골담초(Caragana sinica) 추출물의 생리활성 탐색 (Screening of Biological Activity of Caragana sinica Extracts)

  • 골담초에 함유된 페놀성 물질은 70% 에탄올을 용매로 하여 18시간 추출하였을 때 가장 많이 용출되었다. 항산화 효과 중 전자공여능을 측정한 결과는 꽃과 잎 모두 70% 에탄올 추출물에서 74%로 가장 높은 항산화 활성을 나타내었으며, ABTS radical cation decolorization을 측정한 결과, 꽃의 경우 70~90% 에탄올 추출물에서 60% 이상의 항산화 활성을 나타내었다. 지용성 물질에 대한 항산화력으로 antioxidant protection factor(PF)를 측정한 결과, 70% 에탄올 추출물에서 1.2 이상의 PF를 나타내었다. 활성산소 중 지방산화를 일으키는 hydroxyl radical에 대한 골담초 추출물의 영향은 꽃의 물 추출물과 70% 에탄올 추출물에서 대조구보다 낮은 TBARS 값을 나타내었다. ACE에 대한 골담초 추출물의 저해활성은 꽃의 경우 물과 70% 에탄올 추출물 모두 80~99%의 높은 저해활성을 나타내었고, Xoase 대한 저해활성은 꽃보다 잎 추출물의 저해활성이 높았다. 골담초 추출물의 주름개선과 미백효과를 확인한 결과 꽃과 잎 추출물 모두 페놀성 물질을 0.8 mg/mL의 농도로 처리하였을 때 50% 이상의 elastase 저해활성과 70% 에탄올 추출물에서 40.24%로 가장 높은 tyrosinase 저해활성을 나타내었고, 수렴효과는 같은 농도의 tannic acid보다 높은 수렴효과를 나타내었다.

골담초 열수추출물의 이화학적 특성 및 MIA를 이용한 동물모델에서 골관절염 개선 효과 (The Physicochemical Characteristics and Protective Effect of Hot Water Extracts of Caragana sinica on MIA-induced Osteoarthritis in Rats)

  • The purpose of this study was to examine the physiochemical characteristics of hot water extract of Caragana sinica roots and verified its protective effect on MIA-induced osteoarthritis in rats. The contents of total polyphenol and total flavonoid used in this experiment were 0.40 mg/g and 0.22 mg/g respectively. We found that the weight of all experiment groups which ingested Caragana sinica extract for three weeks evenly increased in rats with MIA-induced osteoarthritis without toxicity in the liver or kidney. In the histopathological test through the Mankin score, the extent of damage of knee joint tissue in the experiment groups to which Caragana sinica extract was administered in 5 mL/kg or 10 mL/kg was significantly lower than that of the negative control groups statistically(p<0.05). As for the grade of osteoarthritis, the extent of tissue damage of the experiment groups to which Caragana sinica extract was administered in 5 mL/kg or 10 mL/kg was lower than that of the negative control groups as well.

골담초 추출액과 흰목이버섯을 이용한 기능성 음료 개발 및 이화학적 특성 (The Development and Physicochemical Properties of Functional Beverages Using Caragana sinica Extract and Tremella fuciformis Berk)

  • The purpose of this study was to develop a functional beverage by using Caragana sinica extract and Tremella fuciformis Berk, which have high physiological effects such as anti-diabetic, antioxidant, anticancer, anti-lipidemic and anti-inflammatory. To this end, we used various mixing ratios of Caragana sinica extract, Tremella fuciformis Berk, and isomalto oligosaccharide, and measured the physicochemical properties of those beverages. The analyses showed the following: moisture content of those beverages using Caragana sinica extract and Tremella fuciformis Berk was 50.51~67.64%, pH was 5.19~5.38, and crude fat content was 0.28~0.52%, crude protein content was 1.26~1.80%, ash content was 0.47~0.80 and sugar content was 8.55~26.98°Brix. The lightness (L) of the beverages was 37.70~58.11, the redness (a) was 6.88~16.94, the yellowness (b) was 13.72~23.68, the total polyphenol content was 2.07~3.46 mg/g, and the total flavonoid content was 0.37~0.42 mg/g. Based on the results, it was confirmed that the nutritional components and total polyphenols of functional beverage using Caragana Sinica Extract, Tremella Fuciformis Berk and isomalto oligosaccharide were high at a mixing ratio of 1:1.3:25%. These results will increase the use of Caragana Sinica Extract and Tremella Fuciformis Berk, as functional materials in the future, and provide a framework for the manufacturing of diluted beverages comprising mineral water and carbonated water.

골담초엽의 플라보노이드 (The Flavonoids from Caragana Chamlagu Leaves)

  • The phytochemical studies of the leaves of Caragana chamlagu were carried out as a sieries of the investigation of medicinal resources. The roots of Caragana chamlagu have been used as neuralgia, arthritis and migraine in the folk medicines of Korea. The methanolic extract of the leaves of Caragana chamlagu was suspended with water and then separated with chloroform. Compound I was isolated from precipitates of these water fraction by recrystalization. The aqueous fraction of MeOH extract was performed to column chromatography on Amberlite XAD-4 and Sephadex LH-20, and three compounds, compound II, compound III, and compound IV were isolated. The structures of the four compounds were elucidated by spectroscopic data of $^1H-NMR$, ^{13}C-NMR$, IR, and FAB-MS. Compound I-IV were tilianine ($acacetin-7-O-{\beta}-D-glycopyranoside$), rutin($quercetin-3-O-{\alpha}-L-rhamnopyranosy(1{\rightarrow}6)-{\beta}-D-glu-copyranoside$), $kaempferol-3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-D-galactopyranoside$, and apigetrin, ($apigenin-7-O-{\beta}-D-glycopyranoside$), respectively.

골담초의 이소후라본 성분 (Some Isoflavones from the Root of Caragana microphylla Lam)

  • Five isoflavones were isolated from the root of Caragana microphylla Lam. and the structures have been identified by means of chemical and physical methods. The isoflavones are 7-hydroxy-4'-methoxyisoflavone(formononetin), $form ononetin-7-O-{\beta}-glucoside(ononin)$, 7-hydroxy-3', 4'-methylenedioxyisoflavone(pseudobaptigenin), $7-O-{\beta}-glucuopyranosyloxy-3'- hydroxy-4'-methoxyisoflavone(calycosin-7-O-{\beta}-gluc-oside)$ and maackiain.

골담초근의 Lipid성분에 관한 연구 (Acylucosyl Sterols from the Roots of Caragana chamlagu)

  • A mixture of acylglucosyl sterols together with ${\beta}-sitosterol$, ${\beta}-sitosterol\;3{\beta}-O-glucoside$ and fatty acids was isolated from the roots of Caragana chamlagu as their acetate forms and the structure elucidated by chemical and spectroscopic means. The major acylglucosyl sterol was ${\beta}-sitosteryl\;3-O-[6'-O-oleoyl]-{\beta}-D-glucopyranoside$ while the minor components were $6'-O-palmitoyl-\;and\;6'-O-stearoyl-{\beta}-D-glucosyl$ sitosterol congeners. The isolation and structure elucidation of these acylglucosyl sterols are reported for the first time from the genus Caragana.

골담초 효모 발효 추출물의 피부 미백 효능에 관한 연구 (Skin Whitening Effects of Caragana sinica Rehder Extract Fermented by Saccharomyces cerevisiae KCTC 7913)

  • 본 연구는 미생물에 의한 골담초(Caragana sinica Rehder) 발효 추출물의 항산화 효과 및 피부 미백 효능에 관한 연구로 골담초에 S. cerevisiae KCTC 7913를 첨가하여 발효를 통해 얻어진 발효 추출물을 B16F10 멜라닌 세포에 처리한 결과, 농도의존적으로 멜라닌 생성 억제 효능이 증가한 것으로 확인하였으며, 추출물 내 유효 성분인 resveratrol의 함량이 발효 전의 추출물에 비해 증가함을 확인하였다. 멜라닌 생성 과정의 주요 단백질인 tyrosinase의 활성 억제 또한 골담초 추출물에 비해 골담초 발효 추출물의 효능이 우수함을 확인하였으며, 골담초 발효 추출물의 피부 자극 또한 일어나지 않는 것으로 확인되어 피부에 안전한 화장품 원료로서의 사용이 가능할 것으로 사료된다.

  • From the roots of Caragana sinica (Buc'hoz) Rehder (Leguminosae), a new saponin named caraganoside A was isolated and elucidated as 3-0-$\beta$-D-xylopyranosyl (1$\rightarrow$2)-[$\beta$-D-glucopyranosyl (1$\rightarrow$3)]-$\alpha$-L-arabinopyranosyl oleanolic acid 28-O-[$\beta$-D-glucopyranosyl ester by means of chemical and spectral studies. Kalopanax-saponin F, hemsloside Ma3 and araloside A were also isolated and characterized. The former two compounds were separated as their methylesters.

골담초 메탄올 추출물의 진통 효과 (Anti-nociceptive Activity of Methanolic Extract of Caragana sinica)

  • Caragana sinica (Leguminosae) is a plant, which has been used as a traditional medicine for the treatment of lots of diseases including neuralgia, goat, hypertension and eczema. However, scientific studies of C. sinica in pharmacological aspects are not carried out. In this study, the anti-nociceptice effect of methanolic extract of C. sinica (MCS) was evaluated using various pain models. Our data represented that MCS significantly delayed the latency time under central pain condition which are arose from thermal stimuli, indicating MCS possess analgesic potential against central nociception. In addition, MCS showed strong and dose-dependent anti-nociceptive activities on acetic acid-induced peripheral pain, compared to positive control such as indomethacin. Further combination studies using naloxone, a non-selective opioid receptor antagonist, have revealed that analgesic activity of MCS was not changed in the presence of naloxone, indicating MCS exerts anti-nociceptive activity independent of opioid receptor. These results suggest that MCS may be an effective medicine in managing pain.

Caragana chamlagu Lamarck의 Sterol 성분에 관한 연구 (Studies on the Sterols of Caragana chamlagu Lamarck)

  • From the dried roots of Caragana chamlagu Lamarck which have been known to be effective against neuralgia, arthritis, migraine, and sputum, etc. in the folk medicines of Korea, a colorless sterol-like substance, C$_{29}H_{50}O$, m.p. $115-117^{\circ}$, was isolated from the non-saponified fraction of methanolic extract. It was confirmed to be a mixture of four sterols, namely cholesterol, brassicasterol, campesterol, and $\beta$-sitosterol by physico-chemical properties, ir, nmr, mass spectra and gas liquid chromatographic method with the comparison of the authentic samples. In mass fragments of four sterols, furthermore, the same ions of m/e 273, 255, 229 and 213 were assigned to fragments of stigmastane skeletons of the unsaturated phytosterol, and the characteristic ions of each sterol were observed 386, 371, 368, 353 in cholesterol, 398, 383, 380, 365, 355, 337 in brassicasterol, 400, 385, 382, 367 in campesterol and 414, 399, 396, 381 in $\beta$-sitosterol, respectively.
