Screening of Antimicrobial Activity from Castanea crenata Sieb. et Zucc. Leaves and Flowers. -II. Screening of Antimicrobial Activities- (밤나무 꽃과 잎의 화학성분 및 항균활성 검색 -제2보. 항균활성 검색-)

  • Chestnut (Casranea crenata S. et Z.) leaves and flowers were extracted with 80% methanol and then fractionated with ethylacetate, methanol and water. Their antimicrobial activities in each fraction were investigated. Methanol fraction of the chestnut leaves and flower showed strong antimicrbial activities against both of Gram positive and Gram negative bacteria. The ethylacetate and water fraction, however, showed only weak antimicrobial activities when the antimicrobial activities were occurred. Minimal inhibitory concentration (MIC) of the methanol extracts of the chestnut leaves and flowers against 5 strains of Gram positive and Gram negative bacteria were at $60\;{\mu}g/disc$. The extracts of the chestnut leaves and flowers inhibited the growth of Bacillus subtilis at 0.5% (w/w) concentration. In order to investigate the effect of extraction methods on the B. subtilis, scanning electron microscope was used. The B. subtilis was damaged when the methanol extracts of the chestnut leves and flowers were at 500 ppm.

Insect Funa of Chestnut Bushes at Paju Area in Korea: -Mainly on Dryocosmus kuriphilus Yasumatsu-

  • 1. Paju area of Kyonggydo has received serious damages by Dryocosmus kuriphilus Yasumatsu in the bushes of Castanea crenata S. et Z. since 1965. I have also found the middle parts of whole country of Korea has damaged by this wasp. 2. Bobwonri was the worst case in its damage by Dryocosmus kuriphilus Yasumatsu with most prosperous parasite, Aletia flavostigma singularis Butler and Dictyoploca japonica Moore. I conclude that this is the result of the lack of beneficial insects and isolated bush formation. 3. The chestnut bush at Bunsuri was recovering from the damage of gall wasp nevertheless it was illustrated big damage at last year's hranches. I conclude that this recovering phenomena is closely related with the insect fauna of this area and this characteristic insect fauna was derived from the bush formation which reveals specific bush arrangement that can be illustrated as surrounded by another bush of Quercus acutissima Carruthers. 4. As a final conclusion, I recommend mixed bush formation of chestnut tree with Quercus species to control insect population dynamics in healthy status.

  • Freezing resistance of ten cultivars of Chestnut (Castanea crenata S. et Z.) collected from four different sites of Kyunggi Province, Korea on March 2, 1975, was measured to find out the differences among tissue parts, and those among cultivars. The freezing and thawing rates were controlled lower than $6^{\circ}C/hr$. which occurs in nature. The resistance to low temperature was in order from lowest to highest; winter bud, cambium, xylum ray parenchyma and bark cortex. The difference in cold hardiness among cultivars was not consistent among tissue parts of twig stem except in cultivar Dan-Taeck of which all tissue parts showed highest cold-hardiness. The importance of the study on the seasonal variation in cold hardiness of different tissue parts was discussed in terms of choosing the most cold resistant Chestnut culitivar in Korea.

Effects of Saligenin Treatment on Acoustical Properties and Adsorption of Soundboard for Traditional Musical Instruments (전통악기 음향판용재의 살리게닌 처리가 음향성 및 흡습성에 미치는 영향)

  • This research was carried out to compare structure, equilibrium moisture content(EMC), adsorption and acoustical properties between 4 retention levels of saligenin(o-hydroxybenzyl alcohol) treated, heat-treated and untreated chestnut(Castanea crenata S. et Z.) and paulownia(Paulownia spp.) woods. The cell structures of untreated wood and heat-treated wood were not different but the cell wall of saligenin treated wood was thicker than that of untreated wood for both species. EMC of untreated wood was the highest, next was that of heat-treated wood, and that of saligenin treated wood was the lowest, and among saligenin treated woods, EMC was decreased as retention level increased for both species. Adsorption curves were parabolically increased. Acoustical properties of saligenin treated wood were most improved, and the next were those of heat-treated wood. Comprehensive acoustical property was improved at retention level 20% and 30% for chestnut wood, and at retention level 80% for paulownia wood.

  • This study explored the efficient dyeing method of aged chestnut wood using the natural dyes extracted from Gardenia jasminoides for, grandiflora, Carthamus tinctorius L., Rhus javanica L., Lithospermum erythrorhizon S. et Z., Caesalpinia sappan L. and Castanea crenata S. et Z.. The color variation of the dyed chestnut woods was also quantitatively evaluated. The wood specimens revealed abundant colors through different dyeing conditions such as dyeing temperature, dye pH, soaking time, and brushing frequency. The chroma and stain concentration of colored woods were calculated with $L^*$, $a^*$ and $b^*$ in order to make color tone measurement. Brushing treatment for colored wood was confirmed as a more economical dyeing method than soaking treatment requiring more time when natural dyeing was done. Furthermore, the desirable tone and shade of color was easily obtained by repetitive brushing treatment using low color tone. During natural dyeing, good color expression was made with high temperature and acidic condition of a dyeing liquor.

Distribution of Epiphytic Bacteria and Acid-Tolerant Bacteria on the Phyllosphere in the Industrial and Clean Areas (공단지역 및 청정지역 식물 잎권의 잎표면세균 및 내산성세균의 분포)

  • Total, direct viable count, and acid-tolerant epiphytic bacterial population sizes were quantified on leaves of chestnut tree (Castanea crenata S. et Z.) near Taejon Industrial Estate affected by acid precipitation and deposition as well as in the clean natural forest area, Mt. Kyejok, in Taejon city from August 1996 to August 1997. Geometric mean numbers of total, direct viable count, and acid-tolerant epiphytic bacteria were $9.9{\times}10^5cell/cm^2$, $1.6{\times}10^6cell/cm^2$, and $7.1{\times}10^3cfu/cm^2$ respectively, being 1.5, 2, and 2.6 times those in the clean area. Acid-tolerant epiphytic bacterial numbers at pH 5.6 by MPN method were $3.3{\times}10^4$ in the industrial area, about the same as the number, $3.4{\times}10^4MPN/cm^2$, of the clean area. Acid-tolerant bacterial number at pH 4.0 was $1.9{\times}10^{-1}MPN/cm^2$ in the industrial area, whereas none was detected in the clean area. Acid-tolerant bacteria at pH 3.0 were not detected at all in the industrial area as well as in the clean area. Epiphytic bacterial population sizes were generally the greatest in May when leaves are emerged and grew hut the lowest in November when defoliation occurs. These results showed that air pollutant deposition on leaves did not cause a decrease of epiphytic bacteria at least and acid deposition on leaves did cause an increase of acid-tolerant bacteria.

Antioxidative and Macrophage Phagocytic Activities and Functional Component Analyses of Selected Korean Chestnut (Castanea crenata S. et Z.) Cultivars (국내산 밤 일부 품종의 기능성 성분분석과 항산화 및 대식세포 활성)

  • Antioxidative and macrophage phagocytic activities and contents of functional component in selected Korean chestnuts (Dantaek, Daebo, Okkwang, Seokchu, Byunggo) were evaluated. Coumarin, gallic acid and catechin in inner skin and whole kernel of selected Korean chestnuts were detected by HPLC. The predominant functional components in inner skin of chestnut were catechin, followed by gallic acid and coumarin. However, the whole kernel had only gallic acid. Thus, the antioxidant properties of gallic acid and catechin were evaluated through DPPH radical-scavenging activity and SOD like activity. Gallic acid and catechin at 6.0 mg/100 g exhibited 69.4% and 38.3% of scavenging activities on DPPH radical, respectively. DPPH radical-scavenging activity of gallic acid increased in a concentration-dependent manner. Gallic acid was found to possess higher DPPH radical-scavenging activity than equivalent amount of catechin at all concentrations, whereas catechin was found to have higher SOD like activity than gallic acid. In addition, pre-incubation of macrophage with white kernel extract from Byunggo resulted in a significant increase of phagocytic activity and yellow kernel extracts from Byunggo, Dantaek, Daebo and Okkwang, leading to an increase in phagocytic activity compared with untreated cells. Yellow kernel extracts was found to have higher phagocytic activity than white kernel extracts. Byunggo had the highest phagocytic activity. The results suggest that the Korean chestnut may provide a natural source of antioxidants and active immunity.

Change of Constituent Components in Selected Korean Chestnut (Castanea crenata S. et Z.) Cultivars by Different Storage Conditions (국내산 밤 일부 품종의 다른 저장 조건들에 의한 성분변화)

  • The aim of this study was to analyze moisture, crude protein, crude lipid, vitamin C and sugar changes in selected Korean chestnut cultivars such as Danteack, Deabo, Seokchu, Okkwang and Byunggo during storage at $4^{\circ}C$ and $-10^{\circ}C$ for 10 months. The moisture contents of selected Korean chestnut cultivars ranged from 49.9 to 57.4%. The moisture content of Seokchu was the highest. The contents of moisture in white kernel were higher than that in yellow kernel. The content of moisture showed decreasing tendency after 10 months of storage. The crude protein and crude lipid contents in whole kernel of selected Korean chestnut cultivars were $3.3{\sim}4.2%$ and $0.3{\sim}1.6%$, respectively. The crude protein content of Deabo was the highest. The crude protein in Danteack, Seokchu, Okkwang and Byunggo was increased during storage at $4^{\circ}C$ for 10 months, while that in whole kernel of Deabo was decreased and no changes in crude protein in yellow kernels of Deabo were observed. The crude protein in Okkwang was increased during storage at $-10^{\circ}C$ for 10 months. The cold storage was found to have higher composition change of crude protein than the freezing storage. The content of crude lipid in Daebo and Byunggo was decreased during storage at $4^{\circ}C$ and $-10^{\circ}C$. Yellow kernels of Deabo, Okkwang and Byunggo were found to have higher crude lipid content than white kernels. The vitamin C content also decreased during storage at $4^{\circ}C$ and $-10^{\circ}C$ and the decrease in vitamin C content was higher at $4^{\circ}C$ than $-10^{\circ}C$. Vitamin C was not detected after 3 months storage at $-10^{\circ}C$. The sugar content increased at the latter period storage at $4^{\circ}C$ and $-10^{\circ}C$. The sugar content of selected Korean chestnut cultivars ranged from $36.2{\sim}44.3%$ and Dantaek had the highest sugar content.
