Melamine과 cyanuric acid의 혼합투여가 붕어(Carassius carassius)의 신장에 미치는 조직병리학적 영향 (Histopathological effect of experimental feeding of melamine and cyanuric acid in kidney of crucian carp (Carassius carassius))

  • Melamine is an organic base and a trimer of cyanamide which contains 66% nitrogen by mass, but it was sometimes illegally added to food products in order to enhance the apparent protein content for the bigger profit. Many Chinese infants and pet animals fed melamine-containing formulas suffered acute kidney failure with renal crystal formation. Fish feeds were also recently found to be contaminated with melamine. This experiment has been designed to determine whether renal crystals can be experimentally induced in crucian carp (Carassius carassius) fed melamine and cyanuric acid (each at 400 mg/kg/day for 3 days) in combination, and to compare experimentally induced crystals with those from pet animals with triazine related renal failure. The results showed that all fish developed gold brown renal crystals arranged in radial spherulites in the renal tubules, similar to those detected in the pet animals.

  • Genomic DNA was extracted from the blood of the freshwater crucian carp(Carassius carassius) from Kunsan in Korea, representing genetic similarity by polymerase chain reaction amplification of DNA as twelve of arbitrary primers. The electrophoretic analysis of polymerase chain reaction-random amplified polymorphic DNAs(PCR-RADP) products showed the high levels of similarity between different individuals in crucian carp.

  • 잉어(Cyprinus carpio) 암컷과 붕어(Carassius auratus) 수컷간 유도된 잡종에 대한 세포 유전학적 연구를 수행하였다. 유도된 잡종의 세포 및 핵의 크기는 양친의 중간값을 나타내었고, 염색체의 경우 잉어, 붕어 및 유도된 잡종 모두 2n=100 으로 나타났다. Flow cytometry를 이용, 잡종의 genome size를 분석한 결과, 유도된 잡종은 3.7pg/cell를 갖음으로서 잉어(3.6pg/cell)와 붕어(3.8pg/cell)의 중간치를 나타내었다.

  • To optimize the mixing ratio of ingredients to obtain the best sensory qualities, a response surface methodology with a central composite design was performed on hot water extracts using crucian carp Carassius carassius treated with diluted vinegar (pH 5.24-5.30). The optimal solutions for four parameters (overall acceptability, odor, flavor, and taste) were obtained using the second-order response surface regression models. The optimal formulation appeared to be 124.15 g of ginger and 96.09 g of garlic with 1 kg of ground crucian carp meat based on the desirability function technique.

  • Goldfish hematopoietic necrosis (GHN) affects hematopoietic organs and causes high mortality in goldfish. The causative agent of GHN is cyprinid herpesvirus 2 (CyHV-2). The purpose of this study was to detect CyHV-2 in ornamental cyprinid fish. CyHV-2 monitoring was conducted on a monthly basis for 1 year using goldfish Carassius auratus and pearlscale goldfish Carassius auratus that had been cultured at a domestic aquafarm and imported from Singapore, respectively. The results of polymerase chain reaction (PCR) assays using specific primers for the CyHV-2 helicase gene showed that 27.8% of goldfish and 33.3% of pearlscale goldfish were positive for CyHV-2. No cytophatic effects were detected in koi fin or common carp brain cells inoculated with tissue homogenates from either fish. Our data provides the useful data for establishing future quarantine and disinfection policies.

붕어육의 단백질 열수추출물이 흰쥐의 혈청중 Insulin-like Growth Factor-I(IGF-I)과 IGF-Binding Proteins에 미치는 영향 (Effects of Carassius carassius Hot-Water Extracts on Serum Insulin-like Growth Factor-I(IGF-I) and IGF-Binding Proteins in Rats)

  • The insulin like growth factors(IGFs) are bound to several binding proteins(IGFBPs) that appear to regulate IGF transfort, receptor binding, and its action. The concentrations of these peptides are regulated by quantity and nutritional quality of dietary proteins. The aim of this study was to compare the effects of two diets, which differed in their protein source, Carassius carassius(CC), Carassius carassius hot water extract(CCHE), for 4 weeks. Body weight was significantly increased in the CC group(74.14$\pm$12.00 to 266.31$\pm$36.62g; p<0.01). Likewise, IGF I concentration of CC group(101.76$\pm$15.90 ng/ml) was significantly higher than that of CCHE group(38.50$\pm$ 11.20ng/ml; p<0.05). By western immunoblot analysis, especially IGFBP 1, 2 levels are increased, whereas IGFBP 3 level was de creased in CCHE group. After extraction of browning material from each samples, the extractive was filtered and absorbance at 420nm was measured. The absorbance of CCHE group was significantly higher than that of CC group. These results suggest that IGF I can be employed as an index of protein metabolism, particulary as a simple index in the assessing the status of protein nutrition.

납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구 (A Study on the Toxicity of Pb and Cu Compound in Carassius auratus(goldfish))

  • In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50}'s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively.   • 수은이 붕어(Carassius carassius L.)의 간, 신장 아가미에 미치는 영향을 규명하기 위하여 붕어를 1 ppm, 2.5 ppm 농도의 $HgCl_2$에 노출시킨 후 전자현미경을 사용하여 그들의 미세구조 변화에 대한 연구를 수행한 바 다음과 같은 결과를 얻었다. 1. 수은에 처리된 간세포에서는 lysosome이 증가하였으며 결정상의 구조물들을 포함한 원형의 lysosome과 히ㅛ\ulcorner두과립이나 mitochondria를 탐식한 lysosome의 2가지 형태가 관찰되었고 mitochondria는 팽대되어 내부 기질의 전자밀도가 감소되었으며 핵에서는 인의 분리현상이 관찰되었다. 2. 신장의 변화로서는 신사구체 기저막의 비후와 기부선회소관에서는 공포들의 증가와 cytoplasmic body들이 출현하였으며 공포형성은 mitochondria와 연관되어 일어났다. 또한 2.5 ppm에서는 핵의 위축이 관찰되었다. 3. Gill lamella의 상피세포에서는 대, 소형의 lysosome이 증가했으며 lamella의 막에 fuzzy한 구조가 관찰되었다. 4. 본 실험에서 관찰된 결과로써 수은에 의해 초래된 세포의 미세구조의 변화는 세포의 해독과정을 활성화시키며 energy대사과정을 손상시키는 것으로 생각된다.

각급 척추동물의 담낭 상피세포에 대한 형태학적 연구 (Morphological Study on the Epithelial Cells in the Gall Blader of Vertebrates)

  • 저자는 붕어, 두꺼비, 유혈목이, 십자매, 소를 재료로 담낭 상피세포의 조직학적인 관찰과 조직화학적인 성상의 관찰로 다음과 같은 결과를 얻었다. 1. 상피세포의 형태는 동물마다 차이를 가지며, 일부의 동물에서 보고된 이른바 통상세포와 간상세포의 2종의 세포를 조직화학적 면에서는 뒷받침 할 수 없었다. 2. 붕어는 단층 원주세포, 두꺼비는 점막주름 상부는 원추형, 주름 측면부는 원주형, 주름 기부는 사다리꼴 세포, 유혈목이는 단층 원주상피로서 높이가 얕은 원주형 내지 입방형 세포 그리고 소는 단층 원주상피로서 원주형이 되었다. 3. 세포질의 eosinophility는 붕어에서는 모든 상피세포의 핵상단부 세포질에서 강하고 균일하였고, 두꺼비에서는 세포질 전체가 약한 세포가 다수 출현하였고, 유혈목이는 핵상부 세포질이 균일하게 강하였으며, 십자매에서는 전 세포질이 균일하였고, 소에서는 세포질 전체가 비교적 균일하였으며 핵 주위 세포질이 환상으로 약하였다. 4. PAS 반응에서 동물에 따른 강약의 차이를 보였고, 세포질이 밝게 보이는 명세포와 어둡게 보이는 암세포로 나눌 수 있었으며, 명세포의 출현은 붕어에서 6.4%, 두꺼비에서 4.3%, 그리고 십자매에서 3.7%였으며 유혈목이와 소에서는 명세포가 출현하지 않았다. 5. Ninhydrin-Schiff에 양성인 단백질 함량은 소, 붕어, 십자매, 두꺼비, 유혈목이의 순이었는데 붕어, 유혈목이, 십자매에서는 점막주름 상부세포들이 풍부하였고, 두꺼비에서는 부위 별 차이가 없을 뿐만 아니라 세포질에 균일하게 함유하였다. 6. 중성 지방은 붕어, 유혈목이, 십자매, 소에서는 세포의 형태에 관계없이 염색성을 보이지 않거나 약한 세포가 출현하였고, 두꺼비에서는 세포 부위의 차이는 있으나 전세포에서 염색성을 나타내었다. 7. RNA 와 DNA는 모든 동물에서 세포의 형태에 관계없이 점막주름 상부세포가 강하고 점막주름 기부 및 측면부의 세포에서는 약한 반응을 보였다.
