Micromorphological changes in waterlogged archaeological woods excavated from Sweden and Germany were investigated. Especially bacterial attacks on those wood samples under near anaerobic conditions were examined by transmission electron microscopy(TEM). The major feature of micromorphological alterations in those wood samples was the preferential destruction of secondary wood cell wall. In contrast, the middle lamella was not extensively degraded. Three distinct degradation patterns by bacteria were observed : erosion, cavitation and tunnelling bacteria. Erosion and cavitation bacteria attacked primarily $S_2$ layer, whereas tunnelling bacteria made the tunnel-like degradation along the $S_1$ layer. Tunnelling bacteria, in some samples, were able to degrade tunnel in the lignin-rich areas, such as middle lamella, suggesting that these bacteria had the capacity to degrade the lignin. IR spectra indicate that hemicellulose and cellulose in the waterlogged woods were preferentially decomposed. Breakdown of the lignin, on the other hand, was much slower.
The three-dimensional turbulent flow in curved pipes susceptible to flow-accelerated corrosion has been analyzed numerically to predict the pressure and shear stress distributions on the inner surface of the pipes. The analysis employs the body-fitted non-orthogonal curvilinear coordinate system and a standard $ {\kappa}-{\varepsilon}$ turbulence model with wall function method. The finite volume method is used to discretize the governing equations. The convection term is approximated by a high-resolution and bounded discretization scheme. The cell-centered, non-staggered grid arrangement is adopted and the resulting checkerboard pressure oscillation is prevented by the application of a modified version of momentum interpolation scheme. The SIMPLE algorithm is employed for the pressure and velocity coupling. The numerical calculations have been performed for two curved pipes with different bend angles and curvature radii, and discussions have been made on the distributions of the primary and secondary flow velocities, pressure and shear stress on the inner surface of the pipe to examine applicability of the present analysis method. As the result it is seen that the method is effective to predict the susceptible systems or their local areas where the fluid velocity or local turbulence is so high that the structural integrity can be threatened by wall thinning degradation due to flow-accelerated corrosion.
In order to investigate the decay process of wood treated with preservative, waterproofing agent and their compound systems, a full-cell process was applied to impregnate the sapwood of poplar (Populus cathay) at paraffin wax emulsion concentrations of 0.5% and 2.0%, Copper Azole (CA) concentrations of 0.3% and 0.5%, and their four compound systems, respectively. Leaching tests and laboratory decay resistance against the white-rot fungus Corious versicolor (L.) Murrill for treated wood were carried out according to the America Standard E11-06 and China Standard GB/T 13942.1-2009. At certain time intervals during the decay test, samples were characterized by Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction technique (XRD) to investigate the time-dependent changes of chemical components and crystalline structure, thus clarifying the decay mechanisms. The results suggested that white-rot fungi degrade hemicellulose and lignin in the wood cell wall first, followed by a simultaneous degradation of polysaccharides and lignin. Besides, CA could not only slower the decomposition of both hemicellulose and lignin, but also reduce the degradation amount of hemicellulose. However, paraffin wax emulsion at high concentration had a negative effect on the impregnation of CA for the compound system treated wood.

The experiment of silage for preservation of fresh Italian ryegrass (Lolium multiflorum) was carried out to examine whether the fermentation quality and microbial degradation in the rumen can be altered by the treatment of amino acids fermentation byproduct (AFB). The plant was ensiled for 40 days with 4 treatments of different ratios of AFB and sugarcane molasses (SCM) mixture. The treatment 2 (T2, AFB:SCM=100:0) and treatment 3 (T3, AFB:SCM=40:60) silages showed higher (p<0.05) concentrations of lactic acids, lower (p<0.05) pH and dry matter (DM) losses than the Control (T1, none additive) and treatment (T4, AFB:SCM=0:100) silages. The treatments 2 and 3 contained higher (p<0.05) DM and crude protein contents in silages compared to treatments 1 and 4 silages. The NDF, ADF and cellulose contents were also lower (p<0.05) in T2, T3 and T4 silages than T1 silage and fresh material before ensiled. The in situ rumen DM, NDF, ADF, hemicellulose and cellulose degradability was also higher (p<0.05) in T2, T3 and T4 silages than T1 silage, while the highest improvement was achieved with addition of AFB:SCM at level of 40:60 at ensiling. The result in this study indicates that the addition of AFB and SCM additives improved the silage fermentation and cell wall degradability of Italian ryegrass silage.
Several researchers have demonstrated that the rumen microbial community rapidly adapts to saponins and proposed interval feeding to prevent this rapid adaptation. An in vivo experiment was carried out to examine the effect of daily versus application every third day (interval feeding) of Sapindus rarak saponins (SE) on rumen fermentation end products, protozoal counts and nutrient digestibility. Thirty sheep were allocated into 5 groups. Sheep were fed daily or every third day with two levels of SE (0.48 and 0.72 g/kg body mass). One group received no saponin and served as control. All sheep received the same diet, a mixture of elephant grass and wheat pollard (65:35 w/w). Independent of the feeding regime and the level of inclusion, the addition of SE decreased protozoal counts and rumen ammonia concentrations (p<0.01). Microbial N supply and N retention were not affected by the high feeding regime. Daily feeding negatively influenced rumen xylanase and cellulase activity, but only when the high level of saponins was fed. However, these negative effects on rumen cell wall degradation were not reflected in decreasing total tract digestibility of the organic matter or the plant cell walls. Our results show that rumen microorganisms do not rapidly adapt to S. rarak saponins.

We investigated the changes of fruit quality parameters, polysaccharide contents and cell wall components during maturation and ripening of two Korean pear cultivar 'Hanareum' and 'Manpungbae' compared with 'Niitaka' pear (Pyrus pyrifolia Nakai) which showed different physiological maturity based on days after full bloom (DAFB). Flesh firmness decreased continuously with fruit development and maturation, reaching a final level of 29.4, 33.5, and 27.4N at maturity in 'Hanareum' (127 DAFB), 'Manpungbae' (163 DAFB), and 'Niitaka' (170 DAFB), respectively. The level of ethylene production was very low in early season 'Hanareum' pear which showed at most 0.39 ${\mu}L{\cdot}L^{-1}$ at maturity and no ethylene was detected in 'Manpungbae' and 'Niitaka' at maturity. Fructose was the most abundant soluble sugar during fruit maturation in the pears tested and an increase of sucrose was observed during fruit ripening in the Asian pears commonly. Ethanol insoluble solids (EIS) content decreased gradually with different levels among the pear cultivars as fruit ripens consisted of 10.79, 12.72, and 12.75 $mg{\cdot}g^{-1}$ FW. The amount of total soluble polyuronides was higher in early season cultivars 'Hanareum' than those of mid-season cultivar 'Manpungbae' and 'Niitaka'. In 'Niitaka' which harvested most late season, the level of 4% KOH soluble hemicelluloses was lower than 'Hanareum' and 'Manpungbae' and maintained constantly during fruit ripening period. Cellulosic residues were determined high level in 'Niitaka' which showed 612.33 ${\mu}g{\cdot}mg^{-1}$ EIS at maturity when compared with 'Hanareum' (408.0 ${\mu}g{\cdot}mg^{-1}$ EIS) and 'Manpungbae' (538.67 ${\mu}g{\cdot}mg^{-1}$ EIS). The main constituents of cell wall neutral sugars which consisted of arabinose, xylose, galactose, and glucose were decreased gradually with onset of fruit ripening regardless of cultivar. Arabinose which was predominant in 'Hanareum' pear decreased at the last stage of ripening, but the changes of cell wall neutral sugar during ripening were not occurred in 'Niitaka' pear. The change of molecular mass distribution in water soluble pectin observed dominantly at the early stage of fruit development. Depolymerization of 4% KOH-soluble hemicelluloses and degradation of xyloglucan showed in early-season cultivar 'Hanareum' during fruit maturation, and degradation of those fractions were detected only at the early stage fruit development in mid-season cultivar 'Manpungbae' and 'Niitaka'. The molecular mass profile of CDTA soluble pectin, $Na_2CO_3$-SP and 24% KOH soluble hemicelluloses showed no significant change during fruit maturation regardless of cultivar.

Objective: Lignin plays a relevant role in the inhibition of cell wall (CW) structural carbohydrate degradation. Thus, obtaining accurate estimates of the lignin content in tropical plants is important in order to properly characterize the mechanism of lignin action on CW degradation. Comparing conflicting results between the different methods available for commercial use will bring insight on the subject. This way, providing data to better understand the relationship between lignin concentration and implications with tropical forage degradation. Methods: Five grass species, Brachiaria brizantha cv $Marand{\acute{u}}$, Brachiaria brizantha cv $Xara{\acute{e}}s$(MG-5), Panicum maximum cv Mombaça, Pennisetum purpureum cv Cameroon, and Pennisetum purpureum cv Napier, were harvested at five maturity stages. Acid detergent lignin (ADL), Klason lignin (KL), acetyl bromide lignin (ABL), and permanganate lignin (PerL) were measured on all species. Lignin concentration was correlated with in vitro degradability. Results: Highly significant effects for maturity, lignin method and their interaction on lignin content were observed. The ADL, KL and ABL methods had similar negative correlations with degradability. The PerL method failed to reliably estimate the degradability of tropical grasses, possibly due to interference of other substances potentially soluble in the $KMnO_4$ solution. Conclusion: ADL and KL methods use strong acid ($H_2SO_4$) and require determination of ash and N content in the lignin residues, therefore, increasing time and cost of analysis. The ABL method has no need for such corrections and is a fast and a convenient method for determination of total lignin content in plants, thus, it may be a good option for routine laboratory analysis.

This study was performed on 16 patients diagnosed with tsutsugamushi disease and cerebral infarction from January 2007 to December 2015. An acute cerebral infarction was diagnosed by brain MRI and MRA. Tsutsugamushi disease was diagnosed using a polymerase chain reaction. To distinguish the difference between the generalized cerebral infarction and infarction with tsutsugamushi disease, the blood pressure and body temperature were measured uponadmission. In general, the blood pressure increases during an acute cerebral infarction. Interestingly, in this study, 12 patients showed a systolic blood pressure less than 130 mmHg uponadmission. The location of the cerebral infarction and whether single or multiple cerebral infarction were examined. Thirteen patients had a cerebral infarction in anterior circulation and 3 patients developed in posterior circulation. To evaluate the coagulation disorders, prothrombin time (PT), activated partial thromboplastin time (aPTT), D-dimer, fibrinogen, fibrin degradation product (FDP). D-dimer, which is generally known to increase in an acute cerebral infarction, showed a significant increase in the 13 patients. Fibrin degradation products (FDP) showed a significant increase in 15 patients. The pathophysiological mechanism of tsutsugamushi disease is known as vasculitis, which may result in an endothelial cell injury and proliferation of the endothelial wall, which may lead to a cerebral infarction accompanied by coagulopathy. Without endothelial cell damage and proliferation, a vasospasm caused by vasculitis may cause vasoconstriction and cerebral infarction.
This study was carried out to investigate changes in the flesh firmness, evolution of ethylene, cell wall components, and degradation and solubilization of polyuronide(PU) and polysaccharide(PS) in green(GP) and mature persimmon(MP) fruits according to testing time of ethylene(50${\mu}\ell$ㆍL$^{-1}$ ). When ethylene was treated in GP and MP, flesh firmness rapidly decreased and it was decreased more GP than MP. When ethylene were treated for 12 hours in GP, production of ethylene began after 3 days. The amount of ethylene product was maximum 16,000 ${\mu}\ell$ㆍL$^{-1}$ at 24 hours of ethylene treatment. However, ethylene was not producted until 7 days after 24 hours ethylene treatment at MP. The content of pectic substances decreased in the distilled- water, 0.05M $Na_2$CO$_3$,4M and 8M KOH-soluble fractions during softening according to increasing time of ethylene treatment. Arabinose and galactose were the major non-cellulosic neutral sugars in the 0.05M CDTA and 0.05M $Na_2$CO$_3$-soluble pectic fractions. Glucose, galactose and xylose were the major non-cellulosic neutral sugars in the 4M KOH- soluble hemicellulosic fraction. High molecular of PU and PS were degraded and solubilized in the distilled-water, 0.05M CDTA 0.05M $Na_2$CO$_3$ and 4M KOH-soluble fractions during time of ethylene treatment.
Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.
