Synergistic Effect of Polygodial with Imidazole Drugs on the Antifungal Activity (천연물 Polygodial과 Imidazole계 화합물의 병용에 의한 항진균 활성의 증진)

  • The fungistatic and fungicidal activities of amphotericin B, fluconazole, miconazole, econazole, and 5-fluorocytosine against Saccharomyces cerevisiae were estimated in the presence of 1/2 minimum inhibitory concentration (MIC) and 1/2 minimum fungicidal concentration (MFC) of polygodial, respectively. Among them, the antifungal activities of miconazoles by polygodial was still shown against several yeast-like fungi including Candida albicans, Candida utilis, Cryptococcus neoformans, except for Candida krusei. The combination of polygodial with imidazole drugs against Saccharomyces cerevisiae was further examined using the macrobroth dilution checkerboard method. The fractional inhibitory concentration (FIC) and the fractional fungicidal concentration (FFC) index between polygodial and miconazole were 0.16 and 0.19, while the combination of polygodial with econazole exhibited the FIC index of 0.19 and the FFC of 0.25, respectively. These results suggest that polygodial and the imidazoles on the fungistatic and fungicidal action are highly synergistic.

Antifungal Activities of Herbal Essential Oils and Combination Effects with Ketoconazole against Candide spp. (수종 허브정유의 Candida속 진균에 대한 억제 활성 및 Ketoconazole과의 병용효과)

  • The antifungal activities of the essential oils from Anthemis nobilis, Ciderus atlantica, Juniperus communis, Lavandula angustifolia, Pelargonium graveolens, Pogestemon patchouli, Rosmarinus officinalis, and Styrax tonkinensis which are recommended for the treatment of microbial infections in aromatherapy and complementary medicines were tested against Candida spp. The activities were measured by broth dilution method and disk diffusion assay. Most of the test oils inhibited growth of Candida albicans, C. utilis and C. tropicalis. Especially, the essential oil from Pelargonium graveolens and its main component, citronellol showed the strongest activity among the herbs except benzoic acid from Styrax tonkinensis which is well-known antimicrobial compound. As a result of checkerboard microtiter test. synergistic effect of citronellol, was shown when the component was combinated with ketoconazole, displaying a fractional inhibiting concentration (FIC) index of 0.37 against C. albicans.

A Study on Methanol-Assimilating Yeast (메탄올자화 효모에 관한 연구)

  • Thirty species of wild yeasts were ioslated and identified from two hundred strains collected from flowers by enrichment techniques using steptomycin. Among them, twnety three species could assimilate methanol and three species, Candida incommunis, cryptococcus aerius and Hansenula ciferrii which showed good biomass yield were selected. These three species assimilated methanol as carbon and energy source without mixture of vitamin and yeast extract. The species grown on methanol media were confirmed to have all essential amino acids in their cellular constituents. The content of total amino acids are as followings ; Candida incommunis : 42.5%, Hansenula ciferrii : 39.9%. Of the essential amino acids lysine and threonine which are usually lacking in grain protein were as much as flour's. The experiments on the growth conditions for the higher biomass yield showed the result that the optimum concentration of methanol ans temeprature wre defined as 1.3% and $30^{\circ}C$ in each species.

  • 인구적 증가와 사료 가격의 상승으로 인한 육류 제품의 가격이 상승하고 있음은 우리나라를 비롯하여 세계적인 현황이다. 이 문제에 대처하는 한 방법으로 새로운 단백질 급원으로써 Single Cell Protein이 개발되었고 이에 대한 여러가지 연구가 되어지고 있다. Single Cell Protein이 인간에게 식용이 될 동물이나 나아가서는 인간에게 직접 식품으로 사용되기 전에 인체나 동물 체내에 끼칠 영향과 그 안전성을 확인하려는 하나의 노력으로 이 연구가 시도되었다. 동물 체내 대사에 미치는 SCP의 악영향의 주원인은 함유된 독성물질로 보고 그 독성물질이 SCP 생산 및 처리 과정 중 어느 시기에 생겨나는가를 규명해 보고자 하였다. Fermentation을 끝낸 SCP의 Supernatant내에 함유된 Inorganic phosphate(I.P.)의 함량을 측정하여 SCP세포의 Viability를 알아보았다. Fermentation을 끝낸 후 10일까지는 I.P.가 차츰줄다가 40일이 지났을 때에는 I.P.의 증가를 보였으나 직후보다는 낮음을 나타냈다. 또한 냉동 건조시킨 뒤에도 SCP Cell이 살아있음을 보였다. 또한 위와 관련지어서 SCP를 Protein급원으로 써서 사육한 흰쥐에 나타난 Protein Efficiency Ratio(PER)를 통하며 SCP의 질을 평가하였다. 실험결과로 나타난 것을 보면 표준 식이에 사용된 Casein의 PER$(\overline{X}=2.58)$이 SCP의 PER$(Dict 1;\overline{X}=0.888$, $Dict 2;\overline{X}=0.893$, $Dict 3;\overline{X}=0.860)$ 보다 유의적으로 높았다. 그리고 총실험기간의 평균치를 볼 때, Fermentation을 끝내고 3일 후와 6일 후에 냉동 건조시킨 SCP의 PER은 시판되는 SCP의 것과 별다른 차이를 보이지 않았다. 그리나 후반기에 있어서는 Fermentation을 끝내고 3일과 6일 후에 냉동 건조시킨 SCP의 PER이 시판되는 SCP의 것보다 약간 저조함을 보였다. 그리고 Casein의 PER은 총 사육기간은 통하여 별 변동이 없음에 반하여 후반기에 SCP의 PER이 급격한 저하를 보임은 SCP 사용상의 문제점을 나타낸 것으로 해석 된다.

  • A method for acid-hydrolysis of agricultural wastes and its utilization was investigated. In order to obtain fermentable sugar solution from cellulosic wastes such as cereal straws and hulls, in particular, of rice, barley and wheat, the chemical compositions were analyzed and optimum conditions of hydrolysis determined. The cereal straws contain 42 to 55 % of crude cellulose including hemicellulose. On the hydrolysis with 1% of sulfuric acid at 40 psig, 35.6% of the reducing sugar based on the weight of dry matter was formed from rice straw, (variety Chinheung) in 30 min. More powerful condition of hydrolysis would appear to decompose the sugar formed into other compounds, for instance, furfural. Under atmospheric pressure with 5% of the acid, rice straw was hydrolyzed to 35% of reducing sugar content in 3 hours. Candida utilis could assimilate the sugars in the hydrolyzate up to more than 97%, and a yield of the yeast cells reached 55% to the utilized sugars.

  • From the on-line mass spectrometric analyese of the exhaust gaseous composition of fermentor and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) and carbon dioxide evolution rate (CER) were calculate using a personal computer (IBM PC-AT) interfaced to a quadrupole mass spectromter. The calculate OUR and CER were used for the indirect estimation of cell and substrate concentrations during the batch culture of Candida utilis. For the estimation of sustrate concentration, the yield model of Pirt was applied. It was found that the cell and substrate (glucose) concentration could be ssatisfactorily estimataed and the results showed the more accurate estimations of both fermentation state variables when OUR data were used than CER data.

  • Continuous on-line monitoring of cell concentration and growth rate in aerobic batch fermentation process was carried out by analyzing the exhaust gas composition of tormentor with a quadrupole mass spectrometer. From the mass spectrometric analyses of major gaseous components, i.e. $N_2$, $O_2$, $CO_2$ and $H_2O$, and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) rind carbon dioxide evolution rate (CER) were instantaneously calculated using a computer (16-bit IBM PC-AT) interfaced to a quadrupole mass spectrometer. The calculated OUR and CER data were used for the estimation of cell concentration and growth rate of Candida utilis during batch culture. It was found that the cell concentration could be satisfactorily estimated from the data of OUR arid CER during the culture and this method could be successfully und for the continuous monitoring of cell growth and growth rate.

  • The pre-hydrolyzed residue of Chestnut-bur (1) was treated with 74% H$_2$SO$_4$, solution at 3$0^{\circ}C$ for 40minutes. Then the solution was diluted to 5% with distilled water and kept for another 40 minutes under steam pressure of 1.5kg/㎤ to complete the post-hydrolzation. In the resulting hydrolyzate, which showed 49.3% in saccharification rate, Candida utilis was grown and found that 62.5% of the reducing sugar had been converted to the yeast cells.

  • Isolation of the toxic substance which was produced by Streptentyces sp. and it's biorhemical characterestics and toxicity to fishes were reported in the previous paper. The present report includes antibiotic activity of the substance, inhibitory activity of the substance on the succinic dehydrogenase of fishes, and its effect on the blood corpuscles of a rabbit. An evident antibiotic activity of this substance was observed on Candida yeasts, but n$ot^1$on molds or bacteria. The substance inhibited the growth of Candida japonica and C. utilis to the 50% at the concentrations of 7.5 and 10.2ug per ml, respectively. The activity of succinic dehydrogenase obtained from various organs of Cyprinous carpio L. was also found to be inhibited by this substance. Original activities of the enzymes from tht brain, kidney, and liver, were inhibited by 75.4%, 38.2%, and 26.2%, respectively, but not the enzymes from the heart and spleen. Neither leukopenia nor leukocytosis was detected after the intravenous administration of the substince to the rabbit at the level of 6 mg per 2 kg body weight.

  • The composition of essential oils in the leaves of three cultivars (Ban-Chung-Gat, Chung-Gat and Dolsan-Gat) of Brassica juncea L. were analyzed and their antifungal activity were investigated in this study. Allyl isothiocyanate, 2-phenyl ethyl isothiocyanate, 4-isothiocyanato-1-butene, 5-methyl isothiazole, benzene acetaldehyde, benzene propane nitrile and beta-ionone have been identified in all of the experimented oils. The main component of the oils from Ban-Chung-Gat and Chung-Gat was 2-phenyl ethyl isothiocyanate while allyl isothiocyanate was the representing compound in the oil of Dolsan-Gat. The antifungal activities of the oils were tested by micro broth dilution method and disc diffusion method. As the result the oils exhibited significant inhibiting activities against Aspergillus niger, A. flavus, Trichoderma viride, Candida albicans, C. utilis, C. tropicalis, Cryptococcus neoformans, Trichosporon mucoides, Trichophyton tonsurans and Geotrichum capitatum.
