Effects of Vitamin E supplement and Perilla oil on the Cytochrome P-450 contents and Fatty acid composition in Rat Hepatocarcinogenesis

  • The effects of vitamin E supplement on 15%(w/w diet) perilla or corn oils were studied in rat hepatocellular chemical carcinogenesis induced by modified Solt & Farber model, which consists of 20mg/kg body weight diethylintrosamine(DEN) injection, 3 weeks feeding of 0.02%2-acetylaminofluorene(2-AAF) and partial hepatectomy. The area of placental glutathione S-transferase(GST-P) positive foci tended to be smaller in perilla oil group had lower thiobarbituric acid reactive substances(TBARS) CONTENT. Fatty acid compositions in microsomal membrane were reflected by dietary fatty acid compositions, and not affected by carcinogen treatment or vitamin E supplement. By vitamin E supplement, linolenic acid contents of perilla oil group were much increased. By carcinogen treatment, membrane stability decreased significantly in corn oil, but maintained in perilla oil groups Vitamin E supplemental effect was noticed only in the corn-carcinogen group. Perilla oil may prevent hepatocarcinogenesis by maintaining membrane stability and by reducing cytochrome P-450 content. Vitamin E supplement did not seem to have the effect on hepatocarcinogenesis, but prevented lipid peroxidation, reduced cytochrome P-450 content and maintained membrane stability.

Studies on the Effect of Stress-Induced Gastric Ulcer on Gastric Carcinogenesis in Rats treated with N-methyl-N'-nitro-N-nitrosoguanidine (랫드에서 스트레스에 의한 위궤양이 위발암화과정에 미치는 영향에 관한 연구)

  • ;ABSTRACT-The effects of gastric ulcer induced by restraint and water-immersion stress on gastric carcinogenesis in Wistar male rats treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) were examined. Rats of group 1 were 2iven stress for 8 hours before they were received MNNG (100 mg/l) in drinking water for 20 weeks. Rats of group 2 were received MNNG first for 2 weeks and then were given stress once a week from 3rd to 12th weeks, with simultaneous MNNG adminitration and followed by MNNG only until 20th weeks. Rats of group 3 were received MNNG only as a positive control and rats of group 4 were not treated with carcinogen. All groups were sacrificed in 20 weeks. Sections of the pyloric mucosa were stained by avidin-biotin peroxidase complex (ABC) immuno-histochemical method. PAPG (pepsinogen isozyme 1 altered pyloric gland), body weight change, gross lesions and histopathological changes were examined. The results obtained from these studies were summarized as follows: 1. The mean body weight gains of the rats fed with carcinogens (group 1, 2, 3) were significantly lower than that of group 4 (control group, without carcinogen. p<0.05). However, the differences of the mean body weight of rats treated with carcinogen were not significant. 2. Stress treatment (group 1 and 2) increased the appearance of the numbers of PAPG (Pepsinogen 1 Altered Pyloric Gland) induced by carcinogen significantly compared with that of group 3 (carcinogen only, p<0.01). 3. The incidence rate of mucosal hyperplasia in pylorus was significantly increased in group 2 compared with group 3 (p<0.05).0.05).

  • Methyl tert-butyl ether (MTBE), an octane booster that is added to the reformulated gasoline, has been a widespread contaminant in aquatic ecosystem. MTBE is a recalcitrant pollutant having low biodegradability. Due to its higher water solubility and low octanol-water partition coefficient, it can be rapidly transported to the surrounding water environment. Also, MTBE is a known animal carcinogen, and is classified as a possible human carcinogen by U. S. Environmental Protection Agency. The adverse effect of MTBE to aquatic biota was widely reported. In Korea, the recent detection of MTBE in groundwater near gasoline filling stations has drawn concern to public health and ecosystem. To address this concern, the effect of MTBE to human health and ecosystem was discussed in this review. Also, ecotoxicity data of MTBE for fish, invertebrates, and algae were extensively compared to estimate the hazard concentration 5($HC_5$) of MTBE as a screening level.

Effects of Ginseng Saponin on DNA Strand Breaks and Replication Inhibition by Benzo(a)Pyrene in CHO-Kl Cells (Benzo(a)Pyrene 유발 DNA 상해 및 복제 억제에 미치는 인삼사포닌의 영향)

  • The effect of saponin extracted from Panax grneng CA Meyer on DNA repair and replicative DNA synthesis were examined in CHO-Kl cells cotreated with benzo(a)pyrene and rat liver S-15 fraction. The DNA strand breaks inititated by benzo(a)pyrene metabolites were measured by alkaline election technique. The addition of ginseng saponin to the culture media resulted in decrease of benzo(a)pyrene-induced DNA strand breaks, and restored the suppressed-semiconservative-DNA-synthesis by the carcinogen. DNA repair synthesis in the damaged cells was also elevated by the ginseng treatment when the repairing activites were measured for the (3H)-thymidine incorporation into the carcinogen damaged cellular DNk Comparative analysis of DNA-adduces of benzo(a)pyrene metabortes in microsomes suggested that ginseng saponin treatment in rats reduced the formation of electrophilic metabolites of benzo (a)-pyrene in the rat liver microsomes.

Management of Cancer Risk Caused by Motor Vehicle in a Large City (대도시 자동차 배출가스의 발암위해 관리 방안)

  • Motor vehicle exhaust is the major cause to the air contamination in Seoul. It includes many toxic chemicals to human health such as aidehyde, PAHs, benzene, xylene, toluene, benzo[a]pyrene, nickel, arsenic and cadmium in gasoline exhaust and formaldehyde, PAHs, 1,3-butadiene, benzene and particulate matter in diesel exhaust. Some chemicals out of them are classified as a human carcinogen. Many large diesel vehicles such as buses and trucks are drivened frequently in Seoul so that the air in Seoul is seriously contaminated by diesel exhaust, especially particulate matter. The amounts of particulate matter from large diesel vehicles may be estimated to be more than 50% of small dust in Seoul. The particles of particulate matter are coated with many toxic chemicals and some of these are considered as a human carcinogen. The cancer risk has to be throughly managed because the population density of Seoul is very high. Government should list hazardous air pollutants in Seoul, assess the exposure of people to toxic pollutants, especially carcinogens and manage human health risk.

  • Presently, mobile diesel equipments contribute a lot to improve the economic feasibility of underground mining and tunneling operations. Even in Korea, a lot of diesel equipments are being applied to the undergroud workings already, but the technology of management and control of them is not sufficient yet. This study handled the production procedure, characteristics and evaluation technology on diesel particulate matte(DPM) which is known as a carcinogen. For easy measurement of DPM using laser dust monitor, conversion known as a carcinogen. For easy measurement of DPM using laser dust monitor, conversion factor(k) to gravimetric concentration has been acquired. It is appeared that the critical material among the diesel exhaust pollutants is becoming DPM instead of NOx from this year of 1996 according to the Government regulation.

Risk Assessment of Airborne Toxic Metals in Thejon Industrial Complex (대전공단지역의 대기중 독성금속에 대한 위해도 평가)

  • The research centers on the concentration profile and risk assessment of toxic metals for ambient air in Taejon industrial complex. Airborne concentrations of each toxic metal for risk assessment were obtained from 2-year sampling by high volume air sampler and analysis by ICP-MS and ICP-AES in the complex. The long-term arithmetic mean of human carcinogen, arsenic, hexavalent chromium and nickel subsulfide was 5.53, 2.16 and $3.46ng/m^3$ while the mean of probable human carcinogen, beryllium, cadmium and lead was 0.08, 2.35, $293.29ng/m^3$, respectively. And the long-term arithmetic mean concentration of non-carcinogenic metal, manganese was $55.91ng/m^3$. The point risk estimate for the inhalation of carcinogenic metals was $3.6{\times}10^{-5}$, which was higher than a risk standard of $10^{-5}$. About 75% of the cancer risk was to the inhalation of human carcinogen, arsenic. Thus, it is necessary to properly manage arsenic risk in Taejon industrial complex. The point hazard index by the inhalation of manganese was 1.1. Therefore, an investigation into Taejon industrial complex is needed to obtain more fine long-term concentration data for airborne non-carcinogenic metals including manganese.

Study on the Characteristics of Carcinogenic Pollutant Emissions and Cancer Incidence Rates in Korea (국내 발암물질 배출량 특성과 암발생 추이에 관한 연구)

  • Objectives: The purposes of the study were to investigate hazardous pollutant emissions changes among group 1 carcinogens. The emissions characteristics were compared with national cancer registration statistics. Methods: A survey of group 1 carcinogen hazardous pollutant emissions (trichloroethylene, benzene, vinyl chloride, formaldehyde, 1,3-butadiene, ethylene oxide, chromium and its compounds, 3,3'-dichloro-4,4-diaminodiphenylmethane, chloromethyl methyl ether, arsenic and its compounds, cadmium and its compounds, o-toluidine) was conducted through a homepage for 2001-2015. The emission of hazardous chemicals and the cancer trend analysis for 2001-2015 were performed using the Korean statistical information service through its homepage as a reference. Results: Emissions of more than 95% of the substances listed as group 1 carcinogens over the last five years were made up of trichloroethylene, benzene, vinyl chloride, formaldehyde, 1,3-butadiene, and ethylene oxide. As a result of the comparison of emission results and cancer incidence rates, carcinogen pollutant emissions showed a tendency to decrease continuously. In addition, the incidence of cancer tended to increase, but showed a tendency to decrease from 2012. Conclusion: The results indicate hazardous pollutant emissions have continued to increase. However, no association between emissions and health effects was shown and more research is needed.
