산화적 스트레스는 세포 및 조직 손상을 통해 피부의 탄력 및 보습 기능 저하, 피부 노화 촉진을 비롯한 다양한 피부질환을 일으킨다. 본 연구의 목적은 인간 피부각질세포 (HaCaT keratinocyte)에서 산화적 스트레스에 대한 붉은 토끼풀 추출물의 효능을 검토하여, 피부에 효과적으로 사용할 수 있는 기능성 소재로서의 활용 여부를 확인하고자 하였다. 본 연구에서는 붉은 토끼풀 추출물이 인간 피부각질세포에서 산화적 스트레스에 따른 세포사를 억제시키는 것을 확인하여, 이를 조절하는 보호기전을 규명하였다. 이는 붉은 토끼풀 추출물이 Caspase-3 비활성, 세포사 촉진단백질 Bax 발현 억제, 세포생존 촉진단백질 Bcl-2 발현 증가 및 MAPK 신호전달계 단백질의 인산화 억제를 통해 H2O2에 의해 유도된 산화적 스트레스를 보호할 수 있다는 것을 확인하였다. 따라서 붉은 토끼풀 추출물은 피부의 산화적 손상을 감소시키는 유용한 소재로 평가되며, 이는 피부보호 및 미용을 위한 다양한 제품 및 산업에 활용 가능성이 높은 것으로 판단된다.
항암 활성 연구를 위한 최적의 모델로서 2-D culture 모델은 여전히 널리 사용되고 있으나, 종양 환경에 더 잘 근접할 수 있는 3D MTS 모델은 시험관 내 모델 연구와 동물 모델 연구 간의 격차를 해소하기 위한 대안이 될 수 있다. Isoalantolactone은 목향(木香: Elecampane, Inula helenium L.)을 포함한 약용 식물에서 발견되는 sesquiterpene lactone 중의 하나로서 항암 활성을 포함한 다양한 약리학적 활성을 가지고 있는 것으로 알려져 있다. 본 연구에서는 HCC Hep3B 세포에서 유래된 2D 모델에서 관찰된 isoalantolactone의 항암 활성이 3D MTS 모델에서도 재현될 수 있는지를 조사하였다. 우리들의 결과에 의하면 isoalantolactone은 처리 농도 의존적으로 MTSs 형성을 억제하였으며, ROS 생성의 증가를 동반하였다. 특히 isoalantolactone 처리 및 배양 시간이 증가하면서 증식성 세포 영역이 세포사멸이 유발된 세포로 대체되었다. 또한, MTSs에서 isoalantolactone은 DR 관련 단백질들의 발현과 caspase-3의 활성을 증가시켰고, Bax/Bcl-2 발현 비율 및 총 PARP 단백질의 발현은 감소시켰다. 그러나 ROS의 생성을 인위적으로 차단하였을 경우, isoalantolactone에 의한 이러한 변화들이 모두 차단되면서 MTSs 구성 세포들의 세포 생존율을 회복시켰다. 따라서 본 연구의 결과는 isoalantolactone에 의한 Hep3B 세포 유래 MTSs의 세포사멸 유도는 외인성 및 내인성 경로의 활성화를 통하여 이루어지며 이는 ROS 의존적임을 시사한다.
$17{\alpha}$-estradiol ($17{\alpha}-E_2$)의 에폽토시스 유도활성에 미치는 종양억제단백질 p53의 조절효과를 조사하고자, $17{\alpha}-E_2$에 의해 유도되는 에폽토시스 현상들을 인체 대장암 세포주 유래 클론인 HCT116 ($p53^{+/+}$) 및 HCT116 ($p53^{-/-}$) 세포에서 비교하였다. HCT116 ($p53^{+/+}$) 및 HCT116 ($p53^{-/-}$) 세포를 $17{\alpha}-E_2$ ($2.5{\sim}10{\mu}M$)로 처리하거나 혹은 HCT116 ($p53^{+/+}$) 및 HCT116 ($p53^{-/-}$) 세포를 $10{\mu}M$$17{\alpha}-E_2$로 시간 별로 처리한 결과, HCT116 ($p53^{+/+}$)에 있어서는 세포독성과 에폽토시스-관련 sub-G1 peak의 비율은 처리농도와 시간에 의존적으로 나타났다. 그러나 HCT116 ($p53^{-/-}$) 세포의 경우는 이러한 현상이 미약하게 나타났다. $17{\alpha}-E_2$에 의해 유도되는 비정상적 유사분열방추사 형성, 중기판 염색체 배열의 미완성, 이에 따른 유사분열정지($G_2/M$ arrest) 등의 현상은 HCT116 ($p53^{+/+}$) 및 HCT116 ($p53^{-/-}$) 세포에서 유사한 수준으로 나타났다. 이에 반해, $17{\alpha}-E_2$에 의해 유도되는 Bak과 Bax의 활성화, 미토콘드리아의 막전위 상실(${\Delta}{\Psi}m$ loss), 그리고 PARP 분해 등의 현상은 HCT116 ($p53^{-/-}$) 세포에 비해 HCT116 ($p53^{+/+}$) 세포에서 훨씬 높은 수준으로 확인되었다. 아울러 $17{\alpha}-E_2$로 처리된 HCT116 ($p53^{+/+}$) 세포에서 확인되는 p53 (Ser-15)의 인산화 및 p53 수준의 증가와 일치하여, 세포 내의 p21및 Bax 수준도 현저히 증가하였다. 이때 $17{\alpha}-E_2$로 처리된 HCT116 ($p53^{-/-}$) 세포에서는 p21 및 Bax의 발현수준이 매우 낮았다. 한편, 에폽토시스 억제단백질인 Bcl-2 단백질 수준은 HCT116 ($p53^{-/-}$) 세포에 비해 HCT116 ($p53^{+/+}$) 세포에서 다소 낮았으나, 이러한 Bcl-2 단백질 수준은 $17{\alpha}-E_2$ 처리 후에도 크게 변화하지 않는 것으로 나타났다. 이러한 결과들은 $17{\alpha}-E_2$ 처리에 의해 유도되는 에폽토시스 유도 경로의 구성원들의 변화, 즉 비정상적 유사분열방추사 형성 및 이에 따른 유사분열정지($G_2/M$ arrest), 뒤이은 Bak 및 Bax의 활성화, 미토콘드리아의 막전위 상실, 그리고 이에 수반되는 caspase cascade 활성화 및 PARP 분해로 진행되는 에폽토시스 현상들 중에서, Bak 및 Bax의 활성화 단계가 종양억제단백질 p53의 에폽토시스 증진 활성에 의해 양성적으로 조절되는 작용 타켓임을 보여준다.
The aim of this study was to evaluate the antitumor activities of Typhae pollen (TP) by confirming in vitro cytotoxicity and in vivo anti-tumor and immune-modulatory effect with anti-cachexia effect. The MTT assay is used in HepG2 cell to detect potential cytotoxic activities of aqueous extract of Typhae pollen (TPe). After HepG2 tumor cell implantation, eight mice per groups were assigned to six groups. Three different dosages of TPe (500, 250 and 125 mg/kg) were orally administered in the amount of $10m{\ell}/kg$ and sorafenib also administered 20mg/kg, every day for 35 days from 28 days after the tumor cell implantation. We observed the changes on body weights, tumor volume and weights, lymphatic organ, serum interferon $(IFN)-{\gamma}$ levels, splenocytes and peritoneal NK cell activity, splenic tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, IL-10 contents. Periovarian fat weights, serum IL-6 levels, thicknesses of deposited periovarian adipose tissue and mean diameters were also detected to monitor the tumor-related anticachexic effects. In tumor masses, the immunoreactivities of cleaved caspase-3 and cleaved poly (ADP-ribose) polymerase (cleaved PARP) - apoptotic marks, cyclooxygenase-2 (COX-2), inducible nitric oxide synthases (iNOS) and tumor necrosis factor $(TNF)-{\alpha}$ were additionally observed by immunohistochemistry. The results were compared with sorafenib. Decreases of COX-2 were demonstrated in sorafenib and TPe treated mice and also increases of iNOS in tumor masses were observed in TPe, not in sorafenib. TPe increased periovarian fat pad weights compared with tumor-bearing controls and sorafenib treated mice. TPe showed increases of splenic $TNF-{\alpha}$, IL-10 and $IL-1{\beta}$, serum $IFN-{\gamma}$ and NK cell activities corresponding to increases of spleen weights, lymph node weights and non-atrophic changes of lymph nodes. Our results show oral treatment of TPe 500, 250 and 125 mg/kg has potent in vitro and in vivo antitumor activities through modest cytotoxic effects, immunomodulatory effects and apoptotic activities in HepG2 tumor cells. In addition, TPe can prevent cancer related cachexia.
Objective: To explore the effect of Withaferin A on A549 cellular proliferation and apoptosis in non-small cell lung cancer (NSCLC). Materials and Methods: NSCLC cell line A549 was selected to explore the effect of Withaferin A on A549 cellular proliferation, apoptosis and the PI3K/Akt signal pathway capable of regulating tumor biological behavior by assessment of cellular proliferation, cellular apoptotic rates and cellular cycling as well as by immuno-blotting. Results: Withaferin A could inhibit A549 cellular proliferation and the control rate was dosage-dependent (P<0.05), which also increased time-dependently with the same dosage of Withaferin A (P<0.05). The apoptotic indexes in A549 cells treated with 0, 2.5, 5.0, 10.0 and 20.0 ${\mu}mol{\cdot}L^{-1}$ Withaferin A for 48 h were significantly different (P<0.05). In addition, the apoptotic rates of each group in both early and advanced stages were higher than those in 0 ${\mu}mol{\cdot}L^{-1}$ (P<0.05), which were evidently higher after 48 h than those after 24 h (P<0.05). A549 cells treated by Withaferin A for 48 h were markedly lower in Bcl-2 level and obviously higher in Bax and cleaved caspase-3 levels than those treated by 0 ${\mu}mol{\cdot}L^{-1}$ Withaferin A (P<0.05), and there were significant differences among 5, 10 and 20 ${\mu}mol{\cdot}L^{-1}$ Withaferin A (P<0.05). The ratios of A549 cells treated by Withaferin A for 48 h in G0/G1 stage were higher than those in 0 ${\mu}mol{\cdot}L^{-1}$, while those in S and G2/M stages were obviously lower than those in G2/M stage, and there were significant differences in 5.0, 10.0 and 20.0 ${\mu}mol{\cdot}L^{-1}$ Withaferin A (P<0.05). Additionally, p-Akt/Akt values were in reverse association with dosage, and the differences were significant (P<0.05). Conclusion: Withaferin A can inhibit the proliferation and apoptosis of A549 cells by suppressing activation of the PI3K/Akt pathways.
Semaphoring is a transmembrane receptor which participates in many cytokine-mediated signal pathways that are closely related to the angiogenesis, occurrence and development of carcinoma. The present study was designed to access the effect of mono-antibody (mAb) guided radioimmunotherapy (RIT) on skin carcinoma and investigate the potential mechanisms. Semaphoring mAb was acquired from mice (Balb/c), purified with rProtein A column; purity, concentration and activity were tested with SDS-PAGE and indirect ELISA; specificity and expression on the cutanuem carcinoma line and tissue were tested by Western blotting; morphology change was assessed by microscopy. MTT assay and colony inhibition tests were carried out to test the influence on the proliferation of tumor cells; Western blotting was also carried out for expression of apoptosis-associated (caspase-3, Bax, Bcl-2) and proliferation-related (PI3K, p-Akt, Akt, p-ERK1/2, ERK1/2) proteins and analyse the change in signal pathways (PI3K/Akt and MEK/ERK). The purity of purified semaphorin mAb was 96.5% and the titer is about $1{\times}10^6$. Western blotting showed semaphoring mAb to have specifically binding stripes with semaphoring b1b2 protein, B16F10, and A431 cells at 39KDa, 100KDa and 130KDa, respectively. Positive expression was detected both in cutanuem carcinoma line and tissue and it mostly located in cell membranes. MMT assay revealed dose-relate and time-relate inhibitory effect of semaphorin mAb on A431 and B16F10. Colony inhibition tests also showed dose-relate inhibitory effects. Western blotting demonstrated the expression of apoptosis and proliferation-related protein and changes in signal pathway. In conclusion, we demonstrated that semaphorin is highly expressed on the tumor cell-surfaces and RIT with semaphorin mAb has effect in i nhibiting proliferation and accelerating apoptosis of tumor cells.
Chungpae-tang is suggested to have the antitumor activity on lung cancer. This study was peformed to investigate apoptotic effect in vitro and antitumor effect and immune response after injection of B16-F10 melanoma cells and Chungpae-tang into a tail vein of C57BL/6 mice and administratition of Chungpae-tang in A549 human lung cancer cell line in vivo, respectively. Experimental studies were obtained by measuring the median survival time and cytokine expression through RT-PCR, and ELISA assay. The results were summarized as follows: 5 mg/ml of Chungpae-tang causing DNA fragmentation, caspase-3 enzyme activation, PARP fragmentation, and cytochrome c release, suggested that Chungpae-tang has in vitro apoptotic effect in A549 human lung cancer cell line in the apoptosis-induced experiment. The median survival time of the Chungpae-tang treated group was 21 days and that of control group was 22 days, suggesting that the median survival time between the Chungpae-tang treated group and the control group was not significant. Cytokine expression between the Chungpae-fang treated group and the control group was noticeable, but was not significant in the RT-PCR. In the ELISA assay, IL-2 productivity in the Chungpae-tang treated group was to increase more than that in the normal group (p<0.05) and was no significant between the Chungpae-tang treated group and the control group. $INF-\gamma$ productivity of the control group decreased more than that of the normal group (p<0.05) and that of the Chungpae-tang-treated group increased more than that of the control group (p<0.05). IL-12 productivity of the control group increased more than that of the normal group (p<0.05) and that of the Chungpae-tang-treated group decreased more than that of the control group (p<0.05) and the normal group. IL-4 productivity of the Chungpae-tang-treated group increased more than that of the normal group and the control group (p<0.05). IL-10 productivity of the Chungpae-tang-treated group increased more than that of the normal group and the control group (p<0.05). Accordingly the results show Chungpae-tang could induce apoptosis in A549 human lung cancer cell line and bring to antitumor effect and immune response against injection of B16-F10 melanoma cells into a tail vein of C57BL/6 mice but it needs more research on the precise mechanism of such effects.
본 연구 목적은 고지방식이 유도 비만 쥐의 피하지방조직에서 라스베라트롤 투여가 대식세포 침윤관련 염증인자에 미치는 영향을 규명하고자 하였다. 본 연구를 위해 정상식이군, 고지방식이군, 고지방식이+라스베라트롤 투여군으로 분류한 후, 라스베라트롤 투여군은 15주간 25 mg/kg 농도로 Dimethyl Sulfoxide에 용해하여 투여하였으며, 비교군은 Dimethyl Sulfoxide 용액만을 투여하였다. 연구결과 고지방식이군은 정상식이군에 비하여 체중이 유의하게 증가하였고, 라스베라트롤 투여군에서 고지방식이 군보다 NLRP3. ASC, Casepase1 mRNA 발현이 감소하였다. 또한 염증마커로 알려진 IL-18 mRNA 발현이 라스베라트롤 투여군에서 정상식이군과 고지방식이군보다 낮게 나타났다. 대식세포 침윤 마커인 F480, CD86 mRNA 발현에서도 라스베라트롤 투여군에서 고지방식이 군보다 유의한 감소를 보였다. 따라서 라스베라트롤 투여는 고지방식이 유도 비만 상황에서 대식세포 침윤 염증과 inflammasome에 긍정적인 영향을 미치는 것으로 보여진다.
본 연구는 MCF-7 유방암 세포에 대한 유자(CJP)와 탱자 (PTP) 과피 추출물의 항암 활성과 환경호르몬에 의해 유도된 암세포의 증식 억제 효과에 대하여 조사하였다. CJP와 PTP를 300 mg/ml 농도에서 72시간 처리하였을 경우, 암세포의 성장을 저해하였고 세포사멸을 유도하였다. MCF-7 유방암 세포의 형태학적 변화는 CJP와 PTP를 500 mg/ml 농도에서 7시간 처리하였을 경우 관측되었고 세포사멸은 capase-3의 활성화에 의하여 유도되었다. 환경호르몬에 의해 유도된 MCF-7 유방암 세포의 증식은 CJP와 PTP의 처리로 인하여 농도 의존적으로 감소하였으며, 300 mg/ml 농도에서는 대조군과 비교하였을 때 각각 70%와 80% 이상 감소하였다.
이상의 결과를 종합해 볼 때, 높은 수준의 포도당 섭취는 공복혈당과 공복혈당면적을 높이고 혈중 인슐린 농도와 아티포넥틴의 수준을 낮추어 혈당조절 능력을 억제시켰다. 반면, 높은 수준의 과당 섭취는 인슐린을 요구하지 않는 과당대사의 특이성으로 인해 혈당조절에는 효과적으로 보인다. 그러나 고과당 섭취는 간 조직 및 혈중 중성지방의 농도를 높이고 염증조절복합체 구성단백질의 발현을 조절하여 전염증인자의 발현을 증가시켰다. 이는 간조직에 있어 과당이 포도당보다 높은 수준의 염증반응을 유도하여 NAFLD의 발병과 진행에 보다 유의적인 영향을 준다는 것을 보여준다. 본 연구의 제한점은 일상에서 과당이나 포도당을 단독으로 섭취하는 경우가 드물다는 점과 3주라는 짧은 중재기간에 의한 실험 결과라는 것이다. 앞으로의 연구는 단순당을 장기간 섭취했을 때 혈청과 간 조직을 포함한 다른 대사 관련조직에서 나타나는 변화에 초점을 맞출 필요가 있으며, 염증조절복합체가 염증인자의 발현을 증가시키는 기전을 구체화하는 것이 요구된다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
