A Comparison of Nutritional Components of Loquat (Eriobotrya japonicaLindl.) Powder in Different Aerial Components (비파 부위별 분말의 영양성분 비교)

  • This study compares the major nutritional components of the leaf, flesh, and seed of dry loquat (Eriobotrya japonicaLindl.). Among proximate compositions, the crude fat, crude ash, and dietary fiber of the leaf exceeded those of the seed and flesh, whereas the carbohydrate content of the leaf was lower than that of the seed and flesh. The main component of free sugars in the leaf, flesh, and seed was fructose. Total amino acids of the leaf, flesh, and seed were 552.43, 63.00, and 260.29 mg%, respectively. Although the amino acid composition of the leaf, flesh, and seed varied, glutamic acid and ${\gamma}$-aminobutyric acid were the major amino acids in the leaf, flesh, and seed. Major fatty acids of total lipid were oleic acid and stearic acid in the leaf and seed, and the major acid was linoleic acid in the flesh. Major organic acids were oxalic acid in the leaf, maleic acid in the flesh, and citric acid in the seed. Vitamin C content was higher in the seed than in the leaf and flesh.

  • The chemical components of Korean pollens and pollen extracts were analysed to provide fundamental data for pollen processing and products development. The contents of crude protein, crude lipid, crude ash, crude fiber and carbohydrate of pollens were $14.0{\sim}22.3%,\;3.0{\sim}8.8%,\;2.2{\sim}3.3%,\;0{\sim}37.5%,\;and\;31.9{\sim}64.4%$ respectively, showing the specially high crude fiber content (37.5%) in Pine pollen. Almost all the carbohydrate, lipid, protein and ash components except fiber were extracted by water and n-hexane. Free sugars identified in water extracts $(10\;^{\circ}Bx)$ of pollens were fructose and glucose as major component and maltose as minor component. The proportions of three free sugars to total soluble solid content of Mixed, Acorn, and Pine water extracts were 73.6%, 85.4% and 47.7% respectively. Potassium and phosphorus content in pollens and pollen water extracts were high, but Ca, Na, Mg, Fe and Zn were not major mineral components. The essential amino acids such as leucine, phenylalanine, methionine, lysine, valine, isoleucine and threonine were contained richly in pollens and those were almost completely extracted by water. Vitamin $B_1$ and C were not detected, but only small amount of vitamin $B_2$ was detected in the pollens and pollen extracts tested. The essential fatty acids such as linoleic acid and linolenic acid were contained richly in pollens. The proportion of unsaturated fatty acids to total fatty acid content in Mixed, Acorn and Darae pollen except Pine was higher than 50%.

Accelerated and restrained effects of gibberellic acid on the growth of Chlorella (Chlorella의 생장에 미치는 gibberellic acid의 촉진 및 억제효과)

  • This investigation was proceeded to define the effects of gibberellic acid on the growth of Chlorella by determining the contents of chlorphyll and changes in various components in Chlorella cells according to the concentration of treated GA. The growth of Chlorella was accelerated with telative low concentrations of GA(10, 40 ppm) and was restrained with relative high concentrations of GA(70, 100, 200 ppm). The synthetic ability of chlorophyll of GA was inhibited generally in proportion to the concentration of treated-GA and the higher the concentration of GA was applied, the longer time was required in the restoration. The contents of RNA, protein and soluble carbohydrate were increased PCA-soluble amino acid and polysacharide were decreased in those cell components between the accelerated and restrained group. Consequently, the effect on accelerated growth in relative low concentrations of GA is considered to be caused by the powerful effet on expansion growth of GA. It is presumed that the effect of restrained growth in relative high concentrations of GA is due to the inhibitory effect on the chlorophyll synthesis.

  • This study was carried out to comparison of the contents of general and mineral components between the original sample and water extract in each part of omija. The content of Carbohydrate was highest in endocarps, and that the crude protein and crude lipid in seeds. The contents of K and Mn in the fruits, endocarps, and seeds were all higher than those of the other cations. The content of K and Zn in endocarps were three to four times as much as those of seeds. and the content of Na, Ca, and Cu in endocarps were 1.5 to 1.75 times of seeds. Mineral contents by water extract in each part were ordered as K, Mg, Ca, and Fe. As the percentage of each ion in water extract on the basis of original sample, Fe was the highest ratio of behavior, and Mn was lowest.

  • In this study, rat hepatocytes known to have active carbohydrate metabolism were obtained by using the liver perfusion technique to examine the hypoglycemic action of red ginseng saponin components [ginsenoside (mixture, $Rb_1$, and $Rg_1$)] and incubated in two different media-one containing insulin and glucagon (control group), and the other containing glucagon only, The specific activities of some regulatory enzymes such as glucokinase, glucose 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and glucose 6-phosphatase, in main pathways which were directly related to the glucose metabolism were compared between these two kinds of hepatocytes cultured in two different media. The effects of red ginseng saponin components [ginsenoside (mixture, $Rb_1$, and $Rg_1$)] under the concentration of $10^3$~$10^6$% on these enzymes In hepatocytes were also investigated, when they were added to these two media. The results were as follows. The specific activity of enzymes such as glucokinase, glucose 6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase related to glucose-consuming pathways of insulin-deficient group was much less than control one, however, their decreased activity was recovered after the addition of ginseng components at all range of concentrations. The increased specific activity of these on - zymes was shown by the addition of ginseng components to the control group. On the other hand, the specific activity of glucose 6-phosphatase related to glucose-producing pathway of insulin-deficient group was much higher than control one, but their Increased activity was decreased after the addition of ginseng components at all range of concentrations. The same results were obtained after the addition of ginseng components to the control group. These results suggest that the red ginseng saponin components might better diabetic hyperglycemia by regulating the activity of enzymes related to glucose metabolism directly and/or Indirectly though more detailed studies were needed.

  • This study has been carried out to investigate the effect of protein components of a diet on the increasing of larval body weight and diet efficiency, as well as the amylase activity of larval digestive juices during the 5th instar. Defending on the amounts of soybean meal as a protein source, the six different kind of artificial diets containing mulberry leaf powder fed to the larvae. The results obtained may be summarized as follows: 1. As the protein components of the diet were increased, the amount of increased larval body weight was also increased. 2. As the protein components of the diet were increased, both the cocoon weight and cocoon layer weight were also increased. 3. As the protein components of the diet were increased, both the amount of diet digested and coefficient of digestibility were also increased. 4. As the protein components of the diet were increased, the diet efficiency of larvae were also increased. 5. But the experimenters could not observe any correlation between the increase of protein components of a diet and the amylase activity of the digestive juices.

Modeling for the Prediction of Liquid Food Density (액체식품의 밀도예측을 위한 모델링)

  • For the development of a general mathermatical model to predict the density of liquid foods based on temperature and composition of each major component, the major components of liquid foods considered in this study were water, protein, fat, carbohydrate, fiber and ash. These samples were subdivided into sixteen pure components. The density of each sample was measured by a volumetric pycnometer at the temperature range of $0^{\circ}C\;to\;100^{\circ}C$ for three different solid content suspensions, The density values of pure component solids were calculated from the assumed model at given temperature, using the experimental values of three different solid content suspensions with known water fraction and density model of water. Using these calculated density data at the temperature range of $0^{\circ}C\;to\;100^{\circ}C$, the coefficients of ther density model for each pure component were determined by the OPT Subroutine Program. The density model developed in this study can be used to predict the density values of liquid foods at given temperature and composition.

  • These studies were conducted to investigate the changes in carbohydrate properties of the endosperm during the stages of maturity. Original moisture continued to decrease while 1,000-kernel weight and test weight increased steadily in all varieties with maturation. Starch content of the endosperm increased continuously by 35 to 40 days after heading. The B-type starch granules synthesis of the the early mature variety, Chokwang, was depressed at the later stages of development. Amylose and amylopectin components of starch both increased as the kernel matured, and amylose-amylopectin ratio also increased during the same period. Amount of pentosan per kernel basis increased throughout the maturation period. Amylograph break-down had a highly negative coefficient correlation with starch and pentosan content of endosperm. Results indicated that wheat maturation was characterized by an increase in the starch and pentosan content of the kernel.

Analysis of Functional Genes in Carbohydrate Metabolic Pathway of Anaerobic Rumen Fungus Neocallimastix frontalis PMA02

  • Anaerobic rumen fungi have been regarded as good genetic resources for enzyme production which might be useful for feed supplements, bio-energy production, bio-remediation and other industrial purposes. In this study, an expressed sequence tag (EST) library of the rumen anaerobic fungus Neocallimastix frontalis was constructed and functional genes from the EST library were analyzed to elucidate carbohydrate metabolism of anaerobic fungi. From 10,080 acquired clones, 9,569 clones with average size of 628 bp were selected for analysis. After the assembling process, 1,410 contigs were assembled and 1,369 sequences remained as singletons. 1,192 sequences were matched with proteins in the public data base with known function and 693 of them were matched with proteins isolated from fungi. One hundred and fifty four sequences were classified as genes related with biological process and 328 sequences were classified as genes related with cellular components. Most of the enzymes in the pathway of glucose metabolism were successfully isolated via construction of 10,080 ESTs. Four kinds of hemi-cellulase were isolated such as mannanase, xylose isomerase, xylan esterase, and xylanase. Five $\beta$-glucosidases with at least three different conserved domain structures were isolated. Ten cellulases with at least five different conserved domain structures were isolated. This is the first solid data supporting the expression of a multiple enzyme system in the fungus N. frontalis for polysaccharide hydrolysis.

Morphological Changes by Submerged Culture Conditions for the Mycelial Optimal Growth of Cordyceps sinensis and Immunological Properties of Hot Water Extract of Mycelium (동충하초 균사체 최적 성장을 위한 심부배양 조건에 따른 형태학적 변화 및 균사체 열수 추출물의 면역학적 특성)

  • The mycelial dispersed growth of Cordyceps sinensis was optimized in submerged batch culture at initial pH of 5.0, 150 rpm, and $25^{\circ}C$. The morphological data showed much more dispersed growth of C. sinenesis at initial pH of 5.0. Also, projected area, main hyphal length and number of tips for the mycelial growth of initial pH 5.0 were higher than those of other initial pHs. The industrial medium for mycelial production of C. sinensis was determined to be molasses of 100 g and crushed brewery yeast of 10 g per liter as carbon and nitrogen sources, respectively. With these culture conditions, the maximum production of mycelia was approximately 30.0 g per liter by batch culture in 5-liter jar fermenter with no controlled pH. This result suggests that large-scale mycelia production of C. sinensis may be possible in submerged batch culture. The hot water extract of mycelia from C. sinensis was mainly composed of 83.0% carbohydrate, 11.8% protein, 1.9% lipid, and 2.4% ash and there were present glucose, mannose, galactose, and arabinose as molar ratio of 8.79 : 2.59 : 1.34 : 1.0 in the carbohydrate, respectively. In the experiment using spleen cell and macrophage, the extract showed potent mitogenic and immuno-stimulating activities and among various components, an important factor that contribute to the immunological activities was turned out to be carbohydrate moiety.
