Effects of Divalent Cations on Alkaline Hydrolysis of Poly(ethylene terephthalate) Fabric (2가 양이온이 폴리에틸렌 텔레프탈레이트 직물의 알칼리 가수분해에 미치는 영향)

  • Inorganic salts have negative or positive effects on the rates of many chemical reactions and also the rates of acidic and alkaline hydrolysis of carboxylic esters. The direction of salt effects on the hydrolysis of ester depends on the charge of esters. It is expected that the rate of the alkaline hydrolysis of Poly(ethylene terephthalte)(PET), polymeric solid carboxytic polyester with carboxyl end group at the polymer end, is also influenced by inorganic salts. In the present work, to clarify the effect of divalent cations on the alkaline hydrolysis of PET, many salts with divalent cations like $MgCl_{2},CaCl_{2},SrCl_{2},BaCl_{2},$ were added to the aqueous alkaline solutions. Then PET was hydrolyzed with aqueous NaOH solution having many salts under various conditions. Some conclusions obtained from the experimental results were summarized as follows. Many salts with various divalent cations increased or decreased the reaction rate of alkaline hydrolysis of PET depending on their electrophilicity, hydration property, ability of ion pair formation, solubility, and the degree of interactions between divalent cations and anions, etc. The hydrolysis was interrupted in the order of $Ca^{+2} and was generally accelerated in the order of $Ba^{+2}. It was inferred from the increase in ΔS$^*$and the decrease in the ΔG$^*$that the divalent cations $Sr^{+2}$ and $Ba^{+2}$attracted by PET increased the collision frequency between carbonyl carbon and $OH^{-}$ion and then accelerated the reaction rate. $Mg^{+2}$and $Ca^{+2}$decreased the reaction rate because of their strong interaction with $OH^{-}$.

Analysis of Amino Acid Residues Involved in Activities of Chitin Deacetylase of Aspergillus nidulans (Aspergillus nidulans에서 분리된 키틴 탈아세틸화 효소활성에 영향을 미치는 아미노산 잔기 분석)

  • Native chitin deacetylase of Aspergillus nidulans was purified to apparent homogeneity by a combination of phenyl-Sepharose and Q-Sepharose column chromatography. In order to analyze the amino acid residues involved in the enzyme activity, the enzyme was chemically modified with chemical agent, which selectively reacted with the specific amino acid residue on the protein. When the enzyme was chemically modified with diethylpyrocarbonate, which specifically reacted with histidine residues on the protein, the activity was eliminated. The chitin deacetylase, chemically modified with 100 ${\mu}M$ modifier at the residue of arginine or tyrosine, has shown to have decreased activities. It was shown that the modification at aspartic acid or glutamic acid did not affect the enzyme activity to a greater extent, which would not implicate that acid amino residues were directly involved in catalytic reaction and would affect on the global structures of the proteins. This results demonstrated that histidine and tyrosine residues of enzyme would participate in an important function of the chitin deacetylase activity.

  • Using six kinds of epoxy acrylates and a conductive carbon black, photosensitive resistor pastes were fabricated and then their developability in alkaline aqueous solution and the resistance values after thermal curing were evaluated. In order to impart the photocurability by UV exposure and the developability on alkaline solution, epoxy acrylate oligomers with carboxyl group, acrylate monomers, a photoinitiator and so forth were used. In addition, an organic peroxide was added into the paste to get a thermally curable composition. As a result, some of the pastes were not developed depending on the kinds of oligomers and, in the developed pastes, the measured resistance showed the different values depending on their compositions, even though they contain the same amount of carbon black. Finally, the optimum oligomer was selected and then, by adjusting the amount of carbon black, the kind of monomer and the curing temperature, the photosensitive resistor paste composition which showed the sheet resistance of about 0.5 $k{\Omega}/sq.$ could be obtained.

Development of Mechanically Expanded Gelatin-AAc-PLLA/PLCL Nanofibers for Vascular Tissue Engineering by Radiation-based Techniques (방사선 기반에 의한 기계적으로 공극을 증가시킨 젤라틴이 도입된 혈관조직공학용 PLLA/PLCL 나노섬유 지지체의 개발)

  • Vascular tissue engineering has been accessed to mimic the natural composition of the blood vessel containing intima, media, and adventitia layers. We fabricated mechanically expanded PLLA/PLCL nanofibers using electrospinning and UTM. The pore size of the meshes was increased the gelatin immobilized AAc-PLLA/PLCL nanofibers ($203.30{\pm}49.62microns$) than PLLA/PLCL nanofibers ($59.99{\pm}8.66microns$) after mechanical expansion. To increase the cell adhesion and proliferation, we introduced carboxyl group, and gelatin was conjugated on them. The properties of the PLLA/PLCL nanofibers were analyzed with SEM, ATR-FTIR, TBO staining, and water contact angle measurement, general cell responses on the PLLA/PLCL nanofibers such as adhesion, proliferation, and infiltration were also investigated using smooth muscle cell (SMC). During the SMC culture, the initial viability of the cells was significantly increased on the gelatin immobilized AAc-PLLA/PLCL nanofibers, and infiltration of the cells was also enhanced on them. Therefore, gelatin immobilized AAc-PLLA/PLCL nanofibers and mechanically expanded meshes may be a good tool for vascular tissue engineering application.

Effects of Carbon Nanotube Addition on the Mechanical Properties of Dental Glassionomer Cement (탄소나노튜브 첨가에 의한 치과용 글라스아이오노머 시멘트의 기계적 특성)

  • The aim of this study was to investigate the effect of multiwall carbon nanotube functionalized with carboxyl group (MWCNT-COOH) on the mechanical properties of dental glassionomer cement (GIC). MWCNT-COOH was prepared by the acid oxidative method. The MWCNT-COOH was incorporated into a commercial GIC powder or liquid at 0.5 wt% or 1.0 wt%. The net setting time of the cements was measured in accordance with ISO 9917 (Dental water-based cement). Specimens for compressive strength ($4mm{\varphi}{\times}6mm$), diametral tensile strength ($6mm{\varphi}{\times}4mm$) and flexure strength with modulus ($2mm{\times}2mm{\times}25mm$) were prepared by mixing with the cement liquid and kept in water bath of $(37{\pm}1)^{\circ}C$. Mechanical tests were conducted in 1 d, 7 d, and 14 days at a cross-head speed of 1 mm/min. Compressive strength of GIC mixed with 0.5 wt% MWCNT-COOH increased significantly at 7 d. However, overall mechanical properties of GIC modified with MWCNT were not significantly increased with a delayed setting time, in comparison with control cement. Overall results indicated that the MWCNT/GIC composite cements showed a limited strengthening effect for dental glassionomer cement.

  • The crystal structure of the potassium salt of penicillin V has been studied by the X-ray crystallographic methods. Crystal data are as follows; potassium 3,3-dimethyl-7-oxo-6-phenoxyacetoamido-4-thia-1- azabicyclo[3.2.0]-heptane-2${\alpha}$-carboxylate, $K^+{\cdot}C_{16}H_{18}N_2O_5S^-$, $M_r$= 388.5, triclinic, Pl, a= 9.371 (1), b= 12.497 (2), c= 15.313 (2) ${\AA},\;{\alpha}= 93.74\;(2),\;{\beta}=99.32\;(1),\;{\gamma}=90.17\;(1)^{\circ},\;V=1765.7\;(2)\;{\AA}^3$, Z=4, $D_m=1.461\;gcm^{-1},\;{\lambda}(Cu\;K{\alpha})=1.5418\;{\AA},\;{\mu}=40.1\;cm^{-1}$, F(000)=808, T=296 K. The structure was solved by the heavy atom and difference Fourier methods with intensity data measured on an automated four-circle diffractometer. The structure was refined by the full-matrix least-squares method to a final R= 0.081 for 3563 observed $[I_0{\geq}2{\sigam}(I_0)]$ reflections. The four independent molecules assume different overall conformations with systematically different orientations of the phenyl groups although the penam moieties have the same closed conformations. There are intramolecular hydrogen bonds between the exocyclic amide nitrogen and phenoxy oxygen atoms. The penam moiety is conformationally very restricted although the carboxyl and exocyclic amide groups apparently have certain rotational degrees of freedom but the phenyl group is flexible about the ether bond despite the presence of the intramolecular N-H${\cdots}$O hydrogen bond. There are complicated pseudo symmetric relationships in the crystal lattice. The penam moieties are related by pseudo 20.5 screw axes and the phenyl groups by pseudo centers of symmetry. The potassium ions, related by both pseudo symmetries, form an infinite zigzag planar chain parallel to the b axis. Each potassium ion is coordinated to seven oxygen atoms in a severely distorted pentagonal bipyramid configuration, forming the infinite hydrophilic channels which in turn form the molecular stacks. Between these stacks, there are only lipophilic interactions involving the phenyl groups.

Study on the Thermal Degradation Behavior of FKM O-rings

  • The degradation mechanism and physical properties of an FKM O-ring were observed with thermal aging in this experiment. From X-ray photoelectron spectroscopy (XPS) analysis, we could observe carbon (285 eV), fluoro (688 eV), and oxygen (531 eV) peaks. Before thermal aging, the concentration of fluoro atoms was 51.23%, which decreased to 8.29% after thermal aging. The concentration of oxygen atoms increased from 3.16% to 20.39%. Under thermal aging, the FKM O-ring exhibited debonding of the fluoro-bond by oxidation. Analysis of the C1s, O1s, and F1s peaks revealed that the degradation reaction usually occurred at the C-F, C-F2, and C-F3 bonds, and generated a carboxyl group (-COOH) by oxidation. Due to the debonding reaction and decreasing mobility, the glass transition temperature of the FKM O-ring increased from $-15.91^{\circ}C$ to $-13.79^{\circ}C$. From the intermittent CSR test, the initial sealing force was 2,149.6 N, which decreased to 1,156.2 N after thermal aging. Thus, under thermal aging, the sealing force decreased to 46.2%, compared with its initial state. This phenomenon was caused by the debonding reaction and decreasing mobility of the FKM O-ring. The S-S curve exhibited a 50% increase in modulus, with break at a low strain and stress state. This was also attributed to the decreasing mobility due to thermal aging degradation.

Raw Inonotus obliquus polysaccharide counteracts Alzheimer's disease in a transgenic mouse model by activating the ubiquitin-proteosome system

  • BACKGROUND/OBJECTIVES: Inonotus obliquus has been used as antidiabetic herb around the world, especially in the Russian and Scandinavian countries. Diabetes is widely believed to be a key factor in Alzheimer's disease (AD), which is widely considered to be type III diabetes. To investigate whether I. obliquus can also ameliorate AD, it would be interesting to identify new clues for AD treatment. We tested the anti-AD effects of raw Inonotus obliquus polysaccharide (IOP) in a mouse model of AD (3×Tg-AD transgenic mice). MATERIALS/METHODS: SPF-grade 3×Tg-AD mice were randomly divided into three groups (Control, Metformin, and raw IOP groups, n = 5 per group). β-Amyloid deposition in the brain was analyzed using immunohistochemistry for AD characterization. Gene and protein expression of pertinent factors of the ubiquitin-proteasome system (UPS) was determined using real-time quantitative polymerase chain reaction and Western blotting. RESULTS: Raw IOP significantly reduced the accumulation of amyloid aggregates and facilitated UPS activity, resulting in a significant reduction in AD-related symptoms in an AD mouse model. The presence of raw IOP significantly enhanced the expression of ubiquitin, E1, and Parkin (E3) at both the mRNA and protein levels in the mouse hippocampus. The mRNA level of ubiquitin carboxyl-terminal hydrolase isozyme L1, a key factor involved in UPS activation, also increased by approximately 50%. CONCLUSIONS: Raw IOP could contribute to AD amelioration via the UPS pathway, which could be considered as a new potential strategy for AD treatment, although we could not exclude other mechanisms involved in counteracting AD processing.

The Kinetics of Delignification in Oxygen·Alkali pulping (효소(酵素)·알칼리 증해(蒸解)의 탈(脱)리그닌에 관(関)한 동역학적(動力學的) 분석(分析))

  • In order to obtain more detailed information concerning the degradation of lignin in the oxygen alkali pulping single stage isothermal delignification of pine wood meal (Pinus koraiensis S. et Z.) was studied in the oxygen alkali system at five temperature level ($110^{\circ}C$, $120^{\circ}C$, $130^{\circ}C$, $140^{\circ}C$, $150^{\circ}C$) for 60 min.. The rate constant, activation energy, oxygen and alkali consumption during the oxygen alkali delignification were determined by the kinetic method. The 2/5 of total lignin was eliminated at the start of the reaction. The delignification rate constant was about 3 times that of caustic soda pulping. The activation energy was about 1/3 lower than in caustic soda pulping. Like oxygen consumption, alkali consumption was also rapid early at the reaction and almost ceased after about 10 min.. The degradation reaction of lignin was strongly dependent upon the pH decrease of the cooking liquor by organic acid generated in pulping. The lignin in the oxygen alkali pulping degraded into lover molecular weight and had more hydrophillic properties. The methoxyl group decreased considerably at the first of oxygen alkali delignification, while the carbonyl, carboxyl and phenolic hydroxyl group increased rapidly.

The Study on Natural Dyeing with Artemisia (쑥을 이용한 천연염색에 관한 연구)

  • Recently the interest in natural dyeing has been increased because of the color clarity, natural grace and reactionism in fashion. Indigo dyeing, safflower dyeing and Gal-ott in Cheju-Do become generally known, so the study about the natural dyeing is continued with national intrust and support. In this study, 1 used artemisia for various dyeing tests because we can get it easily. 1 tested the dyeability in wool as well as cotton and silk with wormwood in natural dyeing material. I also dyed nylon with the same material for the comparison of the molecular structure. The mercerization and the chitosan treatment were done in cotton to improve the low dyeability in the natural dyeing. The result of this study are as follows; We have to dye repeatedly to get deep color in natural dyeing, and mordant treatment brought good result in color difference and dyeing fastness. 1. Compared with silk, wool and nylon, the dyeability of cotton was the worse. The color difference of cotton was 18.81 without mordants , and 24.05 with mordant. The dyeability of cotton was much increased by mordants such as potassiumdichromate, copper sulfate, iron sulfate and salt water. The color of cotton was turned into yellow-green in potassium dichromate and yellow-green with deep green in copper sulfate. The mercerization and the chitosan treatment of cotton made the improvement in color, dyeability, laundering fastness, abrasion fastness. 2. The color difference of silk was 3 times as high as cotton. It showed the similar degree with the chitosan treated-cotton. That is to say, silk had good dyeability because it contains amino group and carboxyl group. The dyebility of silk was increased by a mordant such as iron sulfate, potassium dichromate and salt water, Drycleaning fastness showed 5 grade, abrasion-fastness was high over 4~5 grade and sunlight fastness showed 1 grade in all case. 3. The color difference of wool was the best among four fabrics because of 18 kinds of amino acids. It is considered that the sulfate of wool has the polarity and help metal- mordants get the better dyeability such as potassium dichromate, iron sulfate, copper sulfate, and aluminum chloride. The color was reddish-green in potassium dichromate, yellow.greenin iron sulfate and copper sulfate, and yellow in the rest mordants. Drycleaning fastness of wool showed over 4~5 grade, abrasion fastness 5 grade. Sunlight-fastness 1 grade. 4. The dyeability of nylon was almost same because of the similar molecular structure with silk. The clarity of color was poor. The color was yellow-green in copper sulfate and yellow in the rest mordants. Laundering fastness and abrasion-fastness of nylon was good. Sunlight- fastness represented 1 grade.
