  • This study was conducted to determine grazing intensity of growing Korean native goats(Capra hircus coreanae) on mountainous pasture. It was carried out to obtain basic information for improvement of mountainous pasture management and establishing feeding system of Korean native goat. A total of 20 goats were grouped by feeding systems [A mountainous pasture grazing group (Concentrated feed of 1.5% body weight, treatment 1, T1, n=10) and a barn feeding group (TMR, treatment 2, T2), n=10] to conduct study from April to September. The average forage productivity of the mountain pasture was $500.9{\pm}61.41kg/ha$. The average dry matter intake in T1 was 0.64 and the calculated grazing intensity was 21 head/ha. In productivity, when the two treatments(T1, T2) were compared, the dry matter intake was about two to three times the difference. The average daily gain per day during the experiment was 63.3 in the mountain pasture and 120 g in barn feeding. When grazing, considering mountainous pasture productivity it is necessary to increase the productivity through proper feeding. The feed costs of black goats raised by grazing on the grassland in the same period showed an average 75% reduction compared to barn feeding. As a result of this study, it can be expected that a considerable reduction of feed costs can be expected in the breeding of Korean native black goat using the mountain pasture.

  • Insulin-like growth factor-binding protein-5 (IGFBP-5) is one of the six members of IGFBP family, important for cell growth, apoptosis and other IGF-stimulated signaling pathways. In order to explore the significance of IGFBP-5 in cells of the Inner Mongolian Cashmere goat (Capra hircus), IGFBP-5 gene complementary DNA (cDNA) was amplified by reverse transcription polymerase chain reaction (RT-PCR) from the animal's fetal fibroblasts and tissue-specific expression analysis was performed by semi-quantitative RT-PCR. The gene is 816 base pairs (bp) in length and includes the complete open reading frame, encoding 271 amino acids (GenBank accession number JF720883). The full cDNA nucleotide sequence has a 99% identity with sheep, 98% with cattle and 95% with human. The amino acids sequence shares identity with 99%, 99% and 99%, respectively. The bioinformatics analysis showed that IGFBP-5 has an insulin growth factor-binding protein homologues (IB) domain and a thyroglobulin type-1 (TY) domain, four protein kinase C phosphorylation sites, five casein kinase II phosphorylation sites, three prenyl group binding sites (CaaX box). The IGFBP-5 gene was expressed in all the tested tissues including testis, brain, liver, lung, mammary gland, spleen, and kidney, suggesting that IGFBP-5 plays an important role in goat cells.

  • The morphological studies on the rectal development in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats were investigated by light microscopy and scanning electron microscopy. The rectal wall appeared to be differentiated into the epithelium, lamina propria, tunica muscularis and serosa in 60-day-old fetus. The rectal epithelia were stratified in some areas and simple columnar in others at 60- and 90-day-old fetuses but was only observed simple columnar epithelium at 120-day-old fetus. The goblet cells and the intestinal glands appeared in the rectal mucosa at 90-day-old fetus and gradually increase in number during gestation period. The rectal villi appeared at 60-day-old fetus and increased in length and width until 90-day-old fetus but the villi appeared to be shorter and degenerated after 120-day-old fetus. Only the rudiment trace of the villi remained at birth. The tunica muscularis of the rectum were continuously developed during gestation period. The inner circular and outer longitudinal muscle layers of rectum were observed in the 120-day-old fetuses. Under scanning electron microsoopy, the rectal villi appeared at 60-day-old fetus. The pentagonal/hexagonal microridges and numerous microvilli were observed on the luminal surface of rectal wall. Low and thumb-like rectal villi were developed at 90-day-old fetus. The rectal villi became degenerated after 120-day-old fetus. Only the rudiment of villi was remained at birth. Many circular folds were well developed. Opening portions of intestinal glands appeared like slot.

  • Anaplasma species are obligate intracellular pathogens that can cause tick-borne diseases in mammalian hosts. To date, very few studies of their occurrence in Korean native goats (Capra aegagrus hircus) have been reported. In the present study, we investigated Anaplasma infection of Korean native goats on Jeju Island, Republic of Korea, and performed phylogenetic analysis based on the 16S rRNA gene sequences. Our results showed that Anaplasma infection was found mostly in adult female goats. The phylogenetic tree revealed that the 7 sequences identified in Korean native goats could belong to Anaplasma sp. and were distinct from A. marginale, A. centrale, and A. ovis. The results indicated that the sequences identified to belong to Anaplasma were closely related to sequences isolated from goats in China and were clustered within the same group. To our knowledge, this is the first study to detect Anaplasma sp. infection in Korean native goats.

  • We investigated the cytogenetic characteristics of Korean native goat(Capra hircus). Chromosome slides were prepared from peripheral blood cell cultures. GTG, GBG, RBG and CBG-banding techniques were employed on those slides. The high resolution karyotype of Korean native goat could be made with the incorporation of BrdU. Korean native goat has 60 chromosomes composed of 58 autosomes and XY or XX sex chromosomes. All of autosomes of Korean native goat were acrocentric chromosomes. X chromosome was submetacentric and Y chromosome was metacentric. The GTG, GBG and RBG-band patterns of Korean native goat were similar to those of other goats. CBG-band regions were distinct at the proximal portion of the long arms of all autosomes in Korean native goats. According to our investigation, there was no significant difference in chromosomal band patterns between Korean native goat and other goats. It might be necessary to use molecular genetic markers for clarifying the genetical characteristics of Korean native goat whose biological characteristics are not clearly defined.

  • As a member of a subclass of immunophilins, it is controversial that FKBP38 acts an upstream regulator of mTOR signaling pathway, which control the process of cell-growth, proliferation and differentiation. In order to explore the relationship between FKBP38 and mTOR in the Cashmere goat (Capra hircus) cells, a full-length cDNA was cloned (GenBank accession number JF714970) and expression pattern was analyzed. The cloned FKBP38 gene is 1,248 bp in length, containing an open reading frame (ORF) from nucleotide 13 to 1,248 which encodes 411 amino acids, and 12 nucleotides in front of the initiation codon. The full cDNA sequence shares 98% identity with cattle, 94% with horse and 90% with human. The putative amino acid sequence shows the higher homology which is 98%, 97% and 94%, correspondingly. The bioinformatics analysis showed that FKBP38 contained a FKBP_C domain, two TPR domains and a TM domain. Psite analysis suggested that the ORF encoding protein contained a leucine-zipper pattern and a Prenyl group binding site (CAAX box). Tissue-specific expression analysis was performed by semi-quantitative RT-PCR and showed that the FKBP38 expression was detected in all the tested tissues and the highest level of mRNA accumulation was detected in testis, suggesting that FKBP38 plays an important role in goat cells.

  • This study was carried out to investigate the morphological development of the tongue in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats by light microscopy. In 60-day-old fetuses, the tongue tissues were differentiated into epithelium, lamina propria, and muscle layer. The primordia of filiform, conical, lentiform, fungiform, and vallate papillae appeared by this time, and rudiments of taste buds were observed in the epithelia of the primordia gustatory papillae. The dorsal lingual epithelia were PAS positive. Collagenous fibers and blood vessels were present in the lamina propria. In 90-day-old fetuses, the gustatory glands were moderately positive for PAS and muscle fibers, and connective tissues were developed. In 120-day-old fetuses, many taste buds were observed in the gustatory papillae. The muscle bundles, collagenous fibers, blood vessels and gustatory glands were well developed. In neonates, over 40 taste buds were found in a transverse section of the vallate papillae. Muscle layers, blood vessels, collagenous fibers and gustatory glands were very well developed.

  • The distribution of glucagon-immunoreactive cells in the pancreas during various developmental stages (fetus, neonate, 1-month-old, 6-month-old and adult) of the Korean native goat was investigated by immunohistochemical methods. The varying distribution and frequency of glucagon-immunoreactive cells in the pancreas of the Korean native goat were observed. The glucagon-immunoreactive cells were detected in both exocrine and endocrine portions (pancreatic islets) at all developmental stages and also in ducts of the 6-month-old and adult. The relative frequencies of glucagon-immunoreactive cells increased in the pancreatic islets and ducts with age, but decreased in the exocrine portions. Generally, they were distributed in the interacinar spaces or marginal zone of the pancreatic islets during all stages of development. However, the cell distributions of the pancreatic islets in the neonate divided into two types: 1) ones which were distributed in the inner zone, and 2) others in the peripheral zone.
