Effect of single nucleotide polymorphisms on intramuscular fat content in Hungarian Simmental cattle

  • Objective: To estimate effect of single nucleotide polymorphisms on the intramuscular fat content (IMF) of Hungarian Simmental bulls. Methods: Genotypes were determined on high-density Illumina Bovine DNA Chip. After slaughtering of animals, chemical percentage of intramuscular fat was determined from longissimus dorsi muscle. A multi-locus mixed-model was applied for statistical analyses. Results: Analyses revealed four loci (rs43284251, rs109210955, rs41630030, and rs41642251) to be highly associated ($-{\log}_{10}P$>12) with IMF located on chromosome 1, 6, 13, and 17, respectively. The frequency of their minor alleles was 0.426, 0.221, 0.162, and 0.106. Conclusion: The loci above can be useful in selection programs and gives the possibility to assist selection by molecular tools.

Distribution of the serum Ig G titers to whole cell and leukotoxin of Mannheimia haemolytica Al in Holstein and Korean indigenous cattle slaughtered in abattoir

  • A total of 419 slaughtered cattle were used to investigate the serum Ig G titers to the Mannheimia haemolytica Al whole cell and leukotoxin recognized with important virulence factor in bacterial pathogenesis. Data obtained in this study were represented with average absorbance${\pm}$standard deviation. Serum Ig G titers were detected with the ranges from 0.1 to 0.5 at 490nm. Whole cell titers were higher than leukotoxin antibody on the whole. Antibody titers of slaughtered cattle between races, ages have no significant difference but gradual decrease under aging in dairy cow for whole cell (decline mean titer from 0.29 to 0.27 according to age) was undertaken. Holstein bulls shipped from Seoul province had a significantly lower Ig G titers than those from another ones (p<0.05).

The Effects of Genetic Groups, Nutrition, Finishing Systems and Gender of Brazilian Cattle on Carcass Characteristics and Beef Composition and Appearance: A Review

  • The aim of this review is to address some characteristics that influence meat quality. Genetic groups, nutrition, finishing systems and gender are the major factors that change carcass characteristics, chemical composition and fatty acid profile. Genetic groups that have zebu genes in their composition show higher hot carcass dressing than genetic groups without zebu genes. Genetic groups that have European breeds in their composition have higher marbling scores. On the other hand, genetic groups that have zebu breeds show low marbling scores. Bulls finished in feedlots present higher final weight than steers, cull cows and heifers. Fat thickness is one of the principal parameters that are affected by different gender. Cull cows (4.72 mm) and heifers (4.00 mm) present higher values than bulls (1.75 mm) and steers (2.81 mm). The major effects observed by different systems of termination are fat thickness and marbling. Crude protein presents variation due to nutrition. Nutrition influences variation of fatty acid profile. Genetic groups also influence fatty acid profile. Genetic groups that have zebu genes in their composition show high percentage of PUFA. The major class of fatty acids that is changed with nutrition is PUFA. The better ratios of PUFA/SFA and n-6/n-3 are found in Longissimus muscle of animals finished in pasture systems.

Association of Candidate Genes with Production Traits in Korean Dairy Proven and Young Bulls

  • This study was performed to offer effective basic data for selection and improvement of Korean dairy cattle through identifying distributional properties among candidate genes (bovine butyrophilin, signal transducers and activators of transcription 5a, and prolactin hormone). In this study, polymorphisms of candidate genes were identified and the relationships between loci and production traits of each gene were analyzed using frozen semen of Holstein bulls (19 proven and 77 candidates). In butyrophilin (BTN) locus, polymorphisms information contents (PIC) value of BTN2 (0.372) was higher than those of others (BTN1; 0.155, BTN3; 0.254, BTN4; 0.169). As a result of analysis of genotyping STAT5a, using single stranded conformational polymorphism (SSCP) method and microsatellite locus, PIC values were 0.189 and 0.457, respectively. And PIC value of prolactin hormone gene was 0.176. In the relationships between genotypes and production traits, BTN3 was associated with 305-day production traits (p<0.05). PTAs for B allele were such as 110.43, 88.28 and 75.25 in BTN1, 3, 4 and these values were higher than those of A allele, but in the case of BTN2, A allele with 154.19 was higher than that of B allele. The results obtained from using candidate genes may be used as an useful index for the genetic improvement of dairy cattle population in Korea, and further studies are needed.

Progesterone 측정을 위한 액상(液相) 효소면역측정법(酵素免疫測定法)의 최적조건에 관한 연구 (Optimization of liquid phase enzyme immunoassay for determining of progesterone)

  • This study was carried out to develop an effective liquid-phase double antibody enzyme immunoassay for determining of progesterone. The optimum conditions of assay system, 1st and 2nd antibodies, enzyme conjugate, and time reaction were invested. The bovine plasma progesterone level in dairy cattle and korean native bulls were also analyzed. The results obtained were as follows; 1. The reproducibility of petroleum ether was superior to that of ethyl ether as extract solvent of progesterone in plasma. 2. The optimum dilution rate of 1st and 2nd antibody was 30,000 and 10 times, respectively. Affer the reaction of enzyme conjugate to progesterone 1st antibody, and then 2nd antibody competition reaction was enough for over 1hr. 3. Average plasma progesterone level in 4 pregnant and 9 nonpregnant Holstein was $2.5{\pm}0.5$ and $0.7{\pm}0.2ng/m{\ell}$, respectively. Average plasma progesterone level of 10 Korean native bulls was $0.1{\pm}0.001ng/m{\ell}$ From these results, by using liquid phase double antibody enzyme immunoassay for progesterone is applicable to detect of early pregnancy diagnosis, factorial analysis of reproductive disorder, and also reproductive physiological function such as monitoring of cyclicity during the post-partum period.

Estimation of the genetic milk yield parameters of Holstein cattle under heat stress in South Korea

  • Objective: The objective of this study was to investigate the genetic components of daily milk yield and to re-rank bulls in South Korea by estimated breeding value (EBV) under heat stress using the temperature-humidity index (THI). Methods: This study was conducted using 125,312 monthly test-day records, collected from January 2000 to February 2017 for 19,889 Holstein cows from 647 farms in South Korea. Milk production data were collected from two agencies, the Dairy Cattle Genetic Improvement Center and the Korea Animal Improvement Association, and meteorological data were obtained from 41 regional weather stations using the Automated Surface Observing System (ASOS) installed throughout South Korea. A random regression model using the THI was applied to estimate genetic parameters of heat tolerance based on the test-day records. The model included herd-year-season, calving age, and days-in-milk as fixed effects, as well as heat tolerance as an additive genetic effect, permanent environmental effect, and direct additive and permanent environmental effect. Results: Below the THI threshold (${\leq}72$; no heat stress), the variance in heat tolerance was zero. However, the heat tolerance variance began to increase as THI exceeded the threshold. The covariance between the genetic additive effect and the heat tolerance effect was -0.33. Heritability estimates of milk yield ranged from 0.111 to 0.176 (average: 0.128). Heritability decreased slightly as THI increased, and began to increase at a THI of 79. The predicted bull EBV ranking varied with THI. Conclusion: We conclude that genetic evaluation using the THI function could be useful for selecting bulls for heat tolerance in South Korea.

Effects of Feeding Patterns and Sexes on Growth Rate, Carcass Trait and Grade in Korean Native Cattle

  • The objectives of this study were to examine growth performance and meat quality by three different feeding patterns in Korean native cattle (KNC). In each of 3 years, fifteen KNC were randomly assigned in a (3 feeding management)${\times}$(3 sex) factorial design experiment; thus, in total, there were 5 animals in each of the 9 treatments. The three feeding management treatments were longterm (24 month) restriction feeding (LTFR), long-term restriction feeding-hormone implant (LTFR-tH), and short-term (18 month) nonrestriction feeding (STFNR). Three sexes were bull, steer, and heifer. Concentrate diet was fed restriction-feeding method based on body weight in LTFR and LTFR-tH. However, the diet was fed ad libitum in STFNR. Hormonal implantation was made three times with M-$PO^{TM}$ for bulls and with F-$TO^{TM}$ for heifers at 18, 20, 22 month of age in LTFR-tH. Animal were purchased from the local cattle market and managed in two local farms and at the university research unit. Animals were slaughtered at 24 months for long-term trial and at 18 month for short-term trial. The growth rate was the highest in bulls and the lowest in heifers. However, the differences were diminished in F-$TO^{TM}$ implanted heifers. The average daily gain was high in STFNR due to ad libitum feeding. The carcass grade was similar among the treatments on percentage bases. Hormonal implants improved significantly the meat quality grade in all sexes. Castration increased body fat content and improved meat quality grade by intramuscular fat deposition. In conclusion, long-term feeding and hormone treatment increased meat quality grade more than short-term feeding. However, ADG was higher in the short-term trial although feedefficiency was lower.

REPRODUCTION AND CALF GROWTH IN BRAHMAN CROSSBRED AND SOUTH EAST ASIAN CATTLE IN PAPUA NEW GUINEA

  • Reproduction and growth to weaning were compared for Brahman crossbred (BX) and a local strain of South-East Asian cattle, "Javanese Zebu" (JZ) and their reciprocal crosses at "Erap" in the humid equatorial lowlands of Papua New Guinea. Forty heifers of each breed were mated continuously, half to bulls of each breed, for five years. BX calved first at 35 months while JZ calved at 31 months. Subsequent calving intervals were very short, at 370 and 341 days. JZ cows weighed about two thirds of the BX cattle at each stage of reproduction. Birth weights and growth to weaning were : BX 35 kg and 0.68 kg/d ; BX male $\times$ JZ female 29.3 kg and 0.53 kg/d ; JZ male $\times$ BX female 30.8 kg and 0.61 kg/d ; JZ 25 kg and 0.50 kg/d. The combination of small cow size, short calving interval and rapid calf growth resulted in the BX male $\times$ JZ female being the most efficient producer, in kg of calf weaned per cow mated per year while the reciprocal cross was the least efficient ; both straight-breds were equal and intermediate. These data show that indigenous equatorial cattle may not be inferior under good grazing conditions. For all traits, breed interactions (heterosis) was small and non-significant.

Pedigree Indexing of Imported American Brahman Breeder Cattle in the Philippines

  • Pedigree data were used to predict the performance of 1,166 imported Americas Brahman breeder cattle, composed of 104 bulls and 1,062 heifers with an average age of 24.4 months (range of 17 to 40 months). A total of 13 full sib, 10 maternal half sib, and 228 paternal half sib groups were identified, each with average group size of 2.08, 2.00, and 4.49, respectively. Only 758 (64.9% of total) imported cattle were found to have at least one ancestor with expected progeny differences (EPDs) reported in the Spring 1995 Brahman Sire Summary. Moderate average accuracy values of .71, .69, .52, and .52 for birth weight, weaning weight, yearling weight, and maternal milk, respectively, were noted for EPDs of the ancestors. Prediction equations were derived by multiple regression analysis of available EPDs of sire, paternal grand sire, and maternal grand sire. Based on pedigree indexes that involve various combinations of available ancestral information, the average predicted EPDs (lbs) for imported cattle were $1.76{\pm}0.54$, $14.93{\pm}4.86$, $25.10{\pm}9.50$, and $5.86{\pm}2.08$ for birth weight, weaning weight, yearling weight, and maternal milk, respectively. Significant correlations (p < .05) were also found between sire and son EPDs (+.27) for yearling weight; and between sire and paternal grand sire EPDs for birth weight (+.34), weaning weight (+.51), yearling weight (+.49), and maternal milk (+.55).

한우 Band 3 및 CHS 유전병의 분자유전학적 기초연구 (A Fundamental Genetic Study for Identifying Band 3 and CHS Genetic Diseases in Korean Cattle (Hanwoo))

  • 한국의 소 사육 농가에서 유전병을 monitoring 하는 체계가 현재 존재하지 않기 때문에 정확히 한우에서 어떤 유전병이 존재하는지 알 수가 없는 실정이다. 최근에 일본에서는 흑모화우에 존재하는 유전병을 제거하기 위해 많은 노력을 들이고 있다. 본 연구는 일본의 흑모화우에 존재하는 유전병인 Band 3와 CHS의 존재 유무를 혈연관계가 없는 한우 22개체를 이용하여 PCR-RFLP 방법으로 확인하였다. 비록 한우에서 Band 3와 CHS 유전병을 가진 개체는 나오지 않았으나 본 연구에서 확립된 방법을 한우 보증 종모우와 후보 종모우에서 확인하여 유전병을 가진 유전자가 다음 세대로 전달되는 것을 막을 필요성이 있다.
