Histochemical and Ultrastructural Study on the Digestive Tract of a Land Snail Nesiohelix samarangae (동양달팽이의 소화관에 대한 조직화학적 및 미세구조적 연구)

  • A histochemical and ultrastructural study on the epithelia of some selected digestive tracks such as esophagus, crop, intestine of a land snail N. samarangae was carried out during the period of June 1997 to may 1998. The epithelium of digestive tract are simple columnar epithelium and consisted of five types of columnar cells. Type 1 cell which is majority in number has a brush border with microvilli on the free surface of the cell and contains numerous secretory granules supposed to be neutral mucopolysaccharide. Type 2 cell, elongated conical in shape, is rarely found in the epithelium. This cell also has a brush border with microvilli on its free surface and contains well developed rough surfaced endoplasmic reticulum, Golgi bodies, and secretory granules in various electron densities. This cell seems to produce both of acid and neutral mucopolysaccharides. Type 3 cell, which is morphologically similar to the Type 1 cell, has microvilli and cilia on the free surface and exists in group only in the limited regions of the intestine. Type 4 cell, typical goblet cell containing secretory granules in high electron density. Type 5 cell rarely found in the digestive tract. This cell contain inconspicuous materials.

Effect of Herbal Extract on Helper T Cell activity (소청용탕이 Helper T Cell의 활성에 미치는 영향)

  • SCRT (Sochungyong-tang) has been used for immune disease in human. The purpose of this study was effect of Helper T cell, major regulator of immune system. Spleen cell from 8 week BALB/c mice were cultured in SCRT containing medium without activation for 48 h. The MTS assay and flow cytometry revealed that SCRT treated Iympocyte were non-effect in percentage of CD4+ T cell. Subsequently CD4+ T cell were isolated and cultured in SCRT containing medium. SCRT were non-effective on CD4+ T cell without any involvement of APC. In order to evaluate the direct effect of SCRT on Helper T cell, CD4+ T cell isolated after 48 h of culture in SCRT containing medium and activated with and without anti-CD3/anti-CD28 activation for 48 h. A lower level of CD69 was observed in SCRT treated cells in flow cytometry analysis. Subsequently Using RT-PCR analysis the expression of mRNA for IL-2, INF-γ are upregulated and, IL-4 is downregulated in CD4 T cell. The result suggests that SCRT makes Th1 significantly increased and Th2 relatively inhibited. The results suggest that SCRT potentiate Th1 cell and decrease Th2 development at the same time, which is believed to be bemeficial for IgE-mediated responses.

Histological changes in brain tissue of rat induced neuronal excitotoxicity by NMDA(N-methyl-D-asparate) (NMDA(N-methyl-D-asparate)의 투여에 의해 유발된 신경 과흥분상태에서의 쥐의 뇌조직 변화)

  • Histological changes was investigated in the 4 weeks old rat brain using NMDA (N-methyl-D-asparate) which is capable of mediating excitotoxic events. The changes were occured when the injected NMDA solved in PBS was over $1.0{\mu}g/g$(about 90nM). The necrosis of Purkinje cells in cerebellum and the increasement of coloidal plexus cell number were prevalent. The Purkinje cell number of necrosis were increased according to increasement of amount of injected NMDA. In spite of increasement of degenerated Purkinje cell number, differentiation of new Purkinje cell was not identified because total number of Purkinje cell was not changed. The change of cell number was observed in coloidal plexus cell rather than degeneration of cell. About 5 time increasement was occured. This change may cause increasement of cerebrospinal fluid and the makes mophorogy of brain more round than nomal.

Performance Analysis of Coordinated Random Beamforming Technique in Multi-cell Environments

  • For multi-cell environments, coordinated random beamforming technique in multiuser MIMO(multiple-input multiple-output) broadcast channel is considered. In order to mitigate severe interference at receivers, the multi-cell environments might require complex transmitter and receiver design because the scheduler decision based on full channel state information (CSI) in one cell must be intertwined with decision made by other cells' CSI. With limited CSI, however, this paper considers a scheme of randomizing transmitters' beamforming but being coordinated with other cell transmitters. The transmitters in each cell share random beamforming patterns and schedule data transmission within coherent scheduling period. The corandomized beams allow the users to be selected with the highest SINRs even in multi-cell environments. We analyze the performance of the proposed scheme. And numerical results show that the scheme achieves better performance than the conventional random beamforming when applying to multi-cell environments.

Effect of Distilled Water Supply Method on Performance of PEMWE Typed Hydrogen Generators for Inhalation (흡입용 PEMWE형 수소 발생기에서 증류수 공급 방법이 성능에 미치는 영향)

  • The present study has investigated the performance of hydrogen gas generators for inhalation purposes based on polyelectrolyte membrane water electrolysis (PEMWE). The system applied two watering methods. One is pumped water (pumping system) and the other is gravity-fed water without a pump (non-pumping system). The cell efficiencies were compared by measuring the cell voltage and temperature in the hydrogen gas generator, respectively. The results show that the cell voltage and temperature increase with the cell current. The cell temperature is lower in the pumping system than that in the non-pumping system at a given cell current. Even though the amount of hydrogen production is the same regardless of the pumping system, the cell efficiency of the hydrogen gas generator in the non-pumping system is better than that in the pumping system.

Cell separation from high density culture broths of Alcaligenes eutrophus by using Al-based coagulants (Alcaligens eutrohus 고농도 배양액으로부터 알루미늄(Al)계 응집제를 이용한 세포분리)

  • Cell recovery from high cell density broths of Alcaligenes eutrophus by pretreatment with aluminum-based coagulants such as aluminum sulfate, polyaluminum hydrooxide chloride silicate (PACS), and polyaluminum hydrooxide chloride (Hi-PAX) was carried out. Cells coagulated with coagulants could be successfully recovered above 95-99% by centrifugation or filtration. The optimum initial pH of fermentation broths for cell recovery was in the range of 10 to 12. Optimum coagulants dosage for cell recovery increased with increasing of cell concentrations (21-160 g/L). The optimum coagulant dosages to recover cells with more than 95% cell recovery by centrifugation for the cell concentrations ranged 21-160 g/L were as follows: aluminum sulfate, 416-1708 mg Al/L; PACS, 211-826 mg Al/L; Hi-PAX, 320-960 mg Al/L. At optimum conditions for the coagulation of cells, centrifugal forces for 95% of cell recovery were dependent on the cell concentration. The centrifugal forces at 82 g/L and 160 g/L of cell concentration were only 45${\times}$g and 1600${\times}$g, respectively.

An Empirical Study about the Segmented Cell in Anode Side of PEMFC

  • The present study focused on the segmented cell which has the similar performance to unaltered (not segmented) cell in real operating condition. Many literatures have been made the segmented cell to observe the behavior of local current density distribution in the single cell. However, it has been lack of scheme to segment the cell in that the detailed interpretation of segmenting in analytic point of view was insufficient. Hence, the basic idea of segmenting was introduced to determine the component to be segmented in anode side of unit cell. The electrical contact/bulk resistance was measured by using four wire/probe method through each part of cell components including MEA, GDL, Bipolar Plate and Current Collector. Electron transport mechanism was predicted by comparing resistance values which were obtained from the experiment. As a result, this offered a great benefit to segment the cell efficiently. With this method further experiments would be conducted in research areas which require current density distribution at the same operating condition as unaltered cell.

Studies on the red blood cell sedimention rates in heteroplasma of chicken and goat (닭 및 산양의 이종plasma내에서 적혈구 침강에 대한 연구)

  • In order to study the marked variation of red blood cell sedimentation rate in some species of animals, the packed cell volume, volume percentage of erythrocytes in whole blood, was reshuffled of 20%, 40% and 60% using heteroplasma of chicken and goat, and the red blood cell sedimentation rate was measured in Westergren tubes at $27{\pm}1^{\circ}C$ and $8{\pm}1^{\circ}C$. The results obtained were summarized as follows: 1. The values of packed cell volume(PCV) of goat and chicken were $40.7{\pm}4.1%$ and $30.2{\pm}2.2%$ respectively. 2. The sedimentation rates of reshuffled red blood cell were settled faster at lower PCV than higher PCV, i.${\acute{e}}$. there was a reverse relationship between the sedimention rate and PCV. 3. Red blood cells of chicken settled quickly, where as those of goat settled very slowly. Chicken red blood cell sedimented rapidly even in goat plasma, and goat red blood cell sedimented slowly in chicken plasma. These findings indicate that the plasma is not the only factor determining the rapid red blood cell sedimentation rate of chicken. 4. The sedimentation rate of reshuffled red blood cell of chicken and goat were accelerated at higher temperature than lower temperature.

  • Because of the low temperature operation, proton exchange membrane (PEM) fuel cell has a water phase transition. Therefore, water management is an important operation issue in a PEM fuel cell because the liquid water in the fuel cell causes electrode flooding that can lower the cell performance under high current density conditions. In this study, in order to understand the reactant distributions in the cathode channels of the PEM fuel cell, an experimental technique that can measure the species concentrations of reactant gases by using gas chromatograph (GC) is applied for an operating PEM fuel cell. The oxygen distribution along the cathode flow channels of PEM fuel cell is mainly investigated with various operating conditions. Also, the relations between cathode flooding and oxygen concentrations and oxygen consumption pattern along the cathode channel configurations of the unit cell adopted for this study are discussed using GC measurement and visualization experiment of cathode flooding. It is found that the amount of oxygen consumption is very sensitive to various operating conditions of the fuel cell and was much affected by the flooding occurrence in cathode channels.

The Effects of Loranthus parasiticus Merr. on Cell Cycle and Expression of Related Genes in HepG2 Cell (상기생(桑寄生)이 HepG2 cell의 세포분열 및 관련유전자 발현에 미치는 영향)

  • Objectives : The aim of this study was to evaluate the effects of Loranthus parasiticus Merr. on cell cycle and expression of related genes in HepG2 cells. Methods : The MTT assay, cell counting assay, $[^3H]-Thymidine$ incorporation assay, flow cytometric analysis, quantitative RT-PCR and western blot assay were studied. Results : In the water extract of Loranthus parasiticus Merr., inhibition of cell proliferation and DNA synthesis in HepG2 cells was seen. These inhibitory effects were due to inhibition of G l-S transition in cell cycle. After treatment with the extract, expression of cyclin D1(G1 check point related gene) was inhibited particularly in dose-dependent and time-dependent manners. Conclusion : These results suggest that the inhibition of cell cycle progression by Loranthus parasiticus Merr. in HepG2 cell is due to suppression of cyclin D1(G1 check point related gene) mRNA expression and protein synthesis.
