  • This study was performed to establish a single residue analytical method for determining fungicide carpropamid residues in various agricultural commodities. Korean cabbage, apple, brown rice and green pepper were selected as representative crops. Samples were homogenized, extracted with acetone and purified by liquid-liquid partition and Florisil column chromatography. Carpropamid residues were analyzed at 220 nm with reversed phase HPLC equipped octylsilyl and octadecylsilyl column and confirmed using mass spectrometry. ILOQ (Instrumental limit of quantitation) of carpropamid was 2 ng and MLOQ (Method LOQ) was 0.02 mg/kg. Mean recoveries from four kinds of crop samples fortified at three levels (MLOQ, 10LOQ, 100LOQ) in triplicate were in the range of 84~112%. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types.

  • A rapid and efficient assay to determine melanin biosynthesis inhibition of Magnaporthe grisea, a causal agent of the rice blast, by chemicals was developed. Wells in 24-well plates were loaded with spore suspension of the fungus and three known melanin biosynthesis inhibitors of KC10017, tricyclazole, and carpropamid. Subsequent color changes of mycelia and culture media in the wells were observed 7 days after incubation. The wells treated with KC10017 (an inhibitor of polyketide synthesis step and/or pentaketide cyclization step) became colorless, whereas tricyclazole (an inhibitor of 1, 3, 8-trihydroxynaphthalene reductase) or carpropamid (an inhibitor of scytalone dehydratase)-treated wells exhibited red color. They did not show any inhibitory effect on fungal growth. The inhibition of reaction steps prior to 1, 3, 6, 8-tetrahydroxynaphthalene formation was easily determined by colorless medium and mycelia. However, it was impossible to distinguish between inhibition of reduction steps and inhibition of dehydration steps by colors of the cultures. It was accomplished through HPLC analysis of the melanin biosynthesis-involving pentaketide metabolites accumulated by the inhibitors. Through screening of a number of synthetic chemicals using the in vitro assay, we could find a novel chemical group of melanin biosynthesis inhibitor.

  • Bakanae disease, caused by Gibberella fujikuroi (anamorph Fusarium moniliforme J. Sheldon), a typical seed-borne disease of rice occurs from nursery to paddy fields. Consequently, chemical seed disinfectants is the most efficient control method. Several seed treatment methods with various fungicides were attempted to inhibit disease. Spray and 24 hrs immersion of seeds using prochloraz emulsion reduced disease infection and the control value were 99.3 and 100%, respectively. In contrast, dressing to wet seeds thiophanate-methyl+thiram wp and benomyl+thiram wp reduced disease infection more effectively than 24 hrs immersion of seeds. However, dressing of carpropamid+imidacloprid+fludioxonil wp to wet seeds did not reduced disease as well as wettable liquid of fludioxonil. The results suggest that the bakanae disease might be disinfected effectively by 24 hrs immersion of seeds in prochloraz emulsion and seed dressing of fungicides.

  • To investigate an amount of pesticide residue in rice paddy field soils and greenhouse soil, this monitoring was carried out pesticide detection frequency and concentrations collected samples from 150 rice paddy field soils and 152 greenhouse soils of nationwide in the year of 2007, and 2008, respectively. The detection limit of pesticides of this experiment were ranged 0.001~0.005 ppm. In 2007, One hundred fifty samples were collected from rice paddy field soils in April and monitored for 120 wide-used pesticides. A total of 11 pesticides were detected four fungicides, four insecticides and three herbicides in paddy field soils. The highest concentration levels of pesticide detected were 0.84 ppm as herbicide oxadiazon, 0.81 ppm as fungicide isoprothiolane and 0.50 ppm as insecticide buprofezin. The detection frequencies range were 0~19.3%, and the frequency was 2.7% as isoprothiolane and 19.3% as oxadiazon in paddy field soils. In 2008, One hundred fifty two samples were collected from greenhouse soils in April and monitored for 120 wide-used pesticides. A total of 29 pesticides were detected six fungicides, sixteen insecticides and seven herbicides in greenhouse soils. high concentration levels of pesticide detected levels were 5.09 ppm as insecticide chlorfenapyr, 2.57 ppm as fungicide chlorothalonil and 0.72 ppm as herbicide oxadiazon. The detection frequencies range were 0~38.8%, and high frequencies were 38.8% as insecticide endosulfan, 13.2% as oxadiazone, 10.5% as fungicide hexaconazole and 7.2% as isoprothiolane in greenhouse soils, Total endosulfan and oxadiazon were showed high detection frequency of 38.8% and 13.2%, respectively.

  • In order to develop effective control methods for Bakanae disease caused by Gibberella fujikuroi (Saito) Ito during rearing of rice seedlings, we investigated the disease resistance of 15 rice varieties to G. fujikuroi and control effect of six seed disinfectants, and tried to improve the using methods of the seed disinfectants. Disease resistance was tested by investigating the disease incidence on each rice cultivar grown in rice seedling box infested with or not infested with G. fujikuroi at 30 days after sowing seeds and 20 days after heading date. The results showed that Hwayongbyeo, Dongjinbyeo, Hwoanbyeo, Nonghobyeo, Nampyeongbyeo, and Hwojinbyeo were resistant G. fujikuroi, meanwhile Keulubyeo, Sobibyeo, Odaebyeo, Junambyeo, Samchonebyeo, Sangjubyeo, and Hwabongbyeo were susceptible. Three seed disinfectants, prochloraz, fludioxonil, and carproamid + thiram + fludioxonil controlled Bakanae disease of rice very well, while bonomyl + thiram, thiophanate-methyl + thiram and thiophanate-methyl + triflumizole did not suppress the disease enough. Water temperature was turned to be an important factor for controlling the disease by treating seed disinfectants. Prochloraz showed 61% control value on the disease at $10^{\circ}C$, but it showed above 95% control value at the range of $30-35^{\circ}C$. It was confirmed that the control effect of seed disinfectants increased with increasing water temperature. Meanwhile soaking period of rice seeds in the suspension of seed disinfectants and chemical dosage had no high relation to control the disease. This results suggest that rice varieties, water temperature, and optimal selection of suitable seed disinfectants are very important to control Bakanae disease effectively.
