Acute viral infection or vaccination generates highly functional memory CD8 T cells following the Ag resolution. In contrast, persistent antigenic stimulation in chronic viral infection and cancer leads to a state of T-cell dysfunction termed T-cell exhaustion. We and other have recently identified a novel subset of exhausted CD8 T cells that act as stem cells for maintaining virus-specific CD8 T cells in a mouse model of chronic lymphocytic choriomeningitis virus infection. This stem cell-like CD8 T-cell subset has been also observed in both mouse and human tumor models. Most importantly, in both chronic viral infection and tumor models, the proliferative burst of Ag-specific CD8 T cells driven by PD-1-directed immunotherapy comes exclusively from this stem cell-like CD8 T-cell subset. Therefore, a better understanding of the mechanisms how CD8 T-cell subsets are regulated during chronic viral infection and cancer is required to improve the current immunotherapies that restore the function of exhausted CD8 T cells. In this review, we discuss the differentiation of virus-specific CD8 T cells during chronic viral infection, the characteristics and function of CD8 T-cell subsets, and the therapeutic intervention of PD-1-directed immunotherapy in cancer.
A study on the continuous fermentation with cell recycle by a tower fermentor to produce ethanol has been carried out. ethanol fermentation was conducted with flocculating yeast strain, Saccharomyces cerevisiae TS4, to compare the ethanol productivity with conventional continuous process. Employing a 15% glucose feed, a cell density of 50 g/l was obtaind. The ethanol productivity of the cell recycle system was found to be 26.5g EtOH/1-hr, which was nearly 7.5 times higher than the conventional continuous process without cell recycle. A cell recycle ratio of 7 to 8 resulted in the highest ethanol productivity and cell concentration. Thus the cell recycle ratio was found to be a key factor in controlling the production of clarified overflow liquid. An aeration rate above 3.8 $\times$ 10-3 VVM seemed to decrease the ethanol productivity. The continuous fermentation with cell recycle was successfully used in the separation of cells from fermentation broth with enhancement of mixing in the tower fermentor.

In general, cellular manufacturing system can be constructed by the following two steps. The first step forms machine cells and part families, and the second step determines cell layout based on the result of first step. Cell layout has to be considered when cell is formed becauese the result of cell formation affects it. This paper presents a cell formation algorithm and proposes an integrated mathematical model for cell formation and cell layout. The cell formation algorithm minimizes the number of exceptional element in cellular manufacturing system. New concept for similarity and incapability is introduced, based on machine-operation incidence matrix and part-operation incidence matrix. One is similarity between the machines, the other is similarity between preliminary machine cells and machines. The incapability identifies relations between machine cells and parts. In this procedure, only parts without an exceptional element are assigned to machine cell. Bottleneck parts are considered with cell layout design in an integrated mathematical model. The integrated mathematical model determines cell layout and assigns bottleneck parts to minimize the number of exceptional element and intercell moving distance, based on linearixed 0-1 integer programming. The proposed algorithm is illustrated by using numerical examples.

Although three different research groups have reported successful derivations of human somatic cell nuclear transfer-derived embryonic stem cell (SCNT-ESC) lines using fetal, neonatal and adult fibroblasts, the extremely poor development of cloned embryos has hindered its potential applications in regenerative medicine. Recently, however, our group discovered that the severe methylation of lysine 9 in Histone H3 in a human somatic cell genome was a major SCNT reprogramming barrier, and the overexpression of KDM4A, a H3K9me3 demethylase, significantly improved the blastocyst formation of SCNT embryos. In particular, by applying this new approach, we were able to produce multiple SCNT-ES cell lines using oocytes obtained from donors whose eggs previously failed to develop to the blastocyst stage. Moreover, the success rate was closer to 25%, which is comparable to that of IVF embryos, so that our new human SCNT method seems to be a practical approach to establishing a pluripotent stem cell bank for the general public as well as for individual patients.
In this study, the effect of solar cell cover glass on the solar cell performance is evaluated. Silicon solar cell (active area:4*6cm, efficiency:12.6% at AMO condition) is used for this study. ITO(Indium tin Oxide) film thickness of the ITO/AR/substrate glass/solar cell structure samples are $40{\AA}$, $60{\AA}$, $160{\AA}$, $240{\AA}$ respectively. The solar cell maximum output power on the stacking structure variations showed 465mW in the AR/ITO/substrate glass/solar cell, and minimum output power showed 403mW in the AR/substrate glass/solar cell. The maximum output power of the solar cell on the ITO thickness variations of the ITO/AR/substrate glass/solar cell showed 460mW at $40{\AA}$ then decrease output power as ITO thickness increase. For environment tests, all samples are exposed UV light in the vacuum chanber. The output power degradation of AR(UVR)/substrate glass/solar cell stacking structure is small compared with ITO/AR(UVR)/substrate glass/solar cell stacking structure.
Cord blood (CB) has been used as an important and ethical source for hematopoietic stem cell transplantation (SCT) as well as cell therapy by manufacturing mesenchymal stem cell, induced pleuripotential stem cell or just isolating mononuclear cell from CB. Recently, the application of cell-based therapy using CB has expanded its clinical utility, particularly, by using autologous CB in children with refractory diseases. For these purposes, CB has been stored worldwide since mid-1990. In this review, I would like to briefly present the historical development of clinical uses of CB in the fields of SCT and cell therapy, particularly to review the experiences in Korea. Furthermore, I would touch the recent banking status of CB.
To identify the component(s) involved in cell cycle control in the protozoan Giardia lamblia, cells arrested at the G1/S- or G2-phase by treatment with nocodazole and aphidicolin were prepared from the synchronized cell cultures. RNA-sequencing analysis of the 2 stages of Giardia cell cycle identified several cell cycle genes that were up-regulated at the G2-phase. Transcriptome analysis of cells in 2 distinct cell cycle stages of G. lamblia confirmed previously reported components of cell cycle (PcnA, cyclin B, and CDK) and identified additional cell cycle components (NEKs, Mad2, spindle pole protein, and CDC14A). This result indicates that the cell cycle machinery operates in this protozoan, one of the earliest diverging eukaryotic lineages.
The direct carbon fuel cell (DCFC) has attracted researcher's attention recently, due to its high conversion efficiency and its abundant fuel, carbon. A DCFC mathematical model has developed in two-dimensional, lab-scale, and considers Boudouard reaction and carbon monoxide (CO) oxidation. The model simulates the CO production by Boudouard reaction and additional electron production by CO oxidation. The Boudouard equilibrium strongly depends on operating temperature and affects the amount of produced CO and consequentially affects the overall fuel cell performance. Two different operating temperatures (973 K, 1023 K) has been calculated to discover the CO production by Boudouard reaction and overall fuel cell performance. Moreover, anode thickness of the cell has been considered to find out the influence of the Boudouard reaction zone in fuel cell performance. It was found that in high temperature operating DCFC modeling, the Boudouard reaction cannot be neglected and has a vital role in the overall fuel cell performance.
Autoimmune diseases are caused by a dysfunction of the acquired immune system. In a subset of autoimmune diseases, B cells escaping immune tolerance present autoantigen and produce cytokines and/or autoantibodies, resulting in systemic or organ-specific autoimmunity. Therefore, B cell depletion with monoclonal Abs targeting B cell lineage markers is standard care therapy for several B cell-mediated autoimmune disorders. In the last 5 years, genetically-engineered cellular immunotherapies targeting B cells have shown superior efficacy and long-term remission of B cell malignancies compared to historical clinical outcomes using B cell depletion with monoclonal Ab therapies. This has raised interest in understanding whether similar durable remission could be achieved with use of genetically-engineered cell therapies for autoimmunity. This review will focus on current human clinical trials using engineered cell therapies for B cell-associated autoimmune diseases.
It is known that mast cells (MCs) are increased in H. pylori-infected gastritis and its increase is mediated by stem cell factor (c-kit ligand). To determine the mechanism of mast cell recruitment and activation by stem cell factor, weinvestigated the expression of stem cell factor receptor (c-kit) in H. pylori-positive and -negative gastric mucosa. Biopsy specimens from 16 H. pylori-negative and 20 positive subjects were examined. H. pylori infection in gastric mucosa was examined by the Warthin-Starry method. MC and c-kit were identified by immunohistochemisty, using a monoclonal antihuman MC tryptase antibody and a polyclonal anti-human c-kit antibody. Densities of MC and c-kit positive cell were measured by a computerized image analysis system. MCs were detected in the lamina propria of both H. pylori-positive and -negative gastric mucosa. Densities of MC and c-kit positive cell were significantly greater in H. pylori-positive than -negative subjects. c-kit was located on the surface of MCs. These results indicate that stem cell factors may be one of the factors involved in mast cell increase and that they activate mast cells by binding with c-kit.
