In vitro Alternatives to Skin Irritation Test

  • In vitro cell culture system has been proposed as a promising alternative model to in vivo skin irritation test. These studies were performed to screen the cytotoxicity effects of surfactants using normal human skin fibroblasts. Cell membrane integrity assessed by the leakage of lactate dehydrogenase (LDH) and mitochondrial integrity by MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromides reduction test were affected in a dose dependent manner. The irritation potential of surfactants to human skin patch test, and the changes of capillary permeability by rabbit intradermal safety test were assessed as in vivo methods. Our results suggest that LDH leakage assay and MTT reduction test using cultured human fibroblasts could be predictive for the irritancy of various surfactants in human, and LDH assay is superior correlated with in vivo test (r=0.886) to MTT test with in vivotest (r=0.757).

Comparative Free and Acetylated Polyamine Profiles in the Urine of Normal Subjects and Various Cancer Patients

  • Urinary free and acetylated polyamine profiles have been investigated for their potential usefulness as biochemical markers of cancer in a control of group comprised of healthy volunteers (32 cases) and patients with various types of cancers(48 cases). The nine (5 free and 4 acetylated) endogeneous polyamines were simultaneously determined by a sensitive capillary gas chromatography/nitrogen-phosphorus detector (GC/NPD). The newly modified (simple and convenient) method was developed and the compounds were isolated by adsorption onto silica gel and derivatized by heptafluorobutyric anhydride to enhance their specificity on gas chromatograms. The good quality-control data were obtained through the precision and accuracy test and the recovery range of them was 48.6 ~ 101.2 %. The Korean reference values of urinary polyamines were established and significant differences were found in cancer patients compared with normal subjects. Also, to eliminate subject variations, precursors to product concentration ratios were compared between cancer patients and control group. The ratios of both putrescine to spermidine and total (free plus acetylated) putrescine to total spermidine were significantly greater in cancer patients than in normal subjects.

  • The generation of fine relics of inorganic and metallic materials from precursor suspensions is of significant current interest as it holds the key to the fabrication of display and printed circuit board. Some novel printing methods depositing ceramic particles have been suggested in recent years. When a conducting liquid is supplied to a capillary nozzle at a low flow rate and when the interface between air and the liquid is charged to a sufficiently high electrical potential, the liquid meniscus takes the form of a stable cone, whose apex emits a microscopic jet. This is called as a cone-jet mode. In our experiments, an alumina particles flowing through a nozzle were subjected to electro-hydrodynamic printing in the cone-jet mode. The pattern of 'YONSEI' characters was tested at $10 {\mu}l/min$ of alumina ink flow rate and different applied voltages. At an applied voltage of 6 kV, feature size was in the range of $250 {\mu}m.$

Photodynamic Diagnosis and Therapy for Peritoneal Carcinomatosis from Gastrointestinal Cancers: Status, Opportunities, and Challenges

  • Selective accumulation of a photosensitizer and the subsequent response in only the light-irradiated target are advantages of photodynamic diagnosis and therapy. The limited depth of the therapeutic effect is a positive characteristic when treating surface malignancies, such as peritoneal carcinomatosis. For photodynamic diagnosis (PDD), adjunctive use of aminolevulinic acid- protoporphyrin IX-guided fluorescence imaging detects cancer nodules, which would have been missed during assessment using white light visualization only. Furthermore, since few side effects have been reported, this has the potential to become a vital component of diagnostic laparoscopy. A variety of photosensitizers have been examined for photodynamic therapy (PDT), and treatment protocols are heterogeneous in terms of photosensitizer type and dose, photosensitizer-light time interval, and light source wavelength, dose, and dose rate. Although several studies have suggested that PDT has favorable effects in peritoneal carcinomatosis, clinical trials in more homogenous patient groups are required to identify the true benefits. In addition, major complications, such as bowel perforation and capillary leak syndrome, need to be reduced. In the long term, PDD and PDT are likely to be successful therapeutic options for patients with peritoneal carcinomatosis, with several options to optimize the photosensitizer and light delivery parameters to improve safety and efficacy.

Stem cell-derived exosomes for dentin-pulp complex regeneration: a mini-review

  • This mini-review was conducted to present an overview of the use of exosomes in regenerating the dentin-pulp complex (DPC). The PubMed and Scopus databases were searched for relevant articles published between January 1, 2013 and January 1, 2023. The findings of basic in vitro studies indicated that exosomes enhance the proliferation and migration of mesenchymal cells, as human dental pulp stem cells, via mitogen-activated protein kinases and Wingless-Int signaling pathways. In addition, they possess proangiogenic potential and contribute to neovascularization and capillary tube formation by promoting endothelial cell proliferation and migration of human umbilical vein endothelial cells. Likewise, they regulate the migration and differentiation of Schwann cells, facilitate the conversion of M1 pro-inflammatory macrophages to M2 anti-inflammatory phenotypes, and mediate immune suppression as they promote regulatory T cell conversion. Basic in vivo studies have indicated that exosomes triggered the regeneration of dentin-pulp-like tissue, and exosomes isolated under odontogenic circumstances are particularly strong inducers of tissue regeneration and stem cell differentiation. Exosomes are a promising regenerative tool for DPC in cases of small pulp exposure or for whole-pulp tissue regeneration.

조류생산잠재력조사 방법개발에 의한 육수환경의 부영양화 방지대책에 관한 연구(I) -순수분리종의 형태 및 증식특성- (A Study on Protection Plan of Eutrophication in Fresh Water Environment by Development of Methods for Algal Growth Potential test (I) -Morphology and Growth Characteristics of Isolated algae-)

  • The isolation, morphological study and growth characteristics of the algae were investigated from Lake Chuam. The isolated algae were applied the Agal Growth Potential test. The method of isolation and purification of the algae were used to Agar plating(AP), nutrient enrichment(NE), dilution(DI) and micro capillary technique(MC). Total isolated algae were 21 species. They were composed of Cyanophyceae, Dinophyceae, Bacillariophyceae, Euglenophyceae and Chlorophyceae. The numbers of algal strain by isolation technique were highest in dilution(21 species), and those of the rests were showed in order of NE > MC > AP. The sizes of isolated Selenastrum and Scenedesmus were $1.8\pm 1.4 \mu m$, $3.3\pm 0.9 \mu m$ in diameter and $6.4\pm 2.3 \mu m$, $13.6\pm 1.9 \mu m$ in length respectively. The morphology of isolated algae and NIES-collection strain was very similar each other, but the size was smaller isolated algae than that of NIES-collection. The optimum culture condition of isolated Selenastrum and Scenedesmus was about 30$\circ$C(25$\circ$C-35$\circ$C) in temperature and the maximum growth was appeared between 7,000 lux and 8,000 lux in the light intensity. The comparison of $\mu$(specific growth rate) on the concentration of nutrients such as nitrate and phosphate, isolated Selenastrum was appeared maximum it at 1.0 mg $NO_3-N/l$ but NIES-collection strain was showed 95% of maximum it at same nitrate concentration. Maximum g of isolated algae and NIES-collection strain in Scenedesmus onto nitrate concentration were very similar with the result of selenastrum. The specific growth rates of isolated algae and NIES-collection strain on the gradient concentration of phosphate were showed 0.72/day and 0.70/day at 0.02 mg $PO_4-P/l$ in Selenastrum but those of Scenedesmus were appeared 0.61/day and 0.57/day at same concentration $PO_4-P$.

백서 두개골 결손부의 골 대체물 이식과 홍화씨 섭취 후의 치유양상 (Healing after Implantation of Bone Substitutes and Safflower Seeds Feeding in Rat Calvarial Defects)

  • Many synthetic bone materials have been studied for their potential of regenerative effects in periodontal tissue. Safflower seeds have been traditionally used as a drug for the treatment of fracture and blood stasis in oriental medicines. The purpose of this study was to assess and compare the osseous responses in rat calvarial defects between bone substitutes such as calcium carbonate and bovine-derived hydroxyapatite and feeding of safflower seeds. The calvarial defects were made with 8 mm trephine bur in 24 Sprague-Dawley rats. Two graft materials were implanted in each experimental groups, whereas the control and safflower seed feeding groups were sutured without any other treatment. And then the rats of safflower seed feeding group were supplied with 3 g/day of safflower seeds. Each group was sacrificed at 4 weeks and 8 weeks. To study a histopathology related to bone healing and regeneration, Goldner's Masson Trichrome stain was done at each weeks. The tissue response was evaluated under light microscope. There were more osteoblastic activity, new bone formation, dense bony connective tissues in bovine-derived hydroxyapatite group compared to other groups at 8 weeks. The osseous defect area of safflower seed feeding group was filled with prominent fibrous tissues, where less inflammatory infiltration and new capillary proliferation. In the early phase of bone healing, safflower seed feeding reduces the inflammatory response and promotes the proliferation of connective tissue. These results suggest that natural bovine-derived HA and safflower seed feeding could enhance the regenerative potential in periodontal defects.

미세 채널에서 칼슘이온 물질전달을 이용한 단분산성 알지네이트 하이드로젤 입자의 실시간 젤화 (In situ Gelation of Monodisperse Alginate Hydrogel in Microfluidic Channel Based on Mass Transfer of Calcium Ions)

  • 본 논문은 가교제의 물질전달을 통한 실시간 생체고분자의 젤화 과정으로 단분산성을 갖는 구형의 알지네이트 하이드로젤을 미세유체 채널 내에서 제조하는 방법에 관한 연구이다. 먼저 미세유체 채널 내에서 단분산성 알지네이트 액적들을 형성하고 연속상에 분산된 염화칼슘 분자들의 물질전달 과정을 통해 실시간 젤화과정이 이루어지게 하여 알지네이트 하이드로젤 입자를 제조하였다. 이때, 미세유체 채널에서 형성되는 액적의 크기는 손쉽게 케필러리 수(capillary number)와 분산상의 유속 조절을 통하여 제어할 수 있다. 본 방법은 미세유체 채널 내에서 안정적인 액적을 형성할 수 있고 칼슘 가교제로 제조된 알지네이트 하이드로젤 입자들은 균일한 크기 분포를 가지며(C.V=2.71%) 유속, 점도, 및 계면장력의 조절을 통하여 $30{\mu}m$에서 $60{\mu}m$까지의 다양한 크기의 알지네이트 하이드로젤 입자를 제조할 수 있다. 본 논문에서 제시한 간단한 미세유체 접근방법을 통해 제조되는 단분산성을 갖는 알지네이트 하이드로젤 입자는 생체물질들을 손쉽게 함입(encapsulation)할 수 있으며 이는 식품, 화장품, 잉크 및 약물 등의 전달체로 활용이 가능하고 생체적합성이 뛰어나 세포이식 분야에도 활용될 가능성이 있다.

대두포장(大豆圃場)에서 수분흡수(水分吸收)에 관(關)한 수치해석학적(數値解析學的) 모형(模型) (Numerical Simulation of Water Uptake of Soybean Field)

  • Ohm의 법측(法測)을 이용(利用)한 물 이동(移動) 방정식(方程式)의 해(解)를 수치해석법(數値解析法)으로 구(求)하여 대두(大豆)의 물 소모양상(消耗樣相)을 추정(推定)하여 실측자료(實測資料)와 비교(比較)하였다. 본(本) 모형(模型)은 고속전산기(高速電算器)를 사용(使用)하기 위(爲)하여 FOPTRAN으로 쓰여졌으며 모형(模型)의 검정(檢定)은 미(美) Iowa 주서부농업시험장(洲西部農業試驗場)에서 얻어진 자료(資料)를 사용(使用)하여 시도(試圖)되었다. 모형검정치(模型檢定値)와 실측치간(實測値間)에 잘 맞는 경향(傾向)이었으나 다소(多少)의 오차(誤差)가 시기적(時期的)으로 인정(認定)되었다. 계산(計算)된 토양증발량(土壤蒸發量)은 작물생육초기(作物生育初期)에는 약(約) 2mm/day이었으나 후기(後期)에는 0.4mm/day로 감소(減少)하였으며, 한발기(旱魃期)에 0.1mm/day까지 감소(減少)되었다. 증발량(蒸發量)은 최대(最大) 5mm/day이었다. 토심(土深) 180cm와 210cm 층위(層位)에서 이루어지는 지하배수량(地下排水量)은 약(約) 0.2mm/day이었고, 후기(後期)에는 역전되어 모관상승(毛管上昇)을 보였으며 이 양(量)은 약 0.07mm/day로 근계(根系)의 물 소모량(消耗量)을 충족(充足)시키지는 못하였다. 따라서 심토(深土)의 물 이용(利用)을 위(爲)해서는 근계(根系)의 확장(擴張)이 더 중요(重要)하다.

실내동결시스템을 이용한 노상토의 동상 특성 (The Frost Heaving Characteristics of Subgrade Soils Using Laboratory Freezing System)

  • 겨울에는 시베리아기단의 영향으로 한랭 건조한 대륙성 고기압의 영향을 받아 춥고 건조하여 1월 평균기온이 $-6{\sim}-7^{\circ}C$의 영하의 온도로 낮아져 지반동결시 수분이동으로 동상현상이 발생하여 도로의 불균형 동결팽창을 초래하며, 결국 포장체를 파손시킨다. 동상발생시 토립자는 모관력에 의해 지하수를 흡수하여 아이스렌즈를 형성하며, 이 모관 흡수력은 토립자의 크기에 영향을 받는다. 도로는 다양한 재료와 단면으로 구성된 구조물이기 때문에 환경성과 재료 물성뿐만 아니라 포장체 각 층의 구조적 적정성 또는 지지력을 파악하는 것이 무엇보다 중요하다. 현재 기존 동상방지층 설계법에 따르면, 동상방지층은 포장체의 구조적 적정성과는 무관하게 온도 조건에 따른 동결깊이에 따라 일률적으로 결정되고 있다. 이러한 동결깊이를 포장구조설계에 적용함으로써 포장의 과다설계 우려가 있다. 따라서 본 논문에서는 도로 동상방지층의 효용성 검증 및 설치기준 확립을 위해 실내동결시스템을 활용하여 도로 노상토의 동상 특성에 대한 민감성을 판별, 도로건설 현장 노상토에 대한 역학적 특성과 실내 동결 시험을 수행하였으며, 외부 동결온도의 지속 조건에 대한 시료의 온도변화, 동상팽창압, 동상팽창량, 부동수분 등의 결과값을 통하여 동결 과정에 따른 지반공학적 특성을 평가하였다.
