Change of Inorganic Component, Reducing Sugar, Catalpol and Benzo[α]pyrene Contents of Rehmannia glutinosa Libosch. var. purpurea Makino by Drying Methods (지황의 건조 방법에 따른 무기성분, 환원당, Catalpol 및 Benzo[α]pyrene의 함량 변화)

  • This experiment was carried out in order to collect the basic data on the standardization of the manufacturing process of Rehmannia glutinosa Libosch. var. purpurea Makino drying. By the drying methods of R. glutinosa, the content of water, inorganic components, reducing sugar, catalpol and benzo[${\alpha}$]pyrene were investigated. The water content was 15.6~17.2% when R. glutinosa was dried by cold-warm air moisture absorption drying method (CAMAD) at $60^{\circ}C$ during 6 days. Among of the inorganic components of R. glutinosa the K content was the most followed by P, Na, Ca and Mg. The reducing sugar content of R. glutinosa by the hot air drying method (HAD) was much more than that by the CAMAD. The catalpol content of R. glutinosa was not different by the drying temperature when it was dried by the CAMAD. The catalpol content of the large size tuber (about 50.0 g/unit) showed a tendency to increase from $60^{\circ}C$ until $70^{\circ}C$ drying temperature, but that of the small size tuber(about 4.0 g/unit) was decreased as being a trend as the drying temperature high when R. glutinosa was dried by the HAD, But the catalpol content R. glutinosa had a tendency to drop significantly at drying temperature above $80^{\circ}C$. The benzo[${\alpha}$]pyrene content was little detected when R. glutinosa was dried by both the SLD and the CAMAD, and the sampling by the HAD indicated within the scope of 5 ${\mu}g/kg$ which was the scope to regulate by Korean food and drug administration. In conclusion, it seemed that an appropriate drying temperature of R. glutinosa by the CAMAD and the HAD was about $60^{\circ}C$ and about $70^{\circ}C$, respectively, when we consider the catalpol content and benzo[${\alpha}$]pyrene detection in the manufacturing process of drying R. glutinosa.

Anti-diabetic activities of catalpol in db/db mice

  • The objective was to investigate the hypoglycemic action of catalpol in spontaneous diabetes db/db mice. 40 db/db mice were randomly divided into five groups: model control gourp; db/db plus catalpol 40, 80, 120 mg/kg body wt. groups and db/db plus metformin 250 mg/kg group. Age-matched db/m mice were selected as normal control group. The mice were administered with corresponding drugs or solvent by gavage for 4 weeks. The oral glucose tolerance test was carried out at the end of $3^{rd}$ week. After 4 weeks of treatment, the concentrations of fasting blood glucose (FBG), glycated serum protein (GSP), insulin (INS), triglyceride (TG), total cholesterol (TC) and adiponection (APN) in serum were detected. The protein expressions of phosphorylation-$AMPK{\alpha}$1/2 in liver, phosphorylation-$AMPK{\alpha}$1/2 and glucose transporter-4 (GLUT-4) in skeletal muscle and adipose tissues were detected by western blot. Real time RT-PCR was used to detect the mRNA expressions of acetyl-CoA carboxylase (ACC) and Hydroxymethyl glutaric acid acyl CoA reductase (HMGCR) in liver. Our results showed that catalpol could significantly improve the insulin resistance, decrease the serum concentrations of INS, GSP, TG, and TC. The concentrations of APN in serum, the protein expression of phosphorylation-$AMPK{\alpha}$1/2 in liver, phosphorylation-$AMPK{\alpha}$1/2 and GLUT-4 in peripheral tissue were increased. Catalpol could also down regulate the mRNA expressions of ACC and HMGCR in liver. In conclusion, catalpol ameliorates diabetes in db/db mice. It has benefit effects against lipid/glucose metabolism disorder and insulin resistance. The mechanism may be related to up-regulating the expression of phosphorylation-$AMPK{\alpha}$1/2.

  • Veronicastrum sibiricum (L.) Pennell (Scrophulariaceae) is a perennial herb growing in moist land. It has been used as a korean folk medicine in treating common cold, leucorrhea, cystitis and liver damage and as cholagogue. The present study dealt with the elucidation of the chemical components. From the roots of the plant, five iridoids were isolated and identified as minecoside $(mp\;142{\sim}3^{\circ})$, 6-O-veratryl catalpol ester $(mp\;216{\sim}8^{\circ})$, catalpol $(mp\;204{\sim}6^{\circ})$, aucubin $(mp\;180{\sim}2^{\circ})$ and 6-desoxy-8-isoferuloyl harpagide $(mp\;139{\sim}41^{\circ})$. Furthermore, ${\beta}-sitosteryl-3-O-D-glucoside$, campesteryl 3-O-D-glucoside, ${\beta}-sitosterol$, campesterol, stigmasterol and mannitol were also isolated.

Quality Evaluation of Various Dried Roots of Rehmannia glutinosa (지황(地黃)의 품질평가)

  • The dried roots of Rehmannia glutinosa (RG) have been used in the traditional medicine for treatment of diabetes, fever and dysuria, etc. In order to investigate the quality evaluation of the dried roots of RG, we conducted the physico-chemical and biological evaluation method. The amount of catalpol from commercial samples is very diverse about $0.00{\sim}3.89%$ by using HPLC method, because it is easily decomposed by processing of RG. So, we should try to identify the correlation with the contents of catalpol and biological activities of RG. We chose 3 samples which were a wide difference of catalpol contents between each sample (Sample-I; 3.4%, Sample-II; 2.8%, Sample-III; 0.05%). Sample-I and Sample-II were found to be more effective than Sample-III on the DPPH free radical scavenging effect and inhibitory effect on $H_2O_2-catalyzed$ lipid peroxidation in rat liver in vitro. And, Sample-I and Sample-II exhibited more significant effects than Sample-III on accelerating actions of the small and large intestinal transport, and diuretic action in mice. So, it is suggested that the quantitative determination of catalpol should be required for the tandardization of the dried roots of RG.

Effects of N, P and K Application Rates on the Yield and the Available Constituents Contents in the Rhizoma of Rehmannia glutinosa (질소(窒素), 인산(燐酸), 가리(加里)의 시용량(施用量)이 지황(地黃)(Rehmannia glutinosa) 근경(根莖)의 수량(收量) 및 유효성분함량(有效成分含量)에 미치는 영향(影響))

  • The purpose of this study was centered on investigation of the effects N, P and K application rates on root yield and their available content (catalpol, sugar, ash and extracts) of Ji-whang(Rehmannia glutinosa Lib. var. purpurea Makino), and the current application rates(N 20kg/10a, $P_2O_5\;30kg/10a$, $K_2O\;20kg/10a$) was studied for Ji-whang production in Eui-sung Gun southeastern part of Korea Peninsular. The yields of fresh and dried rhizomas were 941-1,494 kg/10a and 251-385 kg/10a, respectively. The rhizoma yield increased highly with increasing the application amount of nitrogen more than 10 kg/10a, but the effects of phosphorus and potassium were observed only at the application level of 40 kg/10a and 20 kg/10a, respectively. Catalpol contents in rhizoma increased at the plot of 20 kg/10a application of nitrogen, but the effects phosphorus and potassium on the catalpol contents were not observed. Catalpol content in dried root increased with increasing N application more than 10 kg/10a. Fructose contents increased at the plot of 20 kg/10a and more N application, and galactose contents decreased with K application. With increasing N application, ash contents decreased, but ethanol extract contents increased.

Analysis of Iridoid Glycoside and GABA Content in the Roots of the Rehmannia glutinosa Cultivars (지황 품종별 뿌리에서 Iridoid 배당체와 GABA 분석)

  • Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its roots have been utilized as a traditional medicine. The aim of this study was to elucidate the basic information of the roots of the R. glutinosa cultivars and their utilization. Methods and Results: The roots of R. glutinosa cultivars were harvested in the end of March. The two iridoid glycosides, aucubin and catalpol, were analyzed by liquid chromatography/mass spectrometry (LC/MS), whereas ${\gamma}-aminobutyric$ acid (GABA) was analyzed by gas chromatography/mass spectrometry (GC/MS). The aucubin content was the highest in the Dakang cultivar, whereas no aucubin was detected in the five cultivars. All cultivars had more than 12 mg/g catalpol content, and the maximum catalpol content was found in Jihwang 1. The GABA content was the highest in Suwon 1, and it was 40 times more than that in the Yeongang cultivar. Conclusions: The highest aucubin, catapol and GABA contents were detected in the Dakang, Jihwang 1, and Suwon 1, cultivars respectively. This study provides the crucial information regarding the versatile utilization and pedigree selection of R. glutinosa cultivars.

Quality Characteristics of Rehmannia radix Preparata with Pre-soaking Solvents (침지용매에 따른 숙지황의 품질특성)

  • This study was performed to investigate the effects of pre-soaking methods on the preparation of Rehmannia radix Preparata (R.P). The R. radix L (R.L) was soaked in distilled water and traditional Korean wine for 24 hr, then the soaked R.L was treated with a traditional nine-time steaming process. Next, catalpol, 5-hydroxymethyl-2-furaldehyde (5-HMF), polyphenols, flavonoids, antioxidant activities, and ACE inhibition activity were analyzed for the R.P produced by the different methods. The catalpol content of the R.L was 631.4 ppm, but the content decreased as steaming increased to 8-9 times. The 5-HMF, polyphenol, and flavonoid contents of the R.L were 0.12 mg/g, 5.09 mg/g, and 0.83 mg/g, respectively, and these increased gradually with increasing steaming times. As the steaming times of the distilled soaking water increased, the antioxidant activities of 1 mg/mL increased from 19.44% to 75.60% at 14 times of steaming. The ACE inhibition activities of 1 mg/mL of the distilled soaking water increased from 28.70% to 94.78% at 10 times of steaming, but decreased afterward.

Variation of main components according to the number of steaming and drying of Rehmanniae radix preparata

  • Contents of compounds in Rehmanniae Radix change depending on the number of steaming and drying and the drying method. In this study, as an impregnation method for dried Rehmanniae Radix, takju impregnation and cheongju impregnation were carried out and steaming and drying were repeated for 9 times. The changes of 5-HMF and catalpol contents were analyzed according to the number of repetition times to investigate which stage of steaming and drying is preferable. Also, total nitrogen, crude fat, ash, and crude fiber were measured to analyze changes in general components. 5-HMF was not detected in dried Rehmanniae Radix. As a result of repetitive steaming and drying, the content of 5-HMF increased only slightly from 1 to 4-times steaming and drying but increased significantly from 5-times. The catalpol in dried Rehmanniae Radix was not detected after 5 times of steaming and drying. Sucrose, maltose, and glucose were included in dried Rehmanniae Radix before steaming and drying. However, after the process in both Takju impregnation and Cheongju impregnation, galactose and fructose tended to decrease after production and sucrose and glucose tended to decrease after the increase. In this study condition, 6-times and more steaming and drying were appropriate process which met the content criteria (not less than 0.1%) of the Korean Pharmacopoeia (8th edition) for 5-HMF, an index component for quality control of Rehmanniae Radix Preparata.
