Heterologous Gene Expression and Secretion of the Anticoagulant Hirudin in a Methylotrophic Yeast Hansenula polymorpha

  • A heterologous gene expression and secretion system using a methylotrophic yeast, Hansenula polymorpha was developed for the production of anticoagulant hirudin. Hirudin gene was expressed under the control of a strong and inducible methanol oxidase (MOX or AOX) promoter. The mating factor a pre-pro leader sequence of Saccharomyces cerevisiae was employed for hirudin to be secreted into the extracellular medium. Hirudin expression cassette was introduced into three strains of H. polymorpha, A16, HPBl and DLl which have different genetic backgrounds. This expression cassette was stably integrated into the host chromosomal DNA. Biologically active and mature hirudin was efficiently expressed and secreted into the extracellular medium. About 19 mg/L of hirudin was found in the culture supernatant in the case of a two-copy integrant of the strain HPBl under suboptimal culture conditions.

Ergonomic Evaluation and Safety Countermeasures of Personal Cassette Player Noise (휴대용 카세트 소음의 인간공학적 평가 및 안전대책)

  • Recently, noise from personal cassette players (PCP) poses growing concerns together with occupational noise-induced hearing loss. Eighteen male and female volunteer subjects participated to determine preferred noise levels for PCP usage before, during, and after 2-hour subway riding according to sources (types) of PCP listening (language/news, soft music, and hard music). Audiometric tests were conducted before and after the 2-hour exposure of PCP noise under subway riding. Statistical analyses showed some significant hearing losses with the greatest loss of more than 6 dB at 4000 Hz for both ears, indicating that serious noise-induced hearing loss would potentially occurred due to prolonged use of PCPs. Practical safety countermeasures against PCP noise are further discussed.

  • Conventional cancer chemotherapy is seriously limited by tumor cells exhibiting multidrug resistance (MDR), which is caused by changes in the levels or activity of membrane transporters that mediate energy-dependent drug efflux and of proteins that affect drug metabolism and/or drug action. Cancer scientists and oncologists have worked together for some time to understand anticancer drug resistance and develop pharmacological strategies to overcome such resistance. Much focus has been on the reversal of the MDR phenotype by inhibition of ATP-binding cassette (ABC) drug transporters. ABC transporters are a family of transporter proteins that mediate drug resistance and low drug bioavailability by pumping various drugs out of cells at the expense of ATP hydrolysis. Many inhibitors of MDR transporters have been identified, and though some are currently undergoing clinical trials, none are in clinical use. Herein, we briefly review the status of MDR in human cancer, explore the pathways of MDR in chemotherapy, and outline recent advances in the design and development of MDR modulators.

Construction of Recombinant Xanthomonas campestris Strain Producing Insecticidal Protein of Bacillus thuringiensis

  • An insecticidal crystal protein gene, cryIA(c), from Bacillus thuringiensis HD-73 was integrated into the chromosome of a xanthan-producing bacterium, Xanthomonas campestris XP92. The cryIA(c) gene expression cassette was constructed that placed the gene between the trc promoter and rrnB transcriptional terminator. The $lacl^q$ gene was also included to prevent the expression of cryIA(c) gene in X campestris cells. Southem blot analysis confirmed the integration of the cryIA(c) gene expression cassette in chromosome of X campestris XP92 transconjugant. Expression of the insecticidal crystal protein was confirmed by Western blot analysis and bioassay against the larvae of Hyphantria cunea (Lepidoptera: Arctiidae) and Plutella xylostella (Lepidoptera:Plutellidae).

  • In this paper, a program controlling an auto-audio media - cassette deck - by a 16 bit personal computer is studied in order to execute audio and visual learning in CAI. The results of this study are as follows. 1. Audio and visual learning is executed efficiently in CAI. 2. Access rate of voice information to text/image information is about 98% and 60% in "play" and "fast forward" respectively. 3. In "fast forward", quality of a cassette tape affects voice information access rate in propotion to motor driving speed. 4. Synchronizing signal may be mistaken by defects of tape itself.

The effect of semantic, syntactic and lexical factors on the menu selection performance for video cassette recorder operation

  • The product-user interface for consumer electronic products became complex due to the increased variety of functions corresponding to consumer needs. In this study, three types fo design improvement of the menu interface for a video cassette recorder is studied. Three modifications to an existing menu interface was designed using the semantic, the syntactic and the lexical design factors. A series of the validation experiments was performed to test the effect of the three types of task modifications. The results showed that performance time, number of errors and learning effect were improved for all task modification. It also showed that the performance improvement effect of the semantic factors was 8.8%, that of the syntactic factors was 5.9% and that of the lexical factors was 5.6% respectively. It was found out that the performance improvement effect of the semantic design factors were relatively more significant than the syntactic and the lexical design factors.

  • A turbo-fan for the 4-way cassette indoor units of air-conditioners has been investigated. The main purpose of this investigation is the reduction of the turbo-fan noise. In order to reduce the noise level, many design parameters of turbo-fans such as blade section, blade thickness, geometry of blade leading edge, blade width, blade angle and bellmouth depth have been studied. With the experimental data of these parameters, a new turbo-fan was made for our system. The noise level of the new system was at least 3 dB(A) lower than that of the current in use.

  • A new advanced integral reactor of 330 MWt capacity named SMART(System-integrated Modular Advanced ReacTor) is currently under development at KAERI(Korea Atomic Energy Research Institute). One of the major design features of the integral reactor is locating the steam generators(SG) inside reactor vessel and eliminating the possibility of LB LOCA(large Break Loss of Coolant Accident). Orifices are fitted at the low part of steam generator cassette to stabilize and balance coolant flow distribution in the MCP (Main Circulation Pump) pressure header. A sensitivity analysis is conducted to determine the optimum orifice size using computer code 'CFX'.
