Therapeutic Effects of Safflower (Carthamus tinctorius L.) Seed Powder on Osteoporosis (홍화 (Carthamus tinctorius L.)씨 분말의 골다공증 치료효과)

  • The therapeutic effect of safflower seed powder on experimental osteoporosis in the rat induced by ovariectomy was evaluated. Thirty Sprague-Dawley rats were ovariectomized at the age of 12 weeks. Seven weeks postovariectomy, rats were divided into two groups: control and safflower seed powder treated group. Five animals from each group were sacrificed at the following time points: 1, 3, and 5 weeks. Scanning electron microscopic observation and morphometric analysis of the tibiae epiphysis showed that the administration of safflower seed powder significantly prevented reduction of cortical bone width and bone volume compared with the control group. In conclusion, safflower seed powder contains something that prevent bone loss due to estrogen deficiency, and was effective in preventing the osteoporotic decrease of bone mass.

  • Volatile components in flower and seed of safflower were identified. Volatile flavor compounds of safflower (Carthamus tinctorius L.) was extracted by simultaneous steam distillation and extraction method using Likens and Nickerson's extraction apparatus. Concentrated extract was analyzed and identified by gas chromatography and GC-mass spectrometry. Main volatile components in flower were terpene compounds, including p-cymene, limonene, ${\alpha}-phellandrene$, ${\gamma}-terpinene$, camphor, 4-terpineol, selinene, ${\beta}-caryophyllene$, torreyol, ${\beta}-eudesmol$, and 10 acids including 3-methylbutanoic acid, 2-methylbutanoic acid, and acids of $C_{2},\;C_{5}-C_{11}$. Main volatile components in seed and safflower were 20 aldehydes including hexanal (7.17%), (E)-2-heptenal (1.10%), (E,Z)-2,4-decadienal and (E,E)-2,4-decadienal.

Effect of Safflower and Safflower Seed Extract on Osteogenic Differentiation of MC3T3E1 Cells (홍화, 홍화씨 추출물이 MC3T3E1 세포의 골분화 과정에 미치는 영향)

  • Objectives This study investigated the effect of purified safflower (Carthamus tinctorius Linne) and safflower seed (Carthamus tinctorius L. seed; CS) extract, using hot water and ethanol extract methods , on the osteogenic differentiation of MC3T3E1 cells.Methods The safflower and safflower seed were extracted with hot water and ethanol. The samples were concentrated by a rotary evaporator and then freeze-dried using a freeze-dryer. The MC3T3E1 cells were propagated and maintained in DMEM (Gibco) containing 10% FBS and a 1% antibiotic antimycotic solution. To induce osteogenic differentiation, the cells were treated for 14 days with DMEM with 10 mM β-glycerophosphate and 50 μM ascorbic acid. Extract doses were confirmed by the results of an MTT assay, and treatment of the extracts was performed in a differentiation medium every two days. The ALP staining and activity were tested after osteogenic differentiation for five days, and after 14 days, osteogenic differentiation was determined by alizarin red S staining. The mRNA expressions of osteogenic-related genes were quantified using quantitative real-time PCR.Results In the results of the MTT assay, all concentrations of safflower extracts had no toxicity in the MC3T3El cells. But in the groups of 100 ng/ml and 200 ng/ml concentrations of safflower seed extracts, the cell viability was significantly reduced by up to 40-50%. So we fixed the treatment concentration of the extract at 50 ng/ml. In the ALP and alizarin red S staining, all extract groups increased osteogenic differentiation compared with the control group. The water-safflower extract group showed the highest mRNA level of Alp, Runx2, and Dlx5 genes. The mRNA level of Ocn, an osteogenic gene related to late-stage differentiation, in the ethanol-safflower extract group increased the mineralization more significantly than in other groups.Conclusions These data suggest that the extract of safflower increases the osteoblastic differentiation activates of MC3T3E1 cells like the extract of safflower seed. The water-extract and ethanol-extract of safflower have effects on different stages of osteogenesis in MC3T3El. Not only safflower seed but also safflower will be useful therapeutic reagents for age-associated chronic diseases such as osteoporosis.

  • After evaluation of antithrombotic effect of 40 herbs on platelet aggregation induced by ADP(Adenosine diphosphate), these results were obtained as follows: 1. Crude drugs exerting over 30 % of in Chinemys reevesii (Gray)hibition on platelet aggregation induced by ADP were Ganoderma japonicum (Fr.) Lloyd., Panax ginseng C. A. Mey., Gastrodia elata Bl., Thea sinensis, Chinemys reevesii (Gray), Cuscuta chinensis Lam., Cervus nippon Temminck., Biota orientalis (L.) Endl., Coriolus versicolor, Cinnamomum cassia Presl., Sophora flavescens Ait., Amomum villosum Lour., Carthamus tinctorius L., Rubus chingii Hu., Poria cocos (Schw.) Wolf., Laminana japonica Aresch., Ligustrum lucidum Ait., Angelica sineusis (Oliv.), Cyperus rotundas L., Ginkgo biloba L., Zingiber officinale Rosc., Prunus persica (L.) Batsch., Schizandra chinensis (Turcz.) Baill. and Plantago asiatica L.. 2. Of crude drugs having showed over 50% of inhibitory effect on platelet aggregation, at the concentration of $100{\mu}g/m{\ell}$, the inhibitory rates were 82.2% in Ganoderma japonicum (Fr.) Lloyd., 55% in Panax ginseng C. A. Mey., 50.8% in Gastrodia elata Bl., while at the concentration of $200{\mu}g/m{\ell}$, antithrombotic rates were 89.4% in Ganoderma japonicum (Fr.) Lloyd., 59.2% in Panax ginseng C. A. Mey., 57.9% in Thea sinensis, 52.7% in Gastrodia elata Bl.. These results suggest that the study sholuld be necessary on antithrombotic effect of solvent fractions of Ganoderma japonicum (Fr.) Lloyd., Panax ginseng C. A. Mey., Gastrodia elaha B1. and Thea sinensis and isolation of effective compound from above drugs.

Changes in Flavonoid Contents of Safflower Leaf during Growth and Processing

  • Eight flavonoids, apigenin-6-C-β-D-glucopyranosy l-8-C-β-D-glucopyranoside (AGG), quercetin 7-O-β-D­glucopyranoside (QG), luteolin 7-O-β-D-glucopyranoside (LG), quercetin 7-O-(6'-O-acetyl)-β-D-glucopyranoside (QAG), luteolin 7-O-(6'-O-acetyl)-β-D-glucopyranoside(LAG), quercetin (Q), luteolin (L) and acacetin 7-O-β­D-glucuronide (AG) were determined by HPLC in the safflower (Carthamus tinctorius L.) leaf during growth and processing. During growth, levels of five flavonoid glycosides (AGG, QG, LG, QAG, & LAG) in the leaf increased progressively at over time according to growth stages, reached a maximum before June 11, and then decreased sharply, while those of three flavonoid aglycones (Q, L, & AG) increased greatly at the early stage of growth, reached a peak before May 28, and then decreased rapidly. During the steaming process, contents of five flavonoid glycosides increased rapidly with increased steaming time, reached a maximum after 5 min of steaming, and then decreased, whereas those of flavonoid aglycones except for AG decreased sharply with increased steaming time. During the roasting process, contents of three flavonoid glycosides decreased rapidly with increased roasting time, whereas those of two acetylflavonoid glycosides (QAG & LAG) and three flavonoid aglycones increased progressively with increased roasting time, reached a maximum after 3 min of roasting, and then decreased. These results suggest that appropriate steamed and roasted safflower leaves are a rich source of flavonoids, and may be a good source of bioactive components as a functional leaf tea.

Screening of Antiviral Activity from Natural Plants against Feline Calicivirus (Feline calicivirus에서 항바이러스 활성을 가지는 천연식물자원 탐색)

  • In an effort to discover an antiviral substance against noroviruse (NV), which causes gastroenteritis illness world-wide, several plants including spices and herbs were evaluated for their antiviral activities against feline calicivirus (FCV) as a surrogate for NV. Among them, methanolic extract of green tea (Camellia sinensis L.) exhibited significant antiviral activity against FCV. After treatment with green tea extract (3.13 mg/ml) for 1 hr, FCV was completely inactivated. The antiviral activity of green tea extract against FCV was also determined to be dose and time- dependent. The results obtained in this study suggested that green tea will be effective in the prevention of food-borne diseases caused by NV.

[ ${\alpha}$ ]-Amylase and Protein Tyrosine Phosphatase 1B Inhibitory of Some Vietnamese Medicinal Plants Used to Treat Diabetes

  • In this study, the twenty-four ethyl acetate extracts of twenty-two medicinal plants, traditionally used in Vietnam as anti-diabetes agents, were investigated for ${\alpha}$-amylase and protein tyrosine phosphatase 1B (PTP1B) enzymes inhibitory activity in vitro. The results indicated that, twelve materials (50.0%) showed moderate to strong inhibitory activity in ${\alpha}$-amylase inhibitory activity with $IC_{50}$ values ranging from 2.5 to $48.8{\mu}g/mL$; meanwhile, ten extracts (41.6%) could demonstrate PTP1B activity with $IC_{50}$ values less than $30.5{\mu}g/mL$. Some plants presented interesting activities against both of ${\alpha}$-amylase and PTP1B enzymes such as Catharanthus roseus, Carthamus tinctorius, Momordica charantia, Gynostemma pentaphyllum, Glycyrrhiza glabra, Smilax glabra, Psidium guajava (leave), and Rehmannia glutinosa. The study may provide a proof, at least in a part, for the ethno-medical use in diabetes disease of these plants.

Effect of Shifting Planting - time and Different Nitrogen Level on the Yield and Characteristics of Plant Growth in Safflower, Carthamus Tinctorius L. (파종기 추동 및 질소비료수준차이가 홍화의 생육, 수량에 미치는 영향)

  • Effect of shifting planting-time and different nitrogen on the yield and characteristics of plant growth in safflower, Chrthamus Tinctorius L. This study was carried out to investigate the optimum sowing-date and the ecological variations due to differerent amount of nitrogen for safflower on the Experiment Farm of Sang Ji college from March 17 to may 16, 1980, sowing seeds with an interval of 10 days and the amount of nitrogen were applied in 5 levels (Non, half-standard, standard, one and half-ordinary amount-No and twice amount). The triple super phosphate and pottassium chloride were applied only in standard amount. The results obtained are summarized as follows; 1. As sowing-date was delayed, the germination-ratio decreased, germinating speed increased, and the time required to attain the most vigorous germinating stage and days required for germination shortened. 2. Plant height, number of branch and stem diameter showed a decrease as sowing-date was delayed. And at the same time, plant height and number of branch showed a tendency to increase as amount of nitrogen was increased. 3. As sowing-date was delayed, the number of pods and the weight of 1000-grains were decreased. Moreover, the earlier sowing-date was, the more it increased. And as amount of nitrogen was increased, number of pods and yield also were strikingly increased. 4. The fresh-weight of flower with orange and orange-red colour, as influenced by the different sowing-date, was found out to be largest at the begining of April. But increasing amount of nitrogen did not show influence upon the promotion of flowering. 5. Judging from the results reported above, the optimum sowing-date of safflower seemed to be the begining of April; also the culture of safflower seemed to be the effects of much amount of nitrogen.

Effect of Ethanol Extract of Safflower Seed on Bone Loss in Ovariectomized Rat

  • We investigated whether ethanol extracts of the safflower seeds containing phenolic compounds were responsible for the bone-protecting effects. Crude ethanol extract (CEE) of the safflower seeds was fed for 4 weeks at the level of 1% in diet to female Sprague-Dawley rats that had been subjected to bilateral ovariectomy (OVX). The CEE effects (OVX+CEE) were evaluated by comparing results obtained from OVX, Sham, and OVX injected with $17{\beta}$-estradiol ($OVX+E_2$) groups. OVX resulted in a dramatic reduction in the trabecular bone mass of the proximal tibia (approximately 40% of the Sham group) and an increase in fat deposition in bone marrow. In $OVX+E_2$ group, the bone loss was completely prevented as well as marrow adiposity. In OVX+CEE group, approximately 80% of the bone mass was maintained compared with Sham group and fat deposition in the bone marrow was prevented. Meanwhile, the partially purified ethanol extract containing the phenolic compounds stimulated proliferation of the ROS 17/2.8 osteoblast-like cells in a dose-dependent manner, as potently as positive controls of $E_2$ and genistein. The present data demonstrate that the ethanol extracts of safflower seeds reduced bone loss caused by estrogen deficiency. The bone-protecting effect of safflower seeds seems to be mediated, at least partly, by the stimulating effect of the phenolic compounds on the growth of osteoblasts.

Oil Compositions and Antioxidant Properties of Safflower Germplasm Collected from East Asia (동아시아 잇꽃 유전자원의 지방조성 및 항산화 분석)

  • Background: To obtain useful safflower resources for breeding and research of functional materials, the present study was conducted to determine fatty acid compositions and antioxidant activities of 281 safflower accessions collected from East Asia including South Korea. Methods and Results: Lipid contents and fatty acids compositions were evaluated using soxhlet extraction and gas chromatography, respectively. A antioxidant activities were analyzed using a spectrophotometer. The evaluation range of safflower accessions showed very wide variation. In terms of lipid contents, the China accessions were higher than the collection from other regions, whereas antioxidant activities did not differ among regions. The result of the principal component analysis showed that the first and second principal component cumulatively explained 90.6% of the total variation. In clustering the safflower accessions, the tree showed four major clades. Group II (16 accessions) was high in lipid content, oleic acid and linoleic acid, whereas group III (50 accessions) exhibited higher antioxidant activities than other groups. Conclusions: It was recommended that the China collections be utilized as a useful resource for research on functional oil materials. These results provided valuable information for safflower breeders and researchers of functional food.
