Prickly pear cactus cladodes were extracted with hot water and 70% ethanol, followed by fractionation with n-hexane (HF), ethyl acetate (EF), n-butanol (BF), and distilled water. Total phenolics and total flavonoid contents as well as antioxidative and anti-inflammatory activities were then measured. Total phenolic contents were 784, 452, and 220 mg gallic acid equivalents (GAE)/g, whereas total flavonoid contents were 214, 76, and 113 mg quercetin equivalents (QE)/g in EF, BF, and HF, respectively. DPPH and ABTS radical scavenging activities ($IC_{50}$) were 103 and $105{\mu}g/mL$ in EF, 359 and $379{\mu}g/mL$ in BF, and 469 and $605{\mu}g/mL$ in HF, respectively. Oxygen radical absorbance capacity was highest at $391{\mu}M$ TE in EF (in decreasing order of $117{\mu}M$ TE in BF and $64{\mu}M$ TE in HF), whereas superoxide anion radical scavenging activity ($IC_{50}$) was highest at $40{\mu}g/mL$ in EF (in decreasing order of $69{\mu}g/mL$ in BF and $98{\mu}g/mL$ in 70% ethanol extract). Inhibitory activity ($IC_{50}$) of nitric oxide (NO) production induced by LPS-activated RAW264.7 cells was highest at $62{\mu}g/mL$ in HF (in decreasing order of $104{\mu}g/mL$ in EF and $465{\mu}g/mL$ in BF). The selectivity index (ratio of inhibitory activity of NO production to cell cytotoxicity) was highest at 4.63 in EF (in decreasing order of 3.37 in HF and 2.14 in BF). In conclusion, EF showed potent antioxidant and anti-inflammatory effects with high phenolic and flavonoid contents.

In vitro shoot proliferation and rooting were tested for 2-0 seedlings of half-sib families of 4 plus oaks trees. Nodal segments having axillary buds from 37 families(16 of Quercus acutissima, 10 of Q. variabilis, 7 of Q. serrata, and 4 of Q. mongolica) were cultured on WPM(Woody Plant Medium) supplemented with 0.5 mg/l BA (6-benzyladenine) and 0.01 mg/l NAA(${\alpha}$-naphthalene acetic acid) and subcultured at 2-3 weeks of intervals fur 6 months. In vitro rooting was carried out on GD(Gresshoff and Doy) medium supplemented with 0.5mg/l IBA(indole butyric acid). The capacity for shoot proliferation and rooting was highly varied with families. Generally, white oaks(Q. serrata and Q. mongolica) showed poor response than black oaks(Q. acutissima and Q, variabilis) in shoot proliferation and rooting. Among the total of 37 families, 7 of Q. acutissima, each 2 of Q. variabilis, Q. serrata, and Q. mongolica revealed abilities for continuous shoot proliferation, and the others failed to proliferate. Rooting of the selected oak trees also greatly varied among the families. In Q. acutissima, rooting ratio ranged from 10.0%(CB 25. KG 4) to 89.8%(CB 18). Although 26.7% of KG 16 in Q. variabilis, 3.3% of JN 15 in Q. serrata were rooted, Q. mongolica was not rooted at all in this experimental conditions. No relationship between shoot growth and the rooting ability was observed. Present results suggest the possibility of large-scale micropropagation, but further studies on family differences, shoot-tip necrosis, and callusing of rooting junction are still required to develop reliable micropropagation systems.

It is matter of fact processing technology must affects the quality and yield of final product and these also depend on the selection of raw material directly or indirectly. So that the estimation of tile preprocessing condition of shellfish is of great importance for distributors and processors. This study was attempted to establish the basic data for evaluating the processing suitability of baby clam, which is one of the five important shellfishes for domestic use and export. The important results are as follows: 1. The ratio of meat volume and meat weight to the holding capacity by shells may be useful for measuring the condition index of baby clams. 2. Baby clams grown in the beds with the composition of a large quantity of gravel were low in condition index value than in sand and mud. 3. If the green feed phenomenon was the prime consideration for canned baby clams, tile most suitable harvest season if October. In this period, the digestive tract in body was almost colorless. 4. As a whole, seasonal changes of moisture and fat content in baby clams were reversely correlated. Protein content increased from April and slightly decreased for a while from July to August and increased again from September. In March, the content of glycogen was 6.3 to 6.8 percent. From this period to October, glycogen was rapidly decreased. In October, it was only 0.1 to 0.2 percent but increased from November. There were little seasonal changes in pH value and crude ash content. The pH value of meat was 6.0 to 6.2 and crude ash content was about 2 percent. 5. By the results of condition index and chemical composition of baby clams, the suitable harvest season as raw materials for processing was from March to June and from September to October. 6. The steamed baby clam meat was packed with 2 percent salt solution containing 0.15 percent, citric acid or 0.5 percent $Na_2$ EDTA in a round sanitary tin can coated with C-enamel that is 203.9 ml by volume and sterilized for 60 minutes at $112^{\circ}C$. The canned product has shown a good result to reserve the natural characteristics of baby clam through six month storage at room temperature.
To establish the organic cultivation of pepper using green manure crops, this work studied the growth characteristics and yield of green manure crops, mineral composition of green manure crops, mineral uptake in shoots of green manure crops, chemical composition in soil of green manure crops, and the growth characteristics and yield of pepper in vinyl house. Shoot dry weight of green manure crops was higher level in Sorghum bicolor and Sorghum than in Crotalaria juncea and Glycine max. Also, the roots were spread deeply into soil in Sorghum bicolor and Sorghum. Density of root-knot nematodes in rhizosphere of green manure crops was significantly more decrease in Crotalaria juncea and Sorghum than in Glycine max and Sorghum bicolor. Total nitrogen and CaO content of green manure crops was significantly higher in Crotalaria juncea and Glycine max than in Sorghum bicolor and Sorghum. $K_2O$ content was significantly higher in Sorghum bicolor and Sorghum than in Crotalaria juncea and Glycine max. MgO content was not significant difference at all green manure crops. Cations content ratio of $K_2O$ : CaO : MgO was 3.4 : 1.4 : 1. Total nitrogen uptake in shoots of green manure crops was high level in Glycine max, Sorghum bicolor and Sorghum compared with in Crotalaria juncea. $K_2O$ and MgO uptake was significantly higher in Sorghum bicolor and Sorghum than in Crotalaria juncea and Glycine max. Value of pH in soil of green manure crops was more increase in Crotalaria juncea and Glycine max than in Sorghum bicolor, Sorghum and control, but after cultivation of pepper pH in soil was recovered with initial soil pH before seeding of green manure crops. EC value in control, green manure crops, and pepper cultivation decreased by 44%, 15~18%, and 38~61% level, respectively, compared with initial soil of green manure crops treatment. K content in soil of control, Crotalaria juncea and Glycine max cultivation was increased by 14%, but the K content in soil of Sorghum bicolor and Sorghum decreased by 24~38%. Cation exchange capacity (CEC) in soil of Crotalaria juncea and Sorghum bicolor decreased by 11%, but CEC in soil of Glycine max, Sorghum and control increased by 11%. Harvest fruit yield was higher in Crotalaria juncea, Glycine max, and Sorghum bicolor cultivation than in control and Sorghum.
Production of a mature oocyte is a complex process that requires the close association between oocyte and follicular cells. The present study was conducted to investigate the difference between oocytes with and without close junctional communication with cumulus cells and the involvement of two connexins(CXs) in the interactions. Follicles at different sizes(small: 200~400 ㎛; large:>450 ㎛) were mechanically isolated from PMSG-primed mouse ovaries, and punctured to get cumulus-oocyte complex(COC). Oocytes were released themselves(denuded), with partially attached(partial), or with tightly attached(intact) cumulus cells. Maturation and fertilization capacity of the COCs were measured. Expression of CX 37 and CX 43 was examined by RT-PCR and in situ hybridization. The ratio between intact COC and denude/partial oocytes was 30%(SI) and 70%(SPD) in small follicles, while 55%(LI) and 45%(LPD) in large follicles, respectively. Maturation and fertilization rates of the released oocytes were similar among SI, LPD, LI groups, but those were significantly lower in SPD oocytes. In oocytes, CX 37 was the major CX and CX 43 was not expressed, whereas in the cumulus cells, CX 43 was the major, and CX 37 was the next. Results of the present study suggest that 1) immature oocytes from small follicles with intact cell-to-cell communication with cumulus have the similar quality to that of the oocytes from larger follicles, 2) gap junction between oocyte and cumulus cells may be the heterotypic channel, and 3) we could not explain the difference in the cell-to-cell communication between intact and partial/denuded COCs through the expression of the two CXs.
$CO_2$ geological storage plays an important role in reduction of greenhouse gas emissions, but there is a lack of research for CCS demonstration. To achieve the goal of CCS, storing $CO_2$ safely and permanently in underground geological formations, it is essential to understand the characteristics of them, such as total storage capacity, stability, etc. and establish an injection strategy. We perform the impedance inversion for the seismic data acquired from the Ulleung Basin in 2012. To review the possibility of $CO_2$ storage, we also construct porosity models and extract attributes of the prospects from the seismic data. To improve the quality of seismic data, amplitude preserved processing methods, SWD(Shallow Water Demultiple), SRME(Surface Related Multiple Elimination) and Radon Demultiple, are applied. Three well log data are also analysed, and the log correlations of each well are 0.648, 0.574 and 0.342, respectively. All wells are used in building the low-frequency model to generate more robust initial model. Simultaneous pre-stack inversion is performed on all of the 2D profiles and inverted P-impedance, S-impedance and Vp/Vs ratio are generated from the inversion process. With the porosity profiles generated from the seismic inversion process, the porous and non-porous zones can be identified for the purpose of the $CO_2$ sequestration initiative. More detailed characterization of the geological storage and the simulation of $CO_2$ migration might be an essential for the CCS demonstration.
There are around 7,500 manure tanks to treat the manures from pigs in Korea. In the tank, there are too much sediments deposited on the base and wall, which causes low efficiency of stock capacity and manure fermentation. In order to minimize sediments and to ferment manure effectively, we developed a 2-fluid jet mixer for mixing sediments in liquid livestock manure tank. For developing the prototype, we tested a factorial experimental system with various nozzles, and simulated CFD models with two kinds of nozzle arrangement. From the results of factorial experiment and CFD simulation, we concluded the dia. ratio of primary : secondary nozzle should be 1:2 and the nozzles should be arranged at the same distances toward to the circumferential direction. With this results, we manufactured a 2-fluid jet mixer which is consists of four 2-phase nozzles, centrifugal slurry pump and root's type air blower. And, we carried out the performance test of the prototype in the round shaped liquid manure tank in the farm. The performance test results showed that the uniformity of TS (Total Solid) and VS (Volatile Solid) was raised from 21.3 g/L, 13.3 g/L In steady state to TS and VS to 23.0 g/L, 14.1 g/L in the mixing operation. Therefore, we could conclude that the prototype of 2-fluid mixer could make the solid material which could be sediments in the tank not to be deposited in the tank and to be contacted to air bubbles which could enhance the efficiency of the fermentation of livestock manure.
Phosphorus desorption study is essential to understanding P behavior in agricultural and environmental soils because phosphorus is considered as two different aspects, a plant nutrient versus an environmental contaminant. This study was conducted to determine soil P buffering power related to P desorption quantity intensity (Q/I) parameters, $Q_{max}$(an index of P release capacity) and $l_0$(an index of the intensity factor), and to investigate the characteristics of relationship between the P desorption Q/I parameters and the soil properties. Soil samples were prepared with treatments of 0 and $100mg\;P\;kg^{-1}$ applied as $KH_2PO_4$ solution. The P desorption Q/I curves were obtained by a procedure using anion exchange resin beads and described by an empirical equation ($Q=aI^{-1}+bln(I+1)+c$). The P desorption Q/I curves for the high available P (${\g}20mg\;kg^{-1}$ of Olsen P) soils were characteristic concave trends with or without soil P enrichment, whereas for the low available P (${\lt}20mg\;kg^{-1}$ of Olsen P) soils, the anticipated Q/I concave curves could not be obtained without a proper amount of P addition. When the soils were enriched in phosphates, the values of desorbed solid phase labile P and solution P, such as $Q_{max}$ and $I_0$ respectively, were increased, but the ratio of $Q_{max}$ versus $I_0$ was decreased. Thus, the slope of desorption Q/I curve represented as phosphorus buffering power, $|BP_0|$, is decreased. The $|BP_0|$ values of the high available P soils ranged between 48 and $61L\;kg^{-1}$ in the P untreated samples and between 18 and $44L\;kg^{-1}$ in the P enriched samples. Overall $|BP_0|$ values of both low and high available P soils treated with $l00mg\;P\;kg^{-1}$ ranged between 14 and $79L\;kg^{-1}$. The $Q_{max}$, values ranged between 71.4 and $173.1mg\;P\;kg^{-1}$, and the lo values ranged between 0.98 and $3.82mg\;P\;L^{-1}$ in the P enriched soils. The $Q_{max}$ and $I_0$ values that control the P buffering power may be not specifically related to a specific soil property, but those values were complicatedly related to soil pH, clay content, soil organic matter content, and lime. Also, phosphorus release activity, however, markedly depended on the desorbability of the applied P as well as the native labile P.

This research was conducted to investigate the effects of fly ash applications on growth and yield parameters of corn (Zea mays L.), and soil chemical properties. Corn height at silking stage, and height and dry matter ratio at harvesting stage were enhanced by applications of fly ashes derived from bituminous coal and anthracite, respectively. Effects of fly ash treatment on growth parameters of corn were varied with kinds and levels of fly ash application and growth periods, but relatively small without revealing a major negative effect as compared to the control. Yield of corn was increased by applying fly ash of anthracite origin, but other yield components were not influenced negatively by fly ash treatment. Soil total carbon contents, cation exchange capacity, and phosphorus contents of soils sampled after harvest of corn were significantly increased by fly ash treatment, although there were slightly different effects according to kinds and levels of fly ash application. Exchangeable cations of soils were varied within an experimental error range. Phosphorus taken up by corn was enhanced by treating fly ash of the bituminous coal to the soil and there were a positive correlation between phosphorus uptake and soil Phosphorus level. Cation uptake by corn was changed a little, but no significant reduction was observed in cation uptake due to fly ash treatment. It seems to be difficult to figure out the mechanism of fly ash effects on growth and nutrient uptake by corn with one year field experiment, however treatment of fly ash enhanced some parameters of growth and yield, and nutrient uptake by corn without revealing any major negative effects. To determine the value of fly ash as a fertilizer source, continuous researches under various soil and crop conditions were considered to be necessary.

Current study was conducted to investigate the effect of organic selenium supplementation originated from mushroom culture medium on meat quality of Hanwoo sleets. The result showed that organic selenium supplementation of 0.9 ppm (DM based ratio) for 2 (T1), 4 (T2) and 6 (T3) months had no effect on moisture content in longissimus muscle, with 63 to $66\%$ compared to non-supplemented control group Similarly, intramuscular fat content ranged from 11.7 to $16.4\%$ did not differ between the treatment groups (p>0.05). T3 group showed the highest protein content with $20.8\%$ while T2 group had the lowest content with $19.2\%$ The data indicated that organic selenium supplementation to the experimental concentration had indictable effect on proximate composition The treatments similarly had no influence on physical and biological characteristics of longissimus muscle, where cooking loss and shear force ranged from 20 to $21\%$ and from 3.6 to 4.4kg, respectively. On the other hand, muscle pH at 24 h postmortem showed 5.52, 5.57, 5.50, 5.50 for control, T1, T2 and T3, respectively, indicating that the longer feeding period resulted in the lower ultimate un A similar trend was observed from water-holding capacity (63.8, 64.4 and $64.2\%$ for T1, T2 and T3, respectively) which was significantly higher than con01 group of $59.5\%$ For sensory evaluation, juiciness did not differ between the treatment groups, but n and n (5.30 and 5.28, respectively) showed significantly tender mat Particularly, T2 group received significantly higher flavor score among the treatment groups including controls. The data indicated that organic selenium supplementation to the experimental concentration had no effect on beef quality, but the treatment effect on anti-oxidation function is remained for further studies.
