Studies on the overinflation of the Cardiac Stomach in Sterlet Sturgeon Acipenser ruthenus, fingerlings (양식 스털렛 철갑상어, Acipenser ruthenus 치어의 위 팽창 증후군에 관한 연구)

  • Overinflation of cardiac stomach occurred in farmed sterlet sturgeon (Acipenser ruthenus) fingerlings. Affected fish congregated and swam upside-down near the edges of the tanks with abdominal distension. When pressed the dilated abdomen, the fish often exhibited a normal swimming pattern for some time, then resumed their characteristic upside-down swimming patterns. Internally, the fish showed abdominal dropsy and dilated stomach filled with yellowish granular contents. When the contents in the stomach, fish feed, water and excrement in the farm inoculated on Sabouraud destrose agar (SDA) at $25^{\circ}C$ for 48 hr. Candida sp. were isolated except from the feed. Candida sp. in excrement or water in the farm may enter into the stomach with feed and then produce gases in the metabolic process, which in turn may cause the stomach overinflation.

  • Candida is a relatively rare cause of spinal infections that commonly affects immunocompromised patients. A 70-year-old woman, who underwent a lumbar discectomy on L5-S1 two months earlier, was admitted to our department complaining of persistent back and leg pain. Magnetic resonance imaging showed irregular enhancing mass lesion in L5-S1 intervertebral space, suggest of pyogenic discitis with epidural abscess. The surgery was performed via retroperitoneal approach and the infected material at L5-S1 intervertebral space was removed. The histological examination of the specimen revealed chronic inflammation involving the bone and soft tissue, and a culture of the excised material was positive for Candida parapsilosis. The patient received intravenous fluconazole for 4 weeks after surgery and oral fluconazole 400 mg/day for 3 months after surgery. The patient made a full recovery with no symptoms 6 months after surgery. We present a rare case of spondylodiscitis after a lumbar discectomy due to Candida parapsilosis and discuss treatment option with a review of the literatures.

Xylitol Production from D-Xylose by Candida mogii ATCC 18364 (Candida mogii ATCC 18364를 이용한 D-Xylose로부터 Xylitol 생산)

  • Fermentation characteristics of D-xylose into xylitol by Candida mogii ATCC 18364, a potential xylitol producer from rice straw hemicellulose hydrolyzates, were investigated. The influences of the most important operational variables on xylitol production were examined. The best results in xylitol production were obtained in shake-flask fermentations when 3.0 g/L initial cell concentration of 12 hr-old cells grown in D-glucose containing medium were used as inoculum. The oxygen availability is a critical factor in xylose fermentation, therefore, xylose conversion into xylitol was investigated in a 2-L fermenter at different stirring rates. Maximum xylitol production was obtained with an aeration rate of 1 vvm at a stirring rate of 200 rpm.

Purification of Enolase from Candida albicans KNIH10 Isolated in Korea and Application of Immunological Diagnosis (Candida albicans KNIH10으로부터 Enolase의 분리 및 면역진단의 응용)

  • We purified enolase from Candida albicans KNIH10 strain which was isolated from a clinical specimen in Korea. The purified enolase was used to detect anti-Candida antibodies in sera of patients with invasive candidiasis. For purification of enolase from the crude extract prepared by French pressure at 20,000 PSI, the fast performance liquid chromatography (FPLC) using DEAE-sepharose column was used. The elutes at $0.3{\sim}0.4\;M$ NaCl in FPLC was purified with homogenity in SDS-PAGE and its enzymatic activity was confirmed in sera of invasive candidiasis with candidemia patient by immunoblotting. The purified enolase indicated no signal (100% specificity) in 40 normal human sera and 75% (6/8) sensitivity in sera of candidemic patients with suspicious invasive candidiasis by immunoblotting.

  • The effect of phosphate on the production of erythritol and gluconic acid during the batch fermentation of Candida magnoliae SR101 was investigated. In the flask culture experiments, the results showed that phosphate concentration affected the production of erythritol and gluconic acid in Candida magnoliae. In the tormentor experiments, the increase of phosphate concentration of medium up to 10 g/L increased the gluconic acid, while the maximum erythritol concentration was 121.7 g/L from 250 g/L glucose and 3 g/L $KH_2PO_4$

  • The frequency of mucosal and cutaneous fungal infection is increasing worldwide, which is due to the increase of immunocompromised patients. Candida albicans are the principal species associated with human oral mycosis and are known to be the most virulent among pathogenic Candida spp. In this review, oral candidiasis were classified and oral mucosal manifestations of candidiasis were filed. And its diagnosis and management would be reviewed briefly.

In Vitro Antifungal Activity of Equol against Candida albicans

  • In this study, we demonstrate that equol has fungicidal activities against Candida albicans. The minimum inhibitory and minimum fungicidal concentrations of equol against C. albicans were 516 and $1,032{\mu}M$, respectively. Two separate viability assays found that equol changed the integrity of the C. albicans cell membrane, possibly by formation of membrane lesions. Scanning electron microscopy demonstrated ultrastructural changes.

  • Chayacterlstics of the sophorolipid produced from Candida bombiocola were investigated as an emulsifier of oil, a detergent or as a dispersant. Improved emulsification of crude oil was observed at high temperature ($70^{\circ}C$) with less than 1% concentration. Sophorolipid solution produced little foam even at reduced surface tension, however performance as a detergent of soiled cloths was poor. Dispersing and stabilizing abilities of sophorolipid solution were proved to be superior to those of chemical dispersants when examined by dispersing $Fe_2O_3$ or carbon black powders.

  • The purpose of this study was to evaluate the in vitro antifungal effect of several kinds of denture lining materials containing nystatin and silver-zeolite on Candida albicans. Three commercially available tissue conditioners (Soft-Liner, Coe-Comfort, Coe-Soft) and two direct denture relining materials (Tokuso Rebase, Durabase) were selected. In terms of the zone of inhibition and some basic physical properties, I could find the following results ; 1. Nystatin or silver-zeolite included in those relining materials had definite antifungal activities against the Candida albicans. 2. As times went on, both of the antifungal agents's activities decreased gradually. 3. Antifungal agents did not affect the relining materials' basic physical properties. 4 Direct chairside relining materials showed unfavorable dark discoloration with response to silver-zeolite.

IL-33 Priming Enhances Peritoneal Macrophage Activity in Response to Candida albicans

  • IL-33 is a member of the IL-1 cytokine family and plays a role in the host defense against bacteria, viruses, and fungi. In this study, we investigated the function of IL-33 and its receptor in in vitro macrophage responses to Candida albicans. Our results demonstrate that pre-sensitization of isolated peritoneal macrophages with IL-33 enhanced their pro-inflammatory cytokine production and phagocytic activity in response to C. albicans. These macrophage activities were entirely dependent on the ST2-MyD88 signaling pathway. In addition, pre-sensitization with IL-33 also increased ROS production and the subsequent killing ability of macrophages following C. albicans challenge. These results indicate that IL-33 may increase anti-fungal activity against Candida through macrophage-mediated resistance mechanisms.
