differential leucocytes number showed significant increases in neutrophil (N) and decreases in lymphocytes (L) in the highest temperature ($34^{\circ}C$). Different N:L ratio was observed at different thermal conditions which can be used as a reliable alternative to measure stress response. Taken together, these results suggest that higher temperature ($31^{\circ}C$ and $34^{\circ}C$) can interfere the immune system of red spotted grouper by altering the blood cell morphology and number.
Soil acidification is one of major problems limiting crop growth and especially becoming increasingly serious in China owing to excessive use of nitrogen fertilizer. Only the STOP1 of Arabidopsis was identified clearly sensitive to proton rhizotoxicity and the molecular mechanism for proton toxicity tolerance of plants is still poorly understood. The main objective of this study was to investigate the transcriptomic change in plants under the low pH stress. The low pH as a single factor was employed to induce the response of the wheat seedling roots. Wheat cDNA microarray was used to identify differentially expressed genes (DEGs). A total of 1057 DEGs were identified, of which 761 genes were up-regulated and 296 were down-regulated. The greater percentage of up-regulated genes involved in developmental processes, immune system processes, multi-organism processes, positive regulation of biological processes and metabolic processes of the biological processes. The more proportion of down-regulation genes belong to the molecular function category including transporter activity, antioxidant activity and molecular transducer activity and to the extracellular region of the cellular components category. Moreover, most genes among 41 genes involved in ion binding, 17 WAKY transcription factor genes and 17 genes related to transport activity were up-regulated. KEGG analysis showed that the jasmonate signal transduction and flavonoid biosynthesis might play important roles in response to the low pH stress in wheat seedling roots. Based on the data, it is can be deduced that WRKY transcription factors might play a critical role in the transcriptional regulation, and the alkalifying of the rhizosphere might be the earliest response process to low pH stress in wheat seedling roots. These results provide a basis to reveal the molecular mechanism of proton toxicity tolerance in plants.
Arabinoxylan, a complex polysaccharide in cereal cell walls, has recently received research attention as a biological response modifier. The immunomodulating effect of arabinoxylans from rice bran (AXrb) was studied using a combined process of extrusion and commercial hemicellulase treatment in order to elucidate the augmentation mechanism of cell-mediated immunity in vitro. The cytotoxicity of mouse spleen lymphocytes against YAC-1 tumor cells was significantly enhanced by treatment with AXrb at $10-100\;{\mu}g/mL$. In an attempt to investigate the mechanism by which AXrb enhance NK cytotoxicity, we examined the effect of AXrb on cytokine production by spleen lymphocytes. Culture supernatants of the cells incubated with AXrb were collected and analyzed for IL-2 and IFN-${\gamma}$ synthesis by ELISA. IL-2 and IFN-${\gamma}$ production were increased significantly. These results suggest that AXrb may induce Th1 immune responses. Macrophages play an important role in host defenses against tumors by killing them and producing secretory products, which protect against bacterial, viral infection and malignant cell growth. AXrb were examined for their ability to induce secretory and cellular responses in murine peritoneal macrophages. When macrophages were treated with various concentrations ($10-100\;{\mu}g/mL$) of AXrb, AXrb induced tumoricidal activity, as well as increasing phagocytosis and the production of NO, $H_2O_2$, TNF-${\alpha}$, IL-$1{\beta}$, and IL-6. These results indicate that reactive oxygen species, reactive nitrogen species, and inflammatory cytokines are likely to be the major mediators of tumoricidal activity in AXrb-treated macrophages. Therefore, AXrb may be useful in cancer immunotherapy and it is anticipated that AXrb obtained using extrusion and subsequent enzyme treatment can be used as an ingredient in nutraceuticals and cereal-based functional food.
Background: Dengue virus, which belongs to the Flavivirus genus of the Flaviviridae family, causes fatal dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) with infection risk of 2.5 billion people worldwide. However, approved vaccines are still not available. Here, we explored the immune responses induced by alternating prime-boost vaccination using DNA vaccine, adenovirus, and vaccinia virus expressing E protein of dengue virus type 2 (DenV2). Methods: Following immunization with DNA vaccine (pDE), adenovirus (rAd-E), and/or vaccinia virus (VV-E) expressing E protein, E protein-specific IgG and its isotypes were determined by conventional ELISA. Intracellular CD154 and cytokine staining was used for enumerating CD4+ T cells specific for E protein. E protein-specific CD8+ T cell responses were evaluated by in vivo CTL killing activity and intracellular IFN-${\gamma}$ staining. Results: Among three constructs, VV-E induced the most potent IgG responses, Th1-type cytokine production by stimulated CD4+ T cells, and the CD8+ T cell response. Furthermore, when the three constructs were used for alternating prime-boost vaccination, the results revealed a different pattern of CD4+ and CD8+ T cell responses. i) Priming with VV-E induced higher E-specific IgG level but it was decreased rapidly. ii) Strong CD8+ T cell responses specific for E protein were induced when VV-E was used for the priming step, and such CD8+ T cell responses were significantly boosted with pDE. iii) Priming with rAd-E induced stronger CD4+ T cell responses which subsequently boosted with pDE to a greater extent than VV-E and rAd-E. Conclusion: These results indicate that priming with live viral vector vaccines could induce different patterns of E protein-specific CD4+ and CD8+ T cell responses which were significantly enhanced by booster vaccination with the DNA vaccine. Therefore, our observation will provide valuable information for the establishment of optimal prime-boost vaccination against DenV.
Objective : The purpose of this study is to investigated that effect of Bupleuri Radix Herbal-acupuncture solution (BRHS) on the celluar immune response in mice with adjuvant induced arthritis, performed several experimental items : those are paw edema, IL-1b, IL-6, IL-8, PNF-a and $PGE_2$. Methods : All the male Sprague Daeley mice used in this study were bred and maintained in our pathogen-free mouse colony and were 8 weeks of age at the start of the experiment. The experimental model of arthritis was induced by injection of $50{\mu}g/{\mu}{\ell}$ adjuvant(mineral oil mixed Mycobacterium butyricum). Bupleuri Radix Herbal-acupuncture solution (BRHS) was injected into $ST_{36}$(足三理) of mice daily for 21 days. Immunohistological analysis was carried out to assess paw edema, IL-$1{\beta}$, IL-6, IL-8, PNF-${\alpha}$ and $PGE_2$expression in synovial membrance and sera Bupleuri Radix Herbal-acupuncture solution(BRHS) injected. Results : At day 21 post arthritis onset, immunohistological studies using monoclonal antibodies showed that Bupleuri Radix Herbal-acupuncture solution (BRHS) group had decreased expression of IL-$1{\beta}$, IL-6, IL-8, PNF-${\alpha}$ and $PGE_2$ at inflamatory cytokines production and edema compared with control group. Conclusion : Bupleuri Radix Herbal-acupuncture solution (BRHS) inhibited infammatory cytokines production and edema in adjuvant induced arthritic mice. Thus, Herbal-acupuncture solution may have prevention.

경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
