Cellular fatty acid composition of eight strains, indluding six strains of Comamonas terrigena, and two type strains of Pseudomonas acidovorans, and P. testosteroni was determined by gas-liquid chromatography. Almost the same composition was found in all the strains tested, and hexadecanoic acid, hexadecenoic acid, and octadecenoic acid were accounted more than 70% of total fatty acid. However, P. testosteroni differed from C. terrigena and P. acidovorans by the presence of comparatively large amonuts of 2-hydroxy-hexadecanoic acid, and C. terrigena contained three to eight times as much tetradecanoic acid in P. acidovorans and P. testosteroni. According to the similarity values calculated on the basis of fatty acid composition, C. terrigena strains were divided into three groups differentiated in the requirement of growth factors, and C. terrigena, P. acidovorans, and P. testosteroni strains occupied separate position each other in the dendrogram.

The effects of stearic(18 : 0, SA), oleic(18 : 1 cis, OA) and elaidic acid(18 : 1 trans, EA) on the cell growth, contents of cellular lipids, and the fatty acid composition of cellular and medium lipids in Hep-G$_2$cells were evaluated. The cells were incubated in serum-free medium containing 25, 50, 100 and 200$\mu$M of a fatty acid combined with albumin for 2 days. The fatty acid concentration up to 100$\mu$M showed the normal growth, but the cell growth decreased in the presence of 200$\mu$M fatty acid. The treatment of cells with 100$\mu$M of a fatty acid for two days significantly(p<0.05) increased the cellular triglyceride(TG) content in all fatty acid groups compared to control, but TG contents was not significantly different among all treatment group, but total cholesterol(TC) was the highest level in EA group. The level of free cholesterol(FC) and cholesteryl ester(CE) was similar to those of TC in all fatty acid treated group. The cellular phospholipid(PL) contents were similar between the control and all fatty acid groups. The treatment of cells with SA has no notable effects on the fatty acid composition of TG, CE and PL. The OA treatment caused significant increases in CE(51.2%) and PL(29.8%), but not in TG. The EA treatment resulted in 10.1, 10.7 and 7.8% of $C_{18:1\;trans}$ content in cellular TG, CE, and PL. The TG, CE and PL of medium were relatively similar between SA and OA groups. In EA treated group, TG, CE and PL of medium contained 17.0%, 0.7% and 5.6% of $C_{18:1\;trans}$, respectively.

The result of API staph-ident system was compared with cellular fatty acid composition for the identification of Staphylococcus species isolated from cattle. Isolated strains from cattle were correctly identified to S aureus, S intermedius, S hyicus, S simulans, S saprophyticus, S epidemis, S sciuri and S xylosus by API staph-ident system. The correlation between bacterial cellular fatty acid profile and Staphylococcus species isolated to API STAPH-IDENT system were. In conclusion, the result presented indicate that Staphylococci can be indentified to the species level by the cellular fatty acid profiles. Moreover, computerized fatty acid profile correlative anaylsis can be applied for determining identify of Staphylococcus species.
The distribution of Listeria spp. in various foods and its fatty acid composition were examined. A total 60 samples of dairy products(15), seafoods(20), meat products(18), factory waster(2), and salades(5) were tested. Listeria spp. was found 10 samples, showing about 16.7% detection ratio; dairy products 0(0%),,seafoods 1(5%), meat product 7(38.9%), and factory wastes 2(100%). Whereas L. welshimeri was isolated from meat products 1(5.6%) and factory wastes 1(50%). The cellular fatty acid composition determined by gas chromatography was found not to differ among L. innocua isolated from food has similar fatty acid profiles when grown at 3$0^{\circ}C$,24 hrs on the tryptic soy plate with C15 and C17 anteiso branched acids accounting for about 80% of total.
This study was conducted to compare the cellular fatty acid composition and genotypic analysis of Bifidobacterium longum MK-G7 originated from Koreans with other commercial type strains of bifidobacteria. The cellular fatty acid of Bif. longum MK-G7 was shown to be composed of $C_{160FAME},C_{181\;c18DMA},C_{18.1\;CIS9\; FAME},C_{14.0FAME},C_{19\;0cye9,10 DMA},Feature7(C_{17.2 FAME), and Feature 10(C_{181\; Cll/t9/t6 FAME}$. Bif. longum MK-G7 showed 99.9% homology and the highest relatedness with Bif. longum ATCC 15707 type strain. Both Bif. longum MK-G7 and Bif. longum ATCC 15707 showed 153 bp products on RAPD (randomly amplified polymorphic DNA) analysis, however, they showed quite different band patterns on PFGE (pulsed-field gel electrophoresis) analysis. Consequently, our present study showed that Bif. longum MK-G7 was different from any commercial type strains of Bif. longum tested.
This study was performed to compare the growth rates, cellular fatty acid compositions and morphology by using electron microscope of Escherichia coli (E. coli) grown in various conditions including different concentrations of phenol, CdCl₂ and HgCl₂. Ninety eight E. coli strains were isolated from Naktong river and human feces. The content of unsaturated fatty acids, especially 16:1ω7c and 18:1ωc increased as the concentration of phenol and CdCl₂ increased. The content of unsaturated fatty acid increased up to 50 ppb of HgCl₂, but decreased at 75 ppb of HgCl₂. There were more unsaturated fatty acids than saturated fatty acid in the presence of toxic substances. However, the ration was reversed when the affected E. coli was transferred to toxic substance free fresh trypticase soy broth medium. Also, by using transmission electron microscope these cells were observed to various morphological deformation by heavy metals and their deposition on the surface. From these results, we suggested that the changes of major fatty acids composition and morphology of E. coli may be considered to indicate contaminated levels of heavy metals or organic solvents. The information presented here may be useful in predicting effects of heavy-metal and organic solvent contamination in streams and provides a basis for further studies of metal or organic solvent effects on microbial communities.
The membrane fatty acid composition of tumor cell can be modified either in cell by altering the lipid composition of the medium of during growth in animals by changing the dietaty fat composition. These modifications are associated with changes in membrane physical properties and certain cellular functions, including carrier-mediated transport and enzyme contained within the membrane. Such effects influence the transport of nutrients and chemotherapeutic agents in cancer cells .Fatty acid modification also can enhance the sensitivity of the neoplastic cell to chemotherapy. The alteration in plasma membrane composition will be affected through dietary supplementations and the potential value to cancer patients could be a better understanding of the effects of diet on responsiveness of neoplasms to chemotherapy, i.e. cancer patients' chances for a "cure" can be improved by diet changes prior to treatment.
Cellular fatty acid composition of 17 strains of the Pseudomonas aeruginosa was determined by gas-liquid chromatography isolated from environmental and clinical sample in a C university hospital. Straight-chain saturated acid of C16:0 and straight-chain unsaturated acid of C18:1 with a double bond were commonly found in all the strains tested. The presence of 12:0 3OH (3-10%), 16:0 (18-28%), and 18:1w7c (17-37%) showed the characteristics of the species in the Pseudomonas. Bacterial fatty acid composition was considered to be useful for the study of interrelation and for rapid identification of the bacteria.
The cellular fatty acid composition is important for metabolic plasticity in Rhodobacter sphaeroides. We explored the effects of changing the cellular ratio of unsaturated fatty acids (UFAs) to saturated fatty acids (SFAs) in R. sphaeroides by overexpressing several key fatty acid biosynthetic enzymes through the use of expression plasmid pRK415. Bacteria containing the plasmid pRKfabI1 with the fabI1 gene that encodes enoyl-acyl carrier protein (ACP) reductase showed a reduction in the cellular UFA to SFA ratio from 4 (80% UFA) to 2 (65% UFA) and had decreased membrane fluidity and reduced cell growth. Additionally, the ratio of UFA to SFA of the chromatophore vesicles from pRKfabI1-containing cells was similarly lowered, and the cell had decreased levels of light-harvesting complexes, but no change in intracytoplasmic membrane (ICM) content or photosynthetic (PS) gene expression. Both inhibition of enoyl-ACP reductase with diazaborine and addition of exogenous UFA restored membrane fluidity, cell growth, and the UFA to SFA ratio to wild-type levels in this strain. R. sphaeroides containing the pRKfabB plasmid with the fabB gene that encodes the enzyme β-ketoacyl-ACP synthase I exhibited an increased UFA to SFA ratio from 4 (80% UFA) to 9 (90% UFA), but showed no change in membrane fluidity or growth rate relative to control cells. Thus, membrane fluidity in R. sphaeroides remains fairly unchanged when membrane UFA levels are between 80% and 90%, whereas membrane fluidity, cell growth, and cellular composition are affected when UFA levels are below 80%.
The effect of carbon number of hydrocarbon used as a carbon source in the production of cellular lipid of Rhodotorula sp. and its fatty acid composition were investigated. Using Rhodotorula sp. on n-tetradecane and n-hexadecane whose carbon numbers are even, fermentation was carried out in a jar fermentor of 2 liter-capacity at $28^{\circ}C$, with pH range of 4.0∼4.6 and at oxygen flowing rate of 0. 4 vvm and agitation velocity of 1000 rpm. Drying the produced cell after completion of fermentation, cellular lipid was extracted from the cell using soxhlet extractor and examined its fatty acid composition by gas-liquid chromatography. Cellular lipid content in the cell produced on n-tetradecane and n-hexadecane were 12.0 % and 25,8 % on the basis of dry cell weight, respectively and their fatty acids were mostly even numbered in carbon number.
