DNA topoisomerase I Inhibitor 를 생성하는 방선균 분리균주의 수리동정 (Numerical Identification of an Actinomycetes Strain Producing an Antitumor Antibiotic with Inhibitory Activity against DNA Topoisomerase)

  • DNA topoisomerase I have been shown to be important therapeutic target in cancer chemotherapy. Chemotaxonomy and numerical identification were carried out for an isolate strain No.7489 producing an antibiotic that inhibits DNA topoisomerase I activity. The genus of strain No.7489 was determined as Streptomyces sp. from culture, morphological and chemotaxonomic data. Thirty-nine taxonomic unit characters were tested and the data were analyzed numerically using the TAXON program. The isolate was best matched to Streptomyces melanosporofaciens in the major cluster 32 of Streptomyces. Therefore, it was concluded that the isolate was identified to be a member of Streptomyces melanosporofaciens.

  • 마이크로어레이 기술은 한번에 수만 개의 유전자를 동시에 분석할 수 있는 고효율, 고가의 새로운 연구 도구로 자리잡았으며 마이크로어레이 실험 자료의 올바른 분석을 위해서는 실험 목적에 맞는 실험계획법의 확립과 통계분석법의 적용이 중요하다 본 논문에서는 마이크로어레이 자료에서 여러 군 사이에서 발현의 차이를 보이는 유전자를 찾을 수 있는 되돌림 설계를 소개하고 ANOVA 모형을 이용하여 분석하는 방법을 제시한다. 연세대학교 암전이 연구센터의 되돌림 설계를 이용한 백혈병 자료를 MA-ANOVA(Wu et. al.(2003))를 이용하여 분석하였다

Backbone assignment of human Hoxc9DBD

  • Hoxc, or the Homeobox C cluster, is a group of genes that play a crucial role in embryonic development, particularly in patterning the body along the anterior-posterior axis. These genes encode transcription factors, which are proteins that bind to DNA and regulate the expression of other genes. Hoxc9 is specifically involved in the development of the skeletal system, nervous system, and adipose tissue. Hoxc9 overexpression has been linked to the development of various cancers such as leukemia and breast cancer. Here, we assigned the chemical shifts Hoxc9 DNA binding domain (DBD) using heteronuclear NMR techniques. The helical regions of Hoxc9 DBD correspond to the residues T200 - F213 (Helix I), T218 - L229 (Helix II), and T232 - K249 (Helix III). Our result would be helpful for studing the molecular interactions of the Hoxc9 DBD and other proteins.

  • 본 증례에서 환자는 폐렴 치료 중 갑자기 발생한 복통을 주소로 급성 충수염을 진단받고 수술을 시행 받았다. 그 후 조직 검사 결과 소세포암으로 진단되었고 폐병변에 대해 추가적인 검사 결과 소세포폐암이 진단된 경우이다. 저자들은 소세포폐암의 충수 전이를 국내에서 처음으로 경험하여 문헌고찰과 함께 보고하는 바이다.

군집화된 구간 중도절단자료에 대한 치유율 모형의 적용 (Cure Rate Model with Clustered Interval Censored Data)

  • 치유율 모형(cure rate model)은 위험 그룹의 단조 감소에 대한 가정이 부적절한 경우에 적용될 수 있다. 예를 들어, 생존 분석에서 위험 그룹은 시간이 경과함에 따라 점점 감소하여 무한대의 시간대에는 영으로 수렴하며 이는 곧 생존 함수가 영으로 수렴함을 의미한다. 하지만 이러한 가정이 적합하지 못한 자료가 의약학, 사회학, 경제학 등에서 종종 발생된다. 즉, 어느 시점에 이르러 더 이상의 생존함수는 감소하지 않고 평행선을 보여주는 경우에 로그 순위검정(log rank test)과 Cox's 비례위험모형(proportional hazard model)의 적용은 바람직하지 못한 결론을 가져오게 된다. 이러한 자료에 대해 치유율 모형(cure rate model)에서는 사건 발생 취약 그룹(susceptible group)과 비취약 그룹(insusceptible group)으로 나누어 취약그룹에 대해서만 일반적인 생존 분석 방법을 적용하는 혼합 모형(mixture model)을 적용해왔다 (Berkson과 Gage, 1952). 본 연구에서는 이러한 치유율 모형을 군집화 구간 중도 절단 자료(clustered interval censored data)에 적용해 보고자 한다. 최근에 Kim과 Jhun (2008)은 구간 중도 절단자료에 대해 치유율 모형을 적용하였으며 본 연구에서는 그들의 방법을 군집화 자료로 확장할 것이다. 실제 자료 분석의 예로 금연자료를 분석할 것이다.

Evaluation of the Anti-Tumor Effects of Paclitaxel-Encapsulated pH-Sensitive Micelles

  • We evaluated the efficacy of pH-sensitive micelles, formed by methoxy poly(ethylene glycol)-b-poly($\beta$)-amino ester) (PEG-PAE), as carriers for paclitaxel (PIX), a drug currently used to treat various cancers. PTX was successful encapsulated by a film hydration method. Micelles encapsulated more than 70% of the PTX and the size of the PTX-encapsulated micelles (PTX-PM) was less than 150 nm. In vitro experiments indicated that the micelles were unstable below pH 6.5. After encapsulation of PTX within the micelles, dynamic light scattering (DLS) studies indicated that low pH had a similar demicellization effect. An in vitro release study indicated that PTX was slowly released at pH 7.4 (normal body conditions) but rapidly released under weakly acidic conditions (pH 6.0). We demonstrated the safety of micelles from in vitro cytotoxicity tests on HeLa cells and the in vivo anti-tumor activity of PTX-PM in B16F 10 tumor-bearing mice. We concluded that these pH-sensitive micelles have potential as carriers for anti-cancer drugs.

빠른 클러스터 개수 선정을 통한 효율적인 데이터 클러스터링 방법 (Efficient Data Clustering using Fast Choice for Number of Clusters)

  • K-means algorithm is one of the most popular and widely used clustering method because it is easy to implement and very efficient. However, this method has the limitation to be used with fixed number of clusters because of only considering the intra-cluster distance to evaluate the data clustering solutions. Silhouette is useful and stable valid index to decide the data clustering solution with number of clusters to consider the intra and inter cluster distance for unsupervised data. However, this valid index has high computational burden because of considering quality measure for each data object. The objective of this paper is to propose the fast and simple speed-up method to overcome this limitation to use silhouette for the effective large-scale data clustering. In the first step, the proposed method calculates and saves the distance for each data once. In the second step, this distance matrix is used to calculate the relative distance rate ($V_j$) of each data j and this rate is used to choose the suitable number of clusters without much computation time. In the third step, the proposed efficient heuristic algorithm (Group search optimization, GSO, in this paper) can search the global optimum with saving computational capacity with good initial solutions using $V_j$ probabilistically for the data clustering. The performance of our proposed method is validated to save significantly computation time against the original silhouette only using Ruspini, Iris, Wine and Breast cancer in UCI machine learning repository datasets by experiment and analysis. Especially, the performance of our proposed method is much better than previous method for the larger size of data.

  • 셀 카운팅은 세포의 성장을 분석하는 방법으로써 생물학연구에서 가장 많이 사용된다. 최근까지도 다양한 자동 셀 카운팅 기법이 제안되고 있지만 암세포와 같이 분열 속도가 빠르고 군집하려는 성질을 갖는 세포들은 분리 및 검출이 쉽지 않아 세포 이미지 분석을 통하여 셀 카운팅의 신뢰도를 높이기가 어렵다. 본 논문에서는 암 연구의 연구재료로 매우 보편적으로 사용되는 HeLa 세포 이미지 분석을 이용한 자동 셀 카운팅 방법을 제시한다. 세포막 추출 기반의 세포 분할 알고리즘을 통하여 세포의 형태적 상황을 구분하고, 세포 간 경계가 희미한 세포군집 내의 세포 분할을 위하여 역추적 알고리즘을 사용함으로써 셀 카운팅 정확도를 높인다. 실험을 통하여 제안하는 세포 분할 알고리즘이 기존의 세포 분할 알고리즘에 비해 정확함을 입증하였고, 결과적으로 매우 높은 자동 셀 카운팅 정확도를 얻을 수 있음을 확인하였다.

Theoretical Investigations on Structure and Function of Human Homologue hABH4 of E.coli ALKB4

  • Introduction: Recently identified human homologues of ALKB protein have shown the activity of DNA damaging drugs, used for cancer therapy. Bioinformatics study of hABH2 and hABH3 had led to the discovery of a novel DNA repair mechanism. Very little is known about structure and function of hABH4, one of the members of this superfamily. Therefore, in present study we are intended to predict its structure and function through various bioinformatics tools. Materials and Methods: Modeling was done with modeler 9v7 to predict the 3D structure of the hABH4 protein. This model was validated with the program Procheck using Ramachandran plot statistics and was submitted to PMDB with ID PM0076284. The 3d2GO server was used to predict the functions. Residues at protein ligand and protein RNA binding sites were predicted with 3dLigandSite and KYG programs respectively. Results and Discussion: 3-D model of hABH4, ALKBH4.B99990003.pdb was predicted and evaluated. Validation result showed that 96.4 % residues lies in favored and additional allowed region of Ramachandran plot. Ligand binding residues prediction showed four Ligand clusters, having 24 ligands in cluster 1. Importantly, conserved pattern of Glu196-X-Pro198- Xn-His254 in the functional domain was detected. DNA and RNA binding sites were also predicted in the model. Conclusion and Prospects: The predicted and validated model of human homologue hABH4 resulted from this study may unveil the mechanism of DNA damage repair in human and accelerate the research on designing of appropriate inhibitors aiding in chemotherapy and cancer related diseases.

Healthy Life-Style Promoting Behaviour in Turkish Women Aged 18-64

  • Aim: In this study we aimed to investigate the healthy life-style behaviour of Turkish women and establish influencing features. Methods: This descriptive study performed by a questionnaire method was conducted in a primary health care centre, in an urban region in Kayseri, Turkey. Every midwife region belonging to the health care centre was accepted as a cluster, and a sample of 450 women between ages 18-64, was gathered from 9 midwife regions. The Health Promotion Life-style Profile (HPLP) was applied to evaluated the healthy lifestyle behaviour of 421 women that could be reached. T test, Tukey HSD with ANOVA, and chi square tests were used for analysis. Results: The mean total HPLP was $126.8{\pm}19.2$ (interpersonal support subscale, $74.3{\pm}14.1$; nutrition subscale, $73.6{\pm}12.6$; self-actualisation subscale, $70.6{\pm}11.9$; stress management subscale, $63.4{\pm}13.0$; health responsibility subscale, $61.2{\pm}13.2$; and exercise subscale, $47.1{\pm}15.0$). There was no statistically significant variation when evaluated for age, marital state, family type, economic status, and perception of self-health, smoking, and BMI. HPLP was high in people with an education of primary school and lower in university graduates, in people who lived mostly in the city centre and in individuals with chronic diseases. In conclusion, it was established that the health promoting behaviour in Turkish women is, in general, at a medium level, and women should be enlightened in order to develop and increase the habit of health preservation and promotion.
