SELECTIVE TOXICITY OF CHRONIC LEAD INGESTION TO CENTRAL CATECHOLAMINERGIC NERVOUS SYSTEM IN RATS

  • The selective toxicity of lead was tested in central catecholaminergic nervous system of postnatally lead exposed rats. Three groups of animals were prepared; 1) rats exposed to low dose of lead (0.05%PbAc); 2) rats exposed to high dose of lead(0.2%PbAc); 3) age-matched normal control rats. At2, 4, 6 and 8 weeks of age brain and body weight gain, and lead concentrations in brain tissues were measured. At the same ages tyrosine hydroxylase and Na-K ATPase activities were measured in the 4 brain areas of each animal. Body weight gain was decreased after 6 weeks of age in rats exposed to high dose of lead. Concentrations of lead in whole brain tissues were increased from 0.37 to 0.83 (ng/mg wet tissue) in these animals. in lead exposed rats, tyrosine hydroxylase activities were higher but Na-K ATPase activities were lower than those of age-matched control animals. Brain areas where tyrosine hydroxylase activities were detected without concomitant changes of Na-K ATPase activities were pons-medulla (2 weeks of age) and telencephalon (6 weeks of age) in rats exposed to low dose of lead, and those in rats exposed to high dose of lead were midbrain (4 and 6 weeks of age). These data indicate that catecholaminergic nervous system in the brain areas described above could selectively be affected by lead.

선천성 고혈압 쥐에서 시상하부 카테콜아민성 신경계에 미치는 난소 스테로이드 호르몬의 영향 (Effect of Ovarian Steroid Hormones on Catecholaminergic Nervous System in the Hypothalamus of SHR)

  • A question whether abnormal responsiveness of hypothalamic catecholaminergic nervous system to ovarian steoid hormones in spontaneously hypertensive rats (SHR) exist was investigated. Four groups of experimental animals were prepared for SHR and normotensive Wistar rats (NW) respectively: 1) intact, 2) ovariectomized (OVX+V), 3) ovariectomized and estrogen treated (OVX+E), 4) ovariectomized and estrogen plus progesterone treated (OVX+E+P) groups. Hypothalami from experimental animals were dissected out and used for determination of .alpha.-adrenergic receptor binding characteristics and catecholamine contents. Norepinephrine(NE) content and B$_{max}$ of $\alpha_1$-adrenergic receptors in hypothalami were greater in intact SHR than in intact NW, but dopamine(DA) content was lower in SHR than in NW. Neither contents of NE and DA nor binding characteristics of $\alpha_1$-adrenergic receptors were different in OVX+V and OVX+E group from intact group of both SHR and NW. Kd and B$_{max}$ of $\alpha_1$-adrenergic receptors in OVX+E+P was lower than that in intact SHR but not in NW. DA content was lower in OVX+E+P than in intact group of SHR and NW. The result of the present study indicates that there is an abnormal responsiveness of hypothalamic catecholaminergic nervous system to ovarian steroid hormones in SHR which may be one of genetically-determined factors probably not responsible for the development of hypertension.

봉독약침자극이 Catecholamine성 신경세포의 활성변화에 미치는 영향 (Effect of the bee venom aqua-acupuncture on the neuronal activities of catecholaminergic system in brainstem)

  • This study was designed to evaluate the effect of the bee venom(BV) aqua-acupuncture on the neuronal activities of catecholaminergic(tyrosine hydroxylase : TH, dopamine ${\beta}$ hydroxylase : D${\beta}$H) system in the brainstem. After the BV aqua-acupuncture was applied on Chok-Samni(ST36) and the gluteal part(Blank locus) in rats. Also, the number of colocalization between catecholamine containing neurons and Fos immunoreactive neurons were analyzed by using the double immunohistochemical technique. The results of the experiments were summarized as follows : 1. In DR and LC, Chok-Samni group and the Blank locus group showed more significant increase in the number of colocalization between TH containing neurons and Fos immunoreactive neurons than the control group. Furthermore, Chok-Samni group showed more significant increase than the Blank locus group. Also, in Arc, Chok-Samni group showed more significant increase than the Blank locus group and the control group. 2. In LC, Chok-Samni group showed more significant increase in the number of colocalization between D${\beta}$H containing neurons and Fos immunoreactive neurons than the Blank locus group and the control group. Also, in A5, Chok-Samni group and the Blank locus group showed more significant increase than the control group. Chok-Samni group showed more significant increase than the Blank locus group. However, there was no significant change in A7. Consequently, the BV aqua-acupuncture increased more potent the number of Fos immunoreactive neurons and the activity of catecholaminergic neurons. Furthermore, the BV aqua-acupuncture was more effective on Chok-Samni than Blank locus group. These results indicate that the BV aqua-acupuncture is very effective therapy to control pain. The therapeutic effect of BV aqua-acupunture may associated with the endogenous modulatory system such as catecholamine. Those data from the study can be applied to establish the effective treatment of the BV for pain control in the clinical field.

시호(柴胡)와 지골피(地骨皮) 합제(合劑)가 강제수영부하실험에서 HPA Axis와 카테콜라민 시스템에 미치는 영향 (Effects of Mixture of Bupleuri Radix and Lycii Radicis Cortex on the Change of HPA-Axis and Catecholamic System in the Forced Swimming Test)

  • Objectives : The effects of Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex on the change of the HPA-Axis system and the Catecholamic system was investigated. Methods : After performing the Forced Swimming Test(FST), the expressions of corticotropin releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN), and tyrosine hydroxylase(TH) in the ventral tegmental area(VTA) and locus coeruleus(LC) were measured by immunohistochemical method. Results : The duration of immobility in FST was significantly decreased in A 100(Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex, 100mg/kg) and A 400(Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex, 400mg/kg)(p<0.001). The expression of CRF in the PVN was shown the tendency to reduce in A100 and A400. The expression of c-Fos in the PVN was shown the tendency to reduce in A100 and A400. The expression of TH in the VTA was shown the tendency to reduce mildly in A100 and A400. The expression of TH in the LC was significantly reduced in A400(p<0.001). And the dose dependent reduction tendency was shown, respectively. Conclusions : According to the results above mentioned, the immobility, c-Fos and CRF expression was reduced at lower dose and was increased at higher dose. Therefore there is contradictory effects on the HPA Axis system in accordance with the dose. But in the effects on the catecholaminergic system, it significantly reduced the expression of TH in the LC. It was validated that the effect on the catecholaminergic system was ruled by Bupleuri Radix rather than Lycii Radicis Cortex via mainly the noradernergic system.

생지황(生地黃)이 강제수영부하시험에서 HPA Axis System과 Catecholaminergic System에 미치는 영향 (Effects of Rehmanniae Radix on HPA Axis System and Catecholamienrgic System in FST)

  • Object : The effects of Rehmanniae Radix(RR) on HPA Axis system and Catecholaminergic system was investigated. Methods : we performed the Forced Swimming Test(FST). Also the expression of Corticotropin -Releasing Factor(CRF), c-Fos and Tyrosine Hydorxylase(TH) was measured by immunohistochemical method at Paraventricular Nucleus(PVN), Locus Coeruleus(LC) and Ventral Tegmental Area(VTA). Results : 1. The duration of immobility in the Forced Swimming Test was significantly decreased in the Rehmanniae Radix 400 mg/kg groups in comparison with the control group(p<0.05). 2. Comparing to the control group, CRF expression was significantly decreased in the Rehmanniae Radix 400 mg/kg treated group in comparison with the control group(p<0.01). 3. c-Fos expression was significantly decreased at PVN in the Rehmanniae Radix 100 mg/kg treated group in comparison with the control group(p<0.01). 4. TH expression was significantly increased at LC in the Rehmanniae Radix 100 mg/kg and 400 mg/kg treated group in comparison with the control group(p<0.001). 5. TH expression was significantly decreased at VTA in the Rehmanniae Radix 100 mg/kg and 400 mg/kg treated group in comparison with the control group(p<0.001). Conclusion : According to the results, it can be considered that Rehmanniae Radix has antidepressant effect by showing the reduction of immobility in FST through the decreased expression of CRF, c-Fos in PVN and TH in VTA.

  • Channelopathies are a heterogeneous group of disorders resulting from the dysfunction of ion channels located in the membranes of all cells and many cellular organelles. These include diseases of the nervous system (e.g., generalized epilepsy with febrile seizures plus, familial hemiplegic migraine, episodic ataxia, and hyperkalemic and hypokalemic periodic paralysis), the cardiovascular system (e.g., long QT syndrome, short QT syndrome, Brugada syndrome, and catecholaminergic polymorphic ventricular tachycardia), the respiratory system (e.g., cystic fibrosis), the endocrine system (e.g., neonatal diabetes mellitus, familial hyperinsulinemic hypoglycemia, thyrotoxic hypokalemic periodic paralysis, and familial hyperaldosteronism), the urinary system (e.g., Bartter syndrome, nephrogenic diabetes insipidus, autosomal-dominant polycystic kidney disease, and hypomagnesemia with secondary hypocalcemia), and the immune system (e.g., myasthenia gravis, neuromyelitis optica, Isaac syndrome, and anti-NMDA [N-methyl-D-aspartate] receptor encephalitis). The field of channelopathies is expanding rapidly, as is the utility of molecular-genetic and electrophysiological studies. This review provides a brief overview and update of channelopathies, with a focus on recent advances in the pathophysiological mechanisms that may help clinicians better understand, diagnose, and develop treatments for these diseases.

흰쥐의 혈압 및 심박동수 조절에 대하여 Nucleus Tractus Solitarius 부위의 Serotonin성 기전의 역할 (Involvement of Serotonergic Mechanism in the Nucleus Tractus Solitarius for the Regulation of Blood Pressure and Heart Rate of Rats)

  • 혈압 및 심박동수 조절에 대한 serotonin성 기전의 역할을 흰쥐 뇌의 NTS에서 검토하고 catecholamine성 기전과 비교하여 다음과 같은 결과를 얻었다. 1) 5-HT를 NTS에 주사시 혈압 및 심박동수는 감소되었고, 5-HT 300 pmol을 NTS에 주사하였을 때 가장 현저하게 하강하였다. ${\alpha}-MNE$과 clonidine도 마찬가지로 혈압과 심박동수의 감소를 일으켰다. 5-HT의 효과는 5-HT 수용체 길항제인 ritanserin, methysergide 및 ketanserin의 전처치에 의하여 봉쇄되었다. 2) Reserpine과 6-OHDA를 전처치하였을 때는 5-HT에 의한 혈압 및 심박동수 감소가 현저하게 약화되었다. 3) 5,7-DHT 전처치 후에는 5-HT에 의한 혈압 및 심박동수 감소는 증가하였다. 한편 6-OHDA 전처치에 의하여서도 clonidine의 혈압 및 심박동수 감소 현상은 항진하였다. 4) 5,7-DHT와 6-OHDA를 i.c.v.로 전처치하였을 때 phenylephrine과 sodium nitropursside에 의한 압반사 감수성은 현저히 저하하였고, 이들을 NTS로 주사시도 압반사 감수성이 손상되었다. 5) Immunohistochemistry에 의해 NTS에서 catecholamine성 세포체, 신경축색 말단과 serotonin성 신경축색 말단을 관찰하였고, 6-OHDA 주사후에 catecholamine성 신경축색 말단의 varicosity는 현저히 감소하였다. 한편 5,7-DHT처치에 의하여서는 serotonin성 신경축색 말단의 varicosity가 현저히 감소하였다. 이상의 실험 결과, NTS에 있는 serotonin성 수용체의 활성에 의하여 (1) 혈압 및 심박동수 감소가 일어나며, (2) 이는 catecholamine성 신경계와 관련되어 야기되며, (3) serotonin과 catecholamine성 수용체는 NTS의 postsynaptic site에 존재하고, (4) serotonin과 catecholamine성 신경세포는 압반사 조절에 있어서 중요한 역할을 하는 것으로 사료된다.

  • Objectives : In this study the antidepressant effects of mixture of Citri Peticulatae Viride Pericarpium and Lycii Radicis Cortex on the change of HPA-Axis and Catecholamic system was investigated Methods : The forced swimming test(FST) was performed. The expression of corticotropin-releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN), and tyrosine hydroxylase(TH) in the ventral tegmental area(VTA) and locus coeruleus(LC) was measured with immunohistochemical method and the concentration of seum adrenocorticotropic hormone(ACTH) was measured with ELISA method. And the experimental groups were divided into the extraction after mixing(A) and mixture after extraction(B). The effects of both group were compared. Results : The duration of immobility in the forced swimming test was significantly decreased in the A400 group(P<0.01). The expression of CRF in PVN were significantly reduced in the A100, A400, B100, B400groups(P<0.001). but the expression of c-fos in PVN weren't reduced in all groups. And the concentration of ACTH in Plasma were significantly reduced in the A 100 group(P<0.01). The expression of TH in LC were significantly reduced in the A 400, B 100 and B400 groups(P<0.05~P<0.01). Conclusion : Mixture of Citri Peticulatae Viride Pericarpium and Lycii Radicis Cortex has antidepressant effects. But the difference between mixing and extracting methods was not shown.

LPS로 유발된 우울증에 대한 감초(甘草)의 효과 (Effect of Glycyrrhizae Radix Water Extract on the depression induced by LPS in Rats)

  • Objectives : Investigation of the antidepressant effect of Glycyrrhizae Radix (GR) through the anti-inflammatory effect. Methods : Depression in rats was induced by LPS (i,p.3days). The rats were treated with GR100 mg/kg (GR 100) or GR400 mg/kg (GR 400). The depressive immobility was examined with Tail Suspension Test(TST) and Forced Swimming Test(FST). The expression of nuclear factor-${\kappa}B$(NF-${\kappa}B)$, $I{\kappa}B$ was measured with western blotting. The concentration of corticosterone, cytokine in plasma was measured with ELISA. The expression of c-Fos in the paraventricular nucleus(PVN) and tyrosine hydroxylase(TH) in the locus coeluleus(LC) were measured with immunostaining method. Results : In the TST, GR400 group significantly decreased immobility time compared with the LPS group. In the FST, GR100, GR400 group significantly decreased immobility time comparing with the LPS group. c-Fos expression in GR100 and GR400 group was decreased comparing with the lipoplysaccharide(LPS) group. The $I{\kappa}B$ expression of GR100 and GR400 group was increased comparing with the LPS group. The level of corticosterone of GR100 group was decreased comparing with the LPS group. The concentration of cytokine of GR100 and GR400 group was decreased comparing with the LPS group. TH expression in the LC was increased in LPS group, but in GR100 and GR400 group was not shown significant decrease. Conclusion : According to this results obtained, GR has antidepressant effects by the anti inflammatory action through the suppression of HPA axis activity, not through the action against the catecholaminergic system.

측뇌실내 Taurine이 가토의 혈압 및 심박에 미치는 영향 (Influence of Intraventricular Taurine on the Cardiovascular System of the Rabbit)

  • Taurine은 유황 함유 아미노산으로서 뇌내에서 억제작용을 갖는 신경전달물질 역할을 하는것으로 알려져 있다. Taurine을 가토의 측뇌실내에 투여하여 혈압 및 심박에 미치는 영향을 검토하고 그 작용 기전을 규명코자 본 연구를 시행하여 얻은 연구결과는 다음과 같다. Taurine $(0.15{\sim}1.5\;mg)$은 urethane 마취 가토의 측뇌실내로 주사하였을때 혈압강하작용 및 심박감소작용이 현저하고 지속적인 용량반응 곡선을 나타내었다. 이와 동시에 상당한 호흡억제 작용도 관찰되었다. 그러나 동일량의 taurine을 정맥내에 투여시 혈압 및 심박에 아무런 영향을 미치지 못하였다. Taurine의 혈압 하강작용은 chlorisondamine, clonidine, strychnine, bicuculline등으로 전처치시 유의하게 억제되었으나 atropine, 양측 미주신경절단, propranolol 및 metoclopramide등의 전처치로 영향을 받지 않았다. 더우기 taurine은 norepinephrine의 승압 반응에도 영향을 미치지 못하였다. Taurine의 심박감소 작용은 chlorisondamine, propranolol, clonidine, strychnine 및 bicuculline등으로 전처리시 현저히 차단되었으나 atropine, 양측 미주신경절단, metoclopramide 등의 전처리로 영향을 받지 않았다. 이상의 연구한 결과로 보아, taurine은 가토의 측뇌실내 투여시 지속적이며 현저한 혈압하강 및 심박감소를 일으키며 이러한 작용은 뇌내의 catecholamine 뉴론과 관련있는 taurine (glycine) 및 gamma-aminobutyric acid (GABA) 수용체를 통해서 나타나는 것으로 사료된다.
