메기과에 속하는 메기와 미유기의 Carotenoid 색소 성분의 비교 (Comparision of Carotenoid Pigments in Catfish, Parasilurus asotus and Slender catfish, Parasilurus microdorsalis in the Family Siluridae)

  • 미이용자원인 어류 carotenoid에 관한 비교 생화학적 연구의 일환으로서, 메기과의 자연산 및 양식산 메기와 한국 특산의 담수어류인 미유기의 표피 carotenoid를 TLC, column chromatography 및 HPLC로 분리하고 NaBH$_4$에 의한 환원반응, I$_2$에 의한 이성화반응 및 UV-visible spectrophotometer로 비교하고 IR 및 Mass spectrum을 측정하여 carotenoid를 동정한 결과는 다음과 같다. 자연산 메기 표피의 총 carotenoid 함량은 0.27 mg($\pm$0.7)%였으며 carotenoid의 조성은 zeaxanthin 25.8%, diatoxanin 9.7% 및 cynthiaxanthin 9.1%로서 주성분을 이루고 있었으며 그 외 isocryptoxanthin 6.5%, $\beta$-carotene 5.1%, $\beta$-cryptoxanthin 4.3%, lutein 3.4% 및 $\alpha$-cryptoxantin 2.4%의 순으로 함유하고 있었다. 양식산 메기 표피의 총 carotenoid 함량은 0.07 mg($\pm$0.03)%로서 자연산 메기에 비해 다소 낮았으며, carotenoid의 조성은 lutein 48.5%, zeaxanin 13.9% 및 isocryptoxanin 13.3%로서 주성분을 이루고, 그 외 $\beta$-carotene 9.5%, $\beta$-cryptoxantin 1.4% 및 $\alpha$-cryptoxanthin 4.6%의 순으로 함유하고 있었다. 한편 미유기 표피의 총 carotenoid 함량은 0.75 mg($\pm$0.25)%로서 메기과의 다른 어종에 비해 함량이 높았으며, carotenoid 조성은 zeaxanthin 24.5%, 7'8'-dihydro-$\beta$ -carotene-4-ol 24.1%, 7'8'-dihydro-$\beta$-carotene 17.9% 및 7'8'-dihydro-$\beta$-carotene-3-ol 10.8%로서 주성분을 이루고 있었으며 그 외 diatoxanthin 8.7%, cynthiaxanthin 6,7% 및 lutein 5.0%의 순으로 함유하고 있었다. 이러한 결과는 parasiloxanthin과 7',8'-dihyo-parasiloxanthin을 주성분으로 함유하는 biwa호수 메기, P. asotus 표피 carotenoid 조성과 비교하여, 미유기에서는 7'8'-dihydro-$\beta$-carotene의 구조를 가진 carotenoid가 존재하였으나 자연산 및 양식산 메기에서는 발견되지 않아 특이하였다.

Effect of Dietary Genetically Modified ${\beta}$-Carotene Biofortified Rice on Immune in Rats

  • This study aims to examine the effect of Genetically Modified ${\beta}$-Carotene Biofortified Rice rice developed by simultaneous expression technology in NAAS on biological immunity. Accordingly, this study added Genetically Modified ${\beta}$-Carotene Biofortified Rice 25, 50% and general rice 50% as control group into diet and provided rats with the prescribed feeds and then measured the contents of immunoglobulin and cytokine in blood. As a result, male and female IgM, IgE, male IgG1, female IgG2a and TNF-a, IL5 and IL12 showed no significant difference; male IgG2a tended to decrease dependently on the combined concentration of Genetically Modified ${\beta}$-Carotene Biofortified Rice; female IgG1 showed significance with control group, but its association with diet was not found. The higher the dietary mixing ratio, the more the male and female IFN-a and female IL-4 contents, regardless of rice variety, and it was found that female IL6 content decreased significantly, but its association with diet was not found. The risk of beta carotene-enriched rice into environment and human body has not been reported yet. The digestion of Genetically Modified ${\beta}$-Carotene Biofortified Rice can be seen as "safe" as this test result showed no big difference between general rice and Genetically Modified ${\beta}$-Carotene Biofortified Rice, and its usability is full of suggestions.

한국농촌 성인의 혈청 Carotenoids 농도와 이에 영향을 미치는 요인에 관한 연구 (Levels and Factors Affecting Serum Carotenoids Concentrations of Adults Living in Rural Area of Korea)

  • Recent researches suggest that carotenoids are important not only as provitamin A but also for prevention of chronic diseases. This study was conduction to determine levels and factors affecting serum levels of lutein + zeaxanthin, $\beta$-cryptoxanthin, and $\beta$-carotene in 93 adults living in rural area of Korea. Fasting blood samples were collected and serum carotenoid levels were measured by HPLC. Dietary intake was estimated by 24 hour recall method and frequency questionnare of major food groups. Mean serum concentration of lutein + zeaxanthin was 616.32 nmol/L, $\beta$-cryptoxanthin was 856.95nmol/L, and $\beta$-carotene was 242.90nmol/L. Serum $\beta$-carotene levels in study subjects were very low. Both $\beta$-cryptxanthin and $\beta$-carotene were negatively correlated with serum triglyceride and positively correlated with total-choesterol and LDL-cholesterol. Serum levels of female subjects were significantly higher than males in all carotenoids. For age groups, subjects in their 30's were shown to have the highest concentration of all carotenoids. Lutein + zeaxanthin were lowest in subjects in theri 40's while $\beta$-crytoxanthin and $\beta$-carotene levels were lowest in subjects in their 60's. The $\beta$-carotene levels in non-smokers were significantly higher than in drinkers. Lutein+zeaxanthin levels were significantly higher among subjects consuming more green and yellow vegetables by frequency questionnarie. In conclusion, serum carotenoids were affected by sex, age, serum lipids, smoking, and alcohol intake. Intake of vegetables and fruits could affect by sex, serum lipids, smoking, and alchol intake. Intake of vegetables and fruits could affect serum lutein+zeaxanthin level. This data indicated that compared to other studies, Korean adults in rural areas have high lutein+zeaxanthin concentratins and low $\beta$-carotene concentrations in serum. High lutein+zeaxanthin levels may be related to high consumption of vegetables in these subjects.

Tocopherol and Carotenoid Contents of Selected Korean Cooked Combination Foods Consumed by Young Korean Children

  • To more accurately estimate vitamin A and vitamin E intake of Koreans, food composition databases of the tocopherol and carotenoid contents of Korean foods are needed. In this study, the tocopherol $(\alpha-,\;\gamma-,\;and\;\delta-)$ and carotenoid ($\alpha-carotene,\;\beta-carotene,\;\beta-ctyptoxanthin$, lutein, and zeaxanthin) contents of 12 Korean cooked combination foods commonly consumed by children in Kwangju, Republic of Korea, were determined using reversed-phase HPLC. All samples were obtained from 3 different households in Kwangju during summer, 2005. All cooked foods in this study had detectable quantities of $\alpha-tocopherol,\;\gamma-tocopherol$ except for shoegogimugook, and $\delta-tocopherol$ except for myulchibokkeum Doejigogibokkeum had the highest $\alpha-tocopherol$ content (0.64 mg/l00 g edible portion), and $\gamma-tocopherol\;and\;\delta-tocopherol$ contents of gimbab were the highest among the foods (1.01 and 0.26 mg/l00 g edible portion). $\beta-carotene$ was found in all food samples. Gimbab had the highest contents of $\alpha-carotene,\;\beta-carotene$, and lutein among Korean cooked combination foods (158.3, 266.6, and $375.4{\mu}g/100g$ edible portion). Miyukgook contained only $\beta-carotene$ ($2.5{\mu}g/100g$ edible portion). Considerable sample-to-sample variability in tocopherol and carotenoid compositions were observed in several Korean cooked combination foods included in this study. Some of these carotenoids $(\alpha-carotene,\;\beta-carotene,\;and\;\beta-cryptoxanthin)$ are vitamin A precursors. The findings of this study may be valuable for use in Korean databases as well as nutrient consumption research for vitamin A and vitamin E.

DsLCYB Directionally Modulated β-Carotene of the Green Alga Dunaliella salina under Red Light Stress

  • Carotenoids, which are natural pigments found abundantly in wide-ranging species, have diverse functions and high industrial potential. The carotenoid biosynthesis pathway is very complex and has multiple branches, while the accumulation of certain metabolites often affects other metabolites in this pathway. The DsLCYB gene that encodes lycopene cyclase was selected in this study to evaluate β-carotene production and the accumulation of β-carotene in the alga Dunaliella salina. Compared with the wild type, the transgenic algal species overexpressed the DsLCYB gene, resulting in a significant enhancement of the total carotenoid content, with the total amount reaching 8.46 mg/g for an increase of up to 1.26-fold. Interestingly, the production of α-carotene in the transformant was not significantly reduced. This result indicated that the regulation of DsLCYB on the metabolic flux distribution of carotenoid biosynthesis is directional. Moreover, the effects of different light-quality conditions on β-carotene production in D. salina strains were investigated. The results showed that the carotenoid components of β-carotene and β-cryptoxanthin were 1.8-fold and 1.23-fold higher than that in the wild type under red light stress, respectively. This suggests that the accumulation of β-carotene under red light conditions is potentially more profitable.

중년기 성인의 혈중 항산화 물질 분석 및 혈중 지질과 신체계측치와의 상관성 (Analysis of Serum Antioxidant Materials Concentration and Their Relation with Blood Lipids and Anthropometric Indices in Middle-Aged Adults in Korea)

  • 본 연구에서는 우리나라 건강한 중년성인의 혈청 carotenoids류 (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lutein, lycopene)와 ${\alpha}$-tocopherol, ${\gamma}$-tocopherol, retinol의 수준을 파악하고 혈중 지질상태 및 신체계측치와의 상관성을 평가해보고자 하였으며 조사 대상자는 40대 이후 건강한 성인 373명 (남자 231명, 여자 142명)이었다. 우리나라 건강한 중년기 성인의 혈청 중에 존재하는 주요 carotenoids는 ${\beta}$-cryptoxanthin, ${\beta}$-carotene, lutein이었고 lycopene, ${\alpha}$-carotene은 더 낮은 농도로 존재하였다. 성별에 따른 비교에서는 ${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lycopene 등의 carotenoids 물질들의 농도는 여자가 남자에 비해 유의하게 높았고, retinol은 남자가 여자에 비해 유의하게 높았으며 lutein과 tocopherol은 성별에 따른 유의차를 보이지 않았다. 혈중 항산화 물질의 농도는 연령이 높아짐에 따라 증가하는 경향을 보였으며 retinol과 ${\alpha}$-tocopherol 영양상태는 남자가 여자에 비해 우수하였다. 혈청 항산화 물질들의 농도와 혈청 및 소변 중의 지질과산화물 농도 사이에는 뚜렷한 상관성을 발견할 수가 없었다. 혈청 carotenoids들은 상호간에 또 ${\alpha}$-tocopherol 사이에 유의적인 양의 상관관계를 보였으며, 반면 lutein을 제외하고는 retinol과는 음의 상관성을 나타내었다. 혈 중 ${\alpha}$-tocopherol과 retinol 사이에는 유의적인 양의 상관성을 보였다. 혈 중 지질과의 상관성 분석에서 측정된 모든 항산화 물질들은 총콜레스테롤과 양의 상관관계를 나타내었다. 반면 중성지질은 carotenoids 물질과는 음의 상관성을 보였고 tocopherol, retinol과는 양의 상관성을 보였다. 혈청 항산화 물질들의 농도와 신체계측치 사이에는 성별에 관계없이 전반적으로 혈청 carotenoids류는 신체계측치들 및 혈압과 음의 상관성을 보였으며, tocopherol들과 retinol 수준은 양의 상관성을 보였다. 이 중 특히 유의적인 상관성을 보인 것은 남성에서 ${\beta}$-carotene과 lutein의 혈압과의 음의 상관성, 여성에서의 ${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin과 체지방 관련 지표들 (BMI, WHR, 체지방율)과의 음의 관련성이었다. 본 연구 결과 중년 성인의 혈중 항산화 물질들의 농도는 종류에 따라 성별, 연령별로 유의적인 차이를 나타내었으며, 혈중 지질 수준 및 신체 계측치와도 유의적인 관련성을 가지는 것으로 나타났다. 따라서 이러한 상관성의 원인과 의미에 대해서는 향 후 식생활 및 생활습관 등과 관련된 연구를 통해 밝혀져야 할 것이다.

HPLC를 이용한 고추 및 고추 가공품의 비타민 A 함량 측정 (Use of HPLC for the Determination of Provitamin A Carotenoids in Red Peppers)

  • Provitamin A carotenoids($\alpha$- $\beta$-carotene lutein capsanthin) cintents in various red pepper and red pepper products were analyzed by HPLC and UV/VIS spectroscopy. The amounts of $\beta$-carotene and capsanthin were at its highest in powdered red pepper and lutein in red pepper leaves. There are little $\alpha$-carotene present in all samples analyzed. So vitamin A activity was calculated only from $\beta$-carotene content. Capsanthin and $\beta$-carotene contents of red pepper were higher than those of green pepper whereas lutein content of red pepper was lower than that of green pepper. whereas lutein content of red pepper was lower than that of green pepper. Vitamin A activity from colorimetric analysis was higher than that from chromatographic method and results were not generally in accordance with the vitamin A data presented in current food composition tables. Vitamin A activities in retinol equivalent(R. E. ) of green pepper red pepper powdered red pepper fermented red pepper paste and red pepper leaves were 70, 570, 1, 340, 140, 980 R. E respectively.

라드를 이용한 열처리 온도 변화에 따른 ${\beta}-carotene$의 산화 안정성에 대한 연구

  • 0%, 0.0012%, 0.004%의 ${\beta}-carotene$을 lard를 첨가한후 $80^{\circ}C$, $130^{\circ}C$, $180^{\circ}C$의 온도처리로 2분간 열처리를 하고 $50^{\circ}C$에서 보관시키면서 각 기간별로 총 균수검사, 과산화물가(Peroxide Value), TBArs를 측정하여 비교하였다. 1) 미생물 총 균수 실험에서 ${\beta}-carotene$의 첨가량이 0%, 0.0012%, 0.004%로 증가함에 따라 균의 증식이 둔화되었다. 2) Peroxide Value의 측정 실험에 ${\beta}-carotene$의 첨가량이 0.004%일 때가 0%, 0.0012%에서 보다 항산화 능력이 더 높은 것으로 나타났다. 3) TBA가 측정실험에서 control과 Sample과의 비교에서 온도가 증가함에 따라 그 증가폭 커지므로 ${\beta}-carotene$의 항산화 능력이 있다는 것을 알 수 있다.

토마토의 생육과정에 따른 성분 변화 II - Ascorbic acid, Chlorophyll, Carotene, Lycopene 분석을 중심으로 - (The Changes of Components by Maturity Stage of Tomato II)

  • This was aim to study the changes of components by different parts and maturity of tomato. Tomato include the green pigment chlorophyll, the yellow pigment-carotene. the red pigment lycopene and the other component. These colors of tomatoes are reported to have important functional roles to promote health in human. The main objective of this study was to define changes in the content of the four tomato maturation that is ascorbic acid, chlorophyll, carotene, lycopene. A ascorbic acid contents of tomato fruits was the highest at 50th day. Chlorophyll contents of tomatoes fruits was the highest at 10th days after flowering. ${\alpha}$-carotene contained low-quantity while all growth period. ${\beta}$-carotene contained the highest at 50th days after flowering, $1036.97{\mu}g$. Lycopene contained the highest at 50th days after flowering, $5800{\mu}g$.

Identification of Carotenoid Oxidation Products in Pigment Extracts from Star Ruby Grapefruit Pulp at Different Temperatures with Exposure to Light

  • Pigment extracts obtained from Star Ruby grapefruit pulp were stored at different temperatures (4.5$^{\circ}C$, 23$^{\circ}C$) and exposed to light. many carotenoid oxidation products were formed due to light-exposure during storage periods. They were monitored by using HPLC with photodiode array detection and tentatively identified. Including (all-E)-lycopene and trans-$\beta$-carotene as predominant carotenoids in red grapefruit, 5Z-lycopene, 9Z-lycopene, 13Z-lycopene, and 15-cis-$\beta$-carotene were formed at 4.5$^{\circ}C$, 23$^{\circ}C$. Degradation of all-tarns lycopene was more susceptible to light-exposure and temperature a than that of all-trans $\beta$-carotene. The formation of lycopene cis isomers was favored under lighted condition. Respectively, (5Z)-lycopene was formed in greater amounts than other isomers at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$.
