Physio-Chemical Studies on the After-Ripeng of Hot Pepper Fruit -Part VII. Effects of Ethephon on the Major Compoments- (신미종(辛未種) 고추의 추숙(追熟)에 관(關)한 생리화학적(生理化學的) 연구(硏究) -제7보 주요성분(主要成分)에 미치는 Ethephon의 효과(?果)-)

  • The physiological and chemical investigations to explain the after-ripening processes in got green pepper fruit were carried out by treating the fruit with ethephon either alone or with phenylalanine. The studied metabolic changes in fruit during after-ripening period was carbon dioxide and oxygen concentration in interior of the pepper fruit, total carotenoid, ${\beta}-carotene$, total sugar, and free reducing sugar in pepper fruit. These metabolic changes were explained inrelation to the color enhancement judged by the color score to explain the after-ripening processes. Ethephon treatment at 500 ppm significantly accelerated color enhancement as compared to the control and further ethephon treatment increased the number by 20 percent which was not possible in control during same after-ripening period. The oxygen concentration in interior of the pepper fruit during after-ripening period was increased in control when the color score (color enhancement) increased rapidly. However, with ethephon treatment, the oxygen concentration was decreased when the color score increased. Although total and free reducing sugar content were decreased during the after-ripening period total carotenoid and ${\beta}-carotene$ content was increased by 50 and 200 percent, respectively, over control. Also the capsaicin contents was increased by 20 percent by ethephon treatment over control. Although phenylalanine treatment did not affect the capsaicin content, capsaicin content tended to be increased by phenylalanine treatment when treated with ethephon.   • The increasing number of natural disasters resulting from anthropogenic greenhouse gas emissions has prompted the development of a gas separation membrane. Carbon dioxide ($CO_2$) is the main cause of global warming. Organic polymeric membranes with inherent flexibility are good candidates for use in gas separation membranes and poly(dimethyl siloxane)(PDMS) specifically is a promising material due to its inherently high $CO_2$ diffusivity. In addition, poly(vinyl pyrrolidine)(PVP) is a polymer with high $CO_2$ solubility that could be incorporated into a gas separation membrane. In this study, poly(dimethyl siloxane)-poly(vinyl pyrrolidine)(PDMS-PVP) comb copolymers with different compositions were synthesized under mild conditions via a simple one step free radical polymerization. The copolymerization of PDMS and PVP was characterized by FTIR. The morphology and thermal behavior of the produced polymers were characterized by transmission electron microscopy (TEM), thermogravimetric analysis (TGA), and differential scanning calorimetry (DSC). Composite membranes composed of PDMS-PVP on a microporous polysulfone substrate layer were prepared and their $CO_2$ separation properties were subsequently studied. The $CO_2$ permeance and $CO_2/N_2$ selectivity through the PDMS-PVP composite membrane reached 140.6 GPU and 12.0, respectively.

Study on solubilization of sewage sludge using electrolysis techniques (전기분해에 의한 하수슬러지 가용 효과 연구)

  • Although sludge production has been increased due to the number of the wastewater treatment plants expanded, needs of the techniques for the sludge reduction and disposal has been issued importantly because the sludge dumping to ocean is prohibited from 2012 by the London Dumping Convention. Therefore, the sludge solubilization using electrolysis as an alternative techniques for the sludge disposal was carried out in this study. Iridium coated titanium based insoluble electrodes were used and 20 volt was applied to the electrolysis reactor using DC power supply. Supernatants of the treated sludge was monitored: The soluble COD, TN, TP of it was increased to 151%, 22% and 6% respectively. And the sludge floc size distribution was changed, that is, the flocs ranged from 0.1 to 1.0 ${\mu}m$ were increased. All of these results indicate that the cells were lysed and the internal matters bursted out of the cell after electrolysis. As well as the reduction of the sludge production, the soluble organic matters from the cells could be used as an external carbon sources in the advanced wastewater treatment plants.

Bacillus ginsengihumi sp. nov., a Novel Species Isolated from Soil of a Ginseng Field in Pocheon Province, South Korea

  • A Gram-positive, aerobic or facultative anaerobic, non motile, endospore-forming bacterial strain, designated Gsoil $114^T$, was isolated from a soil sample of a ginseng field in Pocheon Province (South Korea), and was characterized taxonomically by using a polyphasic approach. It grew well on nutrient agar medium and utilized a limited number of organic substrates as sole carbon sources, including D-xylose and some other carbohydrates, but did not utilize L-amino acids and organic acids. The isolate was positive for oxidase test but negative for catalase, and negative for degradation of macromolecules such as starch, cellulose, xylan, casein, chitin, and DNA. The G+C content of the genomic DNA was 41.8 mol%. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The major fatty acids were $anteiso-C_{15:0}$ (32.1%), $iso-C_{15:0}$ (30.5%), and $anteiso-C_{17:0}$ (30.2%). Comparative 16S rRNA gene sequence analysis showed that strain Gsoil $114^T$ fell within the radiation of the cluster comprising Bacillus species and joined Bacillus shackletonii LMG $18435^T$ with a bootstrap value of 95%. The highest 16S rRNA gene sequence similarities were found with Bacillus shackletonii LMG $18435^T$ (97.6%), Bacillus acidicola DSM $14745^T$ (96.9%), Bacillus sporothermodurans DSM $10599^T$ (96.5%), and Bacillus oleronius DSM $9356^T$ (96.5%). The phylogenetic distance from any other validly described species within the genus Bacillus was less than 96%. DNA-DNA hybridization experiments showed that the DNA-similarities between strain Gsoil $114^T$ and closest phylogenetic neighbors were less than 39%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $114^T$ (=KCTC $13944^T$=DSMZ $18134^T$) was classified in the genus Bacillus as the type strain of a novel species, for which the name Bacillus ginsengihumi sp. nov. is proposed.

Strength Analyses of New 2- and 3-Axis-Type Small Multiplying Gears in Dental Hand-Pieces (치과드릴 구동용 신 소형 2축 및 3축형 증속기어 강도특성 비교)

  • Two types of very small multiplying gears and arrays have been developed for new dental hand-pieces, and the increased speed ratios, modules, number of teeth, gear diameters, and gear types were calculated based on the dynamics of the machinery. The contacting and bending strengths were evaluated for gear teeth with two design concepts using AGMA equations and finite element analyses, and the contacting stresses on teeth with and without DLC (diamond-like-carbon) coating layers were calculated. Fatigue and tension tests were performed to obtain an S-N curve, the Young's modulus, and the strength of the gear material, and these were utilized in the analyses. Slightly larger stresses were found for 2-axis-type gears than for other types of gears, and the S-N curves showed that a gear lifetime of 109 cycles was satisfied. The contacting stresses in gears coated with DLC were reduced by 30%. A new prototype model of a hand-piece with small gears was successfully fabricated and tested.

Microbial Diversity, Survival and Recovery as Bioindicators in Soils from Different Parent Materials in Korea (생물학적 토양 지표로서의 모재별 미생물의 다양성과 복원율)

  • To develop indicators for soil health evaluation, biological characteristics of native soils from the different parent materials were studied. Survival rate of fluorescent Pseudomonas in soils was the lowest as 0.1% while those of thermophilic Bacillus and alkaliphilic bacteria were over the 90% by the soil drying stress. There was positive relationship between soil microbial biomass and organic carbon exudated from the microbial biomass by the treatment. The average air-drying effect of soils was 39.7% with ranges of 9.7~95.0%. The propagules of mesophilic Bacillus and Gram negative bacteria were increased by the re-wetting of dried soils. Soil pH affected positively to the recovering rate of microbial number. Average recovering rate of microbes was 65.3%, and there was positive relationship between microbial biomass recovery and fluorescent Pseudomonas population.

Effect Naetakchungumsankamibang on Skin tumor induced by 3-MCA and Immunological Response (內托千金散加味方이 3-MCA로 誘發된 皮膚癌 및 免疫調節作用에 미치는 影響)

  • In order to investigate the effect of Naetakchungumsankamibang(NTCGS) water extract on the skin tumor induced by 3-MCA and immunological responses in mice, the cytotoxicity against SK-MEL-2 cells and total number of tumors induced by 3-MCA were measured. The numbers of WBC, platelets and RBC, plaque forming cells, hemagglutinin titer, hemolysis titer, carbon clearance, proliferation of splenocyte by thymidine uptake assay, splenic leukocyte by FACS analysis and $TNF-{\alpha}$ were also measured for the evaluation of the immunological responses. The results were obtained as follows: 1. In cytotoxicity against SK-MEL-2 cells, concentration inhibiting cell growth up to below $20\%$ of control was recognized at 1mg/ml of NTCGS. 2. In Inhibitory effect on the skin tumor induced by 3-MCA, the results showed a strong inhibitory effect of NTCGS. 3. In hematological changes in the tumor bearing mice, the numbers of WBC decreased significantly in NTCGS treated group as compared with control. 4. In hematological changes in the tumor bearing mice, the numbers of platelets increased significantly in NTCGS treated group as compared with control. 5. In hematological changes in the tumor bearing mice, the numbers of RBC increased with no significance in NTCGS treated group as compared with control. 6. Effects of the plaque forming cells in the tumor bearing mice, NTCGS treated group exhibited a significant effect compared with control. 7. In terms of the effects on hemagglutinin titer, NTCGS treated group showed higher level than control, without significance. 8. In terms of the effects on hemolysis titer, NTCGS treated group showed higher level than control, without significance. 9. In terms of the effects on phagocytic index K in Balb/C mice, NTCGS treated group showed significant difference from control. 10. In terms of the effects on proliferation of splenocyte by thymidine uptake assay, NTCGS showed significant effect at the concentration of 0.5mg/ml. 11. In terms of the effects on splenic leukocyte of Balb/C mice by FACS analysis, NTCGS treated group showed significantly higher level of helper T cell, B cell and macrophage than in control. 12. In terms of the effects on the secretion of $TNF-{\alpha}$, the treated group showed significant effect at the concentration of 1mg/ml of NTCGS. Based on the results summarized above, NTCGS is considered to have antitumor activity and immunological responses against skin tumor, and to be usable fur the treatment.

  • A series of laboratory experiments were conducted to find out the populations and identification of soil bacteria, fungi and their B/F ratio in the rhizosphere of intensively cultivatad hot-pepper, garlic, flower plants, chinese cabbage, and round onion. The results obtained are summarized as follows: 1. The number of bacteria, fungi and their B/F ratio are remarkably lower than that of normal paddy soils. 2. Nitrate reducers and bacteria which utilized simple sugars for their sole carbon source are predominated in the rhizosphere of intensively cultivated upland crops. 3. Alkaligenetic bacteria predominate in rhizosphere of garlic and tomato cultivated upland soils. 4. Genera of Pseudomonas, Xanthomonas, Bacillus, Arthrobacter, and Achromobacterium are the most common species in the rhizosphere of intensively cultivated upland crops and flower plants. 5. Phytotoxin producers such as Stachybotris sp. were identified in all rhizospheres of intensively cultivated upland crops and flower plants. 6. Most common and highest population of soil fungi were obtained for the genera of Penicillium, Humicola, Phoma and Aspergillus in the rhizosphere of intensively cultivated upland crops and flower plants.

Analysis on the Linkage between SDGs Framework and Forest Policy in Korea (국내 산림정책과 지속가능발전목표(SDGs)간의 연관성 분석)

  • This study analysed the linkage between national forest policy in Korea, namely the $5^{th}$ National Forest Master Plan, 2016 Korea Forest Service Performance Management Plan, the $3^{rd}$ National Sustainable Development Plan, and UN Sustainable Development Goals (SDGs). The 7 strategies of the $5^{th}$ National Forest Master Plan were related to 11 Goals of SDGs, and 5 strategies of 2016 Korea Forest Service Performance Management Plan were associated with 7 areas of SDGs, and 4 strategies within $3^{rd}$ National Sustainable Development Plan were linked to 7 Goals of SDGs. Among 87 national forest indicators compiled from three respective forest-related policies of Korea, 45 national indicators were related to 18 SDGs indicators. This indicates that 52% of national indicators of Korean forest policy are reflecting the language of SDGs. However, seeing from SDGs perspective, only 18 out of 241, which accounts for 7.8% of SDGs indicators are related to national indicators. The findings imply that a number of national forest-related indicators do not meet the diverse dimension of SDGs which provides potential areas for forest to contribute. Based on the findings, following recommendations were suggested: 1) the term used in forest policy should be aligned to SDGs targets so that it can be embedded in national policies, and 2) indicators should be further contextualized as well as in its assessment system. Lastly, it suggests for leveraging 3) '5 Processes of sub-national climate change adaptation plan' and the core concept of REDD+ MRV which could provide fundamental background for implementing SDGs framework to national forest policy.

Properties Evaluation and flowability of Controlled Low Strength Materials Utilizing Industrial By-Products (산업부산물을 활용한 저강도 고유동 채움재의 유동성 및 물성평가)

  • The purpose of this study is to expand the use of coal ash and coal slag in thermal power plants. In addition, controlled low strength materials was developed to prevent mine settlement. Bottom ash and KR slag are mixed at ratio of 7:3 to expand the use of industrial by-products through carbonate reaction and inhibit the exudation of heavy metals. In order to efficiently fill the abandon mine, workability and physical properties were evaluated according to flow. As a result of elution of harmful substance experiment, it was confirmed that the carbonation reaction inhibited the elution of heavy metals. It was confirmed that the difference in water ratio was the difference in specific surface area of the controlled low strength materials. It was confirmed that the working efficiency is excellent when the flowability is 300mm compared to 260mm. compressive strength measurement result was relatively high at 260mm compared to 300mm because the number of pores due to decrease of water ratio was small.
