Estimation of Environmental Effect and Genetic Parameters for The Carcass Traits in Hanwoo (Korean Cattle) (한우 도체형질의 환경효과 및 유전모수의 추정)

  • This study aims to estimate the genetic parameters for carcass traits on Hanwoo of breeding farmhouses using Animal Products Grading Service’s data of 428,812 cattle from 101 slaughterhouses nationwide from 2000 to 2005. Using carcass traits of carcass weight, eye muscle area, backfat thickness, marbling score, meat color and fat color that greatly influence Hanwoo's grade, the effects of carcass year, carcass season, sex and carcass region were estimated. Based upon carcass traits of carcass weight, eye muscle area, backfat thickness, marbling score and meat color that greatly influence Hanwoo’s grade, the heritabilities and genetic parameters were estimated of 17,578 Hanwoo slaughtered in 2005 with existing herdbook, where EM-REML algorithm was used in estimating genetic parameters. The mean and standard deviation of each carcass trait are 321.42±53.62kg, 76.25±10.43cm2, 9.96± 4.14mm, 3.75±2.00, 4.83±0.48 and 2.99±0.40, for carcass weight, eye muscle area, backfat thickness, marbling score, meat color and fat color, respectively. As a result of analysis on the effects of carcass year, the carcass weight, backfat thickness and meat color came out highest as 359.40±0.181, 9.82±0.017 and 4.90±0.002, respectively in 2004. As a result of analysis on the effects of carcass season, the carcass weight and eye muscle area came out highest as 345.88±0.144 and 79.57±0.033 respectively in spring, and the backfat thickness was highest as 8.78±0.013 in winter, and the meat color and fat color slightly came out higher as 4.88±0.002 and 2.96±0.001 in fall, while the marbling score was highest as 3.29±0.006 in summer. The results of the analysis on the effects of sex indicated that the backfat thickness and fat color were highest as 10.53±0.010 and 3.07±0.001 in cow, the carcass weight came out highest in Hanwoo steer as 368.03±0.068kg, the eye muscle area were highest as 82.96±0.042 in bull, and the marbling score was highest as 4.19±0.007 in steer, and the meat color was highest as 4.89±0.001 in cow.   • To further investigate the regions on porcine chromosome 7 that are responsible for economically important traits, phenotypic data from a total of 287 F2 individuals were collected and analyzed from 1998 to 2000. All animals were genotyped for eight microsatellite loci spanning the length of chromosome 7. QTL analysis was performed using interval mapping under the line-cross model. A permutation test was used to establish significance levels associated with QTL effects. Observed QTL effects were (chromosomewide significance, position of maximum significance in centimorgans): Birth weight (<0.01, 3); Carcass length (<0.05, 80); Longissimus muscle area (<0.01, 69); Skin percentage (<0.01, 69); Bone percentage (<0.01, 74); Fat depths at shoulder (<0.05, 54);Mean fat depth (<0.05, 81); Moisture in m. Longissimus Dorsi (<0.05, 88). Additional evidence was also found which suggested QTL for dressing percentage and fat depths at buttock. This study offers confirmation of several QTL affecting growth and carcass traits on SSC7 and provides an important step in the search for the actual major genes involved in the traits of economic interest.

  • In poultry diets, a requirement of crude protein is one of the most important factors in poultry productivity. Besides, the Pekin duck requirement of crude protein is still not clear. This experiment was conducted to determine the crude protein requirement of Pekin duck on diet formulation by investigation of growth performance, carcass trait, and analysis of blood parameter for a hatch to 21-day (d) of age. A total of 432 male White Pekin ducks were randomly allocated to six levels of crude protein (i.e., 15%, 17%, 19%, 21%, 23%, and 25%) to give six replicate pens per treatment with 12 ducklings per each pen. Body weight and feed intake were measured weekly by calculating feed conversion ratio and protein intake. Two ducklings each pen was euthanized via cervical dislocation for analysis of carcass trait and plasma blood on 21-d of age. Data were applied on both prediction linear-plateau and quadratic-plateau models by estimation of the crude protein requirements. The level of crude protein requirements of Pekin ducks for 21 days after the hatch was estimated to be 20.63% and 23.25% diet for maximum daily gain, and minimum feed conversion ratio, respectively.

Comparison and Correlation Analysis of Different Swine Breeds Meat Quality

  • This study was performed to determine the influence of pig breed and gender on the ultimate pH and physicochemical properties of pork. The correlations between pH and pork quality traits directly related to carcass grade, and consumer's preference were also evaluated. The pH and meat grading scores for cold carcasses of 215 purebred pigs (Duroc, Landrace, and Yorkshire) from four different farms were obtained. Meat quality parameters of the pork loin were analyzed. Duroc and female animals were more affected compared to other breeds and male pigs. Duroc animals had the highest ultimate pH, carcass back fat thickness, marbling scores, yellowness, and fat content (p<0.05). Landrace pigs had the highest color lightness and cooking loss values (p<0.05). Among all trait parameters, marbling scores showed the highest significant differences when evaluating the impact of breed and gender on meat quality characteristics (p<0.001). Ultimate pH was positively correlated with carcass weight (0.20), back fat thickness (0.19), marbling score (0.17), and color score (0.16) while negatively correlated with cooking loss (-0.24) and shear force (-0.20). Therefore, pork samples with lower ultimate pH had lower cooking loss, higher lightness, and higher shear force values irrespective of breed.

  • This aim of this study was to estimate the most reasonable slaughter month for Hanwoo steers according to carcass traits. Carcass trait data on Hanwoo steers (17,249 head) were collected from October 2008 to September 2011. To measure the regression coefficients, Hanwoo steers were divided into two groups according to slaughter age: early (25 to 31 months) and late (32 to 38 months) groups. Regression coefficients of carcass traits according to slaughter age were significantly different (p<0.001) with the exception of PPKG (estimated auction price per 1 kg of carcass weight) in the early slaughter age group and MAR (marbling score, p<0.05) and PPKG (p<0.05) in the late slaughter age group. To obtain a positive gross margin, Hanwoo should be slaughtered at around 28 months of age.

  • A total of 6,804 records for Duroc breed were collected from three farms registered at the Korean Animal Improvement Association (KAIA) from 1998 to 2004 of which both records from two ultrasound modes (A and B) were analyzed to estimate the variance components of carcass traits. Three carcass traits backfat thickness (bf), loin eye muscle area (lma) and lean meat percentage (lmp) were measured. These traits were analyzed separately as bf1, lma1 and lmp1 for ultrasound mode A and bf2, lma2 and lmp2 for ultrasound mode B with multiple trait animal model by using MTDFREML (Boldman et al., 1993). All the traits revealed medium heritability values. Estimated heritabilities for bf1, bf2, lma1, lma2, lmp1 and lmp2 were 0.45, 0.39, 0.32, 0.25, 0.28 and 0.39, respectively. Estimated genetic correlations for traits bf1 and bf2, lma1 and lma2, lmp1 and lmp2 were positive but low. Specifically, genetic correlations between bf1 and bf2 was 0.30 while the estimates for lean traits between lma1 and lma2 and between lmp1 and lmp2 were 0.15 and 0.18, respectively. Conversely, high negative genetic correlations existed between bf1 and the lean traits lma2, lmp2. Likewise, the estimated genetic correlations between lma1 and lma2 and lmp1 and lmp2 were low.

Evaluation of BTA1 and BTA5 QTL Regions for Growth and Carcass Traits in American and Korean Cattle

  • Previously identified QTL regions on BTA1 and BTA5 were investigated to validate the QTL regions and to identify candidate genes for growth and carcass traits in commercial cattle populations from the USA and Korea. Initially, a total of 8 polymorphic microsatellite (MS) markers in the BTA1 and 5 QTL regions were used for Chi-square tests to compare the frequencies of individual alleles between high and low phenotypic groups for the US (Michigan Cattleman's Association/Michigan State University; MCA/MSU) cattle. For a subsequent study, 24 candidate genes containing missense mutations and located within the QTL regions based on bovine genome sequence data were analyzed for genotyping in the two commercial cattle populations. Re-sequencing analyses confirmed 18 public missense SNPs and identified 9 new SNPs. Seventeen of these SNPs were used for genotyping of the MCA/MSU cattle (n = 98) and Korean native cattle (n = 323). On BTA1, UPK1B, HRG, and MAGEF1 polymorphisms residing between BM1312 and BMS4048 were significantly associated with growth and carcass traits in one or both of the MCA/MSU and Korean populations. On BTA5, ABCD2, IL22 and SNRPF polymorphisms residing between BL4 and BR2936 were associated with marbling and backfat traits in one or both of the MCA/MSU and Korean cattle populations. These results suggested that BTA 1 and 5 QTL regions may be segregating in both Korean Hanwoo and USA commercial cattle populations and DNA markers tested in this study may contribute to the identification of positional candidate genes for marker-assisted selection programs.

Effect of Methionine Source and Dietary Crude Protein Level on Growth Performance, Carcass Traits and Nutrient Retention in Chinese Color-feathered Chicks

  • A total of 1,200 LinNan Chinese color-feathered chicks were used to study the effects of methionine source [DL-2-hydroxy-4-methylthio-butanoic acid (HMTBa) or DL-methionine (DLM)] and dietary crude protein (CP) level on growth performance, carcass traits, and whole-body nitrogen and fat retention. The trial was designed as a $2{\times}2$ factorial arrangement, including two CP levels (adequate and low) and two methionine sources (HMTBa and DL-methionine). Diets were formulated for three phases, starter (0-21 d), grower (21-42 d), and finisher (42-63 d). Chicks fed HMTBa had higher daily gain and improved feed efficiency than DLM during the grower phase (p<0.05). A significant two-way interaction was observed for growth performance during the finisher phase and overall (0-63 d). Growth performance was greater for chicks fed HMTBa than DLM on adequate-CP diets (p<0.05), but this was not observed at low-CP level (p>0.05). Chicks fed low-CP diets grew slower, used feed less efficiently during the grower, finisher phase and overall. On d 42, regardless of dietary CP levels, birds fed HMTBa had higher carcass weights, breast and thigh weights than DLM-fed birds (p<0.04). Birds fed low-CP diet had lighter carcass weights and less breast muscle, thigh muscle, and dressing percentage at the end of starter, grower and finisher phases (p<0.05). Whole body composition analyses found that birds fed HMTBa tended to contain more protein and less fat compared to those chicks fed DLM at the end of the starter phase (p<0.10). Low-CP diets increased CP concentration in the whole body at the end of the finisher phase (p = 0.05). HMTBa supplementation increased whole-body N retention rate during the finisher phase and overall (p<0.01), and low-CP diets reduced N intake and whole-body fat retention during the finisher phase and overall (p<0.05). In summary, HMTBa was better than DLM on an equimolar basis for growth performance, carcass traits, and N retention in Chinese color-feathered chicks. Low-CP diets lowered growth performance as well as carcass traits in color-feathered birds, probably due to imbalanced AA profiles.

Effects of Raising Farm on Genetic Evaluation for Carcass Traits in Hanwoo Cows (사육농가의 효과가 한우 암소의 도체형질 유전 평가에 미치는 영향)

  • This research was conducted to analyze the effects of raising farm on the heritability and breeding values of Hanwoo cows for their carcass traits, including cold carcass weight (CWT), back-fat thickness (BFT), eye-muscle area (EMA) and marbling score (MAR). The carcass data and pedigree data were collected from steers raised on Hanwoo farms in Pyeongchang-gun, Gangwon-do, South Korea. Three analytical models were applied for the estimation of heritabilities and breeding values. The first model (model 1) included slaughter house-year-month combination as fixed effects and age at slaughter was fitted as linear and quadratic covariates. The second model (model 2) was similar to model 1, but raising farm was additionally included as random effect. The third model (model 3) was similar to model 1 but farm effects were additionally included as fixed effect. The comparisons between the model 1 and the models including farm effect (model 2 and model 3) revealed that heritability estimates from model 2 or model 3 were smaller to those from model 1 for all carcass traits. Especially, obvious decrease of heritability was observed in CWT where heritability was 0.23 from model 1, 0.15 from model 2 and 0.18 from model 3. The maximum log likelihood of the model 2 and 3 were higher than those of model 1 for all traits. In model 2 that raising farm was included as a random effect, the ratio of farm variance to the total phenotypic variance were ranged from 4% (EMA) to 18% (CWT). Top 10% and bottom 10% of female cows were selected based on the breeding values from model 1, and the Spearman's rank correlation coefficients among models were estimated for each trait within selected group. The correlation coefficients were ranged from 0.57 to 0.95 in top 10% group and from 0.68 to 0.95 in bottom 10% group. These results show that the discrepancies in the rankings of breeding values can be based on the models applied. In conclusion, the results obtained in this study suggest that the herd effect or farm effect should be included in the analytical model when breeding values are estimated with the purpose of improvement of carcass traits of Hanwoo breeding cows.

Identification of Quantitative Traits Loci (QTL) Affecting Growth Traits in Pigs

  • Molecular genetic markers were used to detect chromosomal regions which contain economically important traits such as growth, carcass, and meat quality traits in pigs. A three generation resource population was constructed from a cross between Korean native boars and Landrace sows. A total of 240 F2 animals from intercross of F1 was produced. Phenotypic data on 17 traits, birth weight, body weights at 3, 5, 12, and 30 weeks of age, teat number, carcass weight, backfat thickness, body fat, backbone number, muscle pH, meat color, drip loss, cooking loss, water holding capacity, shear force, and intramuscular fat content were collected for F2 animals. Animals including grandparents (F0), parents (F1), and offspring (F2) were genotyped for 80 microsatellite markers covering from chromosome 1 to 10. Least squares regression interval mapping was used for quantitative trait loci (QTL) identification. Significance thresholds were determined by permutation tests. A total of 10 QTL were detected at 5% chromosome-wide significance levels for growth traits on SSCs 2, 4, 5, 6, and 8.
